Objective To investigate the mechanism of E26 transformation-specific sequence 4(ETV4)affecting the proliferation and migration of colon cancer cells through the nuclear factor-κB(NF-κB)signaling pathway.Methods The expression level of ETV4 in normal colon tissues and cancer tissues was analyzed by the user-friendly interactive cancer transcriptome data analysis resource(UALCAN)database.Reverse transcription quantitative polymerase chain reaction(qRT-PCR)and Western blot were used to detect the expression level of ETV4 in normal intestinal epithelial cells and colon cancer cell lines.After silencing ETV4 in SW480 cells,qRT-PCR and Western blot were per-formed to detect the expression of ETV4 to assess transfection efficiency;colony formation and Tran-swell assays were conducted to explore the effects of ETV4 silencing on the proliferation and migration of colon cancer cells;the Western blot was used to detect the effects of ETV4 silencing on the protein expression of protein 65(p65)and phosphorylated protein 65(p-p65)in the NF-κB pathway.Results The UALCAN database analysis revealed high expression of ETV4 in colon cancer tissues.The qRT-PCR and Western blot showed that ETV4 expression was significantly higher in the colon cancer cell lines SW480,Lovo,Caco-2,and SW620 than in normal intestinal epithelial cells HIEC-6,with the highest expression in SW480 cells(P＜0.001).Colony formation and Transwell assay results indicated that silencing ETV4 significantly inhibited the proliferation and migration of colon cancer SW480 cells(P＜0.001).Western blot results showed that silencing ETV4 significantly inhibited the expression of p-p65 protein in the cells(P＜0.001).Conclusion Silencing ETV4 may inhibit the activation of the NF-κB signaling pathway,thereby inhibiting the proliferation and migration of colon cancer cells.

Objective:To study the effects of various parameters in the structural design and usage methods of insulin pen needles on their performance.Methods:Twenty-one conventional needles and five self-destructive needles were selected. A testing machine was used to clamp the needle and make it vertically puncture the test material with a constant speed of 50, 100, 150, 200, and 250 mm/min, respectively. The influence of the needle insertion speed on the puncture force was analyzed. The deformation of the needle on the small contact surface and the large contact surface at 4 N was recorded by a Photron high-speed digital camera. The penetration depth at 1 N and 3 N was measured by an indirect measurement and a direct measurement method, respectively, and the maximum damage width of the damage area was recorded. The tilt resistance of the needle at 6°, 9° and 12° under 2 N pressure was obtained by the testing machine. SPSS 27.0 software was used for statistical analysis of the experimental data, and the LSD multiple comparison method was used for one-way analysis of variance.Results:The difference in puncture force between the needle insertion speed of 50 mm/min and the other four needle insertion speeds was statistically significant (all P < 0.05), while the differences between the other groups were not statistically significant (all P > 0.05). The small contact surface needle was significantly deformed, with a large penetration depth, a large maximum damage width, and low anti-tilting resistance. The large contact surface needle was deformed slightly, with a smaller penetration depth, a smaller maximum damage width, and greater resistance to tilting. Conclusions:The influence of the usage method on the large contact surface of the needle is relatively small. The hexagonal and cross-shaped needle seats have stronger anti-tilt ability than the circular needle seat. The hexagonal needle seat is not easy to change with the rotation axis, and the performance is optimal. In the structural design, the size of the connection part should be reduced, and the appropriate shape of the needle base should be selected.

Objective: To explore the effect of individualized nutrition intervention mode based on dysphagia screening in postoperative patients with cerebellopontine angle occupying lesion. Methods: By developing nurses training, selecting special screening and evaluation tools, developing screening methods and individualized nutrition intervention measures and meal spectrum, making screening and intervention flow chart, and starting to be implemented in postoperative patients with cerebellopontine angle occupying lesion in July 2017. Forty-six patients with cerebellopontine angle occupying lesion in the previous year were reviewed as the control group, who were given routine treatment and nursing; One year after implementation, Another 48 patients were set as the experimental group, and were given individualized nutritional care based on screening of dysphagia. Results: The incidence rate of dominant aspiration(0), pneumonia (4.17%, 2/48) and diarrhea (2.08%,1/48) in the experimental group was lower than 4.35% (2/46), 21.74% (10/46), 19.57% (9/46) in the control group, especially the difference of incidence rate of pneumonia and diarrhea was statistically significant (pneumonia: χ²=0.010, P=0.013; diarrhea: χ²=0.006, P=0.007). The retention rate of gastric tube in the experimental group (31.30%, 18/48) was lower than that in the control group (58.70%, 27/46), the difference was statistically significant (χ²=7.158, P=0.007). Average retention time of gastric tube in the experimental group (4.47±1.13) d was less than that in the control group (5.11±0.70) d, the difference was statistically significant (t=2.296, P=0,027). The hospitalization time in the experimental group (8.69±1.36) d were less than those in the control group (12.57±2.95) d, the difference was statistically significant (t=8.248, P=0,000). The nutritional status of 7 days after operation of the experimental group was better than that of the control group, the differenc e was statistically significant (albumin: t=4.888, P<0.01; prealbumin: t=5.188, P<0.01; hemoglobin: t=4.039, P<0.01). The knowledge and skills of swallowing of general nurses improved significantly after the work, in particular, the accuracy of screening tests for Wa Tian drinking water increased from 5/8 to 20/20. The difference was statistically significant (χ²=8.148, P=0.017). Conclusions Individualized nutrition intervention based on screening for dysphagia can improve the clinical outcome of patients, improve the comprehensive ability of nurses and achieve a win-win situation of nurse - patient.

