Coronavirus-related diseases pose a significant challenge to the global health system. Given the diversity of coronaviruses and the unpredictable nature of disease outbreaks, the traditional "one bug, one drug" paradigm struggles to address the growing number of emerging crises. Therefore, there is an urgent need for therapeutic agents with broad-spectrum anti-coronavirus activity. Here, we provide evidence that ATV006, an anti-SARS-CoV-2 nucleoside analog targeting RNA-dependent RNA polymerase (RdRp), has broad antiviral activity against human and animal coronaviruses. Using mouse hepatitis virus (MHV) and human coronavirus NL63 (HCoV-NL63) as a model, we show that ATV006 has potent prophylactic and therapeutic activity against murine coronavirus infection in vivo. Remarkably, ATV006 successfully inhibits viral replication in mice even when administered 96 h after infection. Due to its oral bioavailability and potency against multiple coronaviruses, ATV006 has the potential to become a useful antiviral agent against SARS-CoV-2 and other circulating and emerging coronaviruses in humans and animals.
2.Application of virtual simulation experiment technology in Animal and Plant Quarantine courses

Objective:To establish innovative teaching reform methods suitable for food-related majors in medical colleges by leveraging virtual simulation experimental technology.Methods:Based on the course construction and application of the national virtual simulation experiment teaching course sharing platform,a comparative study was conducted between traditional teaching and virtual simulation experiment technology teaching for grade 2021 and grade 2022 students majoring in food quality and safety.The differences between the two teaching methods were analyzed by comparing learning performance.Results:A total of 58 food virtual simulation courses covering 107 courses were constructed in the national virtual simulation experiment teaching course sharing platform,which had been widely recognized in university education.Compared with the traditional teaching method,the experimental teaching using virtual simulation technology significantly improved the students'scores in homework assignments,midterm exams,and final exams.Conclusion:The application of virtual simulation experiment technology in the Animal and Plant Quarantine course enhances students'learning performance and strengthens their practical application abilities,which has become an internal driving force to strengthen the construction of professional level.

Objective:To explore the teaching reform of the Food Analysis experiment course under the concept of employment-oriented education,and to improve students'practical ability and employment competitiveness.Methods:Taking students majoring in Food Quality and Safety at a certain university as the research objects,the control group adopted traditional teaching,while the experimental group carried out reforms by optimizing the curriculum system,innovating the teaching mode,and establishing a diversified evaluation system.The total scores,teaching effectiveness,satisfaction with the teaching mode,and the evaluation of graduates by employers between the two groups were compared.Results:The average total score of the experimental group was significantly higher than that of the control group(P<0.05).The teaching effect evaluations of the experimental group in improving experimental operation skills and enhancing employment position cognition were all higher than those of the control group(P<0.05).The satisfaction of the experimental group with the teaching mode was better than that of the control group(P<0.05).In the evaluation results of employers on graduates,the excellent rate of the experimental group was significantly higher than that of the control group(P<0.01).Conclusion:The reform measures effectively improve the teaching quality and students'comprehensive literacy,and deliver high-quality talents with both practical ability and innovative thinking to the food industry.
5.Effects of rehabilitation nursing based on knowledge-action framework on compliance behavior and coping styles in patients with chronic respiratory failure

