ObjectiveTo investigate the inhibitory effects and mechanisms of Yupingfeng San (YPFS) on high-mobility group box protein 1 (HMGB1)-induced angiogenesis in liver cancer. MethodsAn orthotopic liver cancer model was established in 30 C57BL/6 mice. The mice were randomly divided into a model group, YPFS low-, medium-, and high-dose groups (20, 30, 40 g·kg-1, respectively), a positive control group (sorafenib, 10 mg·kg-1), and a normal control group. After 14 days of medication, tumor and adjacent tissues were harvested. Tumor inhibition rates were calculated based on tumor weight. Microvascular density was determined via immunohistochemistry. HMGB1 and vascular endothelial growth factor (VEGF) expression levels were determined using immunofluorescence. An HMGB1-induced human umbilical vein endothelial cell (HUVEC) proliferation model was established, and cell proliferation was evaluated by a methylthiazolyldiphenyl-tetrazolium bromide (MTT) assay. HUVECs were randomly divided into a blank group, a model group (HMGB1 800 μg·L-1), along with YPFS low-, medium-, and high-dose groups (HMGB1 800 μg·L-1 + YPFS 10, 30, 50 g·L-1, respectively). HUVEC migration ability was evaluated via a scratch assay and a Trans well chamber assay. Tube formation ability was detected by a tube formation assay. Microvascular sprouting ability was evaluated by an aortic ring assay. Furthermore, RAGE/MEK1/2/ERK1/2 pathway-related protein expression in HUVECs was measured using Western blot (WB). ResultsCompared with the model group, all three doses of YPFS significantly reduced tumor weight in mice with orthotopic liver cancer (P<0.01), with inhibition rates of 31.49% ± 8.27%, 49.60% ± 3.77%, and 56.81% ± 9.70%, respectively. Also, YPFS significantly decreased microvascular density in tumor and adjacent tissues (P<0.05), as well as VEGF and HMGB1 expression levels (P<0.01). Compared with the blank group, HMGB1 promoted HUVEC proliferation, migration, tube formation, and microvascular sprouting in aortic rings (P<0.05). After YPFS administration, the HMGB1-induced proliferation rate, migration rate, number of tube formations, and microvascular sprouting in aortic rings were significantly reduced compared with the model group (P<0.01). Additionally, YPFS downregulated receptor for advanced glycation end products (RAGE), mitogen-activated protein kinase kinase 1/2 (MEK1/2), phosphorylated MEK1/2 (p-MEK1/2), extracellular signal-regulated kinase 1/2 (ERK1/2), and phosphorylated ERK1/2 (p-ERK1/2) protein expression in HMGB1-induced HUVECs (P<0.01). ConclusionYPFS exerts anti-angiogenic effects against HMGB1 induction by suppressing the RAGE/MEK/ERK signaling pathway, thereby inhibiting liver cancer tumor growth.

The purpose of this study is to investigate the effect of TPCK trypsin on the proliferation pattern of porcine epidemic diarrhea virus in Vero cells. The TPCK trypsin and conventional tryp-sin were added for virus proliferation, and RT-qPCR technology was used to analyze the changes in virus adsorption and invasion in Vero cells. The replication ability of porcine epidemic diarrhea vi-rus in Vero cells was explored through growth curve drawing,IFA identification, and cell activity detection. The results showed that the optimal concentrations of TPCK trypsin and conventional trypsin were 1 mg/L and 6 mg/L,respectively.The virus showed a decreasing trend with the pro-longation of TPCK trypsin and conventional trypsin pretreatment time.Adding different pancreatic enzymes during the virus proliferation process did not promote the virus invasion in Vero cells.Af-ter 4 h of invasion, the virus particles of each group gradually increased.By plotting the growth curve,it was found that the virus content of the TPCK trypsin group reached its highest level at 24 h(lgTCID50=(6.30±0.14)/0.1 mL),followed by a decreasing trend at 36 h,and the fluorescence intensity produced at 24 h was higher than that of conventional trypsin. In summary,TPCK trypsin has a better promoting effect on the proliferation of porcine epidemic diarrhea virus in Vero cells. It provided theoretical basis for further research on the mechanism of TPCK trypsin affecting porcine epidemic diarrhea virus proliferation, and also provided data support for the isola-tion and purification of porcine epidemic diarrhea virus epidemic strains.

