1.Mechanism of Xiezhuo Jiedu Prescription in Treatment of Ulcerative Colitis by Inhibiting Ferroptosis and Alleviating Intestinal Mucosal Injury Based on Nrf2/SLC7A11/GPX4 Signaling Pathway

ObjectiveTo investigate the mechanism of Xiezhuo Jiedu prescription in the treatment of ulcerative colitis (UC) by inhibiting ferroptosis and alleviating intestinal mucosal injury based on the nuclear factor E2 related factor 2/solute carrier family 7 member/glutathione peroxidase 4 (Nrf2/SLC7A11/GPX4) signaling pathway. MethodsA total of 60 male SD rats were divided into a normal group, a model group, high- and low-dose Xiezhuo Jiedu prescription groups (26.64 and 13.32 g·kg-1, respectively), a ferroptosis inhibitor group (Ferrostatin-1, 0.005 g·kg-1), and a mesalazine group (0.27 g·kg-1), with 10 rats in each group. A UC rat model was established by intrarectal administration of trinitrobenzene sulfonic acid (TNBS)-ethanol. The normal group and the model group were intragastrically administered normal saline. The other groups were given intragastric administration according to the corresponding dosage for 7 d. The general condition, disease activity index (DAI) score, colon length, and mucosal injury index (CDMI) score were observed in each group. The pathological changes of colon tissue in each group were observed by hematoxylin-eosin (HE) staining. The intestinal mucosa and mitochondrial morphology in each group were observed by transmission electron microscopy. The expression levels of Occludin, Claudin-1, mucin 2 (MUC2), and E-cadherin in intestinal tissue were detected by immunofluorescence (IF). Enzyme-linked immunosorbent assay (ELISA) was used to detect the expression levels of serum tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and interleukin-10 (IL-10) in each group, and a lactic acid assay kit or ELISA was employed to detect the expression levels of reactive oxygen species (ROS), ferrous ions (Fe2+), glutathione (GSH), malondialdehyde (MDA), 4-hydroxynonenal (4-HNE), diamine oxidase (DAO), and D-lactate (D-LA). Real-time quantitative polymerase chain reaction (Real-time PCR) was applied to detect the mRNA expression levels of Nrf2, SLC7A11, GPX4, Occludin, Claudin-1, MUC2, and E-cadherin in each group, and Western blot was adopted to detect the protein expression levels of Nrf2, p-Nrf2, SLC7A11, and GPX4 in each group. ResultsCompared with the normal group, rats in the model group exhibited listlessness, sluggish response, and mucopurulent and bloody stools. The model group also showed significantly increased DAI score, colon length, CDMI score, and expression levels of TNF-α, IL-6, ROS, Fe2+, MDA, 4-HNE, DAO, and D-LA (P<0.01). In addition, it presented significantly decreased IF values of Occludin, Claudin-1, MUC2, and E-cadherin and mRNA and protein expression levels of IL-10, GSH, Nrf2, p-Nrf2, SLC7A11, and GPX4 (P<0.01). There were different degrees of improvement in each administration group after treatment, and the improvement was the most significant in the high-dose Xiezhuo Jiedu prescription group (P<0.01). ConclusionXiezhuo Jiedu prescription may alleviate intestinal mucosal injury by inhibiting ferroptosis of intestinal epithelial cells via regulating the Nrf2/SLC7A11/GPX4 signaling pathway, thereby exhibiting efficacy in the treatment of UC.
2.Association of insulin-like growth factor binding protein-3 gene polymorphism at the rs10225396 locus with susceptibility to hepatocellular carcinoma

