1.Research on dynamic monitoring of drug consumption based on seasonal Mann-Kendall trend test

OBJECTIVE To investigate a dynamic monitoring of drug consumption (DMDC) model based on the seasonal Mann-Kendall trend test, aiming to provide scientific evidence for the efficient and macroscopic monitoring of drug use. METHODS A monitoring list of key outpatient drugs was established based on the top 20% of drugs ranked by sales volume in the outpatient pharmacy in October 2024. A DMDC model based on the Mann-Kendall trend test was constructed using the monthly usage data of key outpatient drugs from November 2021 to October 2024, aiming to eliminate the impact of seasonal fluctuations and analyze the temporal trends in drug consumption. Taking mucolytic expectorants, triazole derivatives for dermatophytosis, and single-agent hydroxymethylglutaryl coenzyme A (HMG-CoA) reductase inhibitors as examples, the monitoring effectiveness of the DMDC model was demonstrated, and its performance was compared with that achieved by the traditional sequential growth rate ranking method. RESULTS A total of 215 drug varieties were included in the monitoring list, and DMDC models were successfully established for all of them. Among these, 119 showed a significant increasing trend (P<0.05, S′>0). The model successfully monitored the monthly consumption of mucolytic expectorants, triazole derivatives for dermatophytosis, and single- agent HMG-CoA reductase inhibitors. The precision and recall rates of the DMDC model for identifying abnormal drug use were 60.7% and 85.0%, respectively, both significantly higher than those of the sequential growth rate ranking method (8.3% and 15.0%, respectively) (χ2=20.114, P<0.001; χ2=19.600, P<0.001). CONCLUSIONS DMDC model based on the seasonal Mann-Kendall trend test can effectively identify long-term trends in drug consumption, eliminate seasonal interference, enhance monitoring accuracy and management efficiency, and is suitable for the dynamic monitoring of drug consumption.
2.IGSF11: A Novel Target for Cancer Immunotherapy.

Immune checkpoint blockade therapy has demonstrated remarkable efficacy in treating various malignancies; however, its clinical application remains challenged by low response rates and immune-related adverse events. Immunoglobulin superfamily member 11 (IGSF11), an inhibitory immune checkpoint molecule, serves as a specific ligand for the V-domain immunoglobulin suppressor of T cell activation (VISTA). Through the IGSF11/VISTA axis, it suppresses T cell function and represents a promising novel target for cancer immunotherapy. IGSF11 is widely expressed across multiple tumor types, though its regulatory mechanisms vary depending on the malignancy. Studies have confirmed that blocking the IGSF11-VISTA interaction or specifically inhibiting IGSF11 exerts antitumor effects. While IGSF11 is closely associated with patient prognosis, its prognostic significance differs among cancer types. This review systematically summarizes the structural characteristics of IGSF11, its regulatory mechanisms, interaction with VISTA, and functional role within the tumor microenvironment.

3.Correlation between quantitative airway parameters and disease severity of coal workers' pneumoconiosis based on HRCT

