1.lncRNA DLEU2 regulates IKKα-mediated 131I resistance in thyroid carcinoma TPC-1 cells via the EZH2/H3K27me3 axis

[摘 要] 目的:探讨lncRNA DLEU2通过EZH2/H3K27me3途径调控IKKα介导甲状腺癌(TC)放射性碘抵抗的作用机制。方法:利用TCGA数据库分析TC中DLEU2的表达及其与EZH2的相关性。构建放射性碘抵抗的TPC-1细胞(RR-TPC-1细胞)模型及裸鼠移植瘤模型,通过敲低或过表达DLEU2(si-DLEU2/OE-DLEU2)、抑制EZH2(UNC1999)、过表达IKKα(OE-IKKα)进行干预,采用qPCR、WB、RIP、ChIP、CCK-8、流式细胞术、TUNEL染色及体内成瘤实验检测基因与蛋白表达、表观修饰、细胞增殖、凋亡及肿瘤生长。结果:TCGA分析显示,DLEU2在TC组织中显著上调(P < 0.001),与患者不良预后相关(P = 0.008 4),且与EZH2表达呈正相关(r = 0.390, P < 0.001);RIP证实EZH2与DLEU2存在相互作用/结合(P < 0.05)。体外实验表明,敲低DLEU2可显著下调RR-TPC-1细胞中EZH2、IKKα表达及H3K27me3修饰水平,抑制NF-κB通路活化(P < 0.05或P < 0.01),抑制细胞增殖、促进凋亡(均P < 0.05)。联合敲低DLEU2与抑制EZH2进一步增强上述效应,而过表达IKKα则可部分逆转上述效应(P < 0.05或P < 0.01)。体内实验进一步证实,敲低DLEU2联合抑制EZH2可显著抑制移植瘤生长,增加肿瘤细胞凋亡(均P < 0.01);IKKα过表达则部分逆转上述抗肿瘤效应(P < 0.05或P < 0.01)。结论:lncRNA DLEU2通过招募EZH2催化H3K27me3修饰,间接激活IKKα/NF-κB信号并形成正反馈环路,介导TPC-1细胞131I抵抗。
2.Precision infusion of cryoprecipitate in rare bleeding disorders

The coagulation factors in cryoprecipitate are mainly coagulation factor Ⅷ, fibrinogen, von Willebrand factor (VWF), and coagulation factor ⅩⅢ. This article provides different treatment dosages and plans for different patients with rare coagulation disorders based on their actual conditions referring to international and domestic guidelines and consensus. In order to achieve precise infusion for cryoprecipitate, it is necessary to avoid poor treatment effects caused by insufficient doses, as well as waste of blood components and the risk of thrombosis caused by excessive infusion.
3.Correlation between APTT mixing test results and inhibitor titers in patients with hemophilia A inhibitor

Objective: To study the correlation between activated partial thromboplastin time (APTT) mixing test results and the inhibitor titers in hemophilia A inhibitor-positive patients. Methods: In this cross-sectional study, 41 patients with severe hemophilia A and inhibitors (and negative for lupus anticoagulant) were included from the hemophilia clinic of Shandong Blood Center from February 2022 to February 2024. All patients underwent APTT mixing test. The Rosner's index (RI, including the immediate RI and the RI after 2-hour water bath incubation [water bath 2h RI]), the time-dependent difference (Δ value), and the corrected percentage were calculated based on results of APTT mixing test. The median (interquartile range) of the corresponding indexes were calculated, and the ROC curves for identification of high inhibitor titers using the four indexes (the immediate RI, the water bath 2h RI, the Δ value, and the corrected percentage) were plotted, The correlations between APTT mixing test and inhibitor titers for coagulation factor Ⅷ (Factor Ⅷ, FⅧ) were investigated. Results: The median (lower quartile, upper quartile) of immediate RI, water bath 2h RI, Δ-value and corrected percentage for FⅧ inhibitor positive patients were 11.0 (5.4, 29.3)%, 45.0 (25.7, 75.0)%, 26.2 (7.6, 41.8) s, and 82.2 (58.5, 91.6)%, respectively. The median (lower quartile, upper quartile) of the immediate RI, water bath 2h RI, Δ-value and corrected percentage were 25.2 (13.0, 37.5)%, 64.1 (44.6, 72.6)%, 38.0 (14.3, 38.3) s, and 66.5 (50.1, 82.1)% for the high-titer inhibitor group, and 5.2 (4.2, 9.4)%, 17.9 (8.8, 28.0)%, 13.0 (7.6, 25.4) s, and 92.3 (88.0, 94.3)% for the low-titer inhibitor group. The AUCs of the ROC curves for discrimination between high and low titer inhibitor were: 0.9105 for immediate RI, 0.9118 for water bath 2h RI, 0.8873 for correcter percentage, and 0.6532 for Δ-value. Conclusion: High-titer inhibitors can be highly suspected in hemophiliac patients with an immediate RI >10% and a water bath 2h RI >45%, and the presence of low-titer inhibitors is suspected in patients with a 4-second < immediate RI <10% and a 13% < water bath 2h RI <45%.
4.Acupuncture-synchronized exercise can improve the learning and memory of rats with middle cerebral artery occlusion

