1.Research on mechanism by which Yisui Shengxue Decoction improves bone marrow hematopoietic function in mice with aplastic anemia by regulating Notch signaling pathway

Objective:To investigate the effects of Yisui Shengxue Decoction on the regulation of Notch signaling pathway on bone marrow hematopoiesis in mice with aplastic anemia(AA).Methods:A total of 60 CByB6F1 mice were randomly divided into blank group,model group,positive control cyclosporine(CsA)group,low-dose(YSSX-L)group and high-dose(YSSX-H)group of Chinese herbal compound Yisui Shengxue Decoction,with 12 mice in each group.Except for the blank group,the mice in each group were given 5 Gy X-ray irradiation combined with tail vein infusion of lymphocytes to establish the immune-mediated AA mouse model.The low and high dose groups were given 8.6 and 17.2 g/(kg?d)of Yisui Shengxue Decoction by gavage,the model group and the blank group were given equal volume of saline by gavage,and the positive control group was given 25 mg/(kg?d)of cyclosporine by ga-vage for 14 days.The mice were tested for white blood cell count(WBC),red blood cell count(RBC),hemoglobin(Hb),platelet count(PLT)and bone marrow nucleated cell count in peripheral blood,bone marrow pathological changes and the levels of IFN-γ,IL-4 and IL-5 in peripheral serum,and T-lymphocyte transcription factors T-bet,GATA3,RORγt,FOXP3 mRNA and protein in spleen tissues,as well as the expression levels of Notch signaling pathway-related genes and protein in spleen tissues.Results:Compared with the blank group,the peripheral blood WBC,RBC,Hb,PLT,bone marrow nucleated cell count,expressions of serum IL-4,IL-5 and spleen tissue GATA3,FOXP3 mRNA and protein were significantly lower in the model group(P<0.01),and the expressions of serum IFN-γ,spleen tissue T-bet,RORγt mRNA and protein,and Notch1,Jagged1,DLL4 mRNA and protein were significantly in-creased(P<0.05).Compared with the model group,peripheral blood WBC,RBC,Hb,PLT,bone marrow nucleated cell count,ex-pressions of serum IL-4,IL-5 and spleen tissue GATA3,FOXP3 mRNA and protein were significantly higher in each intervention group(P<0.05),and the expression of serum IFN-γ and spleen tissue T-bet,RORγt mRNA and protein as well as Notch1,Jagged1,DLL4 mRNA and protein were significantly reduced(P<0.05).Conclusion:Yisui Shengxue Decoction can improve bone marrow he-matopoietic function and regulate immune disorders in AA mice,and its mechanism of action may be related to the regulation of Notch signaling pathway.
2.Effects of Tongqiao Pingchuan Decoction on miR-155 and NF-κB-HIF-1α Signaling Pathway in Mice with Combined Allergic Rhinitis and Asthma Syndrome

