1.Research progress on recombinant human DNA damage induced transcription factor 4 in malignant tumors

DNA damage-inducible transcript 4(DDIT4)is a gene activated under stress conditions,par-ticipating in cell growth,apoptosis,metabolism,and oxidative stress.In recent years,significant progress has been made in the research on the mechanism of DDIT4 in tumorigenesis and development.It has been con-firmed that it is involved in regulating processes such as apoptosis,proliferation,migration and invasion of tumor cells,and is also involved in multiple key biological processes such as tumor microenvironment remode-ling,autophagy regulation and drug resistance formation.Targeting DDIT4 may potentially become an effec-tive therapeutic option for inhibiting the tumorigenesis and tumour progression.Targeting DDIT4 is expected to be an effective therapeutic option to inhibit the tumorigenesis and progression.This article reviews the research progress on the role and regulatory mechanisms of DDIT4 in cancer.
2.A cross-sectional study of the effects of well-being of dementia patients and their caregivers

Objective:To study the factors influencing the happiness of dementia patients and their caregivers,provide guidance for improving their well-being.Methods:A total of 94 pairs of patients and their caregivers who were admitted to the neurology department of Peking Union Medical College Hospital from April 2015 to April 2016 were selected, the demographics of each patient and their caregivers were recorded. The Mini-Mental State Examination(MMSE) of patients with dementia, Role of Overload Scale(ROS) of caregivers, Dyadic Relationship Strain(DRS), Quality of Life for Dementia(QOL-D), Self-Evaluation Scale-Depression(CES-D) were recorded. Layered linear model was used to make regression analysis between the influencing factors and the scores of QOL-D and CES-D.Results:The results of the multi-layer linear model of uncontrolled variables in the fixed effect model: the results of QOL-D suggested that the score of patients with dementia was β 1j= 31.01±0.77, and the score of caregivers was β 2j= 35.15±0.88; the results of CES-D suggested that the scores of dementia patients and caregivers were β 1j = 14.55 ± 1.03 and β 2j = 13.11 ± 1.44, respectively. The random effects model suggested that there were statistical differences in the heterogeneity of the QOL-D score and the CES-D score variance component for dementia patients and caregivers (χ 2 values were 98.94-168.06, P<0.01). It indicated that the data was heterogeneous, adjusting the level 2 model, and the final results in the adjusted regression analysis suggested: caregiver relationship pressure (DRS), dementia patient self-awareness assessment (MMSE), caregiver care-related stress (ROS), dementia patient relationship stress (DRS) significantly affected the quality of life score (QOL-D) in both well-being ( β values were -3.22-0.43, P<0.05). Dementia patient relationship stress (DRS), caregiver-related stress (ROS), and caregiver relationship stress (DRS) significantly affected depressive symptoms in both well-being ( β values were 5.34, 3.26, 1.62, P<0.05). Conclusions:A comprehensive assessment of dementia patients and caregivers is needed. The combined family relationship is tense and the pressure associated with caregivers needs to be psychologically counseled.
3.Biomimetic grandient scaffolds for articular cartilage tissue engineering

Due to good mechanical properties and biocompatibility, tissue engineering scaffolds have become the vital method for repairing and regenerating articular cartilage defects. With the continuous development of tissue engineering technology, many scaffolds preparation and formation methods have been developed and tested in the past decade, however, the preparation of ideal regenerative scaffolds remain controversial. As load-bearing tissue inside the body joints, the matrix structure and cell composition of articular cartilage are hierarchical, and there are several smooth natural gradients from the cartilage surface to the subchondral bone layer, including cell phenotype and number, specific growth factors, matrix composition, fiber arrangement, mechanical properties, nutrient and oxygen consumption. Therefore, in the design of regenerative scaffolds, it is necessary to achieve these gradients to regenerate articular cartilage in situ. In recent studies, many new biomimetic gradient scaffolds have been used to simulate the natural gradient of articular cartilage. These scaffolds show different mechanical, physicochemical or biological gradients in the structure, and have achieved good repair effects. The related articles on tissue engineering for the treatment of articular cartilage defects were retrieved by searching databases with key wordsarticular cartilage injury, cartilage repair and gradient scaffolds. In this work,the structural, biochemical, biomechanical and nutrient metabolism gradients of natural articular cartilage were studied and summarized firstly. Then, the latest design and construction of articular cartilage gradient scaffolds were classified. Besides that, the material composition (such as hydrogels, nanomaterials, etc.) and the preparation process (such as electrospinning, 3D printing, etc.) of grandient scaffolds were further enhanced. Finally, the prospect and challenge of biomimetic gradient scaffolds in cartilage engineering are discussed, which provides a theoretical basis for the successful application of gradient scaffolds in clinical transformation.
4.Chemoattractant-mediated in situ tissue engineering for articular cartilage regeneration