Objective:To observe the effect of acupuncture-synchronized exercise training on the learning and memory of rats after middle cerebral artery occlusion (MCAO), and to explore its mechanism.Methods:Among sixty 8-week-old SPF-grade Sprague-Dawley male rats, 6 were randomly chosen to form a sham group, and the rest received MCAO. After successful modeling, 18 rats were assigned at random to either the model, asynchronous or synchronous group, each of 6. The asynchronous group received 20 minutes of acupuncture followed by 20 minutes of treadmill training, while the synchronous group received the acupuncture during their treadmill training. There was no special intervention for the sham and model groups. There were 5 sessions/week for 21 days. Neurological deficit in the rats was assessed using Longa scoring 24h after the modeling and after the 21 days. The localization navigation test was performed on days 17-20 of the experiment, and the latency to escape was recorded. A space exploration experiment was performed the next day, and rats were tested for dwell time and the number of transits of the original platform quadrant. After the intervention, the morphology of hippocampal tissue was observed using HE staining. The mitochondria of the rats′ hippocampal neurons were observed using transmission electron microscopy. MDA and SOD levels were recorded. And the protein expression levels of the Parkin, PINK1, LC3-II/ LC3-Ⅰ, P62 and Beclin-1 genes were detected using western blotting.Results:The model group showed significantly higher average Longa scores than the sham group, with less time spent in the target quadrant and fewer crossings of the platform quadrant. The HE staining revealed numerous degenerating neurons, and swollen mitochondria with vacuolization and sparse cristae. There was also a significant decrease in Parkin, PINK1, LC3-II/LC3-I and Beclin-1 expression and in SOD. But there was a significant increase in P62 expression and MDA, on average. Compared with the model group, both the non-synchronous and synchronous groups showed significantly lower average Longa scores, with more time spent in the target quadrant and more platform crossings. Their neuron morphology was close to normal, with more intact cristae and autophagic bodies in the mitochondria. Those groups showed greater Parkin, PINK1, LC3-II/LC3-I and Beclin-1 expression and had higher SOD levels, on average, with significantly lower P62 expression and less MDA. But compared with the non-synchronous group, the synchronous group spent more time in the target quadrant and had significantly more platform crossings. LC3-II/LC3-I and Beclin-1 expression were higher, on average, as were SOD levels. P62 expression and MDA levels were significantly lower.Conclusions:Acupuncture synchronized with exercise improves the learning and memory of rates after MCAO. The mechanism may be related to reducing oxidative stress levels and activating mitochondrial autophagy.
5.Effects of different storage temperatures and durations on the activity of coagulation factor Ⅷ and Ⅸ in whole blood

Objective: To investigate the effects of different storage temperatures and durations on the activities of coagulation factor Ⅷ (Factor Ⅷ, FⅧ) and coagulation factor Ⅸ (Factor Ⅸ, FⅨ) after whole blood collection, so as to provide data support for the optimal storage conditions. Methods: A total of 16 mL of whole blood was collected from each of the 20 healthy volunteers at our blood center and aliquoted into 8 sodium citrate anticoagulant tubes. Two tubes were immediately centrifuged for the measurement of FⅧ and FⅨ activity levels. The remaining 6 tubes of whole blood were respectively stored under room temperature and low-temperature conditions. At 2, 4, and 6 h, the whole blood samples were centrifuged and analyzed for FⅧ and FⅨ activity levels. The mean values of the two immediately tested tubes were used as the control group, while other tubes were designated as the experimental groups for comparison. Statistical analysis was performed using SPSS 26.0. Results: The activity of FⅧ in whole blood remained stable after 4 hours of storage at both room temperature and low temperature (116.53±25.95 vs 125.22±27.33, 109.77±23.23 vs 125.22±27.33) (P>0.05 for both). However, by 6 hours, FⅧ activity showed a statistically significant decline compared to the control group (108.65±22.92 vs 125.22±27.33, 100.46±20.19 vs 125.22±27.33) (P<0.05 for both), though the room temperature group results were closer to the control values. The activity of FⅨ in whole blood remained stable after 6 hours of storage under both conditions (97.14±19.48 vs 96.76±19.67, 97.10±17.45 vs 96.76±19.6) (P>0.05 for all comparisons). Conclusion: For whole blood samples after collection, storage at either room temperature or low temperature for up to 4 hours does not compromise the accuracy of test results. When stored for 6 hours, FⅨ activity remains stable, whereas FⅧ activity decreases significantly. Notably, FⅧ activity demonstrates better stability at room temperature than under low-temperature conditions within the 6-hour storage.
6.Perioperative nursing care of a child with Angelman syndrome and moderate scoliosis

