1.The predictive value of radiomics for postoperative prognosis of pancreatic ductal adenocarcinoma patients

Objective To explore the predictive value of radiomics for postoperative pancreatic fistula, infection, and long-term survival in patients with pancreatic ductal adenocarcinoma (PDAC). Methods 206 patients who received radical resection of pancreatic cancer in Zhongshan Hospital, Fudan University from January 2014 to December 2020 and were pathologically confirmed as PDAC after surgery were retrospectively selected, all of whom had complete surgical data and long-term follow-up data. Pyradiomics was used to analyze the enhanced CT images of all patients and extract radiomics features. LASSO dimensionality reduction combined with logistic regression analysis was used to construct a predictive model for pancreatic fistula and abdominal infection after PDAC surgery, and evaluating the model’s effectiveness using ROC curves. A long-term survival prediction model for PDAC patients was constructed using LASSO dimensionality reduction combined with Cox regression analysis, and patient risk scores were calculated. The patients were divided into high-risk and low-risk groups based on the median, and the survival curves were compared to evaluate the effectiveness of the model. The imaging omics features with the highest weight were divided into high expression group and low expression group according to the median, and the prognostic differences and clinical features were compared. Radiomics and clinical features were combined to analyze the influencing factors of long-term prognosis and construct a clinical imaging comprehensive model. Results A total of 1 595 radiomics features were extracted. A predictive model for pancreatic fistula and infection after PDAC surgery was constructed, with AUC values of 0.81 and 0.79, respectively. The PDAC long-term survival prediction model was successfully constructed, and the prognosis of the high-risk group was worse than that of the low-risk group (P<0.001). The weight of the radiomics feature “log-sigma-5-mm-3D_glszm_ZonePercentage” was 59.557. The CA19-9 level in the high expression group is higher than that in the low expression group (P=0.017), and there is a statistically significant difference in survival curves between the two groups (P=0.021). The comprehensive clinical imaging model suggested that age, AJCC stage, lymph infiltration, CA19-9 level and imaging characteristics were risk factors for long-term prognosis of PDAC patients (HR=1.028, 4.084, 2.566, 1.232 and 2.536). Conclusions The predictive model based on radiomics has good predictive performance for pancreatic fistula, infection, and long-term prognosis after PDAC surgery. Patients with high expression of the radiomics feature “log-sigma-5-mm-3D_glszm_ZonePercentage” face poorer prognosis.
2.Research progress on the inhibition of photoreceptor cell apoptosis by the pentose phosphate pathway and its molecular mechanism

Photoreceptor cells are a special type of neuroepithelial cells, which play an important role in the generation and transmission of visual signals.They mainly meet the physiological needs by ingesting a large amount of glucose for glucose metabolism.Photoreceptor cell apoptosis is a common cause of blindness in patients with retinal diseases, which is accompanied by changes in oxidative stress and anabolism.Recent studies have found that pentose phosphate pathway, an important branch of glucose metabolism, plays an essential role in the development of the above-mentioned pathology.Its product, nicotinamide adenine dinucleotide phosphate (NADPH), as a crucial hydrogen donor in photoreceptor cells, participates in substance synthesis and metabolism, resists oxidative stress, and then inhibits apoptosis of photoreceptor cells.This review summarized the mechanism of pentose phosphate pathway in inhibiting photoreceptor cell apoptosis from the perspective of view of metabolic activity and molecular pathway to provide reference for clinical scientific research.
3.Simultaneous Determination of 17 Amino Acids in Sarcocarp of Northeast Empetrum nigrum by HPLC-ELSD

OBJECTIVE:To determine the content of 17 amino acid in sarcocarp of northeast Empetrum nigrum simultaneous-ly. METHODS:HPLC-ELSD method was adopted. The determination was performed on Waters Symmetry C18 column with mobile phase consisted of 5.0 mmol/L heptafluorobutyric acid solution(containing 0.7% trifluoroacetic acid)-acetonitrile(gradient elution) at the flow rate of 2.0 L/min,the column temperature was 37 ℃,the drift tube temperature was set at 60 ℃. RESULTS:The line-ar ranges of glycine,serine,aminosuccinic acid,aminopropionic acid,threonine,glutamic acid,cystinol,diaminocaproic acid, histidine,arginine,proline,valine,methionine,oxyphenylaminopropionic acid,isoleucine,aminocaproic acid,phenylalanine were 0.0059-0.1877,0.0082-0.2628,0.0104-0.3328,0.0070-0.2227,0.0093-0.2978,0.0115-0.3678,0.0094-0.3004,0.0114-0.3655,0.0121-0.3880,0.0136-0.4355,0.0090-0.2878,0.0092-0.2930,0.0117-0.3730,0.0142-0.4530,0.0103-0.3280, 0.0103-0.3280 and 0.0129-0.4130 mg/mL(all r>0.9990). RSDs of precision,stability and repeatability tests were not all less than 2.0%. Recoveries were 98.0%-101.2%(RSD<2.0%,n=6). CONCLUSIONS:This method is simple,precision,stable and repeatable,and can be used for simultaneous determination of 17 amino acid in sarcocarp of northeast E. nigrum.
4.In Silico Cloning and Bioinformatics Analysis of AP2/ERF Family Gene from Brassica Oleracea