Immunological Journal 2024;40(5):425-432
This study performed to determine whether OX40L overexpressed by recombinant rabies virus(LBNSE-OX40L)can enhance the innate immune response through activation of dendritic cells and thus activate early antibody production.Bone marrow dendritic cells(BMDCs)were extracted from the femur of Balb/c mice and cultured for 6 days, and the cultured BMDCs were infected with the parental virus LBNSE and the recombinant virus LBNSE-OX40L with the multiplicity of infections(MOI)=1.The effect of each virus on the maturation of BMDCs was analyzed by flow cytometry;ELISA was used to detect the expression of innate immunity-related cytokines such as interferon-α(IFN-α)and interleukin-12p40(IL-12p40)in the supernatants of the infected BMDCs.For in vivo study,Balb/c female mice were injected intramuscularly with 106 FFU of parental virus LBNSE and recombinant virus LBNSE-OX40L in both hind limbs, and the inguinal lymph nodes of mice were collected on day 6 after immunization, and the proportion of mature DCs was detected by flow cytometry. The serum was collected on day 6 after immunization, and the content of virus-neutralizing antibody(VNA)was detected by antiviral neutralizing antibody titration.Mouse serum was collected on day 6 after immunization, and virus neutralizing antibody content was measured by titration of antiviral neutralizing antibody, while IgG antibody in mouse serum was detected by ELISA.IgM antibody subclasses were detected by ELISA on days 2,4,and 6 after immunization.Compared with the parental virus LBNSE,the recombinant virus LBNSE-OX40L was able to activate more BMDCs in vitro and produce significantly higher levels of IFN-α and IL-12p40.Furthermore,the recombinant virus LBNSE-OX40L stimulated the maturation and differentiation of the DCs in vivo, which led to the rapid production of high levels of VNA and RABV-specific IgG and IgM antibodies.Taken together,LBNSE-OX40L activates dendritic cells to promote the body's innate immune response, and in turn enhances early antibody production, thus can be an early effective rabies vaccine candidate.

Objective:To explore the relationship between thyroid parenchymal echogenicity abnormalities and thyroid function evolution in health examination population. Methods:It was a cohort study. According to the inclusion and exclusion criteria, participants were selected from individuals who underwent thyroid color ultrasound and thyroid function tests at the Health Management Center of Tianjin Medical University General Hospital from January to December 2017. Data including age, gender, smoking history, alcohol consumption history, personal medical history, and thyroid function re-examination information were collected. Follow-up was conducted until the occurrence of thyroid function abnormalities or until the end of the follow-up period. The cumulative incidence rate and incidence density of thyroid function abnormalities were evaluated. Cox regression analysis was used to analyze the relationship between thyroid parenchymal echogenicity abnormalities and thyroid function evolution. Results:A total of 6 754 participants were included in this study, with an average age of (45.80±12.12) years, and females accounted for 42.7%. The mean follow-up time was 1.82 years, with a cumulative follow-up duration of 12 263 person-years. During the follow-up period, 154 new cases of thyroid function abnormalities occurred, with a cumulative incidence rate of 2.28% (95% CI: 1.94%-2.66%) and an incidence density of 12.56/1 000 person-years (95% CI: 10.66/1 000-14.69/1 000 person-years). The multivariate adjusted Cox regression analysis showed that individuals with thyroid parenchymal echogenicity abnormalities had a significantly increased risk of developing isolated thyroid stimulating hormone abnormalities and thyroid function abnormalities, with HR(95% CI) of 3.09 (2.02-4.73) and 2.92(1.96-4.33), respectively, both P<0.001. Stratified analysis showed that, except for body mass index <18.5 kg/m 2, current smoking, and current alcohol consumption, all other stratified factors showed a significant increase in the risk of thyroid function abnormalities in individuals with thyroid parenchymal echogenicity abnormalities. Conclusion:Thyroid parenchymal echogenicity abnormalities are important risk factors for the evolution of thyroid function abnormalities. Continuous monitoring of thyroid function should be given due attention.
