1.Application of Zebrafish in Quality Evaluation of Traditional Chinese Medicine

As a new type of model organism, zebrafish is gradually gaining prominence in the field of scientific research. The unique biological characteristics and advantages of zebrafish make them play an increasingly important role in the quality evaluation of traditional Chinese medicine. Compared with other common experimental animals, zebrafish have a fast reproductive and growth speed and high embryo transparency, making them an ideal model for evaluating the quality of traditional Chinese medicine. This provides a new perspective and method for research on traditional Chinese medicine. With the growing global interest in traditional Chinese medicine, it has become crucial to find scientific and accurate methods to evaluate the quality and effectiveness of traditional Chinese medicine. The introduction of the zebrafish model has brought new breakthroughs in the quality evaluation of traditional Chinese medicine. To further promote the application of zebrafish in evaluating the quality of traditional Chinese medicine, this article systematically searched and sorted out the previous studies related to the application of zebrafish for this purpose since 2023. The commonly used disease models and indicators of zebrafish in evaluating the effectiveness of traditional Chinese medicine, as well as the mechanism of zebrafish in exploring the active ingredients of traditional Chinese medicine, were primarily reviewed. The application of zebrafish in evaluating the safety of traditional Chinese medicine and the typical examples in ensuring the quality of traditional Chinese medicine were demonstrated. The limitations encountered by zebrafish models in evaluating the quality of traditional Chinese medicine were highlighted. The resolution of these problems will help further improve the accuracy and reliability of zebrafish in evaluating the quality of traditional Chinese medicine. The article discussed the evaluation of effectiveness, safety, and quality control of zebrafish applied in traditional Chinese medicine, so as to provide a reference for establishing standards for traditional Chinese medicine and promoting its modernization in the future.
2.Application of natural language processing models in cervical cancer staging and risk factors extraction

Objective:To evaluate the accuracy and output stability of online general natural language processing(NLP)models for staging diagnosis and identifying medium-and high-risk factors in cervical cancer patients based on pathology reports.Methods:Surgical pathological reports of 65 patients with cervical cancer who received postoperative adjuvant radiotherapy at Shanghai General Hospital from January 2022 to December 2023 were retrospectively selected.These reports were input into two online NLP models,Kimi and ChatGPT-4o,and their output staging diagnosis results were recorded.Then,the results were classified into 3 categories and scored as follows:correct(2 points),basically correct(1 point)and incorrect(0 point).Each pathologic report was tested 5 times to assess the stability of the outputs form Kimi and ChatGPT-4o,and the consistency between the NLP models and clinical physicians in cervical cancer staging was compared.Prompt-based questioning was used to evaluate Kimi's ability to extract medium-and high-risk factors from the pathology reports of cervical cancer patients.Results:There was no statistical significant difference in the staging diagnosis results between the two NLP models and the clinical physicians(x2=5.740,P=0.057).Kimi and ChatGPT-4o respectively produced 56 and 47 correct results,6 and 15 basically correct results,and 3 and 3 incorrect results.Their mean scores were 7.08±2.70 and 7.97±2.97,and the difference between them was statistically significant(P=0.040).In the extraction of risk factors from 65 cervical cancer patients,involving a total of 390 factors,Kimi made only three false-positive errors,with all other factors correctly identified.Conclusion:Online general NLP models can stably output the stage of cervical cancer patients with diagnostic accuracy comparable to clinical physicians.With the assistance of prompt-based questioning,these NLP models can accurately extract medium-and high-risk factors from pathology reports of cervical cancer patients,demonstrating promising clinical application potential.

4.The interaction between polyphyllin I and SQLE protein induces hepatotoxicity through SREBP-2/HMGCR/SQLE/LSS pathway

Polyphyllin Ⅰ(PPⅠ)and polyphyllin Ⅱ(PⅡ)are the main active substances in the Paris polyphylla.However,liver toxicity of these compounds has impeded their clinical application and the potential hepatotoxicity mechanisms remain to be elucidated.In this work,we found that PPⅠ and PⅡ exposure could induce significant hepatotoxicity in human liver cell line L-02 and zebrafish in a dose-dependent manner.The results of the proteomic analysis in L-02 cells and transcriptome in zebrafish indicated that the hepa-totoxicity of PPⅡ and PⅡwas associated with the cholesterol biosynthetic pathway disorders,which were alleviated by the cholesterol biosynthesis inhibitor lovastatin.Additionally,3-hydroxy-3-methy-lglutaryl CoA reductase(HMGCR)and squalene epoxidase(SQLE),the two rate-limiting enzymes in the choles-terol synthesis,selected as the potential targets,were confirmed by the molecular docking,the over-expression,and knockdown of HMGCR or SQLE with siRNA.Finally,the pull-down and surface plasmon resonance technology revealed that PPⅠ could directly bind with SQLE but not with HMGCR.Collectively,these data demonstrated that PPⅠ-induced hepatotoxicity resulted from the direct binding with SQLE protein and impaired the sterol-regulatory element binding protein 2/HMGCR/SQLE/lanosterol synthase pathways,thus disturbing the cholesterol biosynthesis pathway.The findings of this research can contribute to a better understanding of the key role of SQLE as a potential target in drug-induced hepatotoxicity and provide a therapeutic strategy for the prevention of drug toxic effects with similar structures in the future.
5.Study on Mechanism of Action of Matrine in Treatment of Inflammatory Bowel Disease Based on Zebrafish Model and Network Pharmacology

