1.Regulation of JAK/STAT pathway inhibition on the biological behavior of hepatocellular carcinoma cells and the mechanism

Background and Aims:Hepatocellular carcinoma(HCC)is one of the most common malignancies,with high incidence and mortality.The JAK/STAT signaling pathway plays a crucial role in regulating cell proliferation,apoptosis,and immune responses,and its persistent activation is closely associated with the development of HCC.This study aimed to investigate the effects of the JAK2 inhibitor AG490 and the STAT3-related inhibitor rapamycin(RPM)on the biological behaviors of HCC cells and their underlying molecular mechanisms.Methods:Human hepatoma HepG2 cells were divided into four groups:blank control,AG490,RPM,and AG490+RPM.Cell proliferation,migration,and apoptosis were assessed by MTT assay,scratch test,and flow cytometry,respectively.ELISA and qRT-PCR were used to detect the protein and mRNA expression of Bax,Bcl-2,caspase-3,survivin,and c-Myc.A Western blot analysis was performed to examine the expression and phosphorylation levels of JAK2 and STAT3.Results:Compared with the blank control group,the AG490,RPM,and AG490+RPM groups showed significantly decreased cell proliferation and migration abilities,as well as increased apoptosis,with the most pronounced effects observed in the AG490+RPM group(all P<0.05).Inhibitor-treated groups showed elevated expression of Bax and caspase-3,decreased expression of Bcl-2,survivin,and c-Myc,with the most significant changes in the AG490+RPM group(all P<0.05).In addition,the ratios of p-JAK2/JAK2 and p-STAT3/STAT3 were significantly reduced in all treated groups,with the lowest levels in the AG490+RPM group(all P<0.05).Conclusion:Inhibitors targeting the JAK/STAT pathway significantly suppress proliferation and migration,and induce apoptosis in HCC cells,possibly by downregulating p-JAK2 and p-STAT3,as well as modulating genes related to apoptosis and proliferation.The combined use of AG490 and RPM exhibits superior antitumor effects,suggesting that multi-target blockade of the JAK/STAT pathway may represent a promising therapeutic strategy for HCC.

3.Strategies for Preventing and Treating Allergic Rhinitis in Children with Chinese Medicine

Allergic rhinitis is a common allergic disease in children.Its pathogenesis is complex and it is difficult to achieve radi-cal cure or effective and stable long-term treatment goals.Chinese medicine has obvious advantages in preventing and treating allergic rhinitis in children due to its wide range of targets,long-lasting effects and few adverse reactions.This paper proposes that the onset of allergic rhinitis is mostly caused by the dysfunction of the lung,spleen and kidney,the external wind triggering the latent wind,and the combination of the two winds.A staged prevention and treatment strategy of Chinese medicine should be adopted,which includes dispersing external wind,suppressing latent wind,and promoting lung-qi and clearing nasal orifice during the attack period to treat its symptoms,and preventing external wind,calming down latent wind,and regulating and tonifying the lung,spleen,and kidney during the remission period to treat its root cause;meanwhile,attention should be paid to avoiding the adverse effects of congenital endowment factors and the induction of acquired environmental factors,strengthening the body's health to protect against the evil wind,preventing the transformation of existing diseases and the recurrence of allergic rhinitis in children at all stages.
4.Establishment and Validation of ddPCR Method for Component Proportion of Multivalent Mutant COVID-19 mRNA Vaccine

