1.The regulation and mechanism of hyperthermia combined with iron overload on tumor-associated macrophages in oral squamous cell carcinoma

Objective:To observe the effect of hyperthermia combined with iron overload on the regulation of tumor-associated macrophage polarization and the migration ability of CAL-27 cells and explore its mechanism.Methods:Human monocytic leukemia cell THP-1 was induced and polarized into M2 macrophages. M2 tumor-associated macrophages and CAL-27 cells were divided into the control group (no intervention), hyperthermia group (incubated at 42 ℃ for 1 h, and then incubated at 37 ℃ for 24 h), ferric citrate group (added with 2.5 mg/ml ferric citrate, and cultured in an incubator at 37 ℃for 24 h) and hyperthermia + ferric citrate group (added with 2.5 mg/ml ferric citrate for 1 h, cultured in an incubator at 42℃ for 1 h, and then incubated at 37℃ for 24 h). For M2 macrophage groups, the mRNA relative expression levels of surface markers of M1 macrophage polarization including interleukin (IL)-1β, tumor necrosis factor-α(TNF-α), and those of M2 macrophage polarization including IL-10 and transforming growth factor-β(TGF-β) were detected by real-time reverse transcription polymerase chain reaction. The expression levels of IL-1β and IL-10 were detected by ELISA. The expression levels of CD86 (surface marker of M1 macrophage polarization) and CD206 (surface marker of M2 macrophage polarization) were measured by flow cytometry. The expression levels of signal transducer and activator of transcription 3 (STAT3), Toll-like receptor 4 (TLR4), nuclear factor κB (NF-κB) and phosphorylated (p)-NF-κB were detected by Western blot (WB). The migration ability of CAL-27 cells was assessed by scratch assay.Results:Compared with the control group, the expression levels of IL-1βand TNF-α mRNA, and IL-1βand CD86 proteins were up-regulated, whereas those of IL-10 and TGF-β mRNA, and IL-10 and CD206 proteins were down-regulated in the hyperthermia, ferric citrate and hyperthermia + ferric citrate groups, respectively (all P<0.05). In addition, the changes in the hyperthermia+ferric citrate group were significantly larger than those in the hyperthermia and ferric citrate groups (all P<0.05). WB showed that the expression level of STAT3 protein was down-regulated and those of TLR4, NF-κB and p-NF-κB expression were up-regulated in the hyperthermia + ferric citrate group (all P<0.05). Scratch assay showed that the migration ability of CAL-27 cells was inhibited in the hyperthermia, ferric citrate and hyperthermia + ferric citrate groups ( P<0.001), and the changes in the hyperthermia + ferric citrate group were significantly more pronounced than those in the hyperthermia and ferric citrate groups (both P<0.001). Conclusions:Hyperthermia and iron overload can promote the polarization of M1 macrophages and inhibit the polarization of M1 macrophages into M2 macrophages, thereby suppressing the migration of oral squamous cell carcinoma. The mechanism may be related to inhibiting the expression of STAT3 and activating the TLR4/NF-κB signaling pathway.
2.Application of blepharoplasty by "double-line method" in improving upper eyelid skin relaxation

Objective:To investigate the efficacy of blepharoplasty using the "double-line technique" in addressing upper eyelid skin laxity in middle-aged and elderly patients.Methods:A retrospective analysis was conducted on the clinical data of middle-aged and elderly patients with moderate to severe upper eyelid skin laxity who were admitted to the Plastic Surgery Department of Shaanxi Provincial People’s Hospital between January 2020 and January 2023. The same surgeon performed the "double-line" blepharoplasty to excise excess upper eyelid skin for laxity correction. Patients were positioned flat with their eyes closed; an arcuate line a (5-7 mm above the pupil) and a second line b (aligned with the eyelid margin in an upright position) were marked. The skin between these two lines was excised. Postoperatively, the double eyelid shape, improvement in skin laxity, adverse reactions, and patient satisfaction (categorized as satisfied, basically satisfied, unsatisfied) were assessed.Results:A total of 145 patients were included in the study, comprising 27 males and 118 females. Their ages ranged from 38 to 78 years, with an average age of 52 years. All patients’ incisions healed primarily without complications such as hematoma and infection. During the 3 to 12 months of follow-up after surgery, the double eyelid arcs on both sides appeared symmetrical and natural. The upper eyelid laxity was effectively corrected, obscured vision was improved, crow’s feet were reduced, and the periorbital area was rejuvenated. Patient satisfaction with the surgical results was high, with 112 patients reporting satisfaction, 25 reporting basically satisfied, and 8 reporting dissatisfaction. The overall satisfaction rate reached 94.48%(137/145). Among those dissatisfied, 6 cases cited the double eyelid crease as being too narrow, and 2 cases noted asymmetry in the double eyelid crease.Conclusion:The "double-line method" for blepharoplasty is straightforward to perform and effectively removes excess upper eyelid skin. It is suitable for middle-aged and elderly patients with moderate to severe upper eyelid skin laxity, resulting in favorable surgical outcomes.

