This study aims to prepare nanobodies for the epitopes of spike protein(S)of porcine ep-idemic diarrhea virus(PEDV),and verify its reactivity. The expression vector pCZN1-SN was con-structed by the prokaryotic expression system, and SN fragment was expressed in prokaryotic ex-pression and purified by Ni-NTA chromatography column. The SN nanobodies were displayed u-sing phage display technology and screened from the natural nanobody library. The results showed that SN fragments with a size of 26 kDa were obtained by prokaryotic induction, and the specific nanobodies were obtained through three rounds screening by phage display technology. One strain with the best reactivity was selected for prokaryotic expression and purified. The nanobodies were obtained with a size of 14 kDa by Western blot and demonstrated to have a good binding ability to PEDV SN protein. In summary, nanobodies with epitope fragments of PEDV S protein were suc-cessfully screened and prepared based on the phage display technology, which provided a new way for the in-depth application of nanobodies.

Swine enteric coronaviruses(SeCoV),such as porcine epidemic diarrhea virus(PEDV),transmissible gastroenteritis virus(TGEV),and porcine delta coronavirus(PDCoV),cause severe diarrhea in piglets, resulting in substantial losses in pig farming. In this study we establish a triple fluorescence reverse transcription-quantitative PCR(RT-qPCR)method for the simultaneous de-tection of PEDV,TGEV,and PDCoV.The specific primers and probes for each target virus were designed based on conserved sequences from the PEDV M gene, the TGEV ORF 1b gene, and the PDCoV ORF 1b gene respectively.Following the optimization of parameters and conditions,a triple RT-qPCR method was successfully established to simultaneously detect PEDV,TGEV,and PD-CoV.The developed assay exhibits strong specificity for these three pathogens without any cross-reaction with other common porcine viruses like CSFV,PCV2,PoRVA,PRV,and PRRSV.The de-tection limit of linear templates for pTOPO-PEDV 128,pTOPO-TGEV 116,and pTOPO-PDCoV 125 recombinant plasmids were 16.835,17.610 and 17.020 copies/μL,respectively.The intra group and inter group coefficients of variation were less than 5%,with no significant differences observed(P>0.05).Moreover,the detection consistency rate of the developed RT-qPCR was compared with standard method and showed 100%agreement.Out of 35 small intestine tissue samples,17 tested positive for PEDV,resulting in a positive rate of 48.57%(17/35).The tests for TGEV and PDCoV yielded negative results, and no mixed infections were detected.Based on the above results, the tri-ple RT-qPCR method established is specific, sensitive, stable, and rapid, and can be used for clinical detection and differential diagnosis of PEDV,TGEV,and PDCoV simultaneously, providing a method for the detection and epidemiological investigation of porcine diarrhea coronaviruses.

Objective Based on the VOSviewer software,to systematically analyze and compare the trends and hotspots in existing Chinese and English research on kinesiophobia, aiming to provide references for research in the field of clinical intervention and rehabilitation. Methods Chinese literature data sources were selected from CNKI,Wanfang, and VIP databases, while English literature data sources were selected from PubMed and Web of Science databases.Relevant literature on kinesiophobia was selected, and the VOSviewer1.6.18 software was used to analyze the basic information materials of the literature and to draw maps. Results 760 Chinese literature articles and 4491 English literature articles were included. The peak of publication volume was concentrated between 2020 and 2024.Domestically,the overall publication volume, individual author publication volume, and team collaboration among authors were relatively lower compared to international standards. The common high-frequency keywords in both domestic and international research were"kinesiophobia, pain, rehabilitation exercise, disability, anxiety,"and the frequency of these keywords showed correlation and non-randomness.Conclusion Research on kinesiophobia is on an upward trend.Due to different development focuses, there are similarities and differences in research directions between domestic and international studies.Domestic research can achieve in-depth exploration of kinesiophobia through theoretical integration, technological transformation, population expansion, and methodological innovation.

Objective:To explore the turnover intention of dental outpatient nurses, and analyze its influencing factors. Methods:From August to September 2021, multi-stage sampling was used to select nurses from 20 stomatology hospitals in 7 geographical regions of North China, East China, South China, Central China, Southwest, Northeast, and Northwest China. The survey was conducted using the General Information Questionnaire, Turnover Intention Scale, Nursing Work Environment Scale, Psychological Capital Scale for Nurses, and Work Engagement Scale. Results:A total of 1 513 questionnaires were collected, including 1 135 valid questionnaires, with a valid rate of 75.02% (1 135/1 513). The total score of turnover intention among 1 135 dental outpatient nurses was (2.22±0.56). Pearson correlation analysis showed a negative correlation between nursing work environment, psychological capital, work engagement, and turnover intention ( r=-0.421, -0.340, -0.446; P<0.01). The structural equation model showed that the nursing work environment had a direct negative effect on the turnover intention of dental outpatient nurses (β=-0.401, P<0.01) and had an indirect negative effect on turnover intention through work engagement (β=-0.106, P<0.01), with a mediating effect of 20.91%. Psychological capital had an indirect negative effect on the turnover intention of dental outpatient nurses through work engagement (β=-0.271, P<0.01), with a mediating effect of 52.83%. Conclusions:The turnover intention of dental outpatient nurses is at a high level. Managers can reduce the turnover intention of dental clinic nurses by improving the nursing work environment, developing psychological capital, and increasing work engagement.

Objective:To construct the prediction model of knee degeneration in patients with knee osteoarthritis based on bioelectrical impedance index, and evaluate the prediction performance and application efficiency of the model. Methods:This was a cross-sectional study. From May to July 2021, 248 knee joints of 124 patients with knee osteoarthritis at home from Shijiazhuang Yuqiang Community Health Service Center who participated in physical examination were selected by convenience sampling to establish the model. According to Kellgren-Lawrence (K-L) grading system, the knee joints were divided into four groups, namely K-L1 ( n=19) , K-L2 ( n=103) , K-L3 ( n=96) , and K-L4 ( n=30) . The indicators included in the model were selected through analysis of variance or Kruskal-Wallis test, and a prediction model of knee degeneration was established using support vector machine, and the model was optimized using grid parameter optimization method. The prediction performance of the model was evaluated by the area under the receiver operating characteristic (ROC) curve, sensitivity, specificity, accuracy, positive predictive value and negative predictive value. Results:The indicators in the model included age, complications, lumbar/back/hip pain, high-risk occupation, Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC) -pain, WOMAC-function, capacitive reactance and phase angle. The area under the ROC curve of the training set model was 0.999, the prediction accuracy was 0.920, and the 95% confidence interval was 0.868 to 0.957. The area under the ROC curve of the test set model was 0.833, the prediction accuracy was 0.682, and the 95% confidence interval was 0.572 to 0.780.Conclusions:The prediction model of knee degeneration has good prediction performance and is easy to use, which can be used as a screening tool for knee degeneration in patients with knee osteoarthritis.
