A case of Sifrim-Hitz-Weiss syndrome(Sifrim-Hitz-Weiss syndrome,SIHIWES)is presented.The patient was a 35-year-old male with cryptorchidism,growth retardation,skeletal malformations,muscular atrophy,a wide forehead,special facial features like square face,small low-set and cup-shaped ears since birth.Whole-exon sequencing identified a heterozygous mutation(NM_001273:c.3047A>G(chr12-6701125)(p.K1016R))in CHD4 gene.The clinical significance of this mutation is currently unknown,and has not been previously reported.In light of the patient's symptoms,the case was diagnosed as Sifrim-Hitz-Weiss syndrome.This case represents the first instance of Sifrim-Hitz-Weiss syndrome in an adult patient in China.

3.Molecular Mechanism of KHSRP Promoting Invasion and Metastasis in Esophageal Squamous Carcinoma by JAK1/STAT3 Signaling Pathway

To investigate the malignant progression and molecular mechanism of KHSRP regulating esophageal squamous cell carcinoma(ESCC) through the JAK1/STAT3 signaling axis. Tumor tissues and adjacent non-tumor tissues were collected from 72 patients with ESCC. Human normal esophageal epithelial cells(Het-1A) and multiple ESCC cell lines(EC-9706, TE-7, KYS-450, FLO-1, SK-GT-4, BE-3) were cultured. The expression level of KHSRP in the cells was detected using real-time fluorescence quantitative polymerase chain reaction(RT-qPCR). Through lentiviral transfection technology, stable KHSRP-knockdown EC-9706 and SK-GT-4 cell models(sh-KHSRP group), as well as stable KHSRP-overexpressing BE-3 and KYS-450 cell models(KHSRP group), were established, and corresponding negative control groups(sh-NC group and Vector group) were also established. Cell proliferation, migration, and invasion abilities were assessed using the cell counting kit-8(CCK-8) assay, Transwell migration assay, and Transwell invasion assay, respectively. A total of 62 male BALB/C nude mice aged 4 to 6 weeks were selected for the experiments. Thirty-two nude mice with subcutaneous tumor-loading experiments were randomly divided into four groups: sh-KHSRP 1 group, sh-NC 1 group, KHSRP 1 group, and Vector 1 group, with 8 mice in each group. Thirty nude mice with tail vein injection for lung metastasis model experiments were randomly divided into four groups: sh-KHSRP 2 group( The results of RT-qPCR revealed that, compared with human normal esophageal epithelial cells, the expression of KHSRP was significantly elevated in ESCC cell lines(EC-9706, TE-7, KYS-450, FLO-1, SK-GT-4, BE-3)( KHSRP is upregulated in ESCC and can positively regulate the JAK1/STAT3 signaling axis, potentially promoting the malignant progression of metastasis in ESCC.
4.Study on the Medication Law of Wang Zhongyi in Treating Tic Disorder Based on Data Mining

Objective To study the medication law of Professor Wang Zhongyi in the treatment of tic disorder(TD)based on data mining technology.Methods From January 1,2022 to December 31,2023,the cases treated for TD in Professor Wang Zhongyi's outpatient clinic,which participated in the real-world study were collected.A comprehensive database has been established,screening information related to effective case diagnosis and treatment.Utilizing Excel 2021,R 4.4.2,Origin 2024 and Cytoscape 3.9.1,this study conducted medication frequency analysis,property-flavor-meridian tropism analysis,efficacy analysis,association rule analysis,clustering analysis and co-occurrence network analysis to summarize medication law.Results Totally 640 effective prescriptions were included,involving 208 kinds of Chinese materia medica.The properties were mainly warm,cold,and neutral.The flavors were mainly pungent,bitter and sweet.The meridians were mainly liver meridians.The therapeutic categories were primarily composed of liver-calming and endogenous wind-stopping drugs,along with exterior-resolving prescriptions.Correlation analysis obtained 17 strongly correlated rules.Clustering analysis obtained 5 types of medicinal combinations.The therapeutic categories were primarily composed of liver-calming and endogenous wind-stopping drugs,along with exterior-resolving prescriptions.Conclusion According to the comprehensive statistical analysis,Uncariae Ramulus cum Uncis,Gastrodiae Rhizoma,Haliotidos Concha,Paeoniae Radix alba,Bupleuri Radix,Puerariae Lobatue Radix and Scorpio are the core drugs used by Professor Wang Zhongyi to treat TD.Professor Wang Zhongyi believes that the core pathogenesis of TD is the internal movement of liver wind,and the treatment mainly focuses on calming the liver,calming the wind and stopping spasms,while also nourishing the heart,calming the mind,harmonizing blood and relieving qi.Based on different clinical symptoms of TD,modifications and adjustments are made to the core prescription to treat children with TD.

