Journal of Xinxiang Medical College 2024;41(8):772-775,782
Objective To explore the correlation between Toll-like receptor(TLR)and nuclear factor-KB(NF-κB)levels in monocytes and type 2 diabetic autonomic neuropathy(T2DAN).Methods A total of 112 patients with type 2 diabetes mellitus(T2DM)admitted to the Puyang People's Hospital from April 2021 to December 2022 were selected as the observation group, and 50 healthy volunteers who underwent physical examination in the hospital during the same period were selected as the control group.Reverse transcription polymerase chain reaction(RT-PCR)was used to detect plasma TLR2,TLR4 and NF-κB levels in monocytes of patients in the two groups.According to the presence or absence of autonomic neuropathy, patients in the observation group were divided into the autonomic neuropathy group(n=46)and the non-autonomic neuropa-thy group(n=66),and clinical data of patients in these two groups were compared.Multivariate logistic regression analysis was used on indicators with statistically significant differences to determine the risk factors for T2DAN,and Spearman correlation analysis was used to evaluate the correlation between TLR2,TLR4 and NF-κB levels and T2DAN.Results The TLR2,TLR4 and NF-κB levels in monocytes of patients in the observation group were higher than those in the control group(P<0.05).Univariate analysis showed that there were significant differences in age,monocyte/high-density lipoprotein ratio(MHR),and levels of cystatin C,TLR2,TLR4 and NF-κB between the autonomic neuropathy group and the non-autonomic neuropathy group(P<0.05).Multivariate logistic regression analysis showed that MHR,TLR2,TLR4 and NF-κB levels were risk factors for autonomic neuropathy in T2DM patients(P<0.05).Spearman correlation analysis showed that TLR2,TLR4 and NF-κB levels were positively correlated with T2 DAN(r=0.825,0.822,0.819;P<0.05).Conclusion The expressions of TLR2,TLR4 and NF-κB are up-regulated in T2DM patients, and the expression levels of TLR2,TLR4 and NF-κB are correlated with autonomic neuropathy in T2DM patients.

As a real-time,rapid and non-invasive method for monitoring the occurrence and development of various diseases and evaluating the health status of the body, breath detection has attracted wide attention in the application of early lung cancer screening. However, there is significant variability in the reported specific biomarkers of lung cancer, making it difficult to reach a unified consensus.Meanwhile, the pathways for the generation of volatile organic compounds(VOCs)in exhaled breath of lung cancer patients remain unclear, and further research on the detailed metabolic mechanisms is required. Therefore, this review comprehensively summarizes the VOCs detected in the exhaled breath of lung cancer patients from relevant literature in the past five years, deeply analyzes the production mechanism of these VOCs and the internal correlation with the disease, and evaluates the performance of the lung cancer prediction models constructed based on these VOCs,in order to provide reference for clinical and subsequent researches.

ObjectiveOptimize the latex perfusion technique and apply it to the construction of a murine craniofacial venous vascular model. Methods A total of nine 8-week-old male C57BL/6 mice weighing (25.0±1.3) g were randomly divided into three groups: 60% latex physiological saline group, 60% latex heparin group, and 30% latex heparin group. After completion of the perfusion, the specimens were immersed in 4 °C formalin fixative for 24 h, followed by dissection, observation, and measurement of the extracranial blood vessel diameters. Results After 200 μL latex perfusion solution was injected into the external jugular vein, the supraorbital vein, infraorbital vein, temporal vein, retrofacial vein, masseter vein and external jugular vein were perfused in each group. After comparing the perfusion degree of the distal branches of blood vessels, sublingual vein and tip venule, it was found that the 30% latex heparin group had the best perfusion effect, followed by the 60% latex heparin group, and the 60% latex saline group had the worst perfusion effect.ConclusionThe optimized latex perfusion technique can effectively infuse the veins in the head and face of mice, and this technique can provide a good reference for the study of the direction and morphology of facial veins in mice.

Objective:To establish the quality evaluation method of Perillae caulis formula granules based on the three kind of quality indexes of standard decoction. Methods:Eighteen batches of Perillae caulis were collected from different habitats according to different technical requirements, eighteen batches of standard decoction and three batches of formula granules were prepared and the paste-forming rates were calculated. The content of Caffeic acid and Rosmarinic acid were determined and calculated by Ultra High Performance Liquid Chromatography (UPLC). Then the fingerprints of standard decoction of and formula granules of Perillae caulis were established by UPLC . The similarity values of fingerprints between formula granules and standard decoction were calculated. Results:The average paste-forming rate of standard decoction was (7.16±1.97)%. The paste-forming rates of three batches of formula granules were 5.52%, 5.25% and 5.34%, respectively. The average content of Caffeic acid and Rosmarinic acid in standard decoction was (12.06±3.37)mg/g. The contents of three batches of formula granules were 5.52, 5.82, 5.77 mg/g, respectively. Seven common fingerprint peaks were identified in the fingerprints of standard decoction and formula granules, three of which were identified as Caffeic acid, N-Feruloyl Octopus amine and Rosmarinic acid by comparison of reference substance. The fingerprints similarity of Perillae caulis dispensing granules and standard decoction were 1.000, 0.995 and 0.997, respectively. Conclusions:The quality indexes of three batches of formulation granules are consistent with standard decoction. This method can provide basis for the establishment of quality standard of Perillae caulis dispensing granules.

OBJECTIVE:To optimize the extraction technology for total triterpenes from the leaves of Cornus officinalis . METHODS:Based on the full swelling of the leaves of C. officinalis ,total triterpenes was extracted with heating reflux method. The effects of ethanol concentration ,liquid-solid ratio ,extraction time and extraction times on the contents of total triterpenes from the leaves of C. officinalis were investigated by single factor test. Using oleanolic acid as control ,the contents of total triterpenes were detected by UV spectrometry. On the basis of single factor test ,fixing the times of extraction a s 3 times, taking the contents of total triterpenes as response value ,using ethanol volume fraction ,solid-liquid ratio and extraction time as factors , Box-Behnken design-response methodology was used to optimize the extraction technology of total triterpenes from the leaves of C. officinalis, and the optimized extraction technology was validated. RESULTS :The optimal extraction technology of total triterpenes from the leaves of C. officinalis were as follows as ethanol concentration of 73%,liquid-to-material ratio of 38 ∶ 1(mL/g), extraction time of 60 min. Results of 3 validation tests showed that the contents of total triterpenes from the leaves of C. officinalis were 6.92%,6.91%,6.84%;the average content was 6.89%(RSD=0.63%),relative error of which with the predicted value (7.28%)was 5.36%. CONCLUSIONS :The optimized technology is stable and reliable ,and can be used for the extraction of total triterpenes from leaves of C. officinalis .
