1.Epidemic characteristics and spatiotemporal distribution analysis of human brucellosis in Henan Province, 2005 - 2021

Objective:To analyze the epidemiological characteristics and spatiotemporal characteristics of human brucellosis in Henan Province.Methods:Data of human brucellosis in Henan Province from 2005 to 2021 were collected through the China Disease Prevention and Control Information System, and a descriptive epidemiological method was used to analyze the epidemic profile of brucellosis in Henan Province and the three distribution characteristics. Global and local spatial autocorrelation were used to analyze the spatial distribution and the hot spots of brucellosis in Henan Province, respectively, and spatiotemporal scanning was used to analyze the spatiotemporal clustering regions of brucellosis in Henan Province.Results:A total of 39 862 brucellosis cases were reported in Henan Province from 2005 to 2021, with an average annual incidence of 2.44/100 000, and the number of cases showed an overall increasing trend each year (χ 2trend = 11 127.85, P = 0.001). The onset months were mainly concentrated from March to July, accounting for 59.00% (23 517/39 862), with May as the peak (5 478 cases). Cases of brucellosis were reported in 157 counties (cities, districts) of the province. The ratio of male to female was 2.52∶1.00 (28 542/11 320). Farmers were the main occupation, with 32 985 cases (82.75%). The age of onset was mainly 45 to 65 years old, with 20 226 cases (50.74%). The global spatial autocorrelation analysis showed that the global Moran's I was > 0, Z > 1.96, and P < 0.05 in all years except 2006 - 2008, showing spatial clustering. Further local spatial autocorrelation analysis was performed, and high-high and low-low clustering areas existed in 2012 - 2021 ( P < 0.01). Spatiotemporal scanning analysis showed that there was one spatiotemporal cluster in the high incidence area and two spatiotemporal clusters in the low incidence area. The high incidence cluster was centered in Neixiang County, covering 48 counties (cities, districts) including Song County and Ruzhou City, and the aggregation time was from 2014 to 2021. The two low incidence clusters were centered in Yongcheng City and Boai County, covering 58 and 18 counties (cities, districts), respectively, and the aggregation time was 2016 - 2021 and 2005 - 2012, respectively. Conclusion:The overall incidence of brucellosis in Henan Province is on the rise from 2005 to 2021, with middle-aged and elderly male farmers as the main affected population, and there are spatiotemporal clusters of brucellosis in Henan Province.
2.Epidemiological investigation and genomic characterization of the first case of fever with thrombocytopenia syndrome in Puyang

Objective:To report the first case of sever fever with thrombocytopenia syndrome caused by severe fever with thrombocytopenia syndrome virus (SFTSV) in Puyang city, and to study the epidemiological and molecular characteristics of S, M, L fragments of the SFTSV isolate.Methods:The epidemiological characteristics of this case was analyzed with epidemiological methods. SFTSV was isolated from the patient′s serum sample. Nucleic acid of SFTSV was extracted and detected by fluorescent RT-PCR. A multiplex PCR method was constructed to amplify the nucleic acid sequence of the virus. whole-genome sequencing was performed on the next-generation sequencing platform. MEGA11 and DNAStar was used for homology analysis and a phylogenetic tree was constructed.Results:Epidemiological investigation showed that the patient and his close contacts had no history of travel or tick bite within 14 d, but had a history of fieldwork. The patient′s serum sample was positive for SFTSV nucleic acid. Genetic analysis showed that the S, M, L gene fragments of the first SFTSV isolate in Puyang belonged to genotype E. This isolate shared 94.8%-99.6%, 94.0%-99.8% and 95.7%-99.7% nucleotide sequence homology with the representative strains acquired from GeneBank in S, M, L gene fragments, respectively.Conclusions:This case was the first case of SFTSV-caused severe fever with thrombocytopenia syndrome in Puyang. The SFTSV isolate shared a close homology with domestic isolates, but its genotype was significantly different from the SFTSV strains isolated in Henan in recent years, indicating that it might an imported case from other places in Henan Province or Hubei Province. Disease monitoring and professional training for medical personnel should be strengthened and more attention should be paid to the evolution and mutation of SFTSV.
3.An investigation on serum antibody level of varicella-zoster virus in healthy population in Beijing.