Objective To analyze the clinical and pathological data and prognosis of microalbuminuria IgA nephrology patients with decreased serum C3 level, and investigate the significance of decreased serum C3 level in microalbuminuria IgA nephrology patients. Methods Clinical and pathological data of microalbuminuria IgA nephrology patients confirmed by renal biopsy and followed up more than 6 months were reviewed. The patients were divided into decreased serum C3 level group (34 cases, 25.19%) and normal serum C3 level group (101 cases, 74.81%) according to the serum C3 level. Twenty-four hours urine protein quantitative > 1 g, or normal serum creatinine level turning into abnormal level at renal biopsy, or doubling of serum creatinine level was defined as the end point of follow-up. Renal survival was calculated by Kaplan-Meier survival analysis and risk factors of progression were analyzed by Cox regression models. Results Total of 135 microalbuminuria IgA nephrology patients were followed up successfully, with an average follow-up time (53.4 ± 21.9) months. There were 27 cases (79.41%) and 32 cases (31.68%) in the decreased serum C3 level group and the normal serum C3 level group respectively at the endpoint. Kaplan-Meier survival analysis showed that the median survival time was significantly shorter in decreased serum C3 level group compared with that in normal C3 level group: (46.7 ± 9.1) months vs. (68.4 ± 9.9) months, P =0.014. Cox regression analysis showed that abnormal serum creatinine (RR = 1.147, 95% CI: 1.129-1.395, P = 0.008), decreased serum C3 level (RR=1.028, 95%CI:0.672-1.495, P=0.039), urine protein quantitative>1 g/24 h (RR=2.066, 95%CI:1.242-3.838, P=0.006) and renal biopsy pathological indicators Lee classⅢ-Ⅴ(RR=2.820, 95%CI:1.249-5.638, P=0.041), glomerular sclerosis or adhesions (RR=1.232, 95%CI: 1.065-1.520, P = 0.040), renal interstitial atrophy or interstitial fibrosis (RR = 2.604, 95% CI:1.748- 4.104, P = 0.037), endocapillary cell proliferation (RR = 0.872, 95% CI: 0.491- 1.275, P =0.042), crescentic (RR = 1.528, 95% CI: 1.073- 2.385, P = 0.009) affected prognosis of microalbuminuria IgA nephropathy as the independent risk factors. Conclusions The clinical and pathological features in patients of microalbuminuria IgA nephropathy with decreased serum C3 level is more severe, and the prognosis is poor. The patients should be followed up closely and early intervention treatment and early active control of microalbuminuria should be done at the same time.

Objective To characterize the static balance and limits of stability (LOS) of postmenopausal women and evaluate the relationship between bone mineral density (BMD) and balance.Methods Sixty women more than 12 months past menopause and aged 50 to 60 years were included in the study.Group 1 (n =20) consisted of women with normal BMD,group 2 (n =20) women with osteopenia,and group 3 (n =20) women with osteoporosis.The static balance of all the participants was evaluated in upright postural situations for 60 s with the eyes open and with the eyes elosed.Their LOS was quantified using a force platform.Results There was no significant difference in static balance parameters or LOS among the 3 groups.The static balance parameters were not significantly different in either the eyes-open or eyes-closed situation in the osteoporosis and osteopenia groups.The medial and lateral LOS were greater than the anterior and posterior limits in all 3 groups.The anterior LOS was greater than the posterior limits among the women with osteopenia and normal density.Conclusion In postmenopausal women aged 50 to 60 years,BMD did not show any significant association with static balance or LOS.Postmenopausal women had better postural control in the mediolateral than in the anteroposterior direction.In postmenopausal women with osteoporosis,postural control in the anterior direction deteriorates from age 50 to 60.
