1.Actions and mechanism of argon-helium cryotherapy in inflammatory response in rats with lower extremity atherosclerotic occlusive disease

Objective To investigate the mechanism of argon-helium cryotherapy in regulating the tyrosine-protein kinase 2/signal transducer and activator of transcription 3(JAK2/STAT3)path-way,thereby inhibiting the inflammatory response in rats with atherosclerotic occlusive disease(ASO)of the lower extremities.Methods Forty rats were randomly divided into sham group,lower extremity ASO group,Ar-He Cryo group and Ar-He Cryo+p-JAK group,with 10 rats in each group.The average blood flow velocity of the lower extremity artery,lipid profile and inflammation levels were measured in each group.Hematoxylin-eosin(HE)staining was used to examine histopathologi-cal changes in arterial vessel tissues.Western blotting was performed to detect the expression levels of JAK2,p-JAK2,STAT3,p-STAT3 and suppressor of cytokine signaling 3(SOCS3)proteins in arterial vessel tissues.Results The average blood flow velocity of the lower extremity artery in the lower extremity ASO group was significantly lower than that in the sham group(P<0.05).The average blood flow velocity in the Ar-He Cryo group was significantly higher than that in the ASO group(P<0.05).The average blood flow velocity in the Ar-He Cryo+p-JAK group was significantly lower than that in the Ar-He Cryo group(P<0.05).Serum total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C)levels,tumor necrosis factor-alpha(TNF-α),interleukin-1 β(IL-1β),interleukin-6(IL-6)levels and endothelin-1(ET-1)content in the lower extremity ASO group were significantly higher than those in the sham group,while serum high-densi-ty lipoprotein cholesterol(HDL-C)levels and nitric oxide(NO)content were significantly lower than those in the sham group(P<0.05).In the Ar-He Cryo group,serum TC,TG,LDL-C levels as well as TNF-α,IL-1β,IL-6 levels and ET-1 content were significantly lower than those in the lower extremity ASO group,while serum HDL-C levels and NO content were significantly higher(P<0.05).In the Ar-He Cryo+p-JAK group,serum TC,TG,LDL-C levels as well as TNF-α,IL-1 β,IL-6 levels and ET-1 content were significantly higher than those in the Ar-He Cryo group,while HDL-C levels and NO content were significantly lower(P<0.05).Compared with the sham group,the lower extremity venous vessels in the lower extremity ASO group showed significant steno-sis;compared with the lower extremity ASO group,the degree of stenosis in the lower extremity ve-nous vessels was significantly reduced in the Ar-He Cryo group;the degree of stenosis in the lower extremity venous vessels in the Ar-He Cryo+p-JAK group was similar to that in the lower extremity ASO group and significantly more severe than that in the Ar-He Cryo group.The ratios of p-JAK2/JAK2 and p-STAT3/STAT3 in arterial vessel tissues of the lower extremity ASO group were signifi-cantly higher than those in the sham group,and SOCS3 protein expression was significantly lower than that in the sham group(P<0.05).In the Ar-He Cryo group,p-JAK2/JAK2 and p-STAT3/STAT3 in arterial vessel tissues were significantly lower than those in the lower extremity ASO group,and SOCS3 protein expression was significantly higher than that in the lower extremity ASO group(P<0.05).In the Ar-He Cryo+p-JAK group,p-JAK2/JAK2 and p-STAT3/STAT3 in arte-rial vessel tissues were significantly higher than those in the Ar-He Cryo group,and SOCS3 protein expression was significantly lower than that in the Ar-He Cryo group(P<0.05).Conclusion Ar-gon-helium cryotherapy can reduce lipid deposition in the arteries of rats with lower extremity ASO,inhibit inflammatory responses,and its mechanism may be related to the inhibition of activation of JAK2/STAT3 signaling pathway.

3.Optimization of the Best Proportion of Astragalus membranaceus Injection Combined with Erigeron breviscapus Injection against Cerebral Ischemia-reperfusion Injury in Rats by Baseline Geometric Proportion Increasing and Decreasing Design