ObjectiveTo investigate the association of the single nucleotide polymorphism (SNP) of the insulin-like growth factor binding protein-3 (IGFBP3) gene at rs10225396 (A>G) locus with the susceptibility to hepatocellular carcinoma, as well as its potential molecular regulatory mechanisms, and to provide novel genetic biomarkers and a theoretical basis for early screening and precise targeted therapy for hepatocellular carcinoma. MethodsA total of 192 patients with hepatocellular carcinoma who were admitted to The Affiliated Hospital of Yanbian University and Yanbian Cancer Hospital from January 2009 to August 2016 were enrolled as experimental group, and 190 healthy individuals who underwent physical examination in Physical Examination Center of Yanbian Hospital during the same period of time were enrolled as control group. Peripheral blood samples were collected from all subjects, and after DNA was extracted from whole blood, the DNA samples meeting quality standards were sent to Beijing Genomics Institute Research Center Co., Ltd., for Mass ARRAY mass spectrometry, while genotyping was completed. The independent-samples t test was used for comparison of continuous data between two groups, and the chi-square test was used for comparison of categorical data between two groups. A binary Logistic regression model was used to analyze the association of the SNP of the IGFBP3 gene at rs10225396 locus with the susceptibility to hepatocellular carcinoma, and odds ratio (OR) and 95% confidence interval (CI) were calculated to assess the risk of developing hepatocellular carcinoma in individuals carrying different genotypes. ResultsThere were two alleles (G and A) at the rs10225396 locus of the IGFBP3 gene, yielding the three genotypes of AA, AG, and GG, and its genotype distribution was consistent with the Hardy-Weinberg equilibrium (χ²=0.072, P=0.789). The stratified genetic analysis showed that carriers of the IGFBP3 rs10225396 AA genotype had a significant increased risk of hepatocellular carcinoma among individuals of age <63 years, male sex, smoking history, drinking history, and the Chinese Korean population (all P<0.001). The binary Logistic regression analysis showed that after adjustment for related risk factors in the codominant model, the population with genotype AG and GG had a significant reduction in the risk of hepatocellular carcinoma compared with the population with genotype AA (P<0.001), and in the dominant model, the population with genotype AG+GG had a significant reduction in the risk of hepatocellular carcinoma compared with the population with genotype AA (P<0.001). ConclusionThrough a genotyping analysis of hepatocellular carcinoma patients and healthy individuals in Yanbian Korean Autonomous Prefecture of Jilin Province, China, this study shows that genotype AA at rs10225396 locus of the IGFBP3 gene is significantly associated with the susceptibility to hepatocellular carcinoma, and this genotype can significantly increase the risk of developing hepatocellular carcinoma with the presence of specific risk factors, which provides a potential genetic marker for early screening and precise treatment of hepatocellular carcinoma.

Objective:To evaluate the value of ultrasound radiomics combined with machine learning for early diagnosis of seronegative Hashimoto’s thyroiditis (SN-HT) .Methods:This retrospective study included 164 patients from Liaoning Provincial People’s Hospital , Lixin County People’s Hospital, Linghai Dalinghe Hospital, Fengcheng Phoenix Hospital, who underwent thyroidectomy for solitary nodules with normal thyroid function between Nov. 2016 and Jan. 2024. Postoperative pathology confirmed Hashimoto’s thyroiditis (HT) in some cases, who were further categorized into antibody-positive and antibody-negative groups based on serum antibody status. Patients without Hashimoto’s thyroiditis served as the control group. A total of 298 ultrasound images were analyzed. Radiomics features were extracted from hypoechoic non-nodular areas within 0.5 cm surrounding the tumor. Two senior pathologists and two senior ultrasound physicians independently assessed lymphocytic infiltration, eosinophilic changes of follicular epithelium, and the proportion of hypoechoic areas in pathology and ultrasound images, respectively. A machine learning model, CCH-NET, was developed using linear regression and t-distributed stochastic neighbor embedding (t-SNE) techniques. The dataset was divided into a training set (80%) and a validation set (20%) to compare the diagnostic accuracy of CCH-NET with that of senior ultrasound physicians. Results:In internal validation, CCH-NET achieved a diagnostic accuracy of 88.89% for both antibody-positive and antibody-negative groups, significantly higher than the 66.67% accuracy of senior ultrasound physicians ( P<0.01). In external validation, CCH-NET achieved 75.00% and 66.67% accuracy for the two groups, compared to 50.00% by senior ultrasound physicians. For the control group, both methods achieved 93.33% accuracy. The AUC of CCH-NET was 0.848, outperforming senior ultrasound physicians (0.681) ,demonstrating superior diagnostic performance. Conclusion:The radiomics-based CCH-NET model, using non-nodular hypoechoic areas as a specific indicator, can accurately identify early SN-HT in euthyroid patients. It significantly outperforms senior ultrasound physicians, improving diagnostic accuracy and reducing missed diagnoses.
4.Huanglian Ganjiang Decoction regulates mitochondrial apoptosis through regulation of NF-κB/CXCL1/CXCR2 pathway to improve symptoms in ulcerative colitis mice

