1.The influence of drug-coated balloon catheters on the patency rate of target lesion vessels,coagulation function and vascular endothelial function in patients with restenosis of stents after percutaneous transluminal angioplasty for lower extremity arteriosclerotic occlusion

Objective To explore the effects of drug-coated balloons(DCB)on the patency rate of target lesion vessels,coagulation function and vascular endothelial function in patients with in-stent restenosis(ISR)after percutaneous endovascular angioplasty(PTA)for lower extremity arteriosclerosis obliterans(ASO).Methods A total of 62 patients with ISR and ASO admitted to the hospital were retrospectively enrolled between March 2020 and March 2022.According to different treatment methods,they were divided into DCB group(n=38)and common balloon(SAB)group(n=24).All were followed up for 12 months after surgery.The changes in primary patency rate of target lesion vessel,clinically driven-target lesion revascularization(CD-TLR)rate,late loss of values in the lumen(LLL),ankle-brachial index(ABI),coagulation function indexes[prothrombin time(PT),activated partial thromboplastin time(APTT),fibrinogen(FIB),D-dimer(D-D)]and vascular endothelial function indexes[serum endothelin-1(ET-1),nitric oxide(NO),flow-mediated dilatation(FMD)]were observed,and the occurrence of postoperative complications in the two groups was recorded.Results At 12 months after surgery,primary patency rate of target lesion vessels in DCB group was higher than that in SAB group(86.84%vs 50.00%),CD-TLR rate,LLL and ABI were lower than those in SAB group[13.16%,(1.39±0.52)mm,(0.76±0.12)vs 50.00%,(1.79±0.64)mm,(0.62±0.11);P<0.05].At24h and 2 weeks after surgery,there was no significant difference in PT,APTT,FIB or D-D between the two groups(P>0.05).At 24h and 2 weeks after surgery,levels of serum ET-1 in DCB group were lower than those in SAB group[(66.65±7.12)pg/ml,(65.58±6.98)pg/ml vs(71.74±6.92)pg/ml,(68.84±6.51)pg/ml)],while NO levels were higher than those in SAB group[(32.21±4.17)pg/ml,(34.62±3.32)pg/ml vs(28.53±5.23)pg/ml,(31.21±4.19)pg/ml;P<0.05].At 2 weeks after surgery,FMD in DCB group was higher than that in SAB group[(12.49±5.33)%vs(9.14±4.42)%,P<0.05].There was no significant difference in the total incidence of complications between the two groups(21.50%vs 12.50%,P>0.05).Conclusion Compared with SAB,DCB can effectively protect vascular endothelial function and improve the primary patency rate of ISR after PTA in patients with lower extremity ASO.

3.Study on the level of peripheral serum glial-derived neurotrophic factor in children with attention deficit hyperactivity disorder

Objective To explore the changes of peripheral serum glial-derived neurotrophic factor (GDNF) levels in children with attention deficit hyperactivity disorder(ADHD),and to provide a new mark-er for early identification of ADHD in clinical practice. Methods Totally 42 drug-naive children with ADHD(ADHD group) and 45 healthy children(HC group) were included. General demographic informa-tion,scale scores,and fasting peripheral serum GDNF levels were compared between the two groups. Mean-while,the correlation between GDNF level and symptom severity score of children with ADHD was analyzed. Results There were significant differences in FSIQ,attention deficit, and hyperactivity/impulsivity scores between ADHD group and HC group(FSIQ(105. 26±13. 82) vs (114. 73±9. 93); attention deficit(23. 60± 5. 06) vs (20. 04±2. 85); hyperactivity/impulsivity(19. 43±3. 47) vs (15. 93±2. 42),all P<0. 05). Serum level of GDNF in the ADHD patients was significant lower than that in the HC group ((442. 52± 70. 01) pg/ml vs (554. 02±101. 37)pg/ml,P<0. 01). In addition,GDNF levels were negatively correlated with at-tention deficit scores in children with ADHD (radj=-0. 447,P<0. 01). More importantly,ROC curve results show that GDNF had good diagnostic value (AUC=0. 81). Conclusion GDNF levels decreased child with ADHD. Decreased serum GDNF levels have certain promising value in the diagnosis of ADHD.
4.Expression and clinical significance of proline-rich tyrosine kinase 2 and phospho-protein kinase B in tongue squamous cell carcinoma.