Objective:To observe the changes of small airway parameters in patients with coal workers' pneumoconiosis in different disease stages by high resolution computed tomography (HRCT) , and analyze the correlation between them and the severity of the disease.Methods:From June 2016 to June 2023, 25 healthy volunteers and 71 untreated patients with coal worker's pneumoconiosis in the Fifth People's Hospital of Ningxia were selected as the research objects. The clinical and imaging data of the patients were collected, and the disease stages were performed according to the dust exposure history and high-kilovolt chest X-ray. The patients were divided into 4 groups: control group (25 cases) , coal workers' pneumoconiosis stage Ⅰ group (17 cases) , coal workers' pneumoconiosis stage Ⅱ group (32 cases) and coal workers' pneumoconiosis stage Ⅲ group (22 cases) . Quantitative chest HRCT parameters of each group were collected, including the square root of wall area at 10 mm inner perimeter (AWT-Pi10, Pi10) , airway wall thickness, airway wall volume, airway wall area percentage of the whole lung and the 5th, 6th, 7th and 8th level airways, and low attenuation area percentage (LAA%) of the whole lung. Pulmonary function indicators were collected, including forced expiratory volume in 1 second (FEV 1) and the percentage of its projected value [FEV 1 (%pred) ], the ratio of FEV 1 to forced vital capacity (FEV 1/FVC) and the percentage of its projected value[FEV 1/FVC (%pred) ]. One-way ANOVA or Kruskal-Wallis H test and Spearman rank correlation were used to analyze the difference and correlation. Results:Compared with control group, FEV 1, FEV 1 (%pred) , FEV 1/FVC and FEV 1/FVC (%pred) in stage Ⅱ and Ⅲ coal workers' pneumoconiosis groups were lower ( P<0.05) . In addition, the FEV 1 and FEV 1 (%pred) of the stage Ⅲgroup were lower than those of the stageⅡ group ( P<0.05) , and the FEV 1/FVC and FEV 1/FVC (%pred) of the stage Ⅲgroup were lower than those of the stage Ⅰgroup ( P<0.05) . Compared with stage Ⅰ group, Pi10 in stage Ⅲ group were increased ( P < 0.05) at the 6th and 8th level airways, and airway wall thickness and airway wall volume in the 6th, 7th and 8th level airways of stage Ⅲgroup increased ( P<0.05) . Correlation analysis showed that all pulmonary function indexes were negatively correlated with Pi10 of whole lung and the 6th, 7th and 8th level airways ( P<0.05) , all pulmonary function indexes were negatively correlated with airway wall thickness of the 7th and 8th level airways ( P<0.05) , and FEV 1/FVC (%pred) was negatively correlated with airway wall volume of the 7th and 8th level airways ( P<0.05) . FEV 1, FEV 1 (%pred) , FEV 1/FVC (%pred) were negatively correlated with percentage of airway wall area of whole lung and the 6th, 7th and 8th level airways ( P<0.05) . Conclusion:The quantitative airway parameters of coal workers' pneumoconiosis based on HRCT are correlated with pulmonary function indexes, which can reflect the severity of coal workers' pneumoconiosis.
4.Electrotactic migration and mechanisms of stem cells

BACKGROUND:With the deepening of research on stem cell technology,how to make its accurate homing has become a major problem in clinical application.In addition to the induction of drugs and chemokines,electric fields are also widely used to guide the directional migration of stem cells.They can enhance their migration speed and orientation.OBJECTIVE:To summarize the effect of an electric field on the characteristics of stem cell migration and analyze the possible mechanism of action.METHODS:PubMed and CNKI databases were searched to collect relevant literature up to March 2024,with Chinese and English search terms"stem cells,direct current electric field,pulsed electric field,migration,electric field device,mechanism."Literature that was not available in full text and unrelated to the topic was excluded.RESULTS AND CONCLUSION:A total of 58 articles were included according to the screening requirements,including 15 Chinese articles and 43 English articles.In the literature,the effects of different parameters of the electric field on the migration of adipose-derived mesenchymal stem cells,bone marrow mesenchymal stem cells,neural stem cells,epidermal stem cells,human embryonic stem cells,and human induced pluripotent stem cells and its mechanism were studied in a migration device.(1)As a simple,non-invasive,and stable intervention method,the electric field plays an active role in guiding the directional migration of stem cells.(2)Different types of stem cells had different directions of electrotaxis migration,and the migration speed and directionality of most stem cells increased with the increase of electric field intensity.(3)Different electric field devices have different focuses in observing stem cell migration,and the relevant devices can be selected according to the purpose of the experiment.(4)The mechanism of electrotaxis migration of different stem cells is not completely the same.MAPK pathway,ROCK activation,and PI3K function are involved in the migration process of most stem cells,and other protein complexes and signaling pathways are involved in the regulation of this process.(5)In addition to different electric field parameters,cell senescence and culture environment also affect the results of electrotaxis migration.In summary,as an important signal that affects the migration characteristics of stem cells,the application of electric field combined with other emerging materials has shown certain potential in tissue engineering.It is expected to play a more important role in guiding stem cells to home,promoting bone tissue regeneration and repair,and making greater breakthroughs in the research of the nervous system,autoimmune system,tumors,and other diseases.
5.The predictive value of preoperative CALLY index for the short-term prognosis in patients undergoing radical gastrectomy

