OBJECTIVE To evaluate the quality differences of Alpinia katsumadai from different habitats. METHODS High- performance liquid chromatography(HPLC) was used to establish the fingerprints of A. katsumadai from 18 batches of different habitats, and the quality of A. katsumadai from different habitats was comprehensively evaluated by similarity evaluation, cluster analysis (CA), principal component analysis (PCA), orthogonal partial least squares-discriminant analysis (OPLS-DA) and the content determination results of alpinetin, pinocembrin, cardamonin and alnustone in A. katsumadai. RESULTS The similarity of HPLC fingerprints for 18 batches of A. katsumadai was >0.9. Eleven common peaks were identified from the chromatogram, and four of them were specifically characterized. Both CA and PCA grouped 18 batches of A. katsumadai into 3 categories, extracting 2 principal components (the cumulative variance contribution rate reached 89.798%). OPLS-DA identified 9 quality difference markers, namely the components corresponding to peaks 4, 9, 3, 2, 7 (pinocembrin), 8 (cardamonin), 6 (alpinetin), 10 and 11 (alnustone). The content of alpinetin, pinocembrin, cardamonin, and alnustone ranged from 4.507 1-11.579 7, 5.154 4-14.183 3, 5.109 5-13.588 3 and 4.494 6-11.277 2 mg/g, respectively. CONCLUSIONS The quality of A. katsumadai from different habitats is quite different, and the quality of A. katsumadai from Hainan is the best.

Objective:To construct a prognostic nomogram based on epithelial-stromal interaction protein 1(EPSTI1)and predict the pro-gnosis of clear cell renal cell carcinoma(ccRCC).Methods:A retrospective analysis was performed from January 2012 to December 2015 at The First Affiliated Hospital of Fujian Medical University,on 221 patients with ccRCC who underwent surgical treatment in our center and 533 patients with ccRCC in The Cancer Genome Atlas(TCGA)database.Immunohistochemical(IHC)staining was performed on adjacent nor-mal and cancerous tissues to analyze the expression level of EPSTI1 and its correlation with clinicopathological characteristics.Kaplan-Meier survival analysis was performed for the overall survival(OS)and disease-free survival(DFS)of patients with high and low EPSTI1 expression levels.Univariate and multivariate Cox proportional hazards models were used to analyze the prognostic factors for OS,and a nomogram model was constructed and verified. Results:The IHC scores and mRNA expression levels of EPSTI1 were significantly higher in ccRCC tissues than in normal tissues(all P<0.001).EPSTI1 was expressed at higher levels in cancer tissues at higher T stages(P=0.036,P=0.006).The EPSTI1 protein expression level was related to the maximum tumor diameter and TNM stage(P=0.002,P=0.032,respectively).The OS and DFS were higher in the low-EPSTI1-expression group than the high-EPSTI1-expression group(P=0.046,P=0.003,P=0.001).Univariate and multivariate Cox regression analyses showed that a high EPSTI1 protein expression level,WHO/ISUP grade, and AJCC/TNM stage were independent risk factors for poor prognosis(P=0.009,P=0.039,P<0.001).The prognostic nomogram model constructed based on the above variables was su-perior to the AJCC/TNM stage in predicting the 5-year OS,and the calibration curve showed that the predicted value of the model was con-sistent with the actual value. Conclusions:The nomographic model based on EPSTI1,AJCC/TNM staging and WHO/ISUP staging has a strong predictive ability for the prognosis of renal clear cell carcinoma.

Safety and Health at Work 2024;15(1):118-122
To understand biosafety's current situation in laboratory animal research and risk factors affecting occupational health. Compliance surveys were conducted by questionnaire via Questionnaire Star (an application app on the Internet) in Chinese. Thirty-nine anonymous questionnaires were collected. The surveyed institution has established 24 types of ABSL (Animal Biosafety Laboratory) and biosafety management organizations and systems equipped with safety equipment. Our study also suggests that the principal of the laboratory establishment fails to perform supervision and inspection responsibilities, the inappropriate design of the animal biosafety laboratory, non-standardized personnel training and health management, non-strict waste management, and insufficient emergency management. The administrative department and work units should address certain safety and occupational health risks in laboratory animal research. The author proposes control strategies based on organizational guarantee, personnel management, emergency management, etc., to help prevent risks and ensure occupational health. Due to regional limitations and small sample size, the results may not be generalisable to all parts of the world. However, some of the key common issues may also be present in other regions, so we believe that this research still has some relevance.

