1.TRIM25 inhibits Japanese encephalitis virus replication in U251 cells by up-regulation of the IFN-β and degrading the viral capsid protein

Objective:To investigate the inhibitory effect of tripartite motif-containing 25 (TRIM25) on the replication of Japanese encephalitis virus (JEV) in cells and its molecular mechanism.Methods:Human glioma cells (U251 cells) and Kunming mice were infected with JEV, and then the cells and brain tissue samples were collected. The transcription levels of six TRIM genes were detected by real-time PCR, and the expression of TRIM25 in cells was detected by Western blot. U251 and A549 cells overexpressed with TRIM25 and U251 cells knocked out with TRIM25 gene were constructed. Cells were infected with JEV, and the replication of JEV was detected by viral plaque assay, real-time PCR and Western blot. The interaction of TRIM25 with viral proteins was investigated by co-immunoprecipitation (Co-IP) and indirect immunofluorescence assay. The expression of IFN-β in overexpressed TRIM25 cells was detected by real-time PCR and ELISA.Results:JEV infection promoted the expression of TRIM25 in cells and mouse brain tissues. TRIM25 overexpression restricted JEV replication in U251 and A549 cells, while TRIM25 knockout enhanced JEV replication. TRIM25 overexpression upregulated the level of IFN-β in cells. TRIM25 interacted with JEV capsid protein and promoted the degradation of capsid protein.Conclusion:TRIM25 can inhibit the replication of JEV in cells by upregulating IFN-β and promoting the degradation of JEV C protein.
2.Investigation of radon exposure hazard awareness among non-uranium miners in Chongqing, China

Objective To investigate the awareness of radon exposure hazards among non-uranium miners in Chongqing, China. Methods A survey was conducted among 177 male miners from eight non-uranium metal mines in Chongqing to collect data on basic information, personal habits, and the rate of radon awareness. Factors affecting radon awareness were analyzed using chi-square test and logistic regression model. Results The awareness rate of radon among miners was 23.73%. The chi-square test indicated significant difference in the radon awareness rate among miners with different levels of education (χ2 = 10.28, P < 0.05), while there was no significant difference across different ages, years of work, labor relations, job categories, and types of miners (P > 0.05). Binary logistic regression analysis showed that a college (junior college) or higher level of education, a high school level of education, and working in mines were factors affecting the radon awareness among miners (χ2 = 4.030, 9.150, 11.776, P < 0.05). Conclusion Miners lack awareness of radon, and there is an urgent need to strengthen education and propaganda regarding the hazards of radon.
3.Investigation on radon exposure dose monitoring and health status of non-uranium miners in Chongqing

Objective:To investigate the radon exposure levels and health effects among non-uranium miners in Chongqing.Methods:From June 2022 to December 2023, a survey was carried out on basic information, radon exposure and occupational health status of 110 male miners with good cooperation attitudes in 6 non-uranium metal mines in normal operation in Chongqing. The logistic regression model was utilized to statistically analyze the impact of miners′ individual radon doses on abnormalities in chromosomal aberration analysis, micronucleus examination, tumor marker tests, and pulmonary CT imaging.Results:Monitoring results from passive integrated radon personal dosimeters indicated that three miners had an annual dose exceeding 2 mSv. Logistic regression analysis revealed that abnormalities in pulmonary CT imaging were associated with miners′ age, tenure, and alcohol consumption ( χ2=12.18, 9.63, 4.87, P<0.05), with no significant correlation with smoking or individual radon dose. There was a significant correlation between individual radon dose and abnormalities in tumor markers; for every additional 1 mSv of effective dose from radon and its progeny, the risk of abnormality increased by 2.25 times ( χ2=4.57, P < 0.05). Conclusions:Some miners in Chongqing are exposed to an annual dose exceeding 2 mSv, and there is a correlation between individual radon doses and abnormalities in lung cancer serum tumor markers. It is necessary to focus on monitoring the radon exposure risks for miners, carry out regular health checks, and enhance the health protection for the miner population.

5.Study on the Correlation between Serum CXCL4,CXCR3 Levels and Postoperative Cerebral Vascular Spasm in Patients with Ruptured Intracranial Aneurysms

