Objective To explore the effects of different methods on the culture results of Mycobacterium marinum, including direct inoculation, grinding followed by digestion, and digestion followed by grinding. Methods From June 2022 to March 2024,122 patients with suspected Mycobacterium marinum infection were collected from the Department of Dermatology in this hospital, including 44 cases with superficial skin type and 78 cases with subcutaneous tissue type.Direct inoculation, grinding followed by and digestion followed by grinding were used respectively to analyze the effects of these three methods on the positive detection rate and contamination rate of Mycobacterium marinum culture. Results The culture positive detection rate of Myco-bacterium marinum in superficial skin type and in subcutaneous tissue type were 47.7%(21/44)and 67.9%(53/78),respectively.The use of grinding combined with digestion method for processing skin or subcutane-ous tissue specimens results in a higher positive detection rate and a lower contamination rate compared with the direct inoculation method, with statistically significant differences(P<0.05).For superficial skin type specimens, the grinding followed by digestion method had a higher positive detection and a lower contamina-tion rate than the digestion followed by grinding method, with statistically significant differences(P<0.05).For subcutaneous tissue samples, the grinding followed by digestion method had a higher positive detection rate compared to the digestion followed by grinding method, but the difference was not statistically significant(P>0.05).There were no significant differences in the primary culture time of Mycobacterium marinum a-mong the three pretreatment methods(P>0.05).Conclusion The pretreatment of skin tissue samples with grinding followed by digestion, and the pretreatment of subcutaneous tissue samples with grinding followed by digestion or digestion followed by grinding can significantly improve the positive rate of Mycobacterium mari-num culture and reduce the contamination rate of miscellaneous bacteria.

A series of 5H-benzo[b] carbazole derivatives were synthesized to find new TopⅡ inhibitors. The substitution pattern at C-9 was discussed. All 16 compounds were tested for their antiproliferative activities against 4 tumor cell lines (SMMC-7721,S1,HCT116,and HeLa) by sulforhodamine B assay.Four compounds were found to inhibit the proliferation of all tested cell lines. And compound 10e displayed the best antiproliferative activity against all 4 tested cell lines with IC50 values of 5.06,4.50,5.29,and 6.32 μmol/L,respectively.

ObjectiveToinvestigatetheeffectsofcaveolin1(Cav1)onexpressionsofproinflammatory cytokine interleukin (IL)-1βand IL-6 in the ischemic cortex after permanent middle cerebral artery occlusion in mice. Methods The Cav-1 knockout mice (n=40) and wild-type mice (n=40) were randomly divided into ischemia groups and sham operation groups (n=20 in each group). They w ere also redivided into ischemia or sham operation at 3, 7, 10 and 14 d time points ( n=5 in each time point). A permanent middle cerebral artery occlusion model w as induced by the suture method. Immunohistochemical method w as used to detect the expressions of IL-1βand IL-6 in the ischemic cortex. Results The expression levels of IL-1βand IL-6 in the ischemic cortex at each time point in the ischemia group in Cav1 knockout mice w ere significantly higher than those in the ischemia group in the w ild-type mice ( al P< 0.05 ). Conclusions The upregulations of proinflammatory cytokines IL-1βand IL-6 in the ischemic cortex in Cav1 knockout mice suggests that Cav1 plays an important role in aleviating inflammation after cerebral ischemia.

As a cavola marker protein, caveolin-1 participates in many pathophysiological processes through its scaffolding domain oligomering many celular signal transduction molecules, and also regulates inflammation after cerebral ischemia through different pathways. This article reviews advances in caveolin-1 and inflammation after ischemic stroke in recent years, mainly focusing on its mechanism in regulating inflammation.

Objecfive To investigate possibihty of monitoring myocardial isehemia by eontinuous EASI 12-lead ST-segment trend analysis during orthopaedic surgery and find its rules. Methods Three hundred and eleven consecutive patients who underwent orthopaedic surgery were monitored bv continuous EASI 12-lead ST-segment analysis during surgery. Results Among 311 patients 81 patients(26.0%) had myocardial ischemic events. The ischemie incidence of general anesthetics WaS signifieantly higher than local anesthetics(the spinal or epidural anesthetics and nerve-block anesthetics)(38.1%vs 23.0%.P= 0.01 5).During ischemie events the patients whose heart rate exceeded 90 beat per minute was 71.6% (58/81).Among the 177 isehemie events, the duration ofless than 5 minutes was 79.7%(141/177).ST-seg- ment deviation ofless than 0.15 mV was 71.2%(126/177).Conclusions During orthopaedic surgery EASt 12-lead electrocardiogram is the optimal approach for monitoring myocardial isehemia. The ischemic incidence of general anesthetics is significantly higher than local anesthetics. The myocardial ischemia events correlates with fast heart rate. The intraoperative myocardial ischemia events are more often transient. In most of these ischemic events ST-segment deviation is not very serious.
