1.Reference range of thyroid hormones in Tibetan adults in Baingoin County, Nagqu City

Objective:To conduct a survey and study on thyroid hormone levels in Tibetan adults in Baingoin County, Nagqu City, analyse reference range of thyroid hormones, and provide a basis for subsequent diagnosis of thyroid diseases.Methods:According to the screening requirements of the National Academy of Clinical Biochemistry (NACB) Laboratory Medicine Application Guidelines, 193 local Tibetan adults in Baingoin County were selected as the survey subjects. Among them, 112 adults underwent physical examinations at the outpatient department of Baingoin County People's Hospital from May 2021 to August 2022, and 81 adults participated in rural physical examinations from June to August 2022. Using the Abbott i2000SR chemiluminescence immunoassay analyzer, the levels of thyroid stimulating hormone (TSH), total thyroxine (TT 4), total triiodothyronine (TT 3), free thyroxine (FT 4), and free triiodothyronine (FT 3) were determined. The reference range of thyroid hormones for local residents was established, and the correlation between thyroid hormone levels and age, as well as the differences between different townships were analyzed. Results:A reference range for thyroid hormones in Tibetan adults in Baingoin County, Nagqu City had been established (TSH: 0.53 - 6.16 μU/ml, TT 3: 1.07 - 2.28 nmol/L, FT 3: 3.30 - 5.86 pmol/L, TT 4: 60.68 - 129.96 nmol/L, and FT 4: 9.74 - 16.29 pmol/L). TSH level was positively correlated with age ( r = 0.16, P = 0.026), while TT 3, FT 3, TT 4, and FT 4 levels were negatively correlated with age ( r = - 0.21, - 0.23, - 0.16, - 0.26, P < 0.05). The comparison of TT 3, FT 3, TT 4 levels among 5 townships in Baingoin County showed statistically significant differences ( P < 0.05). Among them, Baoji Township was lower than the other four townships in the TT 4 level, and Deqing Township was higher than the other four townships in the FT 3 level ( P < 0.05). Conclusions:Successfully established a reference range for thyroid hormones in Tibetan adults in Baingoin County, Nagqu City. It is recommended to establish a reference range for thyroid hormones based on age groups and conduct more in depth investigations and studies on iodine nutrition and thyroid function in different townships.

CyTOF has been applied in many fields of medicine, including signaling pathways, growth and development, tumor microenvironments, autoimmune diseases, infections, immunotherapy, and organ transplantation. This article reviews recent progress in the application of CyTOF in the clinical diagnosis, treatment, and research of pediatric diseases to enhance its utility in diagnosis and treatment.

4.Effect of aerobic exercise on spontaneous brain activity in individuals with subthreshold depression based on fractional amplitude of low frequency fluctuations

Objective:To explore the regulation of eight weeks aerobic exercise on brain spontaneous activity in individuals with subthreshold depression.Methods:Through subject recruitment, a total of 44 subjects with subthreshold depression (subthreshold depression group, StD) who scored≥5 according to Patient Health Questionnare-9 (PHQ-9) but did not meet the diagnostic criteria for clinical depression, and 34 healthy subjects (health control group, HC) were included. Two groups of participants were given 8-week moderate-intensity aerobic exercise intervention, and resting state functional magnetic resonance imaging (rs-fMRI) data were collected before and after the intervention to analyze the fractional amplitude of low frequency fluctuations (fALFF) in the two groups. Horizontal two-sample t-tests with years of education as a covariate and longitudinal paired-samples t-tests were performed for fALFF in both groups at baseline and after 8-week moderate-intensity aerobic exercise intervention. The relationship between the fALFF values in significant difference brain regions and psycho-psychological scales were examined by Spearman correlation. Results:After 8 weeks of moderate intensity aerobic exercise, the scores of PHQ-9 and Self-Rating Anxiety Scale (SAS) in StD were significantly lower than baseline ( t=7.08,5.99, both P<0.001). There was no significant difference in fALFF value between StD and HC at baseline. After aerobic exercise intervention, compared with the HC, the fALFF value of the left parahippocampal gyrus in the StD was significantly decreased, and the fALFF value of the left superior frontal gyrus and left precentral gyrus was significantly increased. After aerobic exercise intervention, compared with the baseline, the StD had significantly higher fALFF values in the left inferior parietal lobe (supramarginal gyrus and angular gyrus), right supramarginal gyrus and left anterior cingulate gyrus. In addition, compared with the baseline, the fALFF value of the right precentral gyrus in the HC was significantly decreased. After aerobic exercise intervention, the fALFF value of the left inferior parietal lobe was negatively correlated with the SAS scores ( r= -0.31, P=0.038), the fALFF value of the left anterior cingulate gyrus and the PHQ-9 item 2 (low mood) scores was negatively correlated ( r= -0.41, P=0.006), and the fALFF values of the left inferior parietal lobe, right supramarginal gyrus and the PHQ-9 item 6 (low self-evaluation) scores were negative correlated ( r=-0.39, P=0.010; r=-0.30, P=0.048). Conclusion:Eight weeks of moderate-intensity aerobic exercise can regulate the spontaneous activity pattern of local brain regions in individuals with subthreshold depression, and fALFF analysis can provide objective evidence for the antidepressant effect of aerobic exercise.