Objective To observe the effects of Tongqiao Pingchuan Decoction on miR-155 and NF-κB-HIF-1α signaling pathway in mice with combined allergic rhinitis and asthma syndrome(CARAS);To explore its potential mechanisms in the treatment of chronic inflammation of respiratory tract in CARAS.Methods Totally 60 female BALB/c mice were divided into blank group,model group,miR-155 inhibitor NC group,miR-155 inhibitor group,dexamethasone group and Tongqiao Pingchuan Decoction group using a random number table method,with 10 mice in each group.Except for the blank group,all other groups were sensitized by intraperitoneal injection of ovalbumin combined with aluminum hydroxide gel on days 0 and 7.From days 14 to 30,5%ovalbumin was administered intranasally every other day,followed by continuous nebulization with 5%ovalbumin from days 31 to 37.The miR-155 inhibitor NC group and the miR-155 inhibitor group received intranasal administration of miR-155 inhibitor NC and miR-155 inhibitor solution,respectively,3 hours before nebulization.The blank group was given an equivalent volume of normal saline via intraperitoneal injection,intranasal administration and nebulization.The administration groups were gavaged with the corresponding drug solution 1 hour after nebulization,while the other groups received an equivalent volume of normal saline via gavage.HE staining was used to observe morphology of nasal mucosa and lung tissue,ELISA was used to detect the contents of interleukin(IL)-4,IL-5,and γ-interferon(IFN-γ)in bronchoalveolar lavage fluid,RT-qPCR was used to detect the expression of miR-155,T-bet,GATA-3,NF-κB p65 and HIF-1α mRNA in nasal mucosa and lung tissue,Western blot was used to detect the expressions of T-bet,GATA-3,NF-κB p65 and HIF-1α proteins in lung tissue.Results Compared with the blank group,the model group mice showed shedding of nasal mucosal cilia,tissue congestion and edema,significant glandular hyperplasia and inflammatory cell infiltration,bronchial lumen stenosis,smooth muscle thickening and a large amount of inflammatory substance exudation,the contents of IL-4 and IL-5 increased,while IFN-γ content decreased(P<0.01),the mRNA expression of miR-155,GATA-3,NF-κB p65,HIF-1α in nasal mucosa and lung tissue were increased(P<0.01),while T-bet mRNA expression was decreased(P<0.01),the protein expression of GATA-3,NF-κB p65 and HIF-1α in lung tissue increased(P<0.01),while T-bet protein expression decreased(P<0.01).Compared with the model group,the miR-155 inhibitor group,dexamethasone group and Tongqiao Pingchuan Decoction group showed reduced inflammatory cell infiltration and pathological changes in nasal mucosa and lung tissue of mice,the contents of IL-4 and IL-5 in bronchoalveolar lavage fluid were reduced,while the content of IFN-γ was increased(P<0.01),the expressions of miR-155,GATA-3,NF-κB p65 and HIF-1α mRNA in nasal mucosa and lung tissue were reduced(P<0.05,P<0.01),while the expression of T-bet mRNA increased(P<0.05,P<0.01),the expressions of GATA-3,NF-κB p65 and HIF-1α protein in lung tissue were reduced(P<0.05,P<0.01),the expression of T-bet protein in dexamethasone group and Tongqiao Pingchuan Decoction group mice increased(P<0.05).Conclusion Tongqiao Pingchuan Decoction can effectively inhibit the expression of miR-155,suppress the abnormal activation of NF-κB-HIF-1α signaling pathway,correct the imbalance of Th1/Th2 immunity,and play a therapeutic role in alleviating airway inflammation in CARAS.

5.Predictive value of prognostic inflammatory and tumor score in intrahepatic cholangiocarcinoma

Objective:To compare and analyze the predictive value of different inflammatory factors and tumor markers in intrahepatic cholangiocarcinoma and to develop a new and effective preoperative prognostic scoring system.Methods:102 and 72 cases with intrahepatic cholangiocarcinoma who underwent radical surgery in Tianjin Medical University Cancer Institute and Hospital and the Affiliated Hospital of Weifang Medical University were selected as the experimental group and the validation group, respectively. Clinicopathological and follow-up data were collected. Cox proportional-hazards model was used to analyze the predictive value of different prognostic markers. The relationship between prognostic markers and clinicopathological data was analyzed by rank sum test, χ2 or Fisher's exact test. Results:Among the direct inflammatory factors, tumor markers and combined inflammatory factors, prognostic inflammatory index (PII), carbohydrate antigen (CA) 19-9 and systemic inflammation score (SIS) were the most significant predictive factors for postoperative survival outcomes in patients with intrahepatic cholangiocarcinoma. The prognostic inflammatory and tumor score (PITS) was proposed as a new prognostic scoring system for intrahepatic cholangiocarcinoma. PII and CA19-9 were included into the scoring criteria for prognostic stratification of patients. PITS was an independent predictor of tumor-free survival and overall survival in patients with intrahepatic cholangiocarcinoma. Patients with high-grade PITS had later tumor grade and higher frequency of vascular invasion.Conclusion:PITS is highly effective prognostic scoring system for patients with intrahepatic cholangiocarcinoma. In addition, PITS is recommended for preoperative prognostic stratification in patients with intrahepatic cholangiocarcinoma.

