1.Clinicopathological characteristics of atypical spindle cell lipomatous tumours:report of eight cases

Purpose To explore the clinicopathological characteristics of atypical spindle cell lipomatous tumour(ASLT).Methods The clinical pathology,immunopheno-type,and molecular characteristics of 8 ASLTs diagnosed and treated in Fujian Cancer Hospital from 2020 to 2023 were col-lected.The histological morphology,immunophenotype and mo-lecular characteristics of 8 ASLTs were evaluated by HE stai-ning,immunohistochemistry and FISH.Clinicopathological fea-tures of 8 ASLTs were analyzed with literature review.Results Mixed distribution of spindle cells and adipocytes in different proportions was seen in all 8 cases,with no or mild cell atypia and no necrosis.Immunophenotypically,8 ASLTs were CD34 and S-100 positive to varying degrees,RB was negative,MDM2 and CDK4 were negative or focus positive,and Ki67 prolifera-tion index was low.FISH showed MDM2 amplification was nega-tive in 6 cases.Conclusion ASLT is a newly named benign adipose-derived tumor subtype with unique morphology,immu-nophenotype,and molecular characteristics.It has a good prog-nosis.Correct diagnosis can avoid clinical over-treatment.
2.Construction of multi-epitope gene of periodic Brugia malayi and its expression in eukaryotic cells

Objective:To construct a eukaryotic expression plasmid pcDNA3.1(+)-BmCPI502/BmGAPDH720 of periodic Brugia malayi cysteine protease inhibitor/glyceraldehyde 3-phosphate dehydrogenase (CPI502/GAPDH720) multi-epitope gene and observe its protein expression in Hela cells. Methods:Primers were designed according to the predicted gene sequences of T cell epitopes. The target gene fragment was amplified by reverse transcription (RT)-PCR using plasmid pGEM-T-CPI621 as template. The fragment was cloned into prokaryotic expression plasmid pET-28a(+) to construct prokaryotic expression plasmid pET28a(+)-BmCPI502. The BmCPI502 gene and BmGAPDH720 gene were amplified by RT-PCR, respectively. The gene fragments of pcDNA3.1(+), BmCPI502 and BmGAPDH720 were digested by double enzyme digestion and ligated with the target gene. The eukaryotic expression plasmid pcDNA3.1(+)-BmCPI502/BmGAPDH720 was constructed. The recombinant plasmid was transfected into Hela cells and verified by RT-PCR to obtain the desired target bands. The expression product was detected by sodium dodecyl sulfate polyacrylamide gelelectrophoresis (SDS-PAGE).Results:The recombinant plasmid pET28a(+)-BmCPI502 was obtained, and the 502 bp specific fragment was identified by enzyme digestion, which was in line with the expected value; the eukaryotic expression plasmid pcDNA3.1(+)-BmCPI502/BmGAPDH720 of the multi-epitope gene was successfully constructed, and the fragment size was in line with the expected value. The eukaryotic expression plasmid pcDNA3.1(+)-BmCPI502/BmGAPDH720 was transfected into Hela cells and the recombinant protein was stable expressed. SDS-PAGE analysis showed that the relative molecular weight ( Mr × 10 3) of the recombinant protein was about 50. Conclusions:The eukaryotic expression plasmid of pcDNA3.1(+)-BmCPI502/BmGAPDH720 of periodic Brugia malayi multi-epitope gene is successfully constructed, and the corresponding recombinant protein is obtained in eukaryotic cells. This study has laid a foundation for further study of the purification and biological activity of the recombinant protein.
3.A case of peripartum cardiomyopathy with recurrent ventricular fibrillation rescued by extracorporeal membrane oxygenation

Peripartum cardiomyopathy (PPCM) often develops in the early postpartum period, especially within one month after delivery. However, in recent years, some cases of pregnancy-related cardiomyopathy with earlier or slightly later onset showed similar features with PPCM entirely. One patient with PPCM was admitted to the Department of Intensive Care Medicine of the First Affiliated Hospital of Bengbu Medical College on February 2022. After admission, the patient was exceptionally hemodynamically unstable and had repeated ventricular fibrillation. Apart from extracorporeal membrane oxygenation (ECMO) support, comprehensive treatment with cardiotonic agents and drugs to inhibit the "ventricular electric storm" was given. The cardiac structure and function were gradually improved. ECMO was successfully withdrawn on the 7th day after the onset of symptoms, and the patient was discharged after a full recovery. Early diagnosis and active intervention are the keys to improve the prognosis of patients with PPCM and avoid irreversible changes in cardiac structure.
4.Establishment of an intelligent cervical vertebrae maturity assessment system based on cone beam CT data.