Objective To summarize the perioperative nursing experience of a child with Angelman syndrome(AS)complicated with moderate scoliosis undergoing posterior 3-dimensional osteotomy correction,fusion,and internal fixation under general anesthesia.Methods The clinical data of the child with AS and moderate scoliosis who underwent surgical treatment in our hospital on Aug.4,2023,were analyzed.A multidisciplinary team was established upon admission.Relevant literatures and evidences were reviewed to develop and implement a"1+X"nursing plan,which included 2 components:"1"(core perioperative nursing priorities for scoliosis)and"X"(multiple AS-related nursing issues and corresponding strategies).Results The surgery was successful,with stable postoperative conditions and significant improvement in spinal curvature.The child's height increased by 7 cm.Discharge occurred on postoperative day 10,with 100%follow-up compliance.The child regained preoperative independent walking ability during follow-up.Conclusion The"1+X"nursing protocol for AS complicated with scoliosis can effectively ensure perioperative safety and promote recovery,which providing insights for perioperative nursing care of other rare diseases complicated with scoliosis.
7.Expression and clinical significance of AMOTL1, molecules of Hippo signaling pathway YAP, and TAZ in cervical cancer tissues

Objective:To investigate the expression and clinical significance of angiomotin-like protein 1 (AMOTL1) and downstream related molecules of Hippo signaling pathway yes-associated protein (YAP) and TAZ in different cervical tissues.Methods:From January 2017 and July 2018 ,102 cervical cancer tissues, 50 cervical squamous epithelial high-grade lesions (HSIL) tissues, 45 low-grade lesions (LSIL) tissues and 50 chronic cervicitis tissues, all of which underwent surgical treatment and pathological diagnosis at Xuzhou Medical University Affiliated Hospital from January 2017 to July 2018, were selected. The expression of AMOTL1 and Hippo pathway downstream related proteins YAP and TAZ were evaluated by immunohistochemistry and their relationship with the clinicopathological parameters of cervical cancer were examined in the cervical tissues of each group. The comparison of inter group rates in count data was conducted using the chi square test. Adopting Spearman correlation analysis method to analyze the relationship between AMOTL1, YAP, and TAZ. Single factor survival analysis was performed using Kaplan Meier curves. Kaplan Meier method and multivariate Cox regression analysis were used to investigate the factors affecting the prognosis of cervical cancer patients.Results:The immunohistochemical results showed that the positive rates of AMOTL1 in chronic cervicitis, LSIL, HSIL, and cervical cancer were 12.00% (6/50), 17.78% (8/45), 28.00% (14/50), and 76.47% (78/102), respectively. The positive expression status of AMOTL1 in chronic cervicitis, LSIL, HSIL and cervical cancer were 12.00%(6/50), 17.78%(8/45), 28.00%(14/50), 76.47%(78/102). The positive rates of YAP in chronic cervicitis, LSIL, HSIL, and cervical cancer were 10.00% (5/50), 15.56% (7/45), 30.00% (15/50), and 63.73% (65/102), respectively. The positive expression of YAP in cervical cancer tissues was higher than that in other groups ( χ2=56.66, P<0.001). The positive rates of TAZ in chronic cervicitis, LSIL, HSIL, and cervical cancer were 8.00% (4/50), 17.78% (8/45), 30.00% (15/50), and 71.57% (73/102), respectively. The positive expression of TAZ in cervical cancer tissues was higher than that in other groups ( χ2=74.71, P<0.001), AMOTL1 positive expression is associated with the International Federation of Gynecology and Obstetrics (FIGO) staging ( χ2=16.28, P=0.001), tissue differentiation degree ( χ2=30.16, P<0.001), and lymph node metastasis ( χ2=5.81, P=0.016), YAP positive expression is associated with FIGO staging ( χ2=15.99, P<0.001), differentiation degree ( χ2=25.06, P<0.001), tumor diameter ( χ2=13.63, P=0.003), and lymph node metastasis ( χ2=5.78, P=0.016), and TAZ positive expression is associated with FIGO staging ( χ2=14.49, P<0.001). tissue differentiation degree ( χ2=25.32, P<0.001), and the presence or absence of lymph node metastasis ( χ2=4.95, P=0.026) are related. Spearman correlation analysis showed a positive correlation between AMOTL1 and YAP expression ( r=0.65, P<0.001), and a positive correlation between AMOTL1 and TAZ expression ( r=0.72, P<0.001), There was a positive correlation between YAP and TAZ expression ( r=0.68, P<0.001). Kaplan Meier survival analysis showed that individuals with positive expression of AMOTL1, YAP, and TAZ proteins had shorter survival times than those with negative expression ( χ2 values were 9.84, 8.64, and 18.57, respectively; P values were 0.002, 0.003, and <0.001, respectively). Multivariate Cox regression analysis showed that FIGO staging ( HR=3.18, 95% CI 1.09-9.29, P=0.034), lymph node metastasis ( HR=16.74, 95% CI 3.20-87.45, P=0.010), AMOTL1 positive expression ( HR=13.06, 95% CI 1.41-121.08, P=0.024), and YAP positive expression ( HR=9.75, 95% CI 1.59-59.52, P=0.014) were risk factors for poor prognosis in cervical cancer patients. Conclusion:AMOTL1, YAP, and TAZ may jointly participate in the malignant transformation process of cervical cancer. AMOTL1 and YAP are closely related to prognosis and may become potential prognostic indicators for cervical cancer.

