1.Advances and challenges of 18 kDa translocator protein(TSPO)-PET imaging in neurodegenerative diseases

Neurodegenerative diseases(NDD)are a heterogeneous group of disorders characterized by neuronal degeneration and functional impairment,in which neuroinflammation plays a pivotal role throughout disease onset and progression.The 18 kDa translocator protein(TSPO)serves as an important molecular biomarker of neuroinflammation,with its expression closely associated with activation of microglia and astrocytes.TSPO-PET enables noninvasive and dynamic mapping of spatiotemporal distribution of cerebral neuroinflammation,providing crucial molecular imaging evidence for early diagnosis,disease evaluation and therapeutic response assessment of NDD.The advances and challenges of TSPO-PET imaging in NDD were reviewed in this article.
2.Construction of Evidence-Based Guidelines for the Diagnosis and Treatment of Distal Radius Fractures in Adults (2024) based on the Delphi method

Objective:To report construction of Evidence-Based Guidelines for the Diagnosis and Treatment of Distal Radius Fractures in Adults (2024) using the Delphi method.Methods:Literature related to the study of adult distal radius fractures was fully searched for and evaluated. An expert group was established from representative experts from all over the nation. The related clinical issues were established by consulting the experts in the form of electronic questionnaires, strictly following the Delphi research method. After the first draft of Evidence-Based Guidelines for the Diagnosis and Treatment of Distal Radius Fractures in Adults (2024) was written, an expert consultation questionnaire was designed for the recommendation opinions to determine the recommendation strength.Results:The clinical issues were determined by 2 rounds of correspondence based on the Delphi method. For the both rounds of correspondence, the questionnaire recovery rates were respectively 88.68% (47/53) and 98.11% (52/53), and the expert authority coefficients >0.7. According to the screening criteria based on the importance of clinical issues (mean importance score <3.5 points or a coefficient of variation ≥0.25 points and a full score ratio <30%) and expert opinions, a total of 40 clinical issues were deleted in the first round of determination of clinical issues, and a total of 5 clinical issues deleted in the second round of determination of clinical issues. The reliability analysis of the results of the 2 rounds of questionnaires showed that the Cronbach α coefficient was >0.9. In the questionnaire to determine the recommendation strength, according to the screening criteria for the consistency of recommendation strength (consistency ≥ 70%) and expert opinions, a total of 26 recommendations were screened in the first round. In the second round when the remaining 4 recommendations were investigated, one recommendation reached the consistency of recommendation strength ≥ 70%. Eventually, 27 recommendations were formed.Conclusion:The Evidence-Based Guidelines for the Diagnosis and Treatment of Distal Radius Fractures in Adults (2024) constructed using the Delphi method shows good scientific validity, authority, and reliability, providing methodological references for guideline development and research.

5.Pharmacokinetics of three phenolic acids of xiangdan injection in rats.

OBJECTIVETo establish a HPLC method for the analysis of Danshensu (DSS), protocatechuic acid (PA) and protocatechuic aldehyde (PAL) of Xiangdan injection in rat's plasma, and to study pharmacokinetic characteristics of Xiangdan injection components in rats with m-hydroxybenzoic acid as internal standard.
METHODprotein was precipitated by 10% trichloroacetic acid and extracted by ethyl acetate. The plasma concentration was detected by HPLC, The pharmacokinetics parameters of DSS, PA and PAL were calculated by DAS2.0 software after iv injection.
RESULTDSS, PA and PAL have a good linear relationship in 0.68-44.0 mg x L(-1), 0.43-14.0 mg x L(-1) and 0.38-12.0 mg x L(-1), respectively. The average recoveries were more than 92% and the RSD of precision and stability of the test were between 0.4%-4.8%. DSS, PA and PAL showed a two-compartment open model, the half-life of absorption (t(1/2alpha)) were (6.26 +/- 4.6), (5.93 +/- 4.9), (18.44 +/- 2.4) min, the half-life of elimination (t(1/2beta)) were (64.11 +/- 8.8), (63.28 +/- 0.13), (69.315 +/- 0) min, the area under curve(AUC(0-infinity)) were (852.98 +/- 175.6), (83.84 +/- 58.8), (147.79 +/- 12.3) mg x min(-1) x L(-1).
CONCLUSIONA method with high recovery rate and good stability was established to determine the blood concentration of DSS, PA, PAL in Xiangdan injection and applied in its pharmacokinetics successfully.

6.Pharmacokinetics and Pharmacodynamics of Intravenous Famotidine in Healthy Volunteers

The pharmacokinetics and pharmacodynamics of famotidine were investigated in 10 healthy male volunteers after single intravenous administration of 20 mg. The blood drug levels were determined by a high performance liquid chromatography. The 1gC of famotidine in plasma vs time curve were found to be twcncompartment open model in healthy volunteers. The terminal half-life averaged 3.16h; the total distribution volume 99.40L; the total plasma clearance 392.12ml/min; the area under the plasma concentration curve 1057.45 h?ng/ml. A mathematic equation describing the whole course of blood drug levels in relation to inhibitory effects on intragastric acid output is as follows: E= 100?C2.60/(C2.60+14.712.60). The constant 14.71 is EC50 (ng/ml), the blood drug concentration producing 50% of maximal pharmacological effects. Prediction of pharmacodynamic effects from blood drug level and vice versa becomes possible by using the mathematic equation.
