1.Research Progress in the Mechanism of Shaoyao Decoction in Regulating Cytokines for the Treatment of Ulcerative Colitis

Ulcerative colitis(UC)is a recurrent,chronic,nonspecific inflammatory bowel disease.Shaoyao Decoction has shown good efficacy in the treatment of UC,which mainly regulates the balance of cytokines to achieve the purpose of treating UC.This article reviewed the research progress in the mechanism of Shaoyao Decoction in the treatment of UC from two aspects of pro-inflammatory cytokines and anti-inflammatory cytokines.Research has shown that Shaoyao Decoction can regulate the body's immune response,alleviate intestinal inflammation,protect the intestinal mucosal barrier,restore normal structure and function of the colon mucosa through pro-inflammatory cytokines such as tumor necrosis factor-α,interferon-γ,interleukin(IL)-1β,IL-6,IL-8,IL-17,and anti-inflammatory cytokines such as transforming growth factor-β,IL-4,IL-10,IL-13,which can provide reference for UC related mechanism research and clinical treatment.
2.Current situation and challenges of tumor biotherapy based on alternative splicing

Alternative splicing,a post-transcriptional regulatory mechanism of gene expression,contributes to the diversity of the transcriptome and proteome in eukaryotes.However,aberrant alternative splicing serves as a significant driver of tumor progression,where abnormal splicing in tumor cells,immune cells,and other cell types within the tumor microenvironment collectively results in malignant behaviors of cancer cells,immune evasion,and the immunosuppressive milieu that supports tumor advancement.Targeting tumor-associated spliceosomes,splicing regulatory factors,splicing isoforms and variants,as well as tumor neoantigens generated by aberrant alternative splicing,has emerged as a novel strategy in cancer biotherapy.Some alternative splicing-based antitumor biotherapy programs have progressed to phaseⅠclinical trials.Alternative splicing-based tumor therapy still faces scientific and technological challenges such as safety,optimization of long-read sequencing and bioinformatics algorithm,and nucleic acid drug delivery.Addressing these challenges will provide new tumor therapy strategies and open up new frontiers for precisely screening tumor-related targets and highly immunogenic neoantigens,overcoming drug resistance in traditional therapies,and enhancing the efficacy of immune checkpoint blockade,CAR-T cell therapy,and other treatments.

4.Research of 10-23 DNAZyme inhibit the expression of eIF4E genes.

OBJECTIVE:
To explore the possibility of 10-23DNAzyme becoming a new gene therapy for laryngeal carcinoma treatment at the cell level.
METHOD:
Thiosthorothioate 10-23DNAzyme specific to eIF4E gene mRNA 1059 was designed and synthesized, and its inhibition effects on the expression of eIF4E gene in Hep-2 cells were observed.
RESULT:
The expression of eIF4E gene was remarkable depressed after Hep-2 cells was transfected by DNAzyme. The level of inhibiting eIF4E in hep-2 cells transfected by DNAzyme was lower than that by only lipofectamine 2000 transfected and Hep-2.
CONCLUSION
The expression of eIF4E gene in Hep-2 cells 10-23DNAzyme can be highly blocked. It is a specific and effective gene therapeutic means.

5.Endoscopic thyroidectomy via suprasternal approach

OBJECTIVE To summarize the clinical experience of endoscopic thyroidectomy via suprasternal approach. METHODS Endoscopic thyroidectomy via suprasternal approach was performed in 35 patients with ultrasonic scalpel. RESULTS Operations were successfully performed in 35 patients. The mean operation times were 130 (105~190) minutes in 24 cases with subtotal lobectomy and 4 case with total lobectomy, 60 (50~70) minutes in 2 cases with isthmectomy, 228 (185~270) minutes in 2 case with bilateral subtotal lobectomy, 163 (140~215) minutes in 3 case with subtotal lobectomy and the contralateral ademona resection .The bleeding during operation was 5 to 40ml and the average hospital stay time was 4 (3~5) days. CONCLUSION Endoscopic thyroidectomy via suprasternal approach is a safe way with good cosmetic value.
6.Effects of Shenyanling on chronic glomerulonephritis in rabbits

Objective :To sutdy the effects of Shenyanling on chronic glomerulonephritis in rabbits. Meth-ods:Rat glomerular base membrane (GBM) and Freund's adjuvant were injected into rabbits subcuta-neously or intracutaneously to build the Steblay nephritis model. Shenyanglin was administered. Kidneyfunction was evaluated by measuring urine protein,urea nitrogen ,creatinine ,albumin and globulin in serumas wellas the histopathological changes of kidney were observed. Results:Compared with model group,Shenyanling could dose-dependently reduce the level of the urine protein,urea nitrogen and creatinine inserum and the damage of kidney;while,the serum albumin could maintain the normal level (P<0.05).Conclusion:Shenyanling has protective effects on chronic glomerulonephritis in rabbits.
