1.Effects of Hedyotis diffusa Willd on proliferation,invasion and Th1/Th2 cytokines of renal carcinoma through TGF-β/Smad signaling pathway

Objective:To explore the mechanism by which the extract of Hedyotis diffusa Willd regulates TGF-β/Smad signaling transduction pathway and affects proliferation,invasion of renal cancer cells and expressions of Th1/Th2 cytokines in mice with renal cancer.Methods:Using mouse renal cancer Renca cells as the research object,after intervention with extracts of Hedyotis diffusa Willd,impact on migration ability of mouse renal cancer cells was detected at the cellular level using scratch assay;observing effect of CCK-8 experiment on proliferation ability of mouse renal cancer cells.A mouse kidney cancer model was established by subcutaneous inoculation under the right armpit,and effect of extract of Hedyotis diffusa Willd on proliferation and invasion of renal carcinoma in mice was studied.HE staining was used to observe histomorphology of transplanted tumor mass in mice;Western blot was used to detected expressions of TGF-β/Smad pathway protein TGF-β,Smad2 and p-Smad2 in mouse transplanted tumor tissue;levels of IL-2 and IL-10 in mouse serum were detected by ELISA to investigate whether the extract of Hedyotis diffusa Willd has an inhibitory effect on Th1/Th2 renal carcinoma immune shift.Results:CCK-8 experiment showed that the extract of Hedyotis diffusa Willd inhibited pro-liferation ability of mouse renal cancer Renca cells(P<0.05);scratch test results showed that extract of Hedyotis diffusa Willd effec-tively inhibited the healing ability of mouse renal cancer Renca cells(P<0.05);by constructing a mouse kidney cancer model,it was found that extract of Hedyotis diffusa Willd could inhibit the growth of transplanted tumor volume in mice(P<0.05);HE staining revealed that tumor cells in model group were dense,with numerous mitotic figures and minimal necrosis;whereas in Hedyotis diffusa Willd group and pathway inhibitor group,there was increased necrosis of cancer cells,accompanied by karyopyknosis or lysis;Western blot showed that compared with model group,expression of TGF-β and p-Smad2 in extract of Hedyotis diffusa Willd group and pathway inhibitor group were decreased(P<0.001);ELISA results showed that compared with model group,serum IL-2 expression increased in both Hedyotis diffusa Willd group and pathway inhibitor group(P<0.000 1).IL-10 expression decreased in Hedyotis diffusa Willd group(P<0.05)and pathway inhibitor group(P>0.05).Conclusion:Extract of Hedyotis diffusa Willd may inhibit protein expressions in TGF-β/Smads signaling pathway,further regulate the secretion of Th1/Th2 cytokines,thereby inhibiting proliferation and migration of mouse renal cancer cells both in vitro and in vivo.
2.Euphorbia Lathyrol effects expressions of immune factors IL-7,IL-10 and VEGF in renal clear cell neoplasias mice through TGF-β/Smad signaling pathway

Objective:To investigate the effect of Euphorbia Lathyrol on expressions of IL-7,IL-10 and VEGF in renal clear cell neoplasias(RCCN)mice by regulating TGF-β/Smad signaling pathway.Methods:RCCN xenograft mice were established and divided into model group,Euphorbia Lathyrol group and Cisplatin group.Euphorbia Lathyrol group were given 20 mg/kg Euphorbia Lathyrol solution intragastric administration,model group were given 0.9%NaCl gastric feeding,Cisplatin group were given intraperito-neal injection of 4 mg/kg cisplatin solution,then mice weight and tumor volume were recorded.Expressions of TGF-β1,Smad2 and Smad4 were tracked down through Western blot.Expressions of immune factors IL-7 and IL-10 in serum of xenograft mice were detected by ELISA.Expression of VEGF in tumor mass was detected by IHC.Results:After 14 days of administration,compared with model group,the proliferation of tumor grafts in Cisplatin group and Euphorbia Lathyrol group was inhibited(P<0.05),and the expressions of TGF-β1,Smad2,Smad4 protein and VEGF were observably decreased(P<0.05),expressions of IL-7 in serum was increased,while level of IL-10 in serum was decreased(P<0.05).Conclusion:Euphorbia Lathyrol is able to reduce expressions of IL-7,IL-10,as well as VEGF in RCCN xenograft mice by impeding TGF-β1/Smad signaling transduction system.

5.Effects of ethanol extracts of Panax notoginseng on liver metastasis of B16 melanoma grafted in mice

OBJECTIVE: To observe the effects of ethanol extracts of Panax notoginseng on the tumor and the liver metastasis in experimental mice grafted with B16 melanoma. METHODS: B16 melanoma was transplanted in the spleen of C57BL/6 mice. The effects of different doses of ethanol extracts of Panax notoginseng on the inhibition rate of spleen tumors and the liver metastasis were observed respectively. RESULTS: The high-, medium-, and low-doses of the extracts and the interferon-alpha (IFN-alpha) can improve the quality of life of the experimental mice. The weights of spleen tumor were lower in the low- and medium-dose extracts-treated groups and the IFN-alpha-treated group than that in the normal saline (NS)-treated group (P<0.05 or P<0.01). The liver metastasis was less in the low- and medium-dose extracts-treated groups and the IFN-alpha-treated group than that in the NS-treated group (P<0.01). CONCLUSION: The ethanol extracts of Panax notoginseng can improve the quality of life of the experimental mice and inhibit the growth of tumor and the liver metastasis.