The treatment of articular cartilage (AC) injury caused by various reasons is still a major clinical problem. The emergence of cartilage tissue engineering brings new hope for the treatment of AC injury. In general, AC tissue engineering can be divided into two categories, including cell-based tissue engineering and cell-free tissue engineering. Although cell-based tissue engineering can repair cartilage damage to a certain extent, existing therapeutic strategies still suffer from limited cell sources, high costs, risk of disease transmission, and complex procedures. However, the cell-free tissue engineering avoids these shortcomings and brings hope for in-situ AC regeneration. Non-cellular tissue engineering is mainly used to recruit endogenous stem cells/progenitor cells (SCPCs) to reach the site of cartilage injury, and provide a suitable regenerative microenvironment to promote cell proliferation and chondrogenic differentiation, then the maturation of new cartilage tissue was promoted. Therefore, it is also called as cell-homing in situ tissue engineering. Successful recruitment of endogenous SCPCs is the first step in in-situ cartilage tissue engineering. This review aims to introduce chemokine response of cartilage injury, systematically summarize traditional chemoattractant (chemokines and growth factors etc.) and emerging chemoattractant (functional peptides, exosomes and nucleic acid adapters etc.), evaluate the combination mode between chemoattractant and delivery devices, discuss the prospects and challenges of chemoattractant-mediated in situ tissue engineering and provide theoretical basis for the design of endogenous SCPCs homing-based in situ tissue engineering.
5.Effects of mindfulness-based stress reduction combined with progressive muscle relaxation training on cancer-related fatigue in patients with breast cancer chemotherapy

Objective To explore the effect of mindfulness-based stress reduction (MBSR) combined with progressive muscle relaxation (PMR) training on cancer-related fatigue in patients with breast cancer chemotherapy.Methods Totally 98 patients with breast cancer chemotherapy were randomly divided into the control group and the intervention group.Patients in the intervention group received MBSR therapy combined with PMR training,while the control group received routine nursing care.Cancer-related fatigue,and the level of anxiety and depression in two groups before and after intervention were compared.Results After intervention,the cases of patients in the intervention group without fatigue,and with mild,moderate,severe fatigue were respectively 27,10,10 and 3;in the control group,the cases of patients without fatigue,and with mild,moderate,severe fatigue were respectively 7,12,23 and 6 (Z=-3.994,P<0.05).The level of anxiety and depression in the intervention group was significantly improved compared with the control group (t =29.399,17.960;P< 0.05).Conclusions MBSR therapy combined with PMR training can effectively improve the cancer-related fatigue in patients with breast cancer chemotherapy,and improve anxiety and depression status of patients.
6.Molecular Identification of Metacordyceps Liangshanensis, Its Adulterants and Its Relative Species Based on DNA Barcode

This study was aimed to identify and distinguish Metacordyceps liangshanensis recorded by the Sichuan Province Traditional Chinese Medicine Standard from its adulterants and its relative species by combining ITS and COI barcode sequences in order to study the feasibility of this new method. After extracting DNA of 28 species of Cordyceps samples, DNA were amplified and sequenced. And then, ITS and COI sequences were received. Codon-Code Aligner V3.7.1 and Mega 5.0 were used to analyze the variable site and construct the N-J tree. The results showed that the minimum ITS inter-specific K-2P distance was relatively higher than the maximum intra-specific K-2P distance. The inter-specific sequence divergence between M. liangshanensis and its adulterants exhibited high while intra-specific sequence divergence exhibited low. And COI one was the same case. N-J tree of both ITS and COI indicated that same genus belonged together and each species belonged to relatively independent branch. It was concluded that based on the ITS and COI gene, the technology of DNA barcode can be an excellent identification of M. liangshanensis, its adulterants and its relative species. It provided technical support for the further research on species molecular identification and phylogenetics of Cordyceps .
7.Influence of psychological care and health education on patients with digestive tract cancer receiving chemotherapy

Objective To study the impact of psychological care and health education on patients with digestive tract cancer and abnormal emotion receiving chemotherapy. Methods A retrospective analysis was carried outin 245 patients with malignant digestive tract cancer from July 2005 to July 2009 treated in our hospital, 147 cases of these patients received psychological care and health education besides conventional therapy were set as the intervention group, 98 cases given only basic nursing care were set as the control group. The clinical efficacy was analyzed in the two groups before and after chemotherapy using anxiety and depression indexes as indicators. Results After chemotherapy, SAS and SDS scores were significantly lower, while the survival time was significantly higher in the intervention group than those of the control group. Conclusions Psychological care and health education can reduce abnormal emotion in patients with malignant digestive tract cancer receiving chemotherapy and prolonged their survival time.