Case report form(CRF)is an important tool to obtain clinical research data.Scientifically designed and properly recorded CRF is of great significance to improve the quality of clinical research.The author summarized the common problems existing in the design and recording of CRF in traditional Chinese medicine(TCM)clinical studies,including:①Delayed design time point.②Unstandardized CRF design,recording and modification.③Imperfect records of combined medication/therapy.④Insufficient quantification of syndrome efficacy indicators and objectification of tongue and pulse.⑤ Insufficient protection of subjects' privacy.⑥ Unstandardized records of adverse events.Accordingly,this paper discusses some solutions to provide reference for other TCM researchers,including:①Advance CRF design time.②Standardize CRF design,recording and modification.③Design drug combinations/therapies according to Clinical Data Acquisition Standards Harmonization(CDASH).④Standardization of syndrome and tongue and pulse.⑤Use of subject identification codes to protect privacy.⑥AE records should vary from study to study.
9.Aerobic exercise regulates autophagy to improve myocardial fibrosis in mice with atherosclerosis

Objective:To explore the mechanism of aerobic exercise regulating autophagy to improve myocardial fibrosis in mice with atherosclerosis.Method:Eight weeks old male ApoE-/-knockout mice were randomly divided into model group,aerobic exer-cise group and blank control group after one-week adaption,with nine mice in each group.The atherosclerosis model was established by fed a"Western type"diet for 12 weeks and the aerobic exercise group was re-ceived aerobic training at the same time.After 12 weeks,Oil Red O staining was used to observe the patho-logical status of atherosclerosis in the two groups,and Masson dyeing was used to detect myocardial fibrosis in each group.The expressions and localization of ATG5,LC3BⅡ/Ⅰ,Beclin-1 and p62 in myocardial tissue of mice in each group were detected by immunofluorescence.The protein levels of ATG5,LC3BⅡ/Ⅰ,Beclin-1 and p62 in myocardial tissue of each group were detected by Western Blot.Result:Oil red O staining showed that obvious atherosclerotic plaque formation was found in the model group,while the area of atherosclerotic plaque in the aerobic exercise group was significantly less than that in the model group,indicating the degree of atherosclerosis was significantly improved.Masson dyeing showed that myocardial fibrosis in the aerobic exercise group was significantly improved compared with the model group,and the degree of myocardial fibrosis in both groups was greater than that in the blank control group.Immunofluorescence and Western Blot analysis showed that the expression levels of ATG5,LC3B II/I,and Be-clin-1 in the myocardial tissue of aerobic exercise group mice were significantly increased,while the level of p62 protein was significantly reduced(P<0.05).Conclusion:Aerobic exercise can significantly improve myocardial fibrosis in mice with atherosclerosis,and the mechanism may be related to aerobic exercise activated autophagy-related factors in myocardial tissue of mice.
