OBJECTIVE To systematically evaluate the correlation between dynamic changes in inflammatory markers during treatment with epidermal growth factor receptor-tyrosine kinase inhibitors (EGFR-TKIs) in non-small cell lung cancer (NSCLC) patients and therapeutic efficacy, with the aim of providing evidence-based support for clinical prognosis assessment and treatment strategy adjustment. METHODS Databases including PubMed, Embase, Cochrane Library, CNKI, Wanfang Data, and CBM were searched from the inception to July 20, 2025. Following literature screening, data extraction and quality assessment, descriptive analysis was conducted on the outcomes of included studies. RESULTS A total of eight studies were included to analyze the correlation of 6 inflammatory markers before and after treatment with EGFR-TKIs with therapeutic efficacy. The risk of bias assessment identified six high-quality studies and two moderate-quality studies. Among these studies, seven studies demonstrated that lower levels of neutrophil-to-lymphocyte ratio (NLR), derived neutrophil-to-lymphocyte ratio (dNLR), platelet-to-lymphocyte ratio (PLR), and monocyte-to-lymphocyte ratio (MLR), higher lymphocyte-to-monocyte ratio (LMR) before treatment, as well as decreased NLR and MLR and increased LMR after treatment were associated with longer median progression-free survival. Five studies indicated that lower levels of NLR, dNLR, PLR, and interleukin-6 (IL-6), higher LMR before treatment as well as decreased NLR and dNLR and increased LMR were associated with longer median overall survival. Three studies indicated that lower levels of IL-6 were associated with a higher objective response rate, while the association of these markers after treatment remained controversial; another study showed that an early decline in NLR, MLR, and PLR after treatment may be associated with objective response benefit. CONCLUSIONS Lower inflammatory levels during EGFR-TKIs therapy correlate with better therapeutic efficacy in NSCLC patients.

Objective This study reports on the obligatory parasitism of water mites Arrenurus sp. on Anopheles messeae at the Manzhouli Port, Inner Mongolia, China. Methods Duing July 2024, a survey on the mosquito diversity was conducted at the Manzhouli Port. Captured mosquitoes and their ectoparasites were identified to species level. Results A total of 1840 adult mosquitoes were collected, representing species from three genera: Culex(Cx. modestus, Cx. pipiens pallens), Aedes(Ae. dorsalis, Ae. flavidorsalis, Ae. flavescens), and Anopheles (An. messeae). Among all the mosqutioes specimens,3 out of 150 captured An. messeae were found to carry ectoparasitic mites, with number of 2,4,27 mites separately. Morphological and molecular identification reached the same result as water mites(Hydrachnidiae, Hydracrina). COI gene sequence showed 94% similarity with the closest species Arrenurus truncatellus. Conlusions Literature review suggests water mites are host-specific parasitism of mosquito species and herein with the first record of Arrenurus sp. parasiting on An. Messeae in the most high-latitude region globally.

Objective To explore the effect of microRNA-381-3p(miR-381-3p)/MuRF1 axis on cardiopulmonary injury in hypoxia-induced pulmonary hypertension(HPH)mice and its potential mechanisms. Methods Sixty mice were randomly assigned to four groups:the normal control group(NC),the hypobaric hypoxia-induced pulmonary hypertension(HPH)group, the HPH+agomir control group and the HPH+miR-381-3p agomir analog group(HPH+miR-381-3p agomir),with 15 mice in each group. The HPH mouse model was established using a low-pressure and hypoxic artificial chamber.Three weeks prior to the establishment of the HPH model,miR-381-3p agomir and its corresponding control agomir were prepared by dissolving them in RNA-free phosphate-buffered saline(PBS)according to the experimental requirements. These solutions were administered via tail vein injection at a dose of 10 mg/kg,twice weekly for three consecutive weeks.Right heart function was assessed using echocardiography.Right ventricular systolic pressure(RVSP)was measured via cardiac catheterization.Pulmonary vascular remodeling was evaluated through hematoxylin and eosin(HE)staining.Levels of inflammatory cytokines in bronchoalveolar lavage fluid were quantified using enzyme-linked immunosorbent assay(ELISA).Real-time quantitative fluorescent PCR(RT-qPCR)was employed to analyze the mRNA expression levels of miR-381-3p and MuRF1.Potential targets of miR-381-3p were predicted, and pathway enrichment analysis was conducted.A dual-luciferase reporter gene assay was performed to confirm the direct regulatory effect of miR-381-3p on MuRF1.Results Compared with the NC group, the mRNA expression of miR-381-3p was significantly decreased in both the HPH group and the HPH+agomir control group, whereas the mRNA expression of MuRF1 was significantly increased(P<0.05).In contrast, compared with the HPH group and the HPH+agomir control group, the mRNA expression of miR-381-3p was significantly increased in the HPH+miR-381-3p agomir group, while the mRNA expression of MuRF1 was significantly decreased(P<0.05).Additionally,compared with the NC group,RVSP,right ventricular anterior wall thickness(RVAW),right ventricular hypertrophy index(RVHI),right ventricular collagen volume fraction(CVF),distal pulmonary artery wall thickness ratio(WT),pulmonary artery wall area ratio(WA),as well as IL-1β,IL-6 and TNF-α levels in alveolar lavage fluid were significantly increased in the HPH group and the HPH+agomir control group, whereas the right ventricular diameter(RVID)was significantly decreased(P<0.05).Conversely,compared with the HPH group and the HPH+agomir control group,RVSP,RVAW,RVHI,right ventricular CVF,WT,Wa and RVID were decreased in the HPH+miR-381-3p agomir group, and IL-1β,IL-6,and TNF-α levels of alveolar lavage fluid were significantly decreased(P<0.05).Furthermore,the downstream target genes of miR-381-3p were predicted in the database, and MuRF1 was a potential target, and the Cytoskeleton in muscle cells ranked first in the significant enrichment of target genes.Compared with WT-MuRF1+mimic control group, the luciferase activity was decreased in the WT-MuRF1+miR-381-3p mimic group(P<0.05).There was no significant difference in the luciferase activity between the Mut-MuRF1+mimic control group and the Mut-MuRF1+miR-381-3p mimic group. Conclusion Overexpression of miR-381-3p can improve cardiopulmonary injury in HPH mice, and the mechanism may be related to the targeted inhibition of MuRF1 by miR-381-3p.