Objective To explore the predictive value of the preoperative C-reactive protein-albumin-lymphocyte ratio(CALLY index)for short-term prognosis in patients undergoing radical gastrectomy.Methods The clinical data of 228 gastric cancer patients who underwent radical gastrectomy were retrospectively analyzed.Based on follow-up results,patients were divided into the good prognosis group(n=205)and the poor prognosis group(n=23).The differences in clinical characteristics and laboratory test results between the two groups were compared.Cox proportional hazards regression model was used to analyze the impact of CALLY index on poor prognosis in patients.The receiver operating characteristic(ROC)curve was used to determine the clinical value and optimal cutoff value of CALLY index in predicting poor prognosis of patients.Kaplan-Meier method was used to analyze the difference in disease-free survival(DFS)between patients with different CALLY index levels.Results After a follow-up of 3 to 53 months(median follow-up of 36 months),23 patients developed poor prognosis(including 3 deaths and 20 recurrences or metastases).Compared with the good prognosis group,tumors in the poor prognosis group were larger,the proportion of poorly differentiated tumors was higher and white blood cell count(WBC),neutrophil count(NEU),C-reactive protein(CRP)were higher,and lymphocyte(LYM),albumin(ALB),prealbumin,total cholesterol(TC)and CALLY indexl were lower.Multivariate Cox regression analysis showed that the elevated CALLY index(HR=0.306,95%CI:0.102-0.921,P=0.035)was an independent protective factor for poor prognosis,while larger tumors and poorly differentiated tumors were independent risk factors for poor prognosis.ROC curve analysis showed that the area under the curve for predicting poor prognosis in gastric cancer patients undergoing radical gastrectomy was 0.862(95%CI:0.810-0.904),with a sensitivity of 95.65%,specificity of 69.76%,and an optimal cutoff value of 2.21.Kaplan-Meier analysis showed that patients with a CALLY index>2.21 had a longer disease-free survival than those with a CALLY index≤2.21(mean DFS:48 months vs.42 months,Log-rank χ2=4.123,P=0.042).Conclusion The preoperative CALLY index is a simple,effective and easily accessible predictor for predicting short-term prognosis in patients undergoing radical gastrectomy.A high CALLY index indicates a better prognosis of patient.
6.Study on the mechanism of Actinidia Chinensis Planch Radix in treating colorectal cancer based on network pharmacology

Objective To explore the medicinal mechanism of Actinidia Chinensis Planch Radix(ACPR)in the treatment of colorectal cancer(CRC)by network pharmacology and molecular docking technology.Methods The genes involved in the effects of the main chemical components and disease genes of ACPR were screened from the TCM database and disease database.The main genes were analyzed through protein interaction network analysis,and molecular docking was performed on the main chemical components and key targets.The effects of the drug on tumor cells were measured,and the levels of key proteins in the signaling pathway were detected.Results The primary components of ACPR for treating CRC include quercetin,β-sitosterol,aloe baicalin,and catechin.It targets 144 protein interaction sites and were involved in the protein interaction network,with key genes including AKT1,TP53,MAPK1,JUN,and TNF.The recognition network includes five modules that were involved in various biological processes and signaling pathways.The main components exhibited excellent or good activity when interacting with these targets.At a certain concentration,the drug could inhibit the proliferation,invasion,and migration of colorectal cancer cells and affect the PI3K/AKT signaling pathway.Conclusion ACPR has been used to treat colorectal cancer through multiple pathways and multiple targets,among which the PI3K/AKT signaling pathway may be the mechanism.
7.Comparative Study of International Medication Reconciliation Guidelines and Improvement Strategies in China

Medication reconciliation plays a key role in improving patient medication safety,reducing inappropriate polypharmacy,and promoting the high-quality development of pharmaceutical services.Compared to advanced international guidelines,China's medication reconciliation service standards have deficiencies in areas such as definition and process design,and multidisciplinary team building.There is a need to establish a comprehensive medication reconciliation effect evaluation index system,develop pharmacist-led multidisciplinary teams,promote the advancement of artificial intelligence and big data technologies,and strengthen outpatient and community medication reconciliation coverage,thereby contributing to the high-quality development of pharmaceutical services in China.
8.Analysis of hematological characteristics of patients with three common deletional β-thalassemias and concomitant α-thalassemia in Huizhou, Guangdong province