Objective To explore the mechanism of Huanglian Ganjiang Decoction in regulating the mitochondrial apoptotic pathway through the NF-κB/CXCL1/CXCR2 pathway in ulcerative colitis(UC).Methods Sixty C57BL/6J mice were divided into normal group,model group,Huanglian Ganzhang Decoction groups(low,medium and high doses)and mesalazine group.Except for the normal group,UC models were established in the other groups.The general conditions of the mice of all groups were recorded.HE staining was used to observe the pathological changes of the colon and rectum;electron microscopy was used to observe the mitochondrial condition;ELISA was used to determine the contents of IL-6,IL-10,D-lactic acid and DAO;Real-time PCR was used to detect the mRNA expression of NF-κB p65,CXCL1,CXCR2,Bcl-2 and Bax;Western blot was used to determine the protein expression levels of NF-κB p65,CXCL1,CXCR2,p-JAK2,p-STAT3,Bcl-2 and Bax.Results Compared with the normal group,the model group of mice showed a decline in condition,shortened length of the colon and rectum,and mitochondrial structural damage,furthermore,the levels of IL-6,DAO and D-lactic acid were increased,IL-10 level was decreased,the mRNA expression of NF-κB p65,CXCL1,CXCR2 and Bax were enhanced,Bcl-2 mRNA expression was weakened,the protein expression of NF-κB p65,CXCL1,CXCR2,p-JAK2,p-STAT3 and Bax were enhanced,Bcl-2 protein expression was weakened.Compared with the model group,the indexes mentioned above were reversed in the drug groups,especially in the high-dose group.Conclusion Huanglian Ganjiang Decoction may exert its therapeutic effect on UC in mice by regulating the NF-κB/CXCL1/CXCR2 pathway and mitochondrial apoptotic pathway.
5.Relationship between 24-hour movement behavior and anxiety in middle school students

Objective:To explore the relationship between 24-hour movement behavior and anxiety in middle school students.Methods:Totally 2 690 students from 8 middle schools in Hubei Province were selected as research subjects.The Health Behavior in School-aged Children,Pittsburgh Sleep Quality Index and Self-Rating Anxiety Scale were used to assess 24-hour movement behavior[i.e.,moderate to vigorous physical activity(MVPA),screen time(ST),and sleep duration(SD)]and anxiety symptoms.The binary logistic regression analysis was per-formed to examine the relationship between 24-hour movement behavior and anxiety for middle school students.Results:In middle school students,the compliance rate of 24-hour movement behavior was 4.2%,that of MVPA being 15.8%,that of ST being 27.1%,that of SD being 64.2%,and the detection rate of anxiety was 21.7%.The binary logistic regression analysis showed that,compared with the situation when three factors met with the 24-hour movement guidelines,the anxiety risk of middle school students increased when single factor(i.e.,MVPA,ST and SD)and some two factors(i.e.,MVPA+ST and MVPA+SD)met 24-hour guidelines,with odds ratios being respectively 2.62,2.22,2.80,2.62,2.52.Conclusion:The detection rate of anxiety in middle school students was probably negatively correlated with the compliance rate of 24-hour movement behavior.
6.Three-dimensional finite element analysis of factors affecting stability of anchorage in zygomatic alveolar ridge

Objective To investigate the influence of micro-implant length and screw angle on stress distribution and displacement of micro-implant bone interface when micro-implant was implanted in zygomatic alveolar ridge and bear a certain load,in order to provide certain theoretical basis for micro-implant selection and further optimization design in clinical practice.Methods A three-dimensional finite element model of the jaw in the zygomatic alveolar ridge region was established using ANSYS software.At the same time,35 kinds of micro-threaded implants with length of 8,9,10,11,12 mm and angle of 30°,45°,60°,75°,90°,105°,120° were estab-lished.The micro-implant was simulated to be implanted in the jaw bone,and the implantation model was established.Stress analysis was performed on the bone interface of the micro-implant.Results The maximum stress of micro-implant bone interface occurred in the bone cortex.The maximum stress value of buccal cortical bone,cancellous bone and micro-implant displacement decreased with the increase of screw length.The angle of screw had little effect on the maximum stress and displacement of micro-implant-bone interface.Conclusion The screw length has an impact on stress distribution of micro-implant bone interface.The micro-implant with longer length can improve its stability in zygomatic alveolar ridge.
7.Advances in Etiology of High Incidence of Thyroid Cancer in Women