OBJECTIVE To establish and verify a ddPCR method to determinate the proportion of mRNA components in multivalent mutant COVID-19 mRNA vaccine. METHODS Different types of coronavirus mRNA were used as target genes, and their conserved sequences were selected to design primers and probes. The ddPCR method for measuring the proportion of multivalent mutant COVID-19 mRNA vaccine components was established, and the linear range, repeatability, accuracy, intermediate precision, durability and applicability of the method were verified. RESULTS When the total mRNA concentration ranged from 1 to 150 pg·mL-1, the linearity was good. Repeatability coefficient of variation(CV) was within 10%. The recoveries of accuracy verification were 84%-114%, CV<10%. The intermediate precision and durability coefficient of variation CV were all< 5%, and the CV of the proportion of components in the four different bivalent COVID-19 mRNA vaccines was all≤5%. CONCLUSION The ddPCR method established for detecting the component proportion of multivalent mutant COVID-19 mRNA vaccine has high sensitivity, good stability and strong specificity, which is suitable for detecting the component proportion and content of multivalent COVID-19 mRNA vaccine.
5.Regulation trend of resveratrol on TNFα-,IL-1β, IL-6 expressions in bronchoalveolar lavage fluid of RSV-infected BALB/c mice.

OBJECTIVETo study the regulation trend of resveratrol on TNF-alpha, IL-1beta, IL-6 expressions in bronchoalveolar layage fluid (BALF) of RSV-infected BALB/c mice at different time points.
METHODRSV-induced BALB/c mice were orally administered with resveratrol. Their BALFs were collected at 24, 72 and 144 h after the first nasal drip with RSV to detect the level of TNF-alpha, IL-1P3, IL-6 by EILSA.
RESULTThe expression of TNF-alpha, IL-1Pf and IL-6 in BALF increased significantly compared with the normal group (P <0. 01) after 24 hours of RSV infection, while the expression of TNF-alpha (P < 0.01), IL-1beta (P < 0.05), IL-6 (P < 0.01) in the resveratrol group decreased notably compared with the model group. After 72 hours of infection with RSV, although the expression of TNF-alpha (P < 0.05), IL-1beta (P < 0.01) and IL-6 (P < 0.01) in BALF in model group were higher than those in the normal group, they were much more lower than at 24 h. The expression of IL-1beta and IL-6 (P < 0.05) in the resveratrol groups were down-regulated significantly, but no difference had been shown in TNF-alpha expression compared with the RSV infection group. After infection with RSV for 144 h, the expression of IL-1beta (P < 0.01) and IL-6 (P < 0.05) in BALF in the model group were higher than those in the normal group, but there was no difference in the secretion of TNF-alpha. The expression of TNF-alpha, IL-1beta and IL-6 showed also no remarkable difference between the resveratrol groups and the RSV infection group.
CONCLUSIONResveratrol can inhibit the over expression of inflammatory factors TNF-alpha, IL-1beta, IL-6 in bronchoalveolar lavage fluid of RSV-induced BALB/c mice and keep them at a low level with the passing of infection time.

6.Investigation on Experts' Questionnaires of Curative Effect Evaluation Method of Traditional Chinese Medicine in Treating Pediatrics Viral Pneumonia Based on Delphi Method

Objective: To probe the curative effect evaluation method of traditional Chinese medicine in treating pediatrics virus pneumonia.Methods: On the basis of the outcome of analysis made on the preceding three turns of experts' questonnaires,the fourth turn of questionnaires was prepared to investigate the curative effect evaluation method of traditional Chinese medicine in treating pediatrics viral pneumonia.They were distributed to 30 experts in this field all over the country.The questionnaires were analyzed with the method of Delphi Measurement.Results: Chest X ray,cough,temperature,lung auscultation and so on were very important in disease curative effect evaluation,the ratio of full score higher than 0.95 and the coefficient of variation less than 0.24;cough,lung auscultation,Chest X ray and so on were main components in the syndrome and the ratio of full score and the coefficient of variation were respectively more than 0.90 and less than 0.34;dyspnea with rapid and short breath,lung auscultation and Chest X ray were main final result index and the ratio of full score and the coefficient of variation was respectively 1 and 0.Conclusion: Chest X ray,symptoms,temperature and lung auscultation were very important in disease curative effect evaluation;cough,lung auscultation and Chest X ray were main components in the syndrome;dyspnea with rapid and short breath,lung auscultation and Chest X ray were main final result index.