Attention is an essential cognitive function which was use to perceive the external world, and it is the basis of all cognitive activities.Other cognitive functions such as working memory, executive function, information processing speed and so on, are also adversely affected in hypoprosexia or hyperfocusing state.Patients with schizophrenia have severe attention disorder, which makes patients unable to successfully complete the task of work or study, thus reduce the quality of life.Attention disorder exists throughout the whole course of this disease, and there is no specific drug for it.In recent years, antipsychotic medications have widespread application, but it has not shown up significantly improvement of attention disorder, worse still, its side effects will exacerbate the disease, which results in the impairment of the patients' ability to learn and explore the novelties.Neuroimaging markers of attention disorder in schizophrenia are the focus of current researches.White matter fibers are important pathways that connect attention networks and maintain the three-dimensional structure of the brain.Previous studies have suggested that attention disorder may be related to abnormalities of white matter fibers connecting various encephalic regions.In particular, abnormalities in the integrity of white matter, such as corpus callosum, cingulum bundle, superior longitudinal fasciculus, inferior fronto-occipital fasciculus, have been reported to be significantly associated with attention disorder in patients with schizophrenia.In this study, the relationship between attention disorder and white matter structure in patients with schizophrenia is reviewed through diffusion tensor imaging (DTI) technique.
7.Effects of oral immunotherapy for the prevention of ventilator-associated pneumonia in premature infants: a Meta-analysis

Objective:To systematically evaluate the intervention effect of oral immunotherapy (OIT) on ventilator-associated pneumonia (VAP) of premature infants.Methods:This study searched the Cochrane Library, PubMed, Embase, Web of Science, CINAHL, Chinese Biomedical Literature Service System, China National Knowledge Infrastructure (CNKI) , WanFang Database and VIP Database, and conducted a comprehensive search for randomized controlled trials of OIT applied to premature infants published from database establishment to October 2020. Two researchers independently screened literature, extracted data, and evaluated methodological quality. RevMan 5.3 software was used for Meta-analysis.Results:A total of 8 studies were included, with a total of 351 premature infants in the intervention group and 352 premature infants in the control group. Meta-analysis results showed that OIT could reduce the incidence of VAP in premature infants [ RR=0.36, 95% CI (0.23-0.56) , P<0.05] and the detection rate of pathogenic microorganisms in the tracheal tube [ RR=0.33, 95% CI (0.18-0.59) , P=0.000 2], and could shorten the length of hospital stay [ MD=-6.70, 95% CI (-13.34--0.06) , P=0.05]. However, there was no statistically significant difference in the mechanical ventilation time, detection rate of oropharyngeal pathogenic microorganisms and mortality rate between the two groups after the intervention (all P>0.05) . Conclusions:OIT can reduce the incidence of VAP in premature infants and the detection rate of pathogenic microorganisms in the tracheal tube, and shorten the length of hospital stay to a certain extent. However, high-quality, large-sample, multi-center randomized controlled studies are still needed for further verification in the future.
8.Application of different postoperative analgesia methods in unilateral breast cancer radical surgery

Objective To select the appropriate postoperative analgesia,through the application of different methods in the postoperative analgesia of the patients with breast cancer.Methods Ninety female patients undergoing unilateral breast cancer radical surgery in general anesthesia,aged 35-60 years,weighing 45-80 kg,falling into ASA physical status Ⅰ or Ⅱ,were ran domly divided into simple intravenous postoperative analgesia group (group P,n =30),local infiltration plus intravenous postoperative analgesia group (group R,n =30),ultrasound guided thoracic nerve block plus intravenous postoperative analgesia (group TP,n =30),group P used dexmedetomidine 2μg/kg plus sufentanil 2μg/kg,group R used 0.375% ropivacaine 20 ml local infiltration added dexmedetomidine 2μg/kg plus sufentanil 2 μtg/kg,group TP used 0.375% ropivacaine 20 ml vertebral side block added dexmedetomidine 2μg/kg plus sufentanil 2 μg/kg.The VAS scores and Ramsay score was recorded 6,12,24,48 h after surgery,the deep sleep quality score at preoperative postoperative 24 h and 48 h,the number of postoperative analgesia pump pressure,the use of sufentanil and the nausea,vomiting,itching,bradycardia,respiratory depression happened in 24 h were documented.Results The VAS scores in groups R and TP 6 h after surgery were significantly lower than that of group P.The VAS scores in group TP 12 h after surgery were significantly lower than that of groups R and P (P<0.05).The sedation scores were not statistically significant among the three groups.The deep sleep quality scores at 24 h in group TP were significantly lower than those of groups R and P (P< 0.05).The number of compressions and the use of the sufentanil in 24 h after surgery in group TP were significantly lower than those of groups R and P,and that in group R was significantly lower than that in group P (P<0.05).The incidence of nausea,vomiting,itching,bradycardia,respiratory depression were not statistically significant in 24 h,and there were no postopera tive complications related to neurologic blocking of lateral vertebral nerve.Conclusion Three methods were safe and effective for postoperative analgesia in patients with radical surgery breast cancer.Ultrasound guided by thoracic nerve block combined intravenous postoperative analgesia was obviously better than local infiltration plus infiltration postoperative analgesia and infiltration postoperative analgesia in terms of postoperative analgesia,quality of sleep and the number of sufentanil.
9.IAP-1 gene silencing enhanced the radiosensitivity of liver cancer SMMC-7721 cells