Thyroid cancer is the only non reproductive tumor that mainly occurs in women.The morbidity rate of female thyroid cancer is three times higher than that of male thyroid cancer.However,the ex-isting studies have not systematically summarized the factors that contribute to the high incidence of thyroid cancer in women.This paper reviews all the literatures that mention the high incidence of thyroid cancer in women,and sorts out the main reasons,such as estrogen,obesity,reproductive factors,oxidation antioxidant system imbalance,X chromosome,etc.,which are the main reasons for the current research finding that the in-cidence rate of thyroid cancer in women is higher than that in men.This paper makes a vivid graphic analysis and summary of the reasons,providing a research direction for future researchers to research on gender duality of thyroid cancer,and providing a certain reference for the clinical diagnosis and treatment of gender differences in thyroid cancer.
8.A multicentre retrospective study of house dust mite allergen preparation treating multi-sensitized allergic rhinitis patients

Objective:To investigate, for multi-sensitized allergic rhinitis (AR) patients allergic to dust mites combined with other allergens (pollen, mold, animal dander, etc.), whether the single dust mite subcutaneous immunotherapy (SCIT) can improve the specific symptoms caused by other allergens in the patients, and to analyze the relationship between the effectiveness of symptom improvement in these patients and the type, quantity and severity of the allergens.Methods:A multicenter retrospective study was conducted to collect mul-sensitized AR patients from allergy or respiratory departments of 5 hospitals who received house dust mite allergen preparation SCIT for 12 to 36 months and met other inclusion and exclusion criteria from February to July 2024. General clinical data were collected and the perennial or seasonal symptoms before and after treatment were evaluated with visual analogue scale (VAS) to assess whether there was an perennial or allergen-specific symptom improvement (VAS score decrease ≥30%), by which the patients were divided into effective group and ineffective. R software was used to analyze the differences between groups by using Fisher′s exact test and Mann-Whitney U test. Results:A total of 62 patients were enrolled, and the treatment were effective in 39 of them, with an effective rate of 62.9%. For allergen-specific symptoms, the median age of the effective group was higher than that of the ineffective group (12 years old vs. 8 years old, P=0.039), and the effective rate in dust mite specific immunoglobin E (sIgE) grade ≤5 group was higher than that in sIgE grade >5 group (81.6% vs. 45.5%, P=0.008), and the effective rate of mold sIgE grade ≤2 group was higher than that of sIgE grade >2 group (83.3% vs. 28.6%, P=0.045), and there was no statistically significant correlation between the other allergen grades and the effective rate ( P>0.05). For perennial symptoms, the effective rate in the mold grade ≤2 group was higher than that in the sIgE grade >2 group (91.3% vs. 28.6%, P=0.010), and there was no statistically significant correlation between the other allergen grades and the effective rate ( P>0.05). There was no significant correlation between the treatment effectiveness of perennial or allergen-specific symptoms and the number of combined allergens, the grade of skin test, and the difference between the grade of combined allergens and that of dust mites ( P>0.05). Conclusion:Among the patients with multi-sensitized AR allergic to dust mites included in this study, single dust mite SCIT is effective in some of them, and for allergen-specific symptoms, the effective group was elder, and dust mite sIgE grade 6 and mold sIgE grade ≥2 was related to the low effective rate of SCIT. The present results are insufficient for selecting single or multiple AIT in any type of multi-sensitized patients.
9.Clinical application and efficacy observation of a self-designed transpedicular intravertebral bone grafting device in percutaneous pedicle screw fixation for thoracolumbar fractures