Objective:To investigate the effects of rehabilitation nursing based on knowledge-action framework on compliance behavior and coping styles in patients with chronic respiratory failure (CRF), and provide scientific basis and practical guidance for clinical nursing of patients with CRF.Methods:A historical control study was conducted. A retrospective analysis was performed on the clinical data of patients with CRF in Yiwu Central Hospital between January and April 2024. According to admission time, the patients were divided into the control group (admitted from January to February 2024, received routine rehabilitation nursing) and the study group (admitted from March to April 2024, received rehabilitation nursing based on knowledge-action framework). All patients were intervened for 1 month. The differences in self-care ability, compliance behavior, coping styles, quality of life between the two groups were compared using Exercise of Self-Care Agency Scale (ESCA), Compliance Behavior Questionnaire, Trait Coping Style Questionnaire (TCSQ), St. George′s Respiratory Questionnaire (SGRQ).Results:A total of 60 patients with CRF were included, 30 patients in the study group, with 16 males and 14 females, aged (68.72 ± 4.67) years old; 30 patients in the control group, with 17 males and 13 females, aged (69.21 ± 4.18) years old. Before intervention, there were no significant differences in ESCA score, TCSQ score and SGRQ score between the 2 groups (all P>0.05). After 1 month of intervention, ESCA score in the study group was (134.79 ± 5.62) points, higher than (125.34 ± 5.51) points in the control group, the difference was statistically significant ( t=6.58, P<0.05). The compliance behavior (adherence to medical regimens, reasonable diet, work and rest, regular checking) in the study group were 30, 28, 27, 29 cases, higher than 25, 20, 20, 22 cases in the control group, the differences were statistically significant ( χ2 values were 4.81-6.68, all P<0.05). The positive coping score of TCSQ in the study group was (39.67 ± 4.54) points, higher than (32.19 ± 4.38) points in the control group, and score of negative coping score was (13.55 ± 2.28) points, lower than (18.31 ± 2.43) points in the control group, the differences were statistically significant ( t=6.49, 7.82, both P<0.05). The SGRQ score in the study group was (30.23 ± 3.29) points, lower than (35.87 ± 3.66) points in the control group, the difference was statistically significant ( t=6.28, P<0.05). Conclusions:For patients with CRF, rehabilitation nursing based on knowledge-action framework can effectively improve self-care ability and compliance behavior, form more positive coping styles, improve quality of life.

10.Development and validation of predictive models for esophageal squamous cell carcinoma and its precancerous lesions using terminal motif analysis in circulating cell-free DNA

Objectives:To develop and validate predictive models for esophageal squamous cell carcinoma (ESCC) using circulating cell-free DNA (cfDNA) terminal motif analysis. The goal was to improve the non-invasive detection of early-stage ESCC and its precancerous lesions.Methods:Between August 2021 and November 2022, we prospectively collected plasma samples from 448 individuals at the Department of Endoscopy, Cancer Hospital, Chinese Academy of Medical Sciences for cfDNA extraction, library construction, and sequencing. We analyzed 201 cases of ESCC, 46 high-grade intraepithelial neoplasia (HGIN), 46 low-grade intraepithelial neoplasia (LGIN), 176 benign esophageal lesions, and 29 healthy controls. Participants, including ESCC patients and control subjects, were randomly assigned to a training set ( n=284) and a validation set ( n=122). The training cohort underwent z-score normalization of cfDNA terminal motif matrices and a selection of distinctive features differentiated ESCC cases from controls. The random forest classifier, Motif-1 (M1), was then developed through principal component analysis, ten-fold cross-validation, and recursive feature elimination. M1's efficacy was then validated in the validation and precancerous lesion sets. Subsequently, individuals with precancerous lesions were included in the dataset and participants were randomly allocated to newly formed training ( n=243), validation ( n=105), and test ( n=150) cohorts. Using the same procedure as M1, we trained the Motif-2 (M2) random forest model with the training cohort. The M2 model's accuracy was then confirmed in the validation cohort to establish the optimal threshold and further tested by performing validation in the test cohort. Results:We developed two cfDNA terminal motif-based predictive models for ESCC and associated precancerous conditions. The first model, M1, achieved a sensitivity of 90.0%, a specificity of 77.4%, and an area under the curve (AUC) of 0.884 in the validation cohort. For LGIN, HGIN, and T1aN0 stage ESCC, M1's sensitivities were 76.1%, 80.4%, and 91.2% respectively. Notably, the sensitivity for jointly predicting HGIN and T1aN0 ESCC reached 85.0%. Both the predictive accuracy and sensitivity increased in line with the cancer's progression ( P<0.001). The second model, M2, exhibited a sensitivity of 87.5%, a specificity of 77.4%, and an AUC of 0.857 in the test cohort. M2's sensitivities for detecting precancerous lesions and ESCC were 80.0% and 89.7%, respectively, and it showed a combined sensitivity of 89.4% for HGIN and T1aN0 stage ESCC. Conclusions:Two predictive models based on cfDNA terminal motif analysis for ESCC and its precancerous lesions are developed. They both show high sensitivity and specificity in identifying ESCC and its precancerous stages, indicating its potential for early ESCC detection.