OBJECTIVE:
This study aimed to investigate the expression and clinical significance of proline-rich tyrosine kinase 2 (Pyk2) and phospho-protein kinase B (p-AKT) in tongue squamous cell carcinoma (TSCC) and adjacent nontumor tissues.
METHODS:
The Pyk2 and p-AKT protein levels were detected via immunohistochemistry in 45 cases of TSCC tissues and 30 cases of adjacent nontumor tissues. The relationships of the two protein levels and clinicopathological characteristics were also analyzed.
RESULTS:
Pyk2 and p-AKT levels were significantly higher in the TSCC tissues than in the adjacent nontumor tissues (P<0.05). Nontumor tissues showed poor or no expression. The expression levels of the two proteins were positively correlated (γs=0.412). The expression of Pyk2 was associated with histopathological differentiation type, regional lymph node metastasis, and TNM staging (P<0.05), but not with age and gender. The expression of p-AKT was only related to histopathological differentiation types (P<0.05).
CONCLUSIONS
The abnormal expression of Pyk2 and p-AKT proteins might be closely related to the development and progression of TSCC. Joint detection can be used as an indicator to estimate the degree of TSCC.

5.Significance of multidrug resistance associated protein 1 and protein 2 expressions in peripheral blood mononuclear cells in children with intractable epilepsy

Objective To study the expression of multidrug resistance associated protein 1 (MRP1) and MRP2 in the peripheral blood mononuclear cells of children with intractable epilepsy (IE).Methods During Nov.2010 to Oct.2013,50 children with I E were collected as the experimental group,simultaneously 50 children with epilepsy controlled by the anti-epileptic drugs (AEDs) and 50 healthy children without epilepsy were collected as the control group from the outpatient or inpatient of Xuzhou Children's Hospital.The expressions of MRP1 and MRP2 in the peripheral blood mononuclear cells of children were detected by reverse transcriptase polymerase chain reaction and Western blot.Results 1.The mean relative expression of MRP1 and MRP2 mRNA in the peripheral blood mononuclear cells of children with IE (0.795 ± 0.042,0.804 ± 0.023) were higher than those in epilepsy controlled by AEDs (0.682 ± 0.030,0.675 ± 0.021) and healthy children without epilepsy (0.665 ± 0.031,0.654 ± 0.029) (all P <0.001).2.The mean relative expression of MRP1 and MRP2 protein in the peripheral blood mononuclear cells of children with IE (2.027 ±0.034,1.902 ±0.021) were higher than those in epilepsy controlled by AEDs(1.131 ±0.042,1.086 ± 0.027) and healthy children without epilepsy (1.093 ± 0.023,1.045 ± 0.018) (all P < 0.001).3.There was no difference in the expression of MRP1,MRP2 mRNA and proteins between the children with epilepsy controlled by AEDs and healthy children without epilepsy (all P > 0.05).Conclusions MRP1 and MRP2 over-expression in the peripheral blood mononuclear cells of children with IE may be associated with drug-resistance mechanism for medically intractable epilepsy.

Objective To investigate the changes of the levels of TNF and IL 8 in blood and in gastric mucosa and their mRNA expression after liver ischemia reperfusion (I/R). Methods 130 rats were randomly divided into 3 groups: (1) sham operation group(control group, n=10);(2)liver ischemia reperfusion group (n=60), including ischemia for 20min and 40min, and reperfusion for 1h, 24h and 72h respectively; (3) liver I/R rats treated with famotidine group (n=60).
