1.Thalassemia screening and genotype analysis in 488 sperm donors in Chongqing Human Sperm Bank

Objective:To investigate the carriage of the thalassemia gene in sperm donors in Chongqing,China by analyzing the thalas-semia screening and gene mutation types of sperm donors in Chongqing Human Sperm Bank(CHSB),and to provide a reference for thalassemia screening in sperm donors in Chongqing.Methods:Blood samples were collected from 488 sperm donors in CHSB(donors with all tests qualified during the quarantine)from December 2021 to August 2024.A complete blood count analysis was performed to measure the mean corpuscular volume(MCV)and mean corpuscular hemoglobin(MCH).Polymerase chain reaction(PCR)/agarose gel electrophoresis was used to detect deletional α-thalassemia mutations.PCR and DNA reverse dot blot hybridization were used to detect 3 common non-deletional α-thalassemia mutations and 17 common β-thalassemia mutations.The test results were then analyzed.Results:Among the 488 sperm donors aged 25.64±4.95 years,15(3.07%,all Han nationality)tested positive for thalassemia.This carriage rate was lower than that of the thalassemia population in Chongqing(9.24%).Among the 15 thalassemia carriers,13 had deletional α-gene mutations(-α3.7 genotype in 11 cases[73.33%],-α4.2 genotype in 1 case[6.67%],and-SEA genotype in 1 case[6.67%]),and 2 had non-deletional α-gene mutations(both with the WSM heterozygous mutation).There was only 1 case of abnor-mal MCV and MCH2 indicators among thalassemia carriers(screening positive rate was 6.67%).Conclusion:Thalassemia screening in sperm donors in CHSB reveals a low thalassemia carriage rate,reflecting geographic(predominantly Sichuan and Chongqing ori-gins)and demographic(healthy men of childbearing potential)characteristics.Given the significant impact of thalassemia on the off-spring conceived through sperm donation,it is recommended to conduct thalassemia gene screening among sperm donors in human sperm banks.
2.Mechanism of"treating different diseases with the same method"for chronic obstructive pulmonary disease and diabetes with Shenling Baizhu San based on network pharmacology and molecular docking

Objective To explore the mechanisms of"treating different diseases with the same method"for chronic obstructive pulmonary disease(COPD)and diabetes mellitus(DM)with Shen-ling Baizhu San based on network pharmacology and molecular docking.Methods Comprehensive databases were searched to collect the active ingredients and targets of Shenling Baizhu San,which were then performed standardized process.Relevant targets for COPD and DM were collected from data platforms such as GeneCards and Online Mendelian Inheritance in Man(OMIM).The intersection tar-gets of the drug and diseases were identified using Venn software,imported into the STRING database to construct a protein-protein interaction(PPI)network,and performed visualized analysis using Cyto-scape software.Gene Ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed using R software.Molecular doc-king and visualization analysis were conducted using the CB-Dock2 platform.Results After screening,215 potential targets of Shenling Baizhu San were obtained.Venn diagram analysis showed that there were 70 potential intersection targets for the treatment of COPD and DM with Shenling Baizhu San.Key active ingredients included quercetin,stigmasterol,kaempferol,sitosterol,and luteolin,while core targets included tumor necrosis factor(TNF),interleukin-6(IL-6),interleukin-β(IL-1 β),prostaglandin-endoperoxide synthase 2(PTGS2),and tumor protein p53(TP53).GO functional en-richment analysis yielded 2,599 biological process terms,49 cellular component terms,and 230 molecular function terms.The main pathways obtained from KEGG pathway enrichment analysis in-cluded lipid and atherosclerosis,interleukin-17(IL-17)signaling pathway,TNF signaling pathway,and human cytomegalovirus infection.Molecular docking results showed that stigmasterol had the best docking activity with PTGS2.Conclusion Shenling Baizhu San may achieve the"treatment of different diseases with the same method"for COPD and DM through multiple components,multiple targets,and multiple pathways,and there is a certain correlation in the treatment mechanisms of two diseases,providing a theoretical basis and direction for subsequent research.
3.Case-control study on association of female reproductive factors with risk of papillary thyroid cancer

Background Papillary thyroid cancer is the result of a variety of pathogenic factors. The prevalence of papillary thyroid cancer varies greatly in different regions, and the disease is more harmful to women. Objective This study aims to explore the relationship between reproductive factors and papillary thyroid cancer, and to provide basic data for prevention and control of the disease. Methods A 1∶1 age (±3 years) matched case-control study was conducted in 331 pairs of newly confirmed papillary thyroid cancer cases and controls from two hospitals in Shanghai from November 2012 to December 2013. Comparisons were made in the history of menstruation, pregnancy, gynecological and breast diseases, and other variables between the two groups. Results The results of univariate analysis indicated that the proportions of education below bachelor degree, married, and mainly manual workers in the case group were significantly higher than those in the control group (P<0.05); the proportion of those with a family history of thyroid diseases in the case group was significantly higher than that in the control group (P<0.05); the proportions of the cases with irregular menstruation (77.34%), a history of using oral contraceptive or hormone drugs (24.77%), a history of benign gynecological diseases (31.72%), and a history of gynecological surgery (9.67%) were significantly higher than the proportions in the control group (P<0.05). After adjusting potential confounding factors such as history of CT examination, age, kinds of family salt, total iodine intake every day, education level, occupation, marital status, body mass index, and family history of thyroid diseases, the results of multiple logistic regression analysis showed that irregular menstruation (OR=1.767, 95%CI: 1.122-2.782; P=0.014), surgical menopause (OR=12.787, 95%CI: 3.202-51.057; P<0.001), pregnancy >1 time (OR =2.490, 95%CI: 1.196-5.184; P=0.015), and the history of using oral contraceptive or hormone drugs (OR=2.389, 95%CI: 1.338-4.268; P=0.003) were the risk factors of papillary thyroid cancer. Conclusion Irregular menstruation, surgical menopause, history of pregnancy, and history of using oral contraceptive or hormone drugs might be the risk factors of papillary thyroid cancer. To reduce the incidence of papillary thyroid cancer, strengthened health education and rational use of contraceptives are recommended.