7.Research on the Construction of a New Cultural Indicator System for High-quality Development of Public Hospitals

Objective It constructs a new culture index system for high-quality development of public hospitals to provide scientific tool for evaluating the level of new culture construction of public hospitals and promoting high-quali-ty development.Methods A pool of indicators was established based on policy combing and literature research.Then,an indicator system was formed through Delphi expert consultation and demonstration,and weighting coefficients were assigned using the hierarchical analysis method.Results The indicator system of the new culture in public hospi-tals contains four primary indicators,eight secondary indicators and 23 specific entries.The four primary indicators are hospital culture system construction,strengthening the orientation of patients'needs,caring for medical staff and distinctive hospital culture.The effectiveness of implementation as an additional dimension contains two secondary indicators and eight specific entries.The weights of the primary indicators are balanced.Among the sec-ondary indicators,institutional planning(0.250),staff care(0.170),medical services(0.130),discipline branding(0.125)and professionalism(0.125)have higher weights.Conclusion The new culture index system for high-quality development of public hospitals is in line with Xi Jinping's cultural thought and the requirements of the new culture for high-quality development of public hospitals.It needs to pay attention to the planning of the cultural system,em-phasises the care of employees and the cultivation of professionalism,and focuses on the construction of medical services and disciplinary brands,so as to promote the construction of the new culture in public hospitals in line with the requirements of the national strategies.
8.Regulation of Suaeda salsa in renal AIM and macrophage polarization in rats with diabetes kidney disease

Objective:To explore the regulatory effect of Suaeda salsa on renal apoprosis inhibitor of macrophage (AIM) and macrophage polarization in diabetes kidney disease (DKD) model rats.Methods:A DKD rat model was established using a high-sugar and high-fat diet combined with intraperitoneal injection of streptozotocin (STZ). The rats were divided into model group, metformin group, and Suaeda salsa high-, medium-, and low-dosage groups using a random number table method, with 8 mice in each group. The normal group was set with 8 rats in this group. The metformin group was given 85.71 mg/kg of metformin solution by gavage, while the Suaeda salsa high-, medium-, and low-dosage groups were given 3.08, 1.54, and 0.77 g/kg of Suaeda salsa suspension by gavage (raw dosage). The normal group and model group were given equal volumes of saline by gavage, once a day, administered by gavage for 12 weeks of intervention. Fasting blood glucose (FBG) and glucose tolerance (OGTT) were measured, and the area under the curve (AUC) was calculated; the levels of glycosylated hemoglobin (HbA1c) and glycosylated serum protein (GSP) were detected; urea nitrogen (BUN), serum creatinine (SCr), and 24-hour urine protein (24 hUP) were detected; HE staining was used to observe the pathological morphology of the kidneys; Masson and PAS staining were used to observe renal tissue fibrosis; Western blot method was used for detecting AIM, CD206, CD86, TNF-α and IL-10 protein levels in renal tissue; Immunofluorescence was used to detect the average optical density values of AIM, CD206, and CD86 proteins in renal tissue.Results:Compared with the model group, the FBG, OGTT AUC, HbA1c, GSP of each dosage group of Suaeda salsa decreased ( P<0.01); the expression levels of AIM, CD206, and IL-10 proteins in renal tissue increased ( P<0.01 or P<0.05), while the expression levels of CD86 and TNF-α protein significantly decreased ( P<0.01 or P<0.05); HE, Masson, and PAS staining results showed that compared with the model group, the changes in renal microvasculature and renal fibrosis of rats in each dosage group of Suaeda salsa were improved. Conclusion:Suaeda salsa may regulate AIM, promote polarization of M2 macrophages, improve the inflammatory microenvironment of macrophages, thereby lowering blood lipids of DKD rats, and improving renal pathological damage.
9.Exploration on the effects of Zhiganqing Prescription on insulin resistance and gluconeogenesis in NAFLD mice based on the PI3K/Akt/FoxO1 signaling pathway