Aim To compare the observation results of atomic force microscopy(AFM)and scanning electron microscopy(SEM),and to summarize the main problems and solutions of AFM in observing biological macromolecules, using the observa-tion subjects of protein samples purified by our research group. Methods The protein samples were diluted to 15 nmol·L-1 with PBS,fixed on glass slides, silicon wafers, and mica sheets, dried, and made into solid-phase observation samples.SEM sam-ples were plated with platinum before observation. The surface structures of proteins were observed using AFM and SEM,sample heights were calculated, and differences in results were com-pared.Results Protein samples with positive charges tended to shift to the right during observation due to the repulsion of the AFM probe;mica sheets could effectively eliminate the positive charge of proteins to avoid sample movement;PBS provided a stable environment for protein samples, but the crystallization of PBS salts interfered with probe operation and imaging clarity;SEM samples needed to be plated with platinum before observa-tion and could not achieve the precision of AFM.Conclusions Both AFM and SEM can directly observe protein structures in vitro, with AFM providing higher precision results;when protein sample stability permits, ultrapure water is preferred as the sol-vent carrier, and volatile liquids such as ethanol can also serve as solvent carriers. The application of AFM offers a new approach for pharmacological studies on interactions between biological macromolecules.

Adverse Drug Reactions Journal 2025;27(7):397-402
Objective:To explore the effect of contezolid on platelet count in patients with severe pneumonia and analyze the risk factors. Methods:The study was designed as a retrospective case-control study. The research subjects were selected from patients with severe pneumonia who were admitted to the Department of Respiratory and Critical Care Medicine of Shaanxi Provincial People′s Hospital from July 1, 2022 to November 30, 2024 and were treated with contezolid or linezolid. The clinical data of patients were collected and the incidence of thrombocytopenia [platelet count (PLT)<100×10 9/L after medication], the PLT before and at 1 week of treatments, and the lowest PLT value during treatments were compared in patients treated with contezolid and linezolid. The patients were divided into 2 groups based on whether contezolid- related thrombocytopenia occurred. The clinical characteristics of the patients were compared, and the independent risk factors of contezolid-related thrombocytopenia were analyzed by binary logistic regression method. Results:A total of 175 patients were included, among whom 73 received contezolid and 102 received linezolid. There was no statistically significant difference in PLT between the 2 groups before medication ( P=0.364). Compared with patients treated with linezolid, the incidence of thrombocytopenia in patients treated with contezolid was lower [19.2% (14/73) vs. 40.2% (41/102)], and the PLT at 1 week of treatments and the lowest value of PLT during treatments were higher. The differences were all statistically significant (all P<0.05). Compared with patients without contezolid-related thrombocytopenia(59 patients), patients who developed thrombocytopenia after using contezolid had a longer duration of contezolid medication, lower PLT and creatinine clearance rate before medication, and higher procalcitonin and serum creatinine levels before medication. All these differences were statistically significant (all P<0.05). The results of binary logistic regression analysis showed that lower PLT [odds ratio ( OR)=0.971, 95% confidence interval ( CI): 0.950-0.992, P=0.008] and higher procalcitonin level ( OR=7.292, 95% CI: 1.067-49.814, P=0.043) before medication and longer duration of contezolid medication ( OR=1.165, 95% CI: 1.002-1.355, P=0.046) were the independent risk factors of contezolid-related thrombocytopenia. Conclusions:Compared with linezolid, contezolid has a relatively safer profile in the treatment of patients with severe pneumonia and the risk of thrombocytopenia after medication is lower. Patients with lower PLT and higher procalcitonin levels before medication, and those with a longer duration of contezolid medication have a higher risk of contezolid-related thrombocytopenia and should be closely monitored.
