Objective To analyze the risk factors of preoperative lymph node staging (N-stage) deficiency in gastric cancer and establish a preoperative assessment model to assist in predicting preoperative N-stage. Methods A retrospective method was used to analyze the clinicopathological data of 268 patients with gastric cancer. The patients routinely underwent preoperative thin-section enhanced CT to assess preoperative N-stage. Results The risk factors for preoperative N-stage deficiency were analyzed in combination with postoperative pathological findings. Multifactorial logistic regression analysis was performed to determine influencing factors, and Kaplan-Meier analysis was used to plot the survival curves of preoperative N-stage accurate group and deficiency group. The nomogram plot and ROC curves of the prediction model were drawn using the R package. AUC, 95%CI, sensitivity, and specificity were calculated. Results Age, BMI, poor differentiation, and Lauren's classification as diffuse were independent risk factors for preoperative N-stage deficiency in gastric cancer (P < 0.05). Prognostic survival was significantly worse in the preoperative N stage-inadequate group than that in the accurate group (P=0.041). The AUC area was 0.935, with a sensitivity of 85.9% and specificity of 96.9%. Conclusion Young age, high BMI, poor differentiation, and Lauren's classification as diffuse are independent risk factors for preoperative N-stage deficiency. The established preoperative assessment model based on age, BMI, differentiation degree, and Lauren's classification in this study has relatively high credibility.

Objective To assess the ability of simultaneous amplification and testing ( SAT ) in bacteria detection , bronchoalveolar lavage fluid from children was collected and detected by SAT , combined with in vitro culture experiment , providing theoretical support for the application of SAT in the diagnosis and treatment of mycoplasma pneumonia ( MP ) .Methods A total of 572 bronchoalveolar lavage fluid from children with community acquired pneumonia during October 2015 and December 2017 in Tianjin Children′s Hospital were collected and detected by Mycoplasma pneumonia ( MP ) nucleic acid quantitative assay and SAT technology.Among them, 161 bronchoalveolar lavage fluid were also detected using in vitro culture and the positive ones for MP were analyzed by nucleic acid quantification and SAT after exposing to high concentration of antibiotic .Results The positive rates of MP by nucleic acid quantitative assay and SAT technology in 572 samples were 74.7％ (427/572) and 71.9％ (411/572), respectively.These two detection methods have high consistency (χ2 =1.142,P=0.285).According to the test results of SAT , the positive rates of male and female were 72.7％(224/308) and 70.8％(187/264), respectively.There was no significant difference of positive rate between different sex (χ2 =0.252, P=0.616).The positive rate of MP in 4-14 years old children (78.1％, 317/416) was higher than that in infants (≤3 years) (56.6％, 94/166) (χ2 =26.811, P=0.000).After adding azithromycin (5 MIC) to MP positive medium for 5 days, the result of nucleic acid quantification was positive but SAT was negative .Conclusions SAT technology is a rapid, sensitive and specific method for detection of MP .In addition, SAT technology could identify the "dead" and"live" bacteria and could evaluate the effect of clinical treatment effectively .

In order to improve the precision and robustness in determination of soluble solids content ( SSC) of ‘Fuji ’ apple by NIR spectroscopy and eliminate the effect of origin variability on the accuracy of NIR calibration models for the SSC, sample set partitioning based on joint x-y distances ( SPXY) was used to select representative subset from the apple samples of 4 different origins. As a comparison, partial least square ( PLS) was used to establish local origin and hybrid origin models for the prediction of SSC in apple. Competitive adaptive reweighted sampling ( CARS ) and successive projections algorithm ( SPA ) were implemented to select effective variables of the NIR spectroscopy of SSC of apple. The results indicated that the PLS model established based on the 4 origin apple samples performed better than local origin and other hybrid origin models. The model could be effectively simplified using 16 characteristic variables selected by CARS-SPA method from full-spectrum which had 3112 wavelengths. The correlation coefficient (Rp) and root mean square error of prediction (RMSEP) were 0. 978 and 0. 441 oBrix, respectively for SSC. It was found that the model developed by more samples of different origins combined with effective wavelengths showed good prediction ability for apple sample of unknown origin, which indicated that it could significantly reduce the origin effect on the robustness of NIR models for SSC of apple.