ObjectiveTo explore the effect of Baitouweng Tang (BTWT) on the apoptosis of human colorectal cancer HCT116 cells and decipher the underlying mechanism based on the Hedgehog (Hh) signaling pathway. MethodHCT116 cells were treated with BTWT (25, 50, 100, 200, 500, 750, and 1 000 mg·L-1) for 24 h, and then the cell proliferation was detected by methyl thiazolyl tetrazolium (MTT) colorimetry. Five groups were designed for the treatment of HCT116 cells, including a blank control group, BTWT groups (125, 250, and 500 mg·L-1), and a positive control (5-fluorouracil, 5-FU, 40 mmol·L-1) group. The cell morphology was observed under an inverted microscope. The migration of the cells was detected by scratch test, and the apoptosis by Hoechest 33324/propidium iodide (PI) staining and flow cytometry. Western blot was employed to determine the protein levels of sonic hedgehog (SHh), GLI family zinc finger protein 1 (Gli1), smoothened (Smo), suppressor of fused (SuFu), cellular-myelocytomatosis viral oncogene (c-Myc), and the apoptosis-related proteins B-cell lymphoma-2 (Bcl-2) and Bcl-2-associated X protein (Bax). The quantitative real-time reverse transcription PCR (Real-time PCR) was employed to determine the mRNA levels of Bax, Bcl-2, SHh, Gli1, Smo, SuFu, and c-Myc. ResultCompared with the blank control group, BTWT changed the cell morphology (making the cell become round with dense nucleus), inhibited the proliferation of HCT116 cells in a dose-dependent manner, decreased the ability of migration (P<0.05, P<0.01), and increased apoptotic cells. Compared with the blank control group, BTWT (500 mg·L-1) treatment for 24 h up-regulated the protein and mRNA levels of Bax (P<0.05, P<0.01) and down-regulated the protein and mRNA levels of Bcl-2 in HCT116 cells (P<0.05, P<0.01). Moreover, the treatment down-regulated the mRNA and protein levels of SHh, Gli1, Smo, and c-Myc (P<0.05, P<0.01) and up-regulated the mRNA and protein levels of SuFu (P<0.05, P<0.01). ConclusionBTWT inhibited the proliferation and migration and induced the apoptosis of colorectal cancer HCT116 cells by down-regulating the Hh signaling pathway.

OBJECTIVE:
This study was aimed at investigating the carrier rate of, and molecular variation in, α- and β-globin gene mutations in Hunan Province.
METHODS:
We recruited 25,946 individuals attending premarital screening from 42 districts and counties in all 14 cities of Hunan Province. Hematological screening was performed, and molecular parameters were assessed.
RESULTS:
The overall carrier rate of thalassemia was 7.1%, including 4.83% for α-thalassemia, 2.15% for β-thalassemia, and 0.12% for both α- and β-thalassemia. The highest carrier rate of thalassemia was in Yongzhou (14.57%). The most abundant genotype of α-thalassemia and β-thalassemia was -α 3.7/αα (50.23%) and β IVS-II-654/β N (28.23%), respectively. Four α-globin mutations [CD108 (ACC>AAC), CAP +29 (G>C), Hb Agrinio and Hb Cervantes] and six β-globin mutations [CAP +8 (C>T), IVS-II-848 (C>T), -56 (G>C), beta nt-77 (G>C), codon 20/21 (-TGGA) and Hb Knossos] had not previously been identified in China. Furthermore, this study provides the first report of the carrier rates of abnormal hemoglobin variants and α-globin triplication in Hunan Province, which were 0.49% and 1.99%, respectively.
CONCLUSION
Our study demonstrates the high complexity and diversity of thalassemia gene mutations in the Hunan population. The results should facilitate genetic counselling and the prevention of severe thalassemia in this region.
