Objective This study aimed to investigate the tumorigenic properties of MMTV-PyMT breast cancer transgenic mice at different ages(in weeks)and the changes in the composition of immune cells in the tumor microenvironment. Methods Eight groups of 4,6,8,10,12,14,16 and 18 weeks of age MMTV-PyMT female mice(FVB mice as the background)and one group of 8 weeks of FVB female mice were prepared for routine blood testing, the pathological changes of the mammary gland and lung metastases were observed by histopathological sections, and the immune cells in blood, spleen, and tumor were analyzed by flow cytometry. Results MMTV-PyMT mice showed adenular ductal lesions at 4~6 weeks of age;the ductal portion expanded to the growth boundary at 8~9 weeks of age,and then gradually broke through the glandular boundary to form early breast cancer at 8~12 weeks of age,and advanced breast cancer at 10~14 weeks of age.At 12 weeks of age, metastases were visible in the lungs of some mice, and at 14 weeks of age,the number of metastases in the lungs increased significantly. As the age of the mice increased, the number of white blood cells, neutrophils, and platelets in their blood increased gradually, while the lymphocytes and erythrocytes showed a gradual downward trend.Flow cytometry showed that with the increase in age,the proportion of T cells in the spleen and tumor gradually decreased, the MDSCs in the blood, spleen, and tumor gradually increased, and the NK cells in the tumor also gradually increased. Conclusions This study analyzed routine blood tests, pathology, and immune cells in the tissues of MMTV-PyMT mouse models of different weeks of age, providing a novel perspective on the dynamic alterations of the tumor immune microenvironment during the malignant progression of breast cancer.

Objective To investigate the expression of serum long non-coding RNA X-inactive specific transcript(lncRNA XIST)and microRNA-126-3p(miR-126-3p)in patients with non-small cell lung cancer(NSCLC),and their relationship with recurrence after radical surgery. Methods A total of 108 NSCLC patients who underwent radical surgery and were admitted to General Hospital of Southern Theater Command of the People's Liberation Army of China from February 2019 to October 2020 were selected as the NSCLC group, and 52 healthy volunteers who underwent physical examination in this hospital were selected as the control group.qRT-PCR method was used to detect the relative expression levels of serum lncRNA XIST and miR-126-3p.Pearson method was used to analyze the correlation between serum lncRNA XIST and miR-126-3p expression.Logistic regression analysis was used to analyze the influencing factors of postoperative recurrence in NSCLC patients.Receiver operating characteristic(ROC)curve was used to evaluate the predictive value of serum lncRNA XIST and miR-126-3p on postoperative recurrence in patients. Results The expression level of serum lncRNA XIST in the NSCLC group was higher than that in the control group(P<0.05),while the expression level of miR-126-3p was lower than that in the control group(P<0.05).The expression levels of serum lncRNA XIST and miR-126-3p were related to TNM staging and lymph node metastasis(P<0.05).Bioinformatics website predicted that there was a targeted binding site between lncRNA XIST and miR-126-3p,and serum lncRNA XIST was negatively correlated with miR-126-3p expression(r=-0.579,P<0.05).Compared with the non-recurrent group, the expression level of serum lncRNA XIST in the recurrent group increased(P<0.05),while the expression level of miR-126-3p in the recurrent group decreased(P<0.05).Logistic regression analysis results showed that lncRNA XIST,miR-126-3p,lymph node metastasis, and TNM staging were the influencing factors for postoperative recurrence in NSCLC patients(P<0.05).The area under the curve(AUC)of serum lncRNA XIST,miR-126-3p,and the combination of the two in prediction of postoperative recurrence in NSCLC patients were 0.750,0.886,and 0.933,respectively,the combined prediction of the two was superior to individual prediction(Zcombination vs.lncRNA XIST=4.076,Zcombination vs.miR-126-3p=2.065,P<0.05).Conclusion The expression of serum lncRNA XIST is increased and miR-126-3p is decreased in NSCLC patients, both of which have certain predictive value for recurrence after radical surgery in patients.

This study was aimed at providing basic data for the control and prevention of Yersinia enterocolitica(Ye)infections.Ye isolates from stool samples collected from patients with diarrhea in a Beijing district between January 2019 and June 2024 were studied.Basic patient information and stool samples were collected, and quantitative polymerase chain reaction(qPCR)was applied to enriched cultures.Further analyses included virulence gene detection,whole-genome sequencing, and drug resistance detection. The detection rate of Ye was 0.76%(11/1 439),according to culture methods, thus yielding 12 Ye strains from distinct patients:11 isolated during the study period and 1 from 2017.The 12 Ye positive patients were 6-41 years of age,and their clinical presentations predominantly featured watery stools(66.67%,8/12)and loose stools(33.33%,4/12).The frequencies of nausea, vomiting, and fever were 41.67%(5/12),41.67%(5/12),and 8.33%(1/12),respectively.The drug resistance rates of Ye to TET,AMP,and NAL were 50.00%(6/12),33.33%(4/12),and 25.00%(3/12),respectively.One Ye strain exhibited multidrug resistance to ETP,MEM,TET,CIP,NAL,and AMP.According to qPCR detection of five common virulence genes, two Ye strains were identified as ystA+/ystB-type(ystA+/ystB-/ail+/yadA+/virF+),whereas ten strains were identified as ystA-/ystB+type(ystA-/ystB+/ail-/yadA-/virF-).VFDB database analysis based on genome sequences indicated that 12 Ye strains carried an average of 11 key virulence genes associated with adhesion, invasion, protease activity, and flagellar movement, and predicted 106 virulence genes and 12 virulence gene profiles.Only the two ystA+/ystB-Ye strains contained elements related to the TTSS and ABC transporter function.Detection of ystA-/ystB+Ye in stool isolation and culture of diarrhea cases might potentially have been missed in some cases, thus highlighting the importance of fluorescence PCR screening of fecal growth solutions to enhance isolation efficiency. Moreover, our findings revealed the genetic diversity of Ye isolated from diarrhea cases, thereby indicating the presence of multiple types of virulence genes within this pathogen.

The neonatal mammalian heart has a remarkable regenerative capacity, while the adult heart has difficulty to regenerate. A metabolic reprogramming from glycolysis to fatty acid oxidation occurs along with the loss of cardiomyocyte proliferative capacity shortly after birth. In this study, we sought to determine if and how metabolic reprogramming regulates cardiomyocyte proliferation. Reversing metabolic reprogramming by carnitine palmitoyltransferase 1 (CPT1) inhibition, using cardiac-specific Cpt1a and Cpt1b knockout mice promoted cardiomyocyte proliferation and improved cardiac function post-myocardial infarction. The inhibition of CPT1 is of pharmacological significance because those protective effects were replicated by etomoxir, a CPT1 inhibitor. CPT1 inhibition, by decreasing poly(ADP-ribose) polymerase 1 expression, reduced ADP-ribosylation of dual-specificity phosphatase 1 in cardiomyocytes, leading to decreased p38 MAPK phosphorylation, and stimulation of cardiomyocyte proliferation. Our present study indicates that reversing metabolic reprogramming is an effective strategy to stimulate adult cardiomyocyte proliferation. CPT1 is a potential therapeutic target for promoting heart regeneration and myocardial infarction treatment.
