1.Celastrol attenuates sodium oxalate-induced acute kidney injury and crystal deposition by inhibiting NF-κB

Objective To investigate the role and possible mechanism of celastrol(Cel)in sodium oxalate(NaOx)-induced acute kidney injury(AKI)and crystal deposition in the kidney tissues in mice.Methods Male C57BL/6 mice(aged 8~12 weeks,weighing 22~24 g)were randomly divided into 3 groups.Saline group(control group,intraperitoneal injection with normal saline and drinking water freely),NaOx group(injured group,intraperitoneal injection of 75 mg/kg NaOx,and drinking water containing 50 μmol/L NaOx),and NaOx+Cel group(treatment group,intraperitoneal injection of 1 mg/kg Cel firstly and then 75 mg/kg NaOx in 24 h later,drinking water containing 50 μmol/L NaOx).All specimens were collected in 24 h after NaOx injection.HK-2 cells were randomly divided into 4 groups:Medium group(no treatment),NaOx group(500 μmol/L NaOx),NaOx+Cel group(400 nmol/L Cel pre-treatment for 2 h followed by 500 μmol/L NaOx treatment),and NaOx+Cel+BA group[8 μmol/L betulinic acid(BA,NF-κB agonist)after the interventions as the NaOx+Cel group].Cells of each group were collected in 24 h after corresponding treatments.Von Koosa and cell adhesion assays were used to observe crystal deposition.HE staining was employed to observe renal histopathology and score the damage.CCK-8 assay was utilized to detect cell viability to obtain the optimal concentrations of NaOx and Cel.Serum urea and creatinine levels were detected.Immunohisotochemical assay was conducted to detect the expression of OPN,CD44,KIM-1,NGAL,p65,IL-1β,BAX,and Caspase-3,and Western blotting was performed for protein levels of OPN,CD44,KIM-1,p65,P-p65 and IL-1β.Results The mice in the NaOx+Cel group showed reduced crystal deposition(P<0.0001),attenuated renal tubular damage(P<0.01),decreased serum urea and creatinine levels(P<0.05),and declined expression levels of the renal adhesion molecules OPN and CD44,the kidney injury molecules KIM-1 and NGAL,the inflammation-associated molecules p65 and IL-1β,and the apoptosis related molecules BAX and Caspase-3 when compared with the NaOx group(P<0.05).In in vitro study,the NaOx+Cel group showed reduced crystal adhesion(P<0.0001),decreased expression of the adhesion molecules OPN and CD44(P<0.05),down-regulation of the inflammatory molecule IL-1β and P-p65/p65 ratio(P<0.05),and down-regulation of the renal injury molecule KIM-1(P<0.05)when compared with the NaOx group.In the NaOx+Cel+BA group,crystal adhesion was significantly increased(P<0.0001),the inflammatory molecule IL-1β and the ratio of P-p65/p65 were increased(P<0.05),and the kidney injury molecule KIM-1 was increased when compared with the NaOx+Cel group(P<0.05).Conclusion Cel may reduce NaOx-induced crystal deposition and AKI by inhibiting NF-κB activation.
2.Application of adaptive technology-based speech recognition system in 600 pathological grossing process

Objective:To establish a speech recognition system based on adaptive technology and to evaluate its value in pathological grossing processes.Methods:A total of 600 tissue specimens were collected at the Zhejiang Provincial People's Hospital affiliated to Hangzhou Medical College between October 1, 2020 and December 31, 2020. A speech recognition system based on adaptive technology was used in the pathological grossing processes, and the pathological examination reports were generated and extracted.Results:The speech recognition system based on adaptive technology showed a good recognition rate (overall recognition rate = 77.87%) and helped achieve rapid input and output of pathological examination data.Conclusions:The speech recognition system can reduce the labor costs, improve the work efficiency of pathologists and increase the quality of medical services, which may be valuable for building next-generation smart hospitals.
3.The application of tracheal intubation by the endoscopic-assisted mouth floor-submandibular approach for the general anesthesia and operation of craniofacial multiple fractures