Objective To explore the predictive value of serum levels of CXC chemokine ligand 4(CXCL4),CXC receptor 3 chemokine(CXCR3)for postoperative cerebral vasospasm(CVS)in patients with ruptured intracranial aneurysms.Methods The clinical data of 106 patients with intracranial aneurysms rupture who under went craniotomy and clipping surgery at the Central Hospital of Enshi Tujia and Miao Autonomous Prefecture from October 2020 to December 2022 were collected.The incidence of CVS was observed.Serum CXCL4 and CXCR3 levels were compared at 1,3,7 days after surgery between patients without CVS and patients with CVS.Spearman correlation coefficient was used to analyze the relationship between serum CXCL4,CXCR3 levels and CVS degree.ROC curve was used to analyze the value of serum CXCL4 and CXCR3 levels in predicting postoperative CVS.Results Among 106 patients with ruptured intracranial aneurysms,a total of 38 cases(35.85%)developed CVS after surgery,and 68 cases(64.15%)did not develop CVS.There was no statistically obvious difference in preoperative serum levels of CXCL4 and CXCR3 between the CVS group and the non CVS group(t=0.104,0.141,all P>0.05).The serum levels of CXCL4 and CXCR3 in the non CVS group were lower than those before surgery 1,3,and 7 day after surgery,and the differences were statistically significant(q=14.867,17.525,24.384;16.274,20.251,34.237,all P<0.05).Compated with preoperative levels,the levels of serum CXCL4 and CXCR3 in the CVS group decreased on postoperative 1 and 7 days,and the differences were statistically significant(q=9.486,4.858;10.760,9.104,all P<0.05).Compared with the mild CVS group,the serum levels of CXCL4 and CXCR3 in the moderate and severe CVS groups were obviously increased(q=2.982,5.992;2.961,3.465),and the serum levels of CXCL4 and CXCR3 in the severe CVS group were higher than those in the moderate CVS group(q=3.564,4.036),and the differences were statistically significant(all P<0.05),respectively.Spearman results showed that serum levels of CXCL4 and CXCR3 were positively correlated with the severity of CVS(r=0.491,0.483,all P<0.05).The ROC curve results showed that the AUC of CXCL4 and CXCR3 combined in predicting postoperative CVS in patients with intracranial ruptured aneurysms was higher than that of CXCL4,CXCR3 alone,and the differences were statistically significant(Z=2.937,2.750,all P<0.05).Conclusion The serum levels of CXCL4 and CXCR3 in ruptured intracranial aneurysms patients with postoperative CVS are obviously elevated,and the combined detection of the two can effectively predict the occurrence of CVS.

8.Fear of falling in the initial ambulation day among patients undergoing major abdominal surgery:the latent profile analysis and nursing enlightenment

Objective This study aimed to explore the potential categories and characteristics of the fear of falling in the initial ambulation day among patients who underwent major abdominal surgery,analyze the factors influencing different potential categories,and propose implications for the care of such patients.Methods A total of 213 patients who u nderwent elective major abdominal surgery in the Department of Gastrointestinal Gland Surgery and Hepatobiliary Surgery in the First Affiliated Hospital of Guangxi Medical University from April to August 2023 were selected as the study subjects.The general data questionnaire and the Revised Version of the Fall Fear Scale and Patient Fall Risk Perception Scale were used to investigate the patients on the day they first got out of bed after surgery.The potential profile of the fall fear characteristics of the study subjects was analyzed,and the relevant influencing factors among different categories were explored by univariate analysis,variance analysis,and logistic regression analysis.Results A total of 202 patients who underwent major abdominal surgery were included in this study.The total score of the fear of falling when getting out of bed for the first time was(35.28±8.85).The fear of falling showed 3 potential categories:low-level fear(22.28%),medium-level fear(47.52%),and high-level fear(30.20%).A significant difference was found in the first time of getting out of bed among different types of fear(H=19.292,P<0.001).Results of multi-factor logistic regression analysis showed that patients with high levels of personal activity factors in fall perception and those who underwent pancreatic surgery were likely to develop medium-level fear(P=0.037).Patients with high levels of perceived environmental factors,high levels of perceived personal activity factors,and low self-efficacy were likely to develop high-level fear(P<0.05).Conclusion The fear of falling on the initial ambulation day in patients who underwent major abdominal surgery was distributed in categories,and the proportion of patients with medium and high fear levels was high.Patients'fall risk perception,self-efficacy,and surgical site are important factors influencing fear of falling.Therefore,postoperative nursing staff should focus on developing targeted health education and activity programs to reduce the level of fall fear.
9.Effects of Fuke Qianjin Formula and its modifications on the sequelae of pelvic inflammatory disease in rats