Objective To investigate the correlation between heart development related genes paired-liked homeodomain transcription factor 2 (PITX2) and interval defect congenital heart disease (CHD).Methods To target the investigated single nucleotide polymorphisms (SNPs) by means of dbSNPs data base of National Center of Biotechnology Information(NCBI) website and online bioinformatics software,SNPper.To compared SNPs using 30 interval defect CHD patients and 30 healthy children as control group.Venous blood were collected to detect PITX2 DNA sequencing,then new SNPs were explored.Statistic differences of alleles and genotype frequency distribution between case group and control group were analyzed sequentially.Results Two SNPs in PITX2 genes were rs1051887 and rs28936409,rs1051887:a substitution of cytosine for guanine at nucleotide 954,which suggested the conversion of glutamic acid into glutamine at amino acid residue 106;rs28936409:a substitution of cytosine for guanine at nucleotide 910,which suggested the conversion of arginine into proline at amino acid residue 91.The selected 2 SNPs in the case group and control group showed no polymorphism results.In this research,a new polymorphism 304C ＞ G(Glu102Gln) in PITX2 gene was first identified.Frequencies of CG genetype in case group were higher than control group(x2 =8.531,P ＜0.05),and similar results were found in allelic frequencies(x2 =6.508,P ＜ 0.05).Accordingly,CG genetype increased the risk of interval defect CHD (CG genetype OR =2.833,95 ％ CI:1.297-6.188).Moreover,data showed there was no significant difference in the genotype distribution of SNPs between the case group and control group via Hardy-Weinberg Testing(P ＜ O.05).Conclusions A new SNP 304C ＞ G(Glu102Gln) was found and the CG genetype of the new SNP (304C ＞ G) may increase the risk of interval defect CHD.

Objective To analyze the behavioral characteristics and mental health status for patients with chronic hepatitis B,to explore the correlation between them and analyze the influence of behavioral characteristics of patients with chronic hepatitis B on their mental health.And provide a reference of clinical mental health intervention for patients with chronic hepatitis B.Methods 216 cases of patients with chronic hepatitis B in our hospital from October 2010 to October 2011 were chosen as the research object.Symptom checklist-90(SCL-90)and Eysenck personality questionnaire(EPQ)were used for evaluation.The correlation of behavioral characteristics and mental health status for patients with chronic hepatitis B and the effect of behavioral characteristics of patients with chronic hepatitis B for their mental health were analyzed.Results The SCL-90 score for 216 cases of chronic hepatitis B patients showed that its total score was significantly higher than the norm,the difference was statistically significant.The results of EPQ score showed that the neurotic or emotional stability of personality scores were significantly higher than the norm,the difference was significant.The psychotic scores were significantly lower than the norm,the difference was significant.The analysis of the correlation for nine projects in the SCL-90 scale and four items in the EPQ scale showed that introversion had a significant positive correlation with compulsion,depression,anxiety,fear and paranoia.The nervous or emotion had a significant positive correlation with compulsion,interpersonal sensitivity,paranoid,psychotic.Conclusions The behavioral characteristics of chronic hepatitis B patients have a relation with the mental health.The mental health status changes according to the changes of behavioral characteristics.

Objective To explore the relationship between left ventricular diastolic function and the dynamic changes of myocardial ultrastructure and argyrophilic fiber in diabetic rats on the different periods of lesions(week 4,12 and 24).Methods The diabetes mellitus(DM)in healthy male Sprague-Dawley rats was induced by a single injection of streptozotocin(STZ,Sigma)into intraperitoneal at a dose of 65 mg/kg body weight.The left ventricular diastolic function was measured by Color Doppler Flow Imaging-Pulsed Wave(CDFIPW)and Doppler Tissue Imaging(DTI)echocardiography.Heart tissue at the apex was obtained rapidly for transmission electron microscope study.Argyrophilic staining was used in the study of argyrophilic fiber volume fraction(APFVF)in heart interstitial tissue.Results Diastolic dysfunction of left ventricle was detected in STZinduced diabetic rats by CDFI-PW(E/A ＜ 1)at week 4,and progressed gradually.Pseudonormal filling (E/A ＞ 1)was found in diabetic rats at week 24,which could be identified by DTI(Ea/Aa ＜ 1).Diastolic function of normal rats was not impaired(E/A ＞ 1 and Ea/Aa ＞ 1).Transmission electron microscopy revealed a spectrum of subcellular remodeling in myocardium which was characterized by myofibril content decrease,disorganization,mitochondrial degeneration,sarcoplasmic reticulum,structural disorder.Compared with the control group,APFVF in myocardium was increased significantly in diabetic rats(P ＜ 0.05).Conclusion The diastolic dysfunction in STZ-induced diabetic rats correlates with damage of ultrastructure and increase of myocardial argyrophilic fiber.