Objective:To analyze the hematological characteristics of patients with three common deletional β-thalassemias (β-thal) and concomitant α-thal in Huizhou, Guangdong province.Methods:A total of 1 335 subjects of childbearing age with hemoglobin F (Hb F) ≥5% at the Huizhou First Maternal and Child Health Care Hospital between June 2014 and December 2023 were enrolled as our study cohort. The hematological parameters were determined by blood cell counters and automatic capillary electrophoresis, while liquid phase chip and gap-PCR were employed for the detection of routine thalassemias and the three common deletional β-thal, respectively. The hematological characteristics of patients with the deletional β-thal were analyzed. This study was reviewed and approved by the Ethics Committee of Huizhou First Maternal and Child Health Care Hospital [Ethics No. 20231107(B2)].Results:① A total of 384 cases of the three common deletional β-thal were identified, including 184 cases of Chinese Gγ + ( Aγδβ) 0, 191 cases of Southeast Asian hereditary persistence of fetal hemoglobin (SEA-HPFH), and nine cases of Taiwanese, for a total detection rate of 28.76%. ② Patients who did not meet the established criteria were excluded from the study, leaving 372 cases. All of which presented with hypochromic microcytic anemia and significantly elevated Hb F. Except for normal or decreasing of Hb A 2 levels in patients with Chinese Gγ + ( Aγδβ) 0, the levels of Hb A 2 in patients with the other two deletional β-thal were increased with different degrees. Differential comparison results showed that significant differences were observed in Hb A 2 and Hb F values among the groups of the three common deletional β-thal heterozygotes ( P<0.05). ③ According to the type of gene variation, 180 patients with Chinese Gγ + ( Aγδβ) 0 heterozygotes were divided into three groups, including αα/αα, Chinese Gγ + ( Aγδβ) 0/β N (149), -α/αα, Chinese Gγ + ( Aγδβ) 0/β N (14), and --/αα, Chinese Gγ + ( Aγδβ) 0/β N (17). Similarly, 179 patients with SEA-HPFH heterozygotes were divided into three groups, including αα/αα, SEA-HPFH/β N (150), -α/αα, SEA-HPFH/β N (12), and --/αα, SEA-HPFH/β N (17). Differential comparison results showed that the Hb F levels of the Chinese Gγ + ( Aγδβ) 0 combined with α 0-thal group were significantly lower than those of the Chinese Gγ + ( Aγδβ) 0 combined with α + -thal group and the control group ( P<0.05). The mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), and Hb F values of the SEA-HPFH combined with α 0-thal group were significantly lower than those of the SEA-HPFH combined with α + -thal group and the control group ( P<0.05). Conclusion:The above research results can not only enhance the ability of clinicians to identify deletional β-thal and concomitant α-thal, improve the level of genetic counseling, but also provide data support for the development of deletional β-thal prevention and control programme and the development of prenatal and postnatal care.
9.Role and mechanism of resveratrol in delaying exercise-induced fatigue