To establish an intelligent cervical vertebra maturity assessment system, and to evaluate the reliability and clinical value of the system. Sixty children aged were recruited in the study. Lateral cephalometric radiograph and cone beam CT (CBCT) were taken at the same period. Based on the CBCT data, the system automatically extracted the patient's facial area through Otsu's method, intercepted the sagittal plane by three-dimensional least squares method, captured the second to fourth cervical vertebrae by superpixel segmentation. And then selected points were marked automatically through morphological algorithm and manual method. Consistency test was performed on the two sets of data to compare the reliability of automated cervical morphology capture. According to the parameters of morphological identification, positioning and staging algorithms were designed to form the intelligent cervical vertebra maturity assessment system. The cervical vertebra maturity was also judged manually on the lateral cephalometric radiograph. The weighted Kappa test and the Gamma correlation coefficient were subsequently applied to evaluate the consistency and correlation. The results showed that the cervical vertebra features automatically captured based on CBCT data had a high accuracy on the overall morphological recognition. In the prediction of 8 inflection points out of 13 points, there was no significant difference between automatic and manual method on both X and Y axes (all >0.05). The assessment results of the cervical vertebra maturity of the intelligent system had strong consistency and correlation with the manual recognition results (weighted Kappa value=0.877, Gamma value=0.991, both <0.05). The intelligent cervical vertebrae maturity assessment system based on CBCT data established in this study presents reliable outcome and high degree of automation, indicating that the system may be used clinically.

ABSTRACT:Objective To investigate the effects of the holistic health education system in patients after coronary stent.Methods A total of 720 patients after coronary stent were randomly divided into observation group with 360 cases and control group with 360 cases,the control group was conducted health education at admission and discharge,observation group were carried out whole-course tracking health education based on the control group.Drug therapy compliance, master of secondary prevention knowledge,mastering related diseases,nursing satisfaction,read-mission rates and mortality were compared between the two groups.Results Drug therapy com-pliance,master of secondary prevention knowledge,mastering related diseases,nursing satisfaction in the observation group were better than that in the control group(P <0.05);Readmission rates and mortality were lower than that in the control group (P <0.05).Conclusion Implementation of holistic health education system management in the patients after coronary stent can reduce pa-tient′s risk factors,improve the quality of life,increase the rate of postoperative rehabilitation, strengthen effective drug therapy compliance,correct patient′s bad living habits,reduce readmis-sion rates,and improve nursing satisfaction,so it is worthy of clinical promotion.

8.A survey and analysis of oropharyngeal microbial colonization in elderly patients

Objective To investigate the oropharyngeal microbial colonization in elderly patients.Methods Totally 618 elderly cases and 96 young and middle-aged cases were involved in this investigation.The colonization of bacteria and fungi on the oropharyngeal mucosa were obtained by throat swab cultures.Results There were 85.4% of cases (82 cases) with Streptococcus viridans and Neisseria gonorrheae on the oropharynx mucosa in young and middle-aged group and the pattern of constitute was simple.3 to 5 bacterial species were isolated from the oropharyngeal mucosa in elderly group.There were only 25.7% of cases (159 cases)with Streptococcus viridans and Neisseria gonorrheae on the oropharyngeal mucosa in elderly group.Biodiversity of the constitution pattern in the bacterial colonization was exhibited in the elderly.The colonization rate of Gramnegative bacilli was higher in elderly group than in young and middle-aged group [53.1% (328/618)vs.6.3% (6/96)].Staphylococcus aureus and Streptococcus pneumonia were the common Grampositive bacteria colonization in the elderly.The colonization rate of Candida fungus was 9.1 % (56/618) in the elderly.Conclusions The reduction of commensal bacteria,especially Streptococcus viridans may be the pathological basis of mode changes in bacteria colonization and opportunistic bacteria colonization on oropharyngeal mucosa in the elderly.The colonization rate of oropharyngeal Gram-negative bacili is obviously increased and Klebsiella Pneumoniae is the common bacteria on oropharyngeal mucosa in the elderly.Enterobacteriaceae,Pseudomonas aeruginosa and Acinetobacter baumannii are sensitive to common antibiotics.
9.Construction of periodic Brugia malayi cysteine protease inhibitor and glyceraldehydes-3-phosphate dehydrogenase eukaryotic recombinant plasmid and its immunogenicity