OBJECTIVE: To screen the best proportion of Astragalus membranaceus injection combined with Erigeron breviscapus injection against cerebral ischemia-reperfusion injury in rats. METHODS: Male SD rats were randomly divided into sham operation group, model group and administration group [different A. membranaceus injection-E. breviscapus injection proportion groups, being A(0 ∶ 10), B(2 ∶ 8), C(4 ∶ 6), D(6 ∶ 4), E(8 ∶ 2), F(10 ∶ 0)groups, set by baseline geometric proportion increasing and decreasing design], with 8 rats in each group. Except for sham operation group, reperfusion injury model of middle cerebral artery occlusion were induced by modified suture method in rats. The each administration group was given relevant medicine intraperitoneally once immediately after inducing model, and then given again after 24 hours (medication interval between the two injections of 30 min). Constant volume of normal saline was given to rats in sham operation group and model group. Forty-eight hours after reperfusion, Longa scoring method was used to evaluate neurological impairment of rats, and neurological impairment score was recorded. Serum content of MDA and activity of SOD were measured by colorimetry assay. TTC assay was used to detect cerebral infraction, and cerebral infarction rate was calculated. Kim’s formula was used to calculate the synergistic index (q) of rats in administration groups. RESULTS: Compared with sham operation group, neurological impairment score and serum content of MDA were increased significantly in model group, while activity of SOD was decreased significantly (P<0.01). The area of cerebral infarction increased significantly, and the rate of cerebral infarction increased significantly (P<0.01). Compared with model group, neurological impairment scores and serum contents of MDA were decreased significantly in group A, B, C, D and E; neurological impairment score of group C was significantly lower than those of group A and F; serum contents of MDA in group B, C, D and E were significantly lower than that of group F (P<0.05 or P<0.01). Activities of SOD in group A, B, C, D and E were increased significantly, and group C was significantly higher than group F (P<0.05 or P<0.01). The cerebral infarction area of rats in each administration group was reduced to varying degrees. The cerebral infarction rates of rats in group B, C, D and E were significantly reduced, and group C and D were significantly lower than group F, while group C was significantly lower than group A (P<0.05 or P<0.01). The q values of group B, C, D and E were 0.90, 1.30, 1.00, 0.70 (neurological impairment score) and 0.79, 1.27, 0.98, 0.82 (cerebral infarction rate). CONCLUSIONS: Different ratios of A. membranaceus injection and E. breviscapus injection have certain protective effects on cerebral ischemia-reperfusion injury model rats, can relieve their neurological deficits, alleviate their oxidative stress and reduce their cerebral infarction areas. The effect of the combination of the two drugs is better than that of single use, and the optimum ratio is 4 ∶ 6.
4.ADC measurement in assessment of tumor grade and myometrial invasion of endometrial carcinoma

Objective To investigate the value of the apparent diffusion coefficient (ADC)measurement in assessment of tumor grade and myometrial invasion of endometrial carcinoma (EC).Methods 80 EC patients and 28 cervical cancer patients with normal endometrium were studied retrospectively.1.5T conventional MRI and DWI (b=0,1 000 s/mm2)were performed,and ADC values were calculated by two radiologists.Statistical analyses were performed by using SPSS 19.0 and Medcalc software.Results The mean ADC values (×10-3mm2/s)were 0.851±0.131,0.752±0.099,0.681±0.089 for G1,G2 and G3 EC,respectively.Significant statistical differences were achieved for the three groups (G1 vs G2:P=0.005;G2 vs G3:P=0.03;G1 vs G3:P< 0.000 1).For the prediction of G3,the area under the curve (AUC)of 0.851 and the cut-off value of ≤0.742×10-3mm2/s were identified,with the sensitivity, specificity and accuracy of 88.24%,76.19% and 85%,respectively.Conclusion There are significant statistical differences between histologic grades of EC.ADC measurement may have the potential to select G3 EC patients.
5.MRI characters of 52 cases of septate uterus

Magnetic resonance imaging (MRI) findings of 52 patients with septate uterus were retrospectively analyzed.There were 19 cases of complete septate uterus and 32 cases of partial septate uterus confirmed by hysteroscopy or hysteroscopy combined with laparoscopy,and 1 case without surgery.According to MRI findings,19 cases were diagnosed as complete septate uterus,32 cases as partial septate uterus and 1 case as intrauterine adhesion.The results indicated that MRIcan be used in diagnosis of septate uterus.
6.Carbachol alleviates intestinal inflammation during enteral fluid resuscitation of rats with burn shock

Objective To investigate the effect of carbachol on local gut inflammation during entetal resuscitation of rats with bum shock. Method Thirty-eight Wistar rats were subjected to 35%TBSA full thickness scald injury, and enteral fluid was infused into animal intestines via duodenal stomas 30 minutes post bum. The animals were randomly divided into four groups: no resuscitation (Control, n = 8), enteral resuscitation using either a glucose electrolyte solution (GES, n = 10) or GES plus carbachol (60 μg·kg-1,GES/CAR, n = 10), or carbachol alone (CAR, n = 10) .The volumeof GES infusion was based on the Parkland formula (4 ml· 1% TB-SA-1·Kg-1) - All animals were sacrificed 4 hours post bum, and specimens of jejunal tissue were collected to determine the levels of tumor necrosis factor (TNF)-α, nitric oxide (NO), nitric oxide synthase (NOS) and myeloperoxidase (MPO). Serum assays for plasma diamine oxidase (DAO) activities were also performed. Results There were no statistical differences in the intestinal levels of NOS, NO, TNF-α and MPO, and plasma OAO activities, between the GES group and the control group. Compared to the GES group, the GES/CAR group showed significantly lowered levels of intestinal NOS (1.276 ±0.391 vs. 1.818 ±0.436, P<0.05), NO (0.925 ±0.402 vs. 1.561 ±0.190, P < 0.05, TNF-α (0.87±0.13 vs. 1.94±0.47, P <0.01) and MPO (0.465 ±0.092 vs. 0.832±0.214, P<0.05),and reduction in plasma DAO activites (0.732±0.192 vs. 1.381 ±0.564, P <0.05). The CAR group also showed significantly lowered levels of intestinal NOS, NO, TNF-α and MPO and reduced plasma DAO activites, compared to the GES group. Conclusions Theses results suggest that carbachol significantly inhibits the release of proinflammatory mediator and attenuates local inflammation in gut during enteral fluid resuscitation of rats in rats with bum shock. We postulate that carbachol may exert its and-inflammatory effects via the cholinergic anti-inflammatory pathway.