Objective: To assess the level and trend of varicella-zoster virus (VZV) antibody among healthy population in Beijing in 2017, after the five-year implementation of the two doses varicella vaccination strategy in 2012, and to provide evidence for scientific evaluation of immunization strategy. Methods: A total of 2 144 subjects in ten age groups from 8 districts of Beijing city were recruited in this study using cross-sectional survey based on multi-stage cluster random sampling method. Serum samples were collected and VZV antibody was detected by ELISA. The influencing factors of antibody concentration and positive rate were analyzed and compared with the study in 2012. The antibody concentration and antibody positive rate were analyzed by nonparametric test and χ² test respectively. Results: The ratio of subjects with registered residence in Beijing city to other provinces was 1∶1. The ratio of male to female was 1∶1.08. The median concentration of VZV antibody was 341.4 (78.6, 1 497.8) mIU/ml, and the total antibody positive rate was 71.1% (1 524/2 144). There were significant differences in antibody positive rate (χ²=736.39, P<0.01) and antibody concentration (χ²=740.34, P<0.01) among different age groups. The antibody positive rate generally increased with age (χ²trend=7.32, Ptrend<0.01). Among 862 children under 14 years old, the antibody positive rate of two doses vaccination 72.8% (182/250) was significantly higher than that of one dose vaccination 51.9% (154/297) (χ²=25.14, P<0.01). There was significant difference between 1-4 years old group (χ²=11.71, P<0.01) and 10-14 years old group (χ²=5.95, P=0.02), but not in 5-9 years old group (χ²=3.00, P=0.07). Compared with the study in 2012, the antibody positive rate increased in 5-9 years old group (χ²=14.35, P<0.01) and decreased in 1-4 years old group (χ²=11.51, P=0.01) in 2017. Conclusion: The recommended varicella booster vaccination has significantly improved the VZV antibody level of children in Beijing city. In the future, it is necessary to explore a more optimized two doses varicella vaccination schedule for children in combination with epidemiological evidence.

4.Establishment and evaluation of a genome-wide multiplex PCR sequencing method for severe fever with thrombocytopenia syndrome virus based on high-throughput sequencing technology

Objective:To establish a sequencing method for the genome of severe fever with thrombocytopenia syndrome virus (SFTSV) based on next-generation sequencing (NGS).Methods:SFTSV RNA was extracted from serum samples of patients with severe fever with thrombocytopenia syndrome. SFTSV-specific primers were designed using Primer 5.0 software. A multiplex PCR method was constructed and used to amplify the nucleotide sequence of SFTSV. Whole-genome sequencing was performed on the NGS platform.Results:The whole genes of SFTSV isolates in 28 serum samples were amplified by the multiplex PCR with a coverage over 94%. Sequencing and phylogenetic analysis of those strains revealed that the predominant strains ( n=20) belonged to genotype A, followed by genotypes B ( n=4) and E ( n=3). Conclusions:A high-throughput sequencing method for SFTSV based on multiplex PCR was established in this study. This method was characterized by high specificity and good quality and could improve the sequencing efficiency.
6. Chemical Components and Clinical Application of Abri Herba

Abri Herba,a kind of plant of Leguminosae family,is widely distributed in Guangdong and Guangxi,with rich resources.With a long medication history in China,its whole-plant, apart from plant seeds have been used as medicine, with the effects in removing dampness and removing jaundice, clearing heat and detoxifying,and clearing the liver and relieving pain. By accessing CNKI, Wanfang date,VIP Web, ScienceDirect,FMRS, Pubmed and multiple domestic and foreign databases,recent literatures on chemical constituents and pharmacological action and clinical trials of Abri Herba were collected and summarized in a review.According to a great quantity of relevant domestic and foreign literatures.Abri herba contains abundant chemical constituents, such as betulinic acid and other triterpenoids, catechin and other flavonoids,abrine and other alkaloids, chrysophanol and other anthraquinones, as well as inorganic elements.Abri Herba has a wide range of pharmacological effects due to rich material basis. Abri Herba, as a folk traditional herb, is used to treat jaundice with damp-heat pathogen, distending pain in the stomach, acute mastitisand gall, steatosis hepatis, hepatitis and internal traumatic injury. It also has many other effects in resisting tumor,oxidation,bacteria,virus and inflammation, relieving pain, promoting wound healing and regulating immunity.At present, there is single herb or traditional Chinese medicine compounds of Abri Herba in clinic, and the latter is the majority. Abri Herba can play a better pesticide effect by combining with traditional Chinese medicine. With a wide range of pharmacological effects,cheap price and easy cultivation. Abri Herba has high economic and social benefits, great potential for development and broad market prospects.Based on domestic and foreign studies on Abri Herba in past 30 years, the recent progress in the studies on chemical constituents,biological activities and clinical applications of this plant was reviewed in this paper, in the expectation of providing reference for further development and comprehensive utilization of Abri Herba medicinal resources.