Objective To explore the effect of cellular inhibitor of apoptosis protein1 (cIAP-1) gene on the radiosensitivity of SMMC-7721 cells.Methods We silenced cIAP-1 expressions by the shRNA technology,and then we detected the changes of cell proliferation,cell cycle and cell apoptosis by CCK8 as-say,Western blot,qRT-PCR and flow cytometry after the radiotherapy.Results The cell proliferation rate of liver cancer SMMC-7721 cells at different radiation doses of 1 Gy,4 Gy,7 Gy and 10 Gy was detected.Comparing with control group (pGCsi-H1-control),the cell proliferation in cIAP-1 silencing group (pGCsiH1-shRNA) was significantly reduced at various radiation doses,and the effect was dose-dependent (P < 0.05).G1/G0 phase arrest was observed after radiation (P <0.01),and the proportion of cells in S phase was significantly reduced compared with control (P < 0.01).Compared with control group,G1/G0 phase arrest was detected (P < 0.05),and the percentage of cell apoptosis was increased significantly in cIAP-1 silencing group (P < 0.05).Conclusion cIAP-1 silencing can enhance the radiosensitivity of liver cancer cells,and inhibit cell proliferation by promoting the anti-tumor effect of radiation.
10.Study on the Mechanism of Anti-myocardial Fibrosis of Salvianolic Acid B

OBJECTIVE:To study the effects of salvianolic acid B(Sal B)on angiotensin Ⅱ(Ang Ⅱ)-induced cardiac fibro-blast proliferation,secretion of type Ⅲ collagen,protein expressions of matrix metalloproteinase 9 (MMP-9),Smad2/3,Smad7, and explore its mechanism of anti-myocardial fibrosis. METHODS:Cells were divided into blank control group(culture medium) Ang Ⅱ model group,Sal B low-dose,medium-dose,high-dose groups (12.5,25,50 μmol/L). After cultured 1 h by blank or drug-containing culture,except for blank control group,cells in other groups were added 1 μmol/L Ang Ⅱ to induce proliferation. for 24 h. MTT method and hematoxylin-eosin staining method were adopted investigate the effect of Sal B on proliferation. Western blot method was adopted to detect the effects of Sal B on protein expressions of type Ⅲ collagen,MMP-9,Smad2/3,Smad7. RE-SULTS:Compared with blank control group,cells in Ang Ⅱ model group were significantly proliferated,protein expressions of type Ⅲ collagen,MMP-9,Smad2/3 were obviously enhanced,protein expression of Smad7 was obviously weakened,with statisti-cal significances(P<0.05). Compared with Ang Ⅱ model group,the cell proliferation in Sal B groups were inhibited,protein ex-pressions of typeⅢcollagen,MMP-9,Smad2/3 were weakened,while protein expression of Smad7 was enhanced. Except the pro-tein expression of type Ⅲ collagen in Sal B low-dose and medium-dose groups,the protein expression of Smad2/3 in Sal B high-dose group did not change significantly,other indexes had statistical significances(P<0.05). CONCLUSIONS:The anti-myo-cardial fibrosis effect of Sal B may be associated with inhibiting the proliferation of cardiac fibroblasts,down-regulating protein ex-pressions of typeⅢcollagen,MMP-9,Smad2/3 and up-regulating protein expression of Smad7.