BACKGROUND:Resveratrol is a naturally occurring polyphenolic compound in plants,recognized for its anti-inflammatory,antioxidant,and antiproliferative properties.Its role in preventing and treating various chronic diseases has been extensively documented.Recent evidence suggests that resveratrol may help delay exercise-induced fatigue,although the underlying mechanisms remain to be systematically elucidated.OBJECTIVE:To explore the possible role and mechanism of resveratrol in delaying exercise-induced fatigue,based on the mechanism of exercise-induced fatigue,in order to provide a theoretical basis for the research and application of resveratrol in the field of sports nutrition.METHODS:Chinese and English search terms were"resveratrol,exercise-induced fatigue,oxidative stress,inflammation,energy substance,muscle injury,mitochondrial quality,neurotransmitter."CNKI,WanFang Data,and PubMed were searched for relevant research literature published from January,1992 to August 2024.A total of 73 core related articles were obtained according to inclusion and exclusion criteria.RESULTS AND CONCLUSION:(1)Resveratrol is a naturally occurring polyphenolic compound commonly found in plants such as grape skins,berries,and peanuts.It exhibits multiple biological activities,including antioxidant,anti-aging,anticancer,and anti-inflammatory properties.Resveratrol shows potential for preventing and treating various chronic diseases and is also thought to delay the onset of exercise-induced fatigue.(2)Exercise-induced fatigue is a non-pathological fatigue phenomenon with a complex mechanism.It is related to peripheral mechanisms such as the depletion of energy substrates,the accumulation of metabolic by-products,inflammation,and oxidative stress,as well as central mechanisms,including protective inhibition by the central nervous system and neurotransmitter imbalances.(3)Resveratrol activates the kelch-like epichlorohydrin-associated protein 1/nuclear factor erythroid 2-related factor 2 pathway,upregulating antioxidant-related genes such as catalase and superoxide dismutase while downregulating genes such as malondialdehyde and 4-hydroxy-2-nonenal,ultimately reducing oxidative stress.Resveratrol also activates silent information regulator 1,inhibits the nuclear factor-kappaB and nucleotide-binding oligomerization domain-like receptor thermal protein domain associated protein 3 inflammasome pathways,and downregulates pro-inflammatory cytokines such as tumor necrosis factor-α,interleukin-6,and interleukin-1β,alleviating inflammation.Additionally,resveratrol increases short-chain fatty acid production by the gut microbiota,elevates the lactate/pyruvate ratio in the liver to promote gluconeogenesis,and enhances fatty acid oxidation,thereby improving energy substrate utilization.Furthermore,through silent information regulator 1/peroxisome proliferator-activated receptor-y coactivator-1α/nuclear respiratory factor 1 activation,resveratrol enhances mitochondrial biogenesis in skeletal muscle,promotes mitochondrial fusion and fission for optimal dynamics,and activates mitophagy,improving mitochondrial quality.It also diminishes plasma tryptophan levels,decreases excessive serotonin secretion in the brain,increases dopamine release,and reduces glutamate toxicity,helping regulate neurotransmitter imbalances.Moreover,resveratrol lowers plasma lactate and ammonia levels,promoting metabolic by-product clearance,thereby delaying the onset of exercise-induced fatigue.(4)While current evidence indicates that resveratrol is effective in delaying exercise-induced fatigue,the optimal dose,timing,and duration of supplementation remain uncertain.Moreover,the interactions between resveratrol and other supplements,such as caffeine and vitamins,remain underexplored.Thus,future research should focus on optimizing supplementation strategies for resveratrol.
10.Visualization analysis of macrophage polarization in tissue repair process

BACKGROUND:During tissue repair and regeneration,macrophages exhibit multiple activities such as promoting inflammation,anti-inflammation,fibrosis,and wound healing at various stages of tissue damage.The heterogeneity and balanced polarization of macrophages are decisive in organ repair. OBJECTIVE:To explore the research hotspots and development trends in the field of macrophage polarization in tissue repair through visualization analysis methods,as well as the research level of global scientific and clinical workers in this field. METHODS:Using bibliometric analysis methods,this study employed Citespace literature visualization analysis software and VOSviewer tools,retrieving related literature from 2013 to 2023 in the Web of Science Core Collection's Science Citation Index Expanded(SCI-Expanded)and Social Sciences Citation Index Expanded(SSCI-Expanded)databases.The analysis results were presented in a dynamic map format,revealing the main trends and focuses of the research. RESULTS AND CONCLUSION:The number of publications in this field had dramatically increased from 2013 to 2023,with a significant rise starting in 2017.Chinese researchers had the highest number of publications,with 642 papers,while American researchers began focusing on this field early on.Professor Elisseeff Hennifer H had made a substantial contribution to the research in this area.Shanghai Jiao Tong University had produced the most publications.In recent years,keywords such as"hyaluronic acid"and"regulation"had been prevalent.Macrophage polarization research in tissue repair primarily concentrates on its multifunctional regulatory mechanisms,interactions with other cell types,and its behavior under specific pathological conditions.The main research areas include the role of macrophages in wound healing,cardiovascular diseases,chronic inflammation,tumor microenvironments,and regenerative medicine.A deeper understanding of the multifunctionality and polarization mechanisms of macrophages can lead to the development of new therapeutic strategies to enhance tissue repair and regeneration,thereby improving patient treatment outcomes.