9.Correlations of Four Genetic Single Nucleotide Polymorphisms in Brain-Derived Neurotrophic Factor with Posttraumatic Stress Disorder.

OBJECTIVE: To investigate the correlations of four genetic single nucleotide polymorphisms (SNPs) of brain-derived neurotrophic factor (BDNF) with posttraumatic stress disorder (PTSD). METHODS: A total of 300 patients with sporadic PTSD and 150 healthy subjects (the control group) were selected according to the diagnostic criteria of PTSD (DSM-IV), and the genotypes of the BDNF SNPs G-712A, C270T, rs6265, and rs7103411 were detected by polymerase chain reaction and direct DNA sequencing to determine intergroup differences in the genotypes and allele frequencies; the p values were corrected with the permutation test. RESULTS: The genotypes and allele frequencies of the SNPs G-712A, rs6265, and rs7103411 of BDNF showed no significant intergroup differences (p>0.05). However, the genotype and allele frequencies of C270T showed significant differences between the PTSD group and the control group (p<0.05). CONCLUSION: The SNP C270T of BDNF may be associated with PTSD. Individuals carrying the polymorphic T allele of C270T may be more likely to suffer from PTSD.

10.Effects of Rhubarb, Polygonum cuspidatum on hemodynamics and blood lipid in rats with blood stasis syndrome

Objective To explore the effect of Rhubarb and Polygonum cuspidatum and their combination on blood stasis model rats.Methods Forty female SPF rats were chosen and were randomly divided into blank,model,test I,test Ⅱ,test Ⅲ groups,8 rats in each group.The rats of test I,test Ⅱ,test Ⅲ groups were given 30 mg · kg-1 Rhubarb drug,Polygonum,Rhubarb Polygonum cuspidatum (1:1),qd;blank and model groups were given 0.9% NaCl at 5 mL · kg-1 dose,qd.The rats were treated for consecutive 10 days.After lavage for 1 h,except the blank group,the rats in other groups were given adrennaline hydrochloride 1mg · kg-1 by intraperitoneal injection.After 24 h,they were injected the same dose of adrenaline again.After the acute blood stasis model was prepared,the rats were anesthetized to collect blood from the abdominal aorta After anticoagulation treatment,the hemorheology index and lipid index of all samples were tested by automatic blood rheology tester and automatic biochemical analyzer.Results The blood biscosity in blank and test Ⅲ I groups were (3.26±1.33) and (4.51 ±1.15) mPa · s at 200/s,(4.21 ±1.41) and (5.80 ±1.44) mPa · s at 50/s,(9.75 ±2.97) and (13.27 ±3.14) mPa · s at 5/s,(25.58 ±7.24) and (34.49 ±7.95) mPa · s at 1/s,with significant difference (all P < 0.05).The erythrocyte aggregation index in the blank,test Ⅰ,test Ⅱ groups were (8.04 ± 0.80),(7.39 ± 0.31) and (7.18 ± 0.62),respectively.The difference between the blank and test Ⅰ groups was statistically significant (P < 0.05).The difference between the blank and test Ⅱ groups were highly statistically significant (P <0.01).Triglyceride in model,test Ⅱ groups were (0.92 ±0.39) and (0.61 ±0.15) mmol · L-1,with significant difference (P < 0.05).Conclusion Rhubarb,Polygonum cuspidatum,and Rhubarb-Polygonum cuspidatum combination all had good effect on the rats with blood stasis syndrome,but Rhubarb-Polygonum cuspidatum combination (1:1) did not show the obvious synergistic effect.Polygonum cuspidatum can significantly reduce triglycerides,while Rhubarb alone didn't show the role of significant reduction of blood lipids,and in reducing blood lipids,rhubarb and in this regard,Polygonum cuspidatum compatibility didn't show the synergistic effect.