ObjectiveTo construct the eukaryotic expression plasmids containing cysteine protease inhibitor (CPI) and glyceraldehydes-3-phosphate dehydrogenase (GAPDH) gene from periodic Brugia malayi (Bm),and to observe its cellular immune response in mouse.Methods pcDNA3.1 (+)-BmCPI/BmGAPDH was constructed.The recombinant plasmids were screened and identified by digestion with restriction enzyme.BALB/c mice were injected intramuscularly with a dosage of 100 μg purified recombinant plasmid DNA with GpG oligodeoxynucleotide (CpG ODN) and two same doses were administrated at 2-week intervals.pcDNA3.1 (+) and phosphate buffered solution (PBS) were used as controls.The tissue of muscles at 4 weeks after the third injection was collected and the target gene was detected by reverse transcription-polymerase chain reaction (RTPCR).Two weeks after the third immunization,the stimulation index (SI) of spleen lymphocytes of immunized mice was measured by methylthiazolyldiphenyl-tetrazolium bromide (MTT) method and the serum levels of interleukin (IL)-4 and interferon (IFN)-γ were detected by enzyme-linked immunosorbent assay (ELISA).The data were analyzed by t test.ResultsBmCPI/BmGAPDH gene in the injected muscle of the immunized mice was detected by RT-PCR. At 6 weeks after immunization,the SIot spleen T lymphocytes in pcDNA3.1 (+)-BmCPI/CpG group and pcDNA3.1 (+)-BmCPI/BmGAPDH/CpG group were 1.466 ± 0.635 and 1.610 ± 0.112,respectively,which were both higher than PBS group and pcDNA3.1( +)-CpG group (1.004 ± 0.019 and 1.078 ± 0.129,respectively) (t=64.438,45.318,42.749 and 34.314,respectively; all P<0.05).At 4 weeks after immunization,the serum levels of IL-4 and IFN-γ of mice in pcDNA3.1 ( + )-BmCPI/BmGAPDH/ CpG group were significantly higher than those in pcDNA3.1 (+)-CpG group (t=288.053 and 76.453,respectively; both P<0.05),while the serum level of IFN-γ was also higher than that in pcDNA3.1 (+)-BmCPI/CpG group (t=129.642,P<0.05). ConclusionThe recombinant eukaryotic plasmid pcDNA3.1 (+)-BmCPI/BmGAPDH could be expressed in mice,and could elicit specific cellular immune responses in immunized mice.
10.Effects of endothelial progenitor cell combined with allogeneic bone marrow transplantation on hematopoietic reconstitution in mice

Objective To explore a proper dose of endothelial progenitor cells (EPCs)administration that can achieve optimal hematopoietic improving effectiveness in a murine allogeneic hernatopoietic stem cell transplantation (allo-HSCT) model.Methods Female Balb/c mice were lethally irradiated with 60Co source,and then were injected intravenously with 5 106 bone marrow cells from C57BL/6 mice (bone marrow transplantation group).In co-transfer experiments,5 × 104,1 ×105,5 × 105 or 1 × 106 donor EPCs (EPCs treated groups) were injected simultaneously with bone marrow cells.The recipients were monitored for survival,peripheral white blood cells,hematopoietic stem cells (HSCs) and bone marrow histology.Results Compared with bone marrow transplantation group,all EPCs treated groups had accelerated recovery of peripheral white blood cells (P<0.05),platelets (P<0.05) and HSCs (P<0.05).When infused with less than 5 × 105 EPCs,these effective hernatopoietic improving phenomena showed a positive correlation with the administrated doses of EPCs.However,when infused with 1 × 106 EPCs,the mice showed lower survival rate (P<0.05)and slower recovery of peripheral white blood cells (P<0.05),platelets (P<0.05) and HSCs (P<0.05) than 5 × 105 EPCs treated grpup.Bone marrow histopathology analysis confirmed the above findings.Conclusion Co-transfer with donor EPCs can improve survival rate and hematopoietic reconstitution of recipient mice in allo-HSCT,and 5 × 105 EPCs should be a proper dose to achieve the best effectiveness.