In previous study we isolated a gram-positive bacterial strain, designated Niu-O16, from bovine rumen gastric juice. The growing cells of bacterial strain Niu-O16 is capable of biotransforming isoflavone daidzein into dihydrodaidzein efficiently under anaerobic conditions. In this study we investigated the optimal bioconversion conditions for the resting cells of bacterial strain Niu-O16 to convert daidzein into dihydrodaidzein. Single factor test showed that the optimal conditions for the initial pH of phosphate buffer, the concentration of the resting cell and the concentration of the substrate daidzein were 6.0-8.0, 32-64 mg/mL (wet weight) and 0.8-1.2 mmol/L, respectively. Orthogonal experiments were used to determine the optimal combination of the resting cell concentration, substrate concentration and biotransformation time. The results showed that the optimal combination included resting cell concentration 32 mg/mL, substrate concentration 0.8 mmol/L and the biotransformation time 24 h. Furthermore, the biotransformation kinetics under optimal conditions were studied, under which conditions the highest bioconversion rate was 63.9% in the resting cell system. The results might provide information for resting cell biotransforming of anaerobes as well as its industrial application.

Sertoli cell (SC) is intrinsic to the testis and provides an appropriate growth environment for the germ cells. It was separated from rat's testis and identified by hematoxylin and eosin staining(HE) and immunocytochemical reaction, then cultivated in vitro. Culture conditions such as pH, osmotic pressure and metabolic parameters that include consumption rates of glucose, glutamine, amino acids and formation rates of lactic acid, ammonium ion were investigated. It was showed that adhesion process of SCs was accomplished within 2-4 hours after inoculation. It was also observed that the SCs entered into the decline phase when the concentration of ammonium ion and lactic acid were above 2.3 mmol/L and 14 mmol/L, respectively, which caused osmotic pressure above 326 mosm/kg and pH below 6.8 in the medium. As the changes of amino acids during culture were concerned, Glu and Ala accumulated rapidly, while Val, Leu, Ile reduced slightly and at the same time Ser, Arg, and Gly were stable. The restrict factors for SCs grown in static culture might be high osmotic pressure and low pH, which were generated when glutamine and glucose were metabolized into lactic acid. The findings could be fundamental in the process optimization of large scale Sertoli cells in vitro culture.

Objective To evaluate the development of fetal cerebellar vermis using 3D transabdominal ultrasound,and provide evidence for prenatal screening fetal cerebellar vermis anomalies.Methods Totally 387 normal fetuses at 20～36 gestation weeks were examined by three-dimensional extended imaging(3DXI) to observe the fetal cerebellar vermis.The width,anteroposterior diameter,craniocaudal diameter of the cerebellar vermis,the angle between brain stem and vermis,a ratio between the area of anterior vermis and posterior vermis were measured,and the relationship between the gestational age and parameters mentioned above were analyzed.Results The multi slice view of 3DXI and 3D reconstructed sagittal section well evaluated the integrity of vermis morphous and vermis size,identified characteristic signs of vermis:the fourth ventricle apex and vermis crack.In normal fetus,brain stem and vermis were almost parallel,the angle between them was 3.97°±1.65°.There was no significant correlation between the angle and the gestational age.The area of the anterior vermis was smaller than that of posterior vermis,with a ratio of 0.76±0.06,which was also not related to gestational age.The width,anteroposte rior diameter and craniocaudal diameter of the cerebellar vermis were positively correlated with gestational age.Conclusions The multi slice view of 3DXI and 3D reconstructed sagittal section should help evaluate the development of the cerebellar vermis,accurately show the cerebellar vermis and its surrounding strutures,and provide a new way to evaluate the fetal cerebellar vermis.