5.Study on the effects of thermotherapy combined with gemcitabine on the biological behavior of tongue squamous cell carcinoma cells

Objective:To explore the impacts of thermotherapy combined with gemcitabine on the biological behavior of tongue squamous cell carcinoma cells.Methods:Human tongue squamous cell carcinoma Tca8113 cells were divided into the control group (blank control), gemcitabine group, thermotherapy group (heated in an incubator at 43℃ for 1 h and then incubated at 37℃ for 24 h) and thermotherapy + gemcitabine group. The proliferation ability of Tca8113 cells was assessed by EdU staining and CCK-8 assay. Cell apoptosis and cell cycle of Tca8113 cells were detected by flow cytometry. The invasion of Tca8113 cells was determined by Transwell chamber assay. The expression levels of cyclin D1 (CyclinD1), Bcl-2-associated X protein (Bax), matrix metalloproteinase (MMP)-9, phosphorylated histone H2AX (γH2AX) and Nijmegen breakage syndrome 1 (NBS1) proteins in Tca8113 cells were measured by Western blot. The changes of tumor mass and volume were detected by xenograft tumor in vivo test in nude mice. Multi-group comparison was performed by one-way ANOVA. Two group comparison was conducted by SNK- q test. Results:Compared with the control group, EdU positive cell percentage, OD 450 value, invasive cell number, CyclinD1, MMP-9 and NBS1 protein expression of Tca8113 cells were decreased, whereas the apoptosis rate, the expression of Bax and γH2AX proteins were increased in the gemcitabine, thermotherapy and thermotherapy + gemcitabine groups ( q=4.45-72.06, all P<0.001). Compared with the control group, the proportion of G 0/G 1 phase cells was decreased, whereas the proportion of S and G 2/M phase cells was increased in the gemcitabine and thermotherapy + gemcitabine groups, the proportion of G 0/G 1 phase cells was decreased and the proportion of G 2/M phase cells was increased in the hyperthermia group ( q=10.36-61.09, all P<0.001). Compared with the gemcitabine and thermotherapy groups, EdU positive cell percentage, OD 450 value, G 0/G 1 phase cell proportion, invasive cell number, CyclinD1, MMP-9 and NBS1 protein expression of Tca8113 cells were decreased, whereas apoptosis rate, S, G 2/M phase cell proportion, Bax and γH2AX protein expression were increased in the thermotherapy + gemcitabine group ( q=4.45-28.73, all P<0.001). Xenograft tumor in vivo test in nude mice showed that the tumor volume and mass of nude mice in the gemcitabine, thermotherapy, and thermotherapy + gemcitabine groups were decreased compared with those in the control group ( q=5.58-73.02, all P<0.001). Compared with the gemcitabine and thermotherapy groups, the tumor volume and mass in the thermotherapy + gemcitabine group were decreased ( q=5.58-21.45, all P<0.001). Conclusion:The combination of thermotherapy and gemcitabine can inhibit the proliferation and invasion, block the cell cycle, and induce cell apoptosis of Tca8113 cells.
6.Research progress in factors influencing the polarization of macrophages by hyperthermia

Hyperthermia, as an adjunct to radiotherapy and chemotherapy, can change the immune condition of tumor microenvironment and enhance the effect of tumor treatment without damaging normal tissues. Tumor-associated macrophages are the main immune cells in the tumor microenvironment, and there are two subtypes: M1 phenotype can inhibit the growth of tumor cells, and M2 phenotype can promote the occurrence and metastasis of tumor cells. Therefore, repolarization of M2 phenotype into M1 phenotype is a new research direction. In this paper, the mechanisms of hyperthermia and tumor-associated macrophage repolarization were reviewed based on the current research progress at home and abroad, aiming to provide novel ideas for tumor therapy.
7.Early efficacy of posterior Dynesys corrective internal fixation surgery for Lenke1 and Lenke5 adoles-cent idiopathic scoliosis