Objective:To observe the protective effects of Zhiganqing Prescription on the liver of C57BL/6J non-alcoholic fatty liver disease (NAFLD) mice induced by high fat diet and its effects on PI3K/Akt/FoxO1 signaling pathway, insulin resistance (IR) and gluconogenesis.Methods:A total of 48 8-week-old male C57BL/6J mice were divided into control group ( n=8) and modeling group ( n=40) according to random number table method. The control group was fed with ordinary diet, and the model group was fed with high-fat diet. The NAFLD model was established after 8 weeks of feeding. The modeling group was divided into model group, Pioglitazone group, Zhiganqing Prescription low-, medium-, and high-dosage group ( n=8 in each group) according to random number table method, and drug intervention lasted for 8 weeks. The body mass of mice was measured regularly during administration. Fasting blood glucose (FBG) levels were measured at 0 and 8 weeks of administration, and oral glucose tolerance tests (OGTT) were conducted. After the experiment, serum levels of GPT, GOT, TC, TG, LDL-C, HDL-C, FINS and C-P were detected and HOMA-IR was calculated. The pathological morphology of liver was observed by HE and PAS staining. The expression levels of PI3K and p-Akt were detected by IHC staining. The protein expression levels of PI3K, Akt, p-Akt, FoxO1, p-FoxO1, G6PC and PCK1 were detected by Western blot. Results:Compared with model group, the body weight of mice in each administration group decreased at 4, 6 and 8 weeks ( P<0.05 or P<0.01). At the 8th week of administration, the levels of FBG and OGTT AUC in each administration group decreased ( P<0.05 or P<0.01), the levels of GPT, TC, TG and LDL-C decreased ( P<0.01), and the GOT levels in Zhiganqing Prescription medium- and high-dosage groups decreased ( P<0.01). The HDL-C level in Zhiganqing Prescription medium-dosage group decreased ( P<0.05 or P<0.01), and the HOMA-IR level in Zhiganqing Prescription low- and medium-dosage groups decreased ( P<0.05 or P<0.01). The levels of FINS and C-P in each administration group increased ( P<0.05 or P<0.01), and the expressions of PI3K protein and p-Akt/Akt, p-FoxO1 /FoxO1 protein in liver tissues increased ( P<0.05 or P<0.01). The protein expressions of G6PC and PCK1 decreased ( P<0.05 or P<0.01). Conclusion:Zhiganqing Prescription can effectively control the body mass, blood glucose, liver function and blood lipids of NAFLD mice, improve IR and gluconeogenesis, the mechanism of which may be related to the activation of PI3K/Akt/FoxO1 signaling pathway.
10.Next-generation sequencing-based minimal residual disease detection reveals clonal evolution in pediatric acute B-lymphoblastic leukemia: a case report and literature review

Minimal residual disease (MRD), a crucial biomarker for assessing efficacy and predicting recurrence, refers to residual tumor cells remaining in the body of patients with hematological malignancies who achieved complete remission after treatment. This study aimed to conduct a retrospective analysis of the clinical diagnosis, treatment, and MRD monitoring of a pediatric patient with multiple acute B-lymphocytic leukemia relapses, alongside a review of relevant literature. In this case, Ig rearrangement based on next-generation sequencing (NGS) was more accurate in assessing the MRD level, compared with the traditional method of MRD detection, indicating the risk of earlier relapse and guided interventions in time. Additionally, NGS-MRD detected clonal evolution, providing new ideas to further investigate the intrinsic factors of disease development.