ObjectiveTo study the mechanism of matrine in the treatment of inflammatory bowel disease (IBD) based on the zebrafish model and network pharmacology. MethodThe IBD model of zebrafish was established using 2,4,6-trinitro-benzenesulfonicacid (TNBS), and the intestinal phagocytic function, goblet cell secretion, and neutrophil aggregation were evaluated using neutral red staining, alcian blue staining, and neutrophil number changes. Changes in tumor necrosis factor (TNF)-α and cholecystokinin (CCK) content in zebrafish were determined by using relevant reagent kits. Network pharmacology and molecular docking techniques were used to predict the potential mechanism of matrine in the treatment of IBD. Gene expression of relevant targets was verified through Real-time polymerase chain reaction (Real-time PCR). ResultCompared with the model group, the matrine administration group can increase the neutral red staining area in a dose-dependent manner and improve intestinal phagocytic function(P<0.05,P<0.01). It can reduce the staining area of alcian blue and affect the secretion of intestinal goblet cells(P<0.01). It can reduce the number of neutrophil granulocytes, relieve its aggregation, significantly reduce TNF-α content(P<0.01), and increase the CCK content. Network pharmacology analysis identifies 28 potential targets for matrine in the treatment of IBD. The top five targets by protein-protein interaction (PPI) network analysis are CHRNA7, DRD1, CHRNA4, SLC6A3, and GRM5. The Kyoto encyclopedia of genes and genomes (KEGG) results show that the treatment of IBD with matrine may be related to neuroactive ligand-receptor interaction, cholinergic synapse, and neutrophil extracellular trap formation. Real-time PCR results show that matrine can affect the expression level of related target genes. Conclusionmatrine has a certain therapeutic effect on IBD and can affect the inflammatory response of IBD. Its therapeutic effect may be related to neuroactive ligand-receptor interaction and other pathways.
6.Sex differences in systemic lupus erythematosus (SLE): an inception cohort of the Chinese SLE Treatment and Research Group (CSTAR) registry XVII.

BACKGROUND:
The onset and clinical presentation of systemic lupus erythematosus (SLE) are sex-related. Few studies have investigated the distinctions in clinical characteristics and treatment preferences in male and female SLE patients in the initial cohort. This study aimed to improve the understanding of Chinese SLE patients by characterizing the different sexes of SLE patients in the inception cohort.
METHODS:
Based on the initial patient cohort established by the Chinese SLE Treatment and Research Group, a total of 8713 patients (795 men and 7918 women) with newly diagnosed SLE were enrolled between April 2009 and March 2021. Of these, 2900 patients (347 men and 2553 women) were eligible for lupus nephritis (LN). A cross-sectional analysis of the baseline demographic characteristics, clinical manifestations, laboratory parameters, organ damage, initial treatment regimens, and renal pathology classification was performed according to sex.
RESULTS:
In the SLE group, as compared to female patients, male patients had a later age of onset (male vs. female: 37.0 ± 15.8 years vs. 35.1 ± 13.7 years, P = 0.006) and a higher SLE International Collaborative Clinic/American College of Rheumatology damage index score (male vs. female: 0.47 ± 1.13 vs. 0.34 ± 0.81, P = 0.015), LN (male vs. female: 43.6% vs. 32.2%, P < 0.001), fever (male vs. female: 18.0% vs. 14.6%, P = 0.010), thrombocytopenia (male vs. female: 21.4% vs. 18.5%, P = 0.050), serositis (male vs. female: 14.7% vs. 11.7%, P = 0.013), renal damage (male vs. female: 11.1% vs. 7.4%, P < 0.001), and treatment with cyclophosphamide (CYC) (P < 0.001). The frequency of leukopenia (male vs. female: 20.5% vs. 25.4%, P = 0.002) and arthritis (male vs. female: 22.0% vs. 29.9%, P < 0.001) was less in male patients with SLE. In LN, no differences were observed in disease duration, SLE Disease Activity Index score, renal biopsy pathological typing, or 24-h urine protein quantification among the sexes. In comparisons with female patients with LN, male patients had later onset ages (P = 0.026), high serum creatinine (P < 0.001), higher end-stage renal failure rates (P = 0.002), musculoskeletal damage (P = 0.023), cardiovascular impairment (P = 0.009), and CYC use (P = 0.001); while leukopenia (P = 0.017), arthritis (P = 0.014), and mycophenolate usage (P = 0.013) rates were lower.
CONCLUSIONS
Male SLE patients had more severe organ damage and a higher LN incidence compared with female SLE patients; therefore, they may require more aggressive initial treatment compared to female patients.