Objective:To investigate the application of tracheal intubation by the endoscopic-assisted mouth floor-submandibular approach(EAMFA) for general anesthesia and the operation of craniofacial multiple fractures.Methods:8 patients with complex craniofacial fractures and associated with the contraindication of nasal trachea cannula underwent EAMFA general anesthesia and operation.Blood pressure(BP),saturation of blood oxygen(SPO2),heart rate(HR) and electrocardiogram(ECG) were monitored timely.Results:The intubation and anesthesia were successfull in all patients;BP,SPO2,HR and ECG were normal in all operation procedure in the patients.No complication was observed.Conclusion:EAMFA is an effective anesthesia approach for the surgical treatment of craniofacial multiple fractures.
4.Application of Q-Syte separated membrane sealed without pin connector in children with blood diseases

Objective To explore the effect of Q-Syte separated membrane sealed without pin connector in children with blood disease. Methods Using random digit table, the children using Q-Syte separation membrane enclosed needle-free connector and heparin cap were divided into the observation group and the control group with 260 children in each group. The two groups were compared in terms of the incidence of using Q-Syte membrane closure cap pin connector and heparin. Results The result showed a significant positive relation between the two groups (P<0.05). The incidence of using Q-Syte separation membrane sealed without pin connector was lower than that of the control group. Conclusion Q-Syte separation membrane sealed without pin connector can reduce the incidence of infusion, which is easy, safe and worth clinical application.
5.Role of Renal Artery Doppler Ultrasonography in the Evaluation of Function of Transplanted Kidney

Objective:To evaluate the clinical value of renal artery Doppler ultrasonography in the diagnosis of renal transplant dysfunction by detecting blood perfusion in transplanted kidney with ultrasound .Methods:A total of 107 renal transplantation patients ,aged (38 .8 ± 1 .6) years in average ,including 68 new cases and 39 follow‐ups ,were recruited .Peak systolic velocity (PSV) ,end‐diastolic velocity (EDV) and resistance index (RI) ,in main renal artery ,renal segmental arteries and interlobar arteries of transplanted kidneys ,were examined by color Doppler ultrasonography .Results:Hemorheologic alterations in renal segmental arteries and interlobar arteries were closely correlated with renal function .Correlation indexes between PSV and clearance of creatinine (Ccr) were r=0 .072(P>0 .05) ,r=0 .224 (P=0 .014) and r=0 .250 (P=0 .006) ,respectively ,in main renal artery ,renal segmental arteries and interlobar arteries .Correlation indexes between RI and Ccr were r= -0 .398 (P<0 .001) ,r= -0 .400(P<0 .001) and r= -0 .436(P<0 .001) ,respectively ,in main renal artery ,renal segmental arteries and interlobar arteries .Correlation indexes between EDV and Ccr were r=0 .446(P<0 .001) ,r=0 .659(P<0 .001) and r=0 .529(P< 0 .001) ,respectively ,in main renal artery ,renal segmental arteries and interlobar arteries .The above results indicated that ,the maintenance of diastolic blood flow in renal arteries was important for renal function .The increase of RI and decrease of EDV in arteries of transplanted kidney was characteristic manifestation of chronic allograft nephropathy and vasculitic rejection .Conclusions:Renal artery Doppler ultrasonography can effectively measure the hemorheologic alterations of transplanted kidney ,thus it is an important adjuvant diagnostic tool for clinical renal transplantation .
6.The correlation between point mutation of MBL ExonI and its plasma concentration in North Huis

Objective:The major aim of this study is to analyze the point mutation at the codon 54th of MBL in healthy North Huis,and to measure the plasma levels of MBL, and to analyze the association between the mutation frequency and plasma MBL concentrations.Methods:PCR-RFLP was used to detect MBL point mutation.MBL plasma concentrations were measured using MBL Oliger ELISA kit.Results:Frequency of point mutation at the codon 54th of MBL in healthy Huis was 0.15. The plasma MBL concentration was (3.40?2.55)mg/L. There was negative correlation between MBL concentrations and gene mutation frequency in huis(r=-0.67).Conclusion:The relationship between frequency of mutation at codon 54 of MBL gene and the plasma MBL concentrations in healthy Huis is negative correlation.