Objective:To obtain AP2/ERF genes from Brassica oleracea by using in silico cloning. Methods: AP2/ERF genes were cloned by retnieving the EST database and using the bioinformatics software with the Arabidopsis thaliana AP2/ERF as a querying probe. Results:Two AP2/ERF family transcriptional regulators (BoAP2/ERF1 and BoAP2/ERF2) were isolated from Brassica olera-cea by the in silico cloning method. Some characters of AP2/ERF protein were analyzed and predicted by the tools of bioinformatics in the following aspects including the composition of amino acid sequence, hydrophilicity and hydrophobicity, subcellular localization, secondary and tertiary structure of protein and function. Conclusion:Bioinformatical analysis shows BoAP2/ERF1 and BoAP2/ERF2 gene encode 40. 85kDa and 39. 44kDa protein with 371 and 352 amino acids. The domain is predicted to locate on nucleus. Sequence analysis indicates the protein may be involved in signaling transducer and stressing response roles in plantbiotic stresses.
5.Patent applications in hospitals of Guangdong Province since 2000

The relation between patent applications and published papers in hospitals of Guangdong Province since 2000 on China Intellectual Property Network was analyzed , which showed that the quality and quantity of patents were lower in hospitals of Guangdong Province than in those of Shanghai and Beijing , the quality of patents was re-lated with the medical service level in hospitals, the patent applications were centralized in large class A hospitals of Guangzhou , and inventive patents were those of medical preparations .Suggestions were thus put forward for hos-pitals of Guangdong Province to improve their awareness of patent application, the quality of patents, transformation of patents, cooperation with patent service organizations, research of patent application, and balance of patent dis-tribution in different areas.
6.Analysis on gene sequence of HIV isolated from men who have sex with men in Beijing

Objective To analyze genetic characteristics of HIV isolated from men who have sex with men(MSM) in Beijing and predict the epidemic trend in this population.Methods All of the HIV gene sequences in our laboratory obtained from MSM in Beijing were used,which were aligned with all of the HIV gene sequences from MSM and other populations in China downloaded from Los Alamos HIV Database.Phylogenetic trees were constructed by using software PhyML 3.0,based on which the relationships of prevalent HIV strains between Beijing MSM and other populations in China were further explored.The evolution rate,the time of most recent common ancestor (tMRCA),the epidemic parameters,the reproductive number (R0) were calculated by using software BEAST to predict HIV evolution and epidemic characteristics.Results Multiple HIV subtypes,including subtype B,CRF01_AE and CRF07_BC,were found to be prevalent among MSM in Beijing.In ML tree constructed based on strains from the whole country,three clusters including B-1,CRF01_AE-1,and CRF01_AE-2 were found among the MSM in Beijing (accounting for 40%).At least three independent introduction of B 1 cluster strains into Beijing MSM were found,which were at March 1991 (July 1984-February 1997),January 1994 (January 1989-January 1998),April 1991 (August 1984-January 1996).For CRF01_AE strains,two clusters including CRF01_AE-1 and CRF01_AE-2 were introduced into the population at December 2000 (March 1998-January 2003) and December 2001 (January 2000-July 2003) respectively.The population epidemiology of HIV in Beijing MSM was reconstructed based on sequences.The CRF01_AE-1 cluster spread more quickly than the other two clusters,and the evolution rate was higher.Conclusion Multiple HIV subtypes were found prevalent among MSM in Beijing.Although subtype B strain was introduced into Beijing MSM earlier than CRF01_AE strain,CRF01_AE strain increased more quickly than subtype B strain.More research and control of the CRF01_AE prevalence will be helpful for prevention and control of HIV epidemic in MSM in Beijing.
