1.Exosomes derived from umbilical cord mesenchymal stem cells carrying miR-296-3p improve symptoms of rat model with gestational diabetes mellitus

Objective To investigate the improvement of exosomes(Exo)derived from umbilical cord mesenchymal stem cells(MSCs)carrying miR-296-3p on blood lipid metabolism and immune function in gestational diabetes melli-tus rats.Methods After transfection of MSCs with miR-296-3p-NC and miR-296-3p mimics,Exo was extracted to obtain MSCs-Exo-miR-NC and MSCs-Exo-miR-296-3p,respectively.RT-qPCR was used to detect the expression of miR-296-3p;Pregnant mice were prepared and induced to establish a GDM model by tail vein injection of strep-tozotocin.Then they were randomly grouped into a model group,MSCs-Exo-miR-NC group,and MSCs-Exo-miR-296-3p group with 10 each.In addition,10 pregnant mice were prepared as the control group.The weight of mother and fetal mice,four items of blood lipids and immune organ index in each group were measured after intervention with MSCs-Exo-miR-NC and MSCs-Exo-miR-296-3p.ELISA was applied to measure the level of serum immuno-globulin and immune inflammatory factors.Flow cytometry technology was applied to detect the immune function of peripheral blood T lymphocytes of rats in various groups,and their CD4+and CD8+values were compared.Results Compared with the control group,the maternal rat weight and fetal rat weight,serum triglycerides(TG),total cho-lesterol(TC),low-density lipoprotein(LDL-C),IL-6 and IL-8 levels and CD8+ratio of rats in model group in-creased(P<0.05),the spleen and thymus index,serum high-density lipoprotein(HDL-C),IgA,IgG,IgM,IL-2 and IL-10 levels and CD4+ratio decreased(P<0.05).Compared with the model group,the maternal rat weight and fetal rat weight,serum TG,TC,LDL-C,IL-6 and IL-8 level,and CD8+ratio of rats from MSCs-Exo-miR-NC group and MSCs-Exo-miR-296-3p group decreased(P<0.05),the spleen and thymus index,serum HDL-C,IgA,IgG,IgM,IL-2 and IL-10 levels and CD4+ratio all increased(P<0.05);MSCs-Exo-miR-296-3p had a stronger effect on various indicators of GDM rats(P<0.05).Conclusions Exo derived from MSCs carrying miR-296-3p improves blood lipid metabolism and immune function of GDM rats.
2.Clinical observation of bevacizumab combined with chemotherapy in elderly patients with gastrointestinal neoplasms

Objective To investigate the clinical efficacy and toxicity of bevacizumab combined with chemotherapy in elderly patients with gastrointestinal neoplasms.Methods A total of 64 elderly patients with gastrointestinal neoplasms were selected.The clinical data of patients treated with chemotherapy or chemotherapy plus bevacizumab were analyzed.Patients were randomly divided into the observation group and the control group (n=32,each).The observation group was treated with bevacizumab (7.5 mg/kg) in combination with XELOX (OXA+Xeloda),and the control group was treated with XELOX only (21 weeks as a cycle).The clinical efficacy and toxicity of bevacizumab were evaluated according to WHO short-term efficacy standards and standards of anticancer drugs in acute or subacute adverse reactions.Results In observation group,complete remission was found in 4 cases,partial response in 18 cases,stable disease in 6 cases,progressive disease in 4 cases,and the total efficiency was 68.8%.In control group,partial response was found in 14 cases,stable disease in 11 cases,progressive disease in 7 cases,and the total efficiency was 43.8%.The incidences of leucopenia,hemoglobin desaturation and gastrointestinal reactions were lower and the incidence of thrombocytopenia was higher in observation group than in control group,which had no significant differences between the 2 groups.Phase Ⅳ toxicity was not found in observation group.Conclusions Bevacizumab combined with chemotherapy is safe and effective for elderly patients with gastrointestinal neoplasms.Most patients can tolerate the side effects.It is worth promoting in clinical medicine.
3.Simultaneous determination of multicomponent in Xuefuzhuyu granule with different UV-wavelength by HPLC.

OBJECTIVETo establish the method for the quality control of Xuefuzhuyu granule.
METHODThe content of Semen Armeniacae Amarum PE, Hydroxysafflor yellow A, paeoniflorin and ferulic acid in Xuefuzhuyu granule were determined by HPLC under multi-wavelength of 210, 403, 230, 316 nm.
RESULTThe calibration curve of Semen Armeniacae Amarum PE. hydroxysafflor yellow A, paeoniflorin and ferulic acid were linear within 0.1154-0.9232 (r = 0.9998), 0.0219-0.1754 (r = 0.9994), 0.3952-3.1616 (r = 0.9995), and 0.0426-0.3408 microg (r = 0.9998), respectively. The average recoveries were 101.78% (RSD was 1.7%), 99.60% (RSD was 2.9%), 98.90% (RSD was 2.0%), and 100.31% (RSD was 1.8%), respectively.
CONCLUSIONThe method of quantification is accurate, rapid and reliable for the quality control of Xuefuzhuyu granule.