7.Current status of application of resources on medical exposure in Dongying, China

Objective To investigate the current status of application of resources on medical exposure in Dongying, China, and to provide fundamental data for setting up the health information management system of occupational radiation diseases and implementing health supervision and management strategies. Methods According to the requirements of monitoring program on radiation health of Dongying in 2021, a general survey was carried out in all medical institutions (other than dental clinics) providing radiodiagnosis and radiotherapy services by the field investigation method using the unified questionnaires and survey content, and a descriptive analysis was conducted on the results. Results There were 92 institutions of radiodiagnosis and radiotherapy in Dongying, 1112 radiation workers who accounted for 5.9% of all workers on duty, including 679 (61.1%) males and 433 (38.9%) females, and 364 pieces of radiation equipment, including 339 for X-ray diagnosis, 8 for radiotherapy, 2 for nuclear medicine, and 15 for interventional radiology. The allocation of personal protective equipment for interventional radiology staff in tertiary hospitals was higher than that in secondary hospitals, in which there were 0.36 and 0.23 pieces of lead-rubber aprons, 0.43 and 0.30 pieces of lead-rubber hats, 0.50 and 0.35 pieces of lead-rubber collars, 0.05 and 0 pairs of lead-rubber gloves, and 0.31 and 0.17 pairs of lead protection glasses per capita, respectively. The allocation rates of lead-rubber collars and lead protection glasses in the nuclear medicine staff were higher than those in the interventional radiology staff, with 0.63 collars and 0.88 pairs per capita, respectively. The annual frequency of medical exposure was calculated to be 864.3 person-times per 1000 population for radiodiagnosis and 5.2 patients per 1000 population for radiotherapy. Conclusion The allocation and application of resources on medical exposure in Dongying is unbalanced, so it is demanded to set up a health information management system of occupational radiation diseases for continuously mastering the application of medical exposure and adopting differentiated regulatory measures such as rating classification in Dongying.
8.MTERFD1 promotes the proliferation of ovarian cancer cells through regulating IL-6

Objective:To investigate the role of mitochondrial transcription termination factor domain 1 (MTERFD1) in regulating the malignant biological behavior of ovarian cancer (OC) cells and preliminarily reveal its molecular mechanism.Methods:RT-qPCR was used to detect the expression levels of MTERFD1 mRNA in human OC cell lines SKOV3, HEY, and OVCAR3, human embryonic kidney epithelial cells HEK-293, and normal ovarian tissues. After transfecting these cells with MTERFD1 overexpression plasmid and small interfering RNA, MTT analysis was used to detect the cell proliferation, and Annexin V/PI double staining assay was used to detect apoptosis. ELISA was used to check the effect of MTERFD1 level on the expression of IL-6. MTT analysis was used to evaluate the effect of IL-6 level changing on the proliferation of OC cells with different MTERFD1 expressions.Results:The MTERFD1 mRNA levels in OC cell lines SKOV3, HEY, and OVCAR3 were higher than those in HEK-293 cell line and normal ovarian tissues with statistically significant differences ( P<0.05). In human OC OVCAR3 and HEK-293 cell lines, the cell proliferation in the group of MTERFD1 overexpression plasmid was higher than that in the blank plasmid group; after the MTERFD1 being knocked out, the HEY and SKOV3 cell proliferations obviously decreased and the apoptosis rates were higher than those in the negative control group; all differences were statistically significant ( P<0.05). The IL-6 levels in OC cells were positively correlated with MTERFD1 expressions, and the IL-6 blocking antibody could reverse the promoting effect of MTERFD1 on the proliferation of OC cells. Conclusion:The promoting effect of MTERFD1 on the proliferation of ovarian cancer cells may be achieved through regulating IL-6.

10.Relationship between cognitive functions and medication compliance in patients with schizophrenia at stable phase

Objective To elucidate the effect of cognitive functions on medication compliance of schizophrenia patients at stable phase, and provide clinical cognitive coping strategies to improve the medication compliance of schizophrenia patients. Methods Two hundred and thirty-eight schizophrenia patients at stable phase, admitted to psychiatry department of the 3rd affiliated hospital of Sun Yat-sen University from May 2017 to October 2017, and all study subjects were divided into without medication compliance group (the scores of medication compliance scale≤4, n=32) and medication compliance group (the scores of medication compliance scale>4, n=206) by Medication Adherence Rating Scale (MARS). MATRICS Consensus Cognitive Battery (MCCB) and Brief Psychiatric Rating Scale (BPRS) were used to evaluate the differences of cognitive performance and disease severity between groups. The factors influencing medication compliance of schizophrenia patients with stable phase were explored by multiple linear regression analysis. Results (1) The total scores of MCCB and it,s subscales scores (except for visual learning) of schizophrenia patients with medication compliance were higher than that of patients without medication compliance. The scores of symbol coding, spatial span and emotional intelligence ability in medication compliance group were significantly higher than those of patients without medication compliance (P<0.05). (2) Symbol coding (neurocognition ability), emotion management (social cognition ability), severity of illness, and antipsychotic drug dose were the independent influencing factors of medication compliance of schizophrenia patients with stable phase (P<0.05). Conclusion The medication compliance of schizophrenia patients with stable phase is not only affected by antipsychotics dose and severity of illness, but also affected by neurocognitive and social cognitive functions.