Objectives:To investigate the application effect of a self-designed transpedicular intravertebral bone grafting device in percutaneous pedicle screw fixation for thoracolumbar fractures,and to evaluate its safety and the role in preventing the"shell"phenomenon.Methods:A retrospective analysis was performed on 49 patients with thoracolumbar fractures who underwent percutaneous pedicle screw fixation combined with intravertebral bone grafting at our hospital from January 2018 to January 2023.Among them,25 patients were treated with a self-designed transpedicular intavertebral bone grafting device[Self-designed device group:8 males,17 females,aged 48.0±9.7 years;Body mass index(BMI)21.6±2.7kg/m2;T11 3 cases,T12 9 cases,L17 cases,L2 6 cases;Thoracolumbar injury classification and severity(TLICS)score 4.3±0.5;AO classification:A3 18 cases,A4 7 cases].The other 24 patients were treated with a conventional technique bone grafting funnel(Conventional technique group:11 males,13 females,aged 48.2±10.7 years;Body mass index 21.4±2.7kg/m2;T11 6 cases,T12 6 cases,L1 8 cases,L2 4 cases;TLICS score 4.3±0.5;AO classification:A3 17 cases,A4 7 cases).There were no statistically significant differences between the two groups in age,gender ratio,body mass index,fracture level,AO classification,or TLICS score(P>0.05).The total operativetime,graft implantation time,fluoroscopy frequency during bone grafting,intraoperative blood loss,hospital stay,and in-traoperative complications were compared between the two groups.Low back pain was assessed using visual analogue scale(VAS)score,lumbar function was evaluated using Oswestry disability index(ODI)and Japanese Orthopaedic Association(JOA)score preoperatively,at 3 months and 1 year postoperatively.The local kyphotic angle(LKA)and anterior vertebral height ratio(AVHR)were measured and compared preoperatively,immediately postoperatively,at 3 months and 1 year postoperatively.Immediate postoperative CT scan was performed to determine bone graft distribution,assess iatrogenic injuries to the pedicle or anterior vertebral wall,and iden-tify the"shell"phenomenon.Follow-up CT at 1 year postoperatively was performed to evaluate bony fusion and the outcomes of the"shell"phenomenon.Results:All the patients successfully completed the surgery and were followed up for over 12 months(17.0±3.1 months).The self-designed device group was significantly lower in fluoroscopy frequency during bone grafting(11.0±1.5 vs.13.5±1.7,P<0.05)and longer in graft implan-tation time(15.0±1.7min vs.12.7±1.9min,P<0.05)than the conventional technique group,but there was no significant difference in total operative time between the two groups(P>0.05).In the conventional technique group,3 cases(12.5%)had pedicle injuries,9 cases(37.5%)had anterior vertebral wall injuries,and 12 cases(50.0%)had bone graft material leakage;In contrast,the self-designed device group had no pedicle or ante-rior wall injuries,and there were 4 cases(16%)of bone graft leakage.The difference in complication rates be-tween the two groups was statistically significant(P<0.05).Both groups showed significant improvement in LKA and AVHR immediately postoperatively,at 3 months,and at 1 year compared to preoperative values(P<0.05).VAS score,ODI,and JOA score also significantly improved at 3 months and 1 year postoperatively compared to preoperative scores(P<0.05).The"shell"phenomenon occurred in 5 cases in the self-designed device group,with 1 case healed at the 1-year follow-up,which occurred in 8 cases in the conventional technique group,with 2 cases healed at 1 year,and the difference was no statistically significant(P>0.05).Conclusions:The application of the self-designed minimally invasive transpedicular vertebral bone grafting device in percu-taneous pedicle screw fixation combined with vertebral bone grafting for thoracolumbar fractures can reduce intraoperative fluoroscopy frequency and complications,therefore to improve surgical safety.

For the control group, both methods achieved 93.33% accuracy. The AUC of CCH-NET was 0.848, outperforming senior ultrasound physicians (0.681) ,demonstrating superior diagnostic performance. Conclusion:The radiomics-based CCH-NET model, using non-nodular hypoechoic areas as a specific indicator, can accurately identify early SN-HT in euthyroid patients. It significantly outperforms senior ultrasound physicians, improving diagnostic accuracy and reducing missed diagnoses.