Objectives:To evaluate the early efficacy of Dynesys corrective internal fixation system in the treatment of Lenke1 and Lenke5 adolescent idiopathic scoliosis(AIS).Methods:A retrospective analysis was conducted on 15 AIS patients(4 males and 11 females)who underwent Dynesys corrective and internal fixa-tion treatment in our department from November 2019 to September 2023.The average age of the patients was 15.5±2.2 years(12-21 years).Among the patients,there were 5 cases of Lenke1 type and 10 cases of Lenke5 type,mainly characterized by thoracolumbar deformities.The operative time,bleeding volume,and complications were recorded by reviewing medical records,and imaging data were measured and analyzed.The measurement indicators included Cobb angle,thoracic kyphosis,lumbar lordosis,and lumbar range of mo-tion at preoperation,first follow-up within 1 week after surgery,and final follow-up to evaluate the corrective effect of posterior Dynesys corrective internal fixation on scoliosis.Results:All the patients successfully com-pleted the surgery and were followed up for 3-48 months.The average operative time was 280.8±45.8min,average bleeding volume was 194.0±145.5mL,the number of main curvature segments was 4.7±1.1,and the number of operative segments was 4.7±1.1.The Cobb angle of the coronal plane was 40.8°±5.5° before surgery,5.5°±4.5° after surgery,and 7.6°±5.5° at final follow-up.Coronary scoliosis improved significantly af-ter surgery(P<0.05),with a total correction rate of 86.9%for the main curvature,89.8%for Lenke1 type,and 85.4%for Lenke5 type.There was no significant statistical difference between the two types.At final follow-up,the range of motion of the thoracolumbar fixed segment was 8.2°±3.1°.All patients did not experience any operation-related complications,and no implant complications such as loose screws or broken ropes were found during follow-up.Conclusions:Dynesys corrective internal fixation surgery can effectively correct Lenke1 and Lenke5 AIS,with minimal surgical trauma,fast postoperative recovery,preservation of partial spinal motor function,which has satisfactory early postoperative outcomes.
8.Effect of hyperthermia on biological behavior of squamous cell carcinoma of the tongue by regulating ferroptosis pathway

Objective:To investigate the regulation and possible mechanism of hyperthermia (HT) on the ferroptosis of squamous cell carcinoma of the tongue cell line CAL-27.Methods:Half maximal inhibitory concentration (IC 50) of Fer-1, an inhibitor of ferroptosis, was detected by CCK-8 assay and used for subsequent experiments. CAL-27 cells were divided into the HT, control, Fer-1 and HT+ Fer-1 groups according to experimental design. Reactive oxygen species (ROS) levels and iron ion concentration were determined by corresponding detection kits. The p53 and TfR1 mRNA levels were detected by real-time reverse transcription PCR. Cell migration was detected by cell scratch test and cell apoptosis was detected by flow cytometry. Results:HT significantly up-regulated the ROS levels ( P<0.01) and iron ion concentration ( P<0.001), and significantly increased the expression levels of p53 and TfR1 mRNA (both P<0.01). The cell migration ability was decreased ( P<0.001), whereas cell apoptosis rate was increased by HT ( P<0.01). In the HT+Fer-1 group, the ROS levels ( P<0.001), iron ion concentration ( P<0.001), expression levels of p53 and TfR1 mRNA (both P<0.01) were significantly down-regulated, the cell migration ability was recovered ( P<0.01), and cell apoptosis rate was decreased ( P<0.01) compared with those in the HT group, respectively. Conclusions:HT may induce the ferroptosis of CAL-27 cell line, inhibit cell migration ability and promote cell apoptosis by activating the p53/TfR1 pathway.
9.Research progress on the effect of hyperthermia on vascular endothelial growth factor in tumor microenvironment

Vascular endothelial growth factor (VEGF) plays an important role in promoting tumor vascular growth and changing vascular wall permeability. With the in-depth study of tumor hyperthermia and tumor microenvironment, more and more studies have shown that hyperthermia exerts multiple regulatory effects on VEGF in tumor microenvironment. Combined with current research progress in China and abroad, this article reviews the effect of hyperthermia on VEGF and its related cells and factors in tumor microenvironment, aiming to provide new ideas for the clinical application of tumor hyperthermia combined with immune or targeted therapy.