The work explored the DEHP migration parameters in PVC infusion in clinic,based on the previous research on the test model of DEHP migrated from PVC infusion,to assess the safety of PVC infusion.The leaching solution samples in different conditions were evaluated by analysis of the DEHP in leaching solution using GC-MS under simulated clinical transfusion way.The release behavior of DEHP was significantly affected by the storage time,storage temperature,surrounding temperature,dripping speed,sterilization process,volume of the leaching solution,and the property of the leaching solution.

7.Research on Experimental Model of Security Assessment of DEHP Migration from PVC-Tubes Medical Devices.

As we all know, DEHP is seriously harmful to human health and consequently has been acquired critical attention. DEHP is able to migrate from PVC medical devices for the non-chemically bound to PVC, thus contact with user and patient. The DEHP migration is influenced by various parameters. In order to assess the security of PVC-tubes medical devices scientifically of DEHP migration, we develop an experimental model by analyzing the parameters comprehensively and systematically, taking into account the clinical practices. For example, assessing the security of DEHP migration from infusion sets by utilizing this model.

8.Application of 3.5 mm superfine bronchoscope in children′s difficult airway

Objective To discuss the practicability and safety of 3.5 mm superfine bronchoscope in children's difficult airway.Methods Twenty-eight children with difficult airway,aged 4 months to 8 years were selected.Intravenous anesthesia was used and autonomous respiration was maintained.The 3.5 mm superfine bronchoscope was used to guide tracheal intubation by nose(mouth).Results Among 28 cases, 1 case had oral bleeding and succeeded after suction and pressurized oxygen supply, and the rest was successful in the first time.The intubation time was(1.50 ± 0.25)min.There was no damage to the whole airway.Success rate was 100%.Conclusions The 3.5 mm superfine bronchoscope in guiding tracheal intubation of children's difficult airway has obvious advantages and security, and it deserves clinical attention and promotion.
9.Harringtonine induces apoptosis in NB4 cells through down-regulation of Mcl-1.

OBJECTIVETo investigate the growth inhibition effect, cytotoxicity and apoptotic induction of harringtonine (HT) in human acute promyelocytic leukemia (APL) NB4 cells,and the related mechanism.
METHODSNB4 cells were treated with HT. Total cell numbers were counted by hemocytometer, and cell viabilities were determined by trypan blue exclusion. Apoptotic cells were determined by fluorescence microscopy and FACS after staining with AO and EB or PI, respectively. The cleavage of PARP and the activation of Bax and the expression of anti-apoptotic proteins were determined by Western Blot. siRNA was used to silence the expression of target genes. Primary cells were isolated following Ficoll-Hypaque density gradient centrifugation method.
RESULTSHT inhibited cell growth and induced apoptosis of NB4 cells in a dose- and time-dependent manner. Apoptosis induced by HT was correlated with the down-regulation of Mcl-1 and the cleavage of PARP, while HT did not affect the protein level of Bax and Bak or change the protein level of Bcl-2. The silence of Bcl-XL sensitized HT-induced apoptosis in NB4 cells.Apoptosis induced by HT in primarily cultured APL cells was also correlated with the down-regulation of Mcl-1.
CONCLUSIONHT inhibits cell growth and induces apoptosis in NB4 cells and primarily cultured APL cells, which may be associated with down-regulation of Mcl-1.
