Inflammatory diseases (IDs) are a general term of diseases characterized by chronic inflammation as the primary pathogenetic mechanism, which seriously affect the quality of patient′s life and cause significant social and medical burden. Current drugs for IDs include nonsteroidal anti-inflammatory drugs, corticosteroids, immunomodulators, biologics, and antioxidants, but these drugs may cause gastrointestinal side effects, induce or worsen infections, and cause non-response or intolerance. Given the outstanding performance of metal polyphenol network (MPN) in the fields of drug delivery, biomedical imaging, and catalytic therapy, its application in the diagnosis and treatment of IDs has attracted much attention and significant progress has been made. In this paper, we first provide an overview of the types of IDs and their generating mechanisms, then sort out and summarize the different forms of MPN in recent years, and finally discuss in detail the characteristics of MPN and their latest research progress in the diagnosis and treatment of IDs. This research may provide useful references for scientific research and clinical practice in the related fields.

Aim To invepredict the mechanism of ac-tion and targets of the traditional Chinese medicine compound Zheng Gan Decoction against hepatocellular carcinoma based on bioinformatics, and to experimen-tally verify the mechanism of anticancer action of Zheng Gan Decoction against DEN-induced hepatocellular carcinoma in a rat model. Methods The compounds of CZLF were collected from TCMSP and HERB Herbal Histology Database, and the potential targets of CZLF were predicted from Uniprot Protein Database, and the disease targets of hepatocellular carcinoma were searched with the help of OMIM,TTD,and Gene Cards databases, and then Venny analysis was per-formed on the targets of the disease-drugs,and then the protein-drug interactions(PI)of CZLF were mapped by String database, which was used for the study. After that,we used String database to draw pro-tein-protein interaction(PPI)network,R 4.1.3 soft-ware and Metascape database to enrich gene ontology(GO)and analyze KEGG signaling pathway for the core intersected targets, and finally,we used Cyto-scape 3.8.2 software for visualization to construct"compound-pathway-target-disease".Lastly,Cytoscape 3.8.2 software was used to visualize and construct a"compound-pathway-target-disease"network diagram, which was finally verified by Western blot experiment. Results Bioinformatics results showed that there were 171 common targets between Zheng Gan Decoction and diseases, and Zheng Gan Decoction might exert its an-ticancer effect through the main active ingredients such as lignans,quercetin,β-glutosterol,ivy saponin, etc.,as well as through the core protein targets such as CASP3,BCL2,AKT1,IL6,etc.,and the modula-tion of the Hippo signaling pathway and PI3K-Akt sig-naling pathway. The results of animal experiments showed that the expression content of apoptosis-related proteins caspase-3,BCL-2 and Bax was detected by Western blot, and the expression of Bax and caspase-3 proteins was up-regulated and the expression of Bcl-2 protein was down-regulated in the treatment group of Zheng Gan Decoction. Conclusions This study sug-gests that Zheng Gan Decoction may inhibit the growth of hepatocellular carcinoma cells and promote the apop-tosis of hepatocellular carcinoma cells by regulating the apoptosis-related proteins of caspase-3,Bcl-2,and Bax,so as to achieve the effect of anti-hepatocellular carcinoma.

Chinese Pharmacological Bulletin 2024;40(8):1591-1598
Aim To explore the feasibility of the cloud exposure system for in vitro exposure experiments on inhalation toxicology. Methods Calu-3 cells cultured at the air-liquid interface(ALI)were exposed to three concentrations of lipopolysaccha-ride(LPS):high,medium,and low(800,400,200 mg·L-1)by the cloud exposure system, and phosphate buffer solu-tion(PBS)was used as a negative control group for one expo-sure,while the high concentration of LPS was used to expose Calu-3 cells for five times.Calu-3 cells were exposed to phos-phate buffer solution(PBS)once as negative control group and to high concentration of LPS solution for five times, and the ac-tivity of Calu-3 cells, the release of lactate dehydrogenase(LDH),TEER,mucin MUC5AC,and the expression of inflam-matory factors IL-6,IL-8 and TNF-α were detected 3 h and 24 h after the end of the exposure, respectively. Results Compared with the PBS-negative control group, after exposure to the Calu-3 cell model at the air-liquid interface with three concentrations of LPS,high, medium, and low, there were no significant changes in the activity and LDH release, but the cellular electrical resist-ance value was reduced, and the barrier function of the cells was impaired;with the increase of the exposure concentration, the LPS promoted the expression of the cellular mucin MUC5AC,which led to a decrease in the expression of cellular IL-6,IL-8,and a decrease in the expression of TNF-α.Expression of IL-6 and IL-8 decreased and TNF-α expression increased;as the fre-quency of exposure increased,LPS inhibited the expression of mucin and increased the expression of IL-6;an increase in the frequency of exposure along with a prolongation of post-exposure assay time resulted in an increase in the expression of cellular IL-8 and TNF-a.Conclusions The ALI cloud exposure ap-proach can effectively reflect the cellular response to positive subjects, and this in vitro exposure can be used in subsequent exposure experiments to evaluate the inhalation toxicity of com-pounds.

Jinqi Jiangtang Capsule (JQJTC) is clinically used for the prevention and treatment of type 2 diabetes, but the contents of its main chemical components are not yet clear. In this study, an ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) method was established for the determination of 15 components in JQJTC, including new chlorogenic acid, chlorogenic acid, cryptochlorogenic acid, formononetin, ononin, calycosin, calycosin-7-glucoside, astragaloside IV, berberine, epiberberine, berberrubine, coptisine, jatrorrhizine, palmatine and magnoflorine. The method was used to determine the contents of 15 components in the capsule and then to investigate the influence of excipients on the contents of the components in JQJTC. The separation was performed on a ACQUITY UPLC BEH C18 column (100 mm × 2.1 mm, 1.7 μm) with a mobile phase consisting of 0.1% acetic acid and 5 mmol·L-1 ammonium acetate (A) and acetonitrile (B) with gradient elution at a flow rate of 0.3 mL·min-1 and a column temperature at 40 ℃. Electron spray ionization was used for mass spectrometry in positive ion mode. The established method meets the requirements of methodology of content determination in Chinese pharmacopoeia. The contents of 15 components in JQJTC varied from high to low. The top 5 contents were berberine, chlorogenic acid, magnoflorine, coptisine, and cryptochlorogenic acid, accounting for 87.31% of the total content. The contents of 10 components, including the alkaloids of coptidis rhizoma (berberine, epiberberine, berberrubine, coptisine, jatrorrhizine, palmatine and magnoflorine) and the organic acids of honeysuckle (new chlorogenic acid, chlorogenic acid, and cryptochlorogenic acid) in the whole formula extract without excipients was significantly lower than that in the capsule. These components accounted for 99.20% of the determined component contents. In this experiment, an accurate, sensitive and efficient UHPLC-MS/MS method for the determination of multi-components in JQJTC was established, which stably and reliably detected the contents of 15 components in the capsule and could provide the basis for more comprehensive quality analysis. It was also found that excipients had an increasing effect on the contents of detected alkaloid and organic acid components, which may be beneficial to the effectiveness of the capsules.
