1.Study on the mechanism of tetramethylpyrazine pretreatment umbilical cord mesenchymal stem cell transplantation in the treatment of ischemic stroke

Objective To investigate the therapeutic effect,underlying mechanism,and key genes involved in tetramethylpyrazine-pretreated umbilical cord mesenchymal stem cell (ucMSC) transplantation in a rat model of ischemic stroke. Methods The rat MCAO model was established,and umbilical cord-derived mesenchymal stem cells (ucMSCs) pretreated with or without tetramethylpyrazine were transplanted via the tail vein. Neurological function scores,TTC staining,and infarct rates were assessed. Localization of ucMSCs in brain tissue was observed. Experimental groups were analyzed using chip technology,and sample data were standardized. Bioinfor-matics analysis was employed to identify differential genes,which were subsequently validated by PCR. Results The treatment effect in ucMSCs pretreated with tetramethylpyrazine group was significantly superior to that of the untreated group,as evidenced by a significant reduction in neurological function score,infarct rate,and infarct area observed through TTC staining. Moreover,the treated group exhibited a significantly higher number of ucMSCs located within brain injury tissues compared to the untreated group. Subsequently,2905 differential mRNA were screened based on predetermined criteria,including 1754 up-regulated and 1151 down-regulated genes. Among these differentially expressed genes related to the chemokine signaling pathway (identified using a multiple change value ≥ 2.0 and P value ≤ 0.05),we identified 27 genes of interest. Notably,our analysis revealed activation of four genes closely associated with cell migration:Ccr6,Ccr3,Cxcr1 and Ccl6 respectively. Random verification experiments further confirmed a significant increase in gene expression for both Ccr3 and Cxcr1. Conclusions Pretreatment of umbilical cord-derived mesenchymal stem cells (ucMSCs) with tetramethylpyrazine significantly augmented the therapeutic efficacy in a rat model of ischemic stroke. Following pretreatment,there was a substantial increase in the migration of ucMSCs towards the site of brain injury. Our analysis suggests that this effect may be attributed to the activation of multiple chemokines,including Ccr6,Ccr3,Cxcr1,and Ccl6,by tetramethylpyrazine.

3.Survey on servant leadership behavior of head nurses from hospitals of different levels in Shanghai

Objective To understand the whole level of head nurses ’ servant leadership behavior in Shanghai hospitals , so as to provide theoretical basis for optimal management .Methods A total of 2 000 nurses were chosen from two first-level hospitals , two second-level hospitals and two third-level hospitals from February to March 2012 , by the methods combined with step sampling and cluster sampling .Scale for head nurses ’ servant leadership behavior was used to investigate the whole level .SPSS 19.0 was used to analyze the data , descriptive analysis was used to analyze the scores , one-way anova or t test was used to compare perception of leadership behavior between different clusters , and LSD-t was used for pairwise comparison .P<0.05 indicated statistical significance .Results A total of 2 000 questionnaires were handed out and 1 511 effective questionnaires were recovered ,with the rate of 75 .6%.The average score of head nurses ’ servant leadership behavior in Shanghai hospitals was (84.54 ±17.98), which was higher than the median of the scale which was 60.The recognition degree about easy-going was the highest while the recognition degree about authorization was the lowest.The scores of head nurses from first-level hospitals, second-level hospitals and third-level hospitals were(91.62 ±9.40),(90.53 ±11.49),(80.53 ±17.98), respectively, and the difference was statistically significant (F=61.88,P <0.01).There was statistically significant difference in perception toward head nurses’ servant leadership behavior from nurses of different ages , degrees, titles and genders (P<0.05).And there was no statistically significant difference in perception toward head nurses ’ servant leadership behavior from nurses of different working years and marriage status (P >0.05).Conclusions The level of head nurses’ servant leadership behavior in Shanghai hospitals is comparatively good , but there is still room for improvement , especially for head nurses in third-level hospitals .Evaluation of head nurses ’ servant leadership behavior should consider and control the initialization of nurses , in order to prevent differences of perception from affecting the accuracy .