AIM To study the effects and possible mechanism of Fuke Qianjin Formula and its modifications on sequelae of pelvic inflammatory disease(PID)in rats.METHODS The SD rats were randomly divided into the normal group,the sham operation group,the model group,Kangfuyan group(FYK,0.756 g/kg),Fuke Qianjin Formula group(FKQ,2.7 g/kg),Yijun Formula group(YJ,2.7 g/kg),Kangyan Formula group(KY,2.7 g/kg)and Buyi Formula group(BY,2.7 g/kg).After 28 days of corresponding administration by gavage,the rats were put to death to procure their uterus,spleen,thymus and liver for the calculation of the organs indices levels;the observation of the general morphology of uterus and oviduct and histopathological examination of uterus and oviduct;the measurement of.the serum levels of IL-1β,IL-18 and IL-6 by ELISA;the detection of the uterine expressions of TGF-β1,caspase-1,NLRP3,IL-18 and CTGF by Western blot;and the detection of the expressions of CD4+and CD8+in spleen lymphocytes by flow cytometry.RESULTS Compared with the model group,FKQ and YJ group,KY group and BY group displayed decreased indices levels of the uterus,spleen and thymus(P<0.01);FKQ group and KY group displayed reduced uterine swelling,increased endometrial glands,and reduced inflammatory exudation(P<0.01),and lower serum levels of IL-1β,IL-18 and IL-6(P<0.01);FKQ group,KYgroup and BY group displayed decreased protein expressions of TGF-β1 and CTGF(P<0.05,P<0.01);FKQ group and KY group displayed decreased protein expressions of NLRP3,caspase-1 and IL-18(P<0.05,P<0.01);and FKQ group and BY group displayed increased CD4+/CD8+ratio(P<0.01).CONCLUSION Being protective to sequelae of PID rat models,Fuke Qianjin Formula and its modifications may contribute their anti-inflammatory efficacy by inhibiting the activation of NLRP3 body to hinder the activation of caspase-1,reduce the secretion and release of pro-inflammatory factor IL-18,and thus inhibit pyroptosis.
10.Antibacterial magnesium oxide-calcium phosphate composite coating prepared by combining electrodeposition and sol-gel impregnation

BACKGROUND:Calcium phosphate(CaP)coatings are widely used to improve the integration of titanium implants into bone but these coatings are associated with risks of infection.It is thus desirable to confer antibacterial properties to CaP coatings. OBJECTIVE:To prepare CaP-MgO composite coatings by impregnating magnesium oxide(MgO)sol into CaP coatings and assess the in vitro antibacterial activities and cytocompatibility. METHODS:An electrolyte was determined by titration and used for CaP coating electrodeposition on titanium(referred to as Ti-CaP).MgO was impregnated into the coating by immersing in an MgO sol with different mass fractions(15%,30%,50%)and subsequently calcined to form MgO-CaP composite coatings,which were recorded as Ti-CaP-15Mg,Ti-CaP-30Mg and Ti-CaP-50Mg,respectively.Microstructure,tensile properties,critical load,and Mg2+ release of coatings in vitro were characterized.Antibacterial activity was assayed using spread plate method by culturing S.aureus on the pure titanium sheet surface and Ti-CaP,Ti-Cap-15mg,Ti-Cap-30mg and Ti-Cap-50mg surfaces for 24 and 48 hours.Mouse osteoblast suspension was inoculated on pure titanium sheets and Ti-CaP,Ti-CaP-15Mg,Ti-CaP-30Mg and Ti-CaP-50Mg coated titanium sheets,respectively.Cell proliferation was detected by CCK-8 assay,and cell survival rate was calculated.The morphology of composite coating soaked in DMEM was also observed. RESULTS AND CONCLUSION:(1)Homogeneous,microporous CaP coatings consisting of octacaclium phosphate crystal flakes were prepared on titanium by electrodeposition.After sol impregnation-calcination,MgO aggregates were filled into the inter-flake voids.The extent of MgO filling and Mg concentration in the coating increased with the number of sol impregnation procedures.When immersed in phosphate buffered saline,all composite coatings actively released Mg2+ within 1 day;subsequently,the Mg2+ release slowed down on day 3.A small amount of Mg2+ release was still detected on day 7.The yield strength,tensile strength and fracture growth rate of Ti-CaP-30Mg coated titanium were not significantly different from those of pure titanium(P>0.05).There was no significant difference in the critical load of Ti-CaP,Ti-CaP-15Mg,Ti-CaP-30Mg and Ti-CaP-50Mg groups(P>0.05).(2)Except that pure titanium sheet and Ti-CaP had no antibacterial properties,the other samples had good antibacterial properties,and the antibacterial rate increased with the increase of MgO content in the coating.(3)After 1 and 3 days of co-culture,the cell survival rate of Ti-CaP-15Mg,Ti-CaP-30Mg and Ti-CaP-50Mg groups was lower than that of pure titanium group and Ti-CaP group(P<0.05).After 5 and 7 days of culture,there was no significant difference in cell survival rate among five groups(P>0.05).The content of MgO in the coating decreased gradually with the time of immersion in the medium.(4)The MgO sol impregnation added antibacterial properties to the CaP coatings while retained their biocompatibility.
