Network pharmacology has gained widespread application in drug discovery, particularly in traditional Chinese medicine (TCM) research, which is characterized by its "multi-component, multi-target, and multi-pathway" nature. Through the integration of network biology, TCM network pharmacology enables systematic evaluation of therapeutic efficacy and detailed elucidation of action mechanisms, establishing a novel research paradigm for TCM modernization. The rapid advancement of machine learning, particularly revolutionary deep learning methods, has substantially enhanced artificial intelligence (AI) technology, offering significant potential to advance TCM network pharmacology research. This paper describes the methodology of TCM network pharmacology, encompassing ingredient identification, network construction, network analysis, and experimental validation. Furthermore, it summarizes key strategies for constructing various networks and analyzing constructed networks using AI methods. Finally, it addresses challenges and future directions regarding cell-cell communication (CCC)-based network construction, analysis, and validation, providing valuable insights for TCM network pharmacology.

Soy is a vital source of plant carbohydrates. However,it poses significant allergenic risks, particularly to young children and animals.Among the various proteins in soy,β-conglycinin,which con-stitutes approximately 30%of total soy carbohydrates,is a primary allergen.Undigested β-conglycinin can lead to intestinal damage by inhibiting cell growth, disrupting the cytoskeleton, and inducing apopto-sis.It can also enter the lymphatic and circulatory systems, triggering allergic reactions.Conventional ELISA methods for detecting β-conglycinin rely on polyclonal or monoclonal antibodies, which are limited by their large molecular weight, difficulty in accessing the protein core, and sensitivity to acidic and bas-ic conditions. To address these limitations, this study aimed to develop nanobodies(Nbs)against β-con-glycinin.Nbs,derived from the variable regions of heavy-chain antibodies found in camelids, have a mo-lecular weight approximately one-tenth that of conventional antibodies. They offer advantages such as small size, stable structure, high specificity, and strong affinity.A female alpacas was immunized five times using β-conglycinin,which showed a heavy chain antibody potency of 1∶16 000 by ELISA.Pe-ripheral blood lymphocytes were subsequently isolated and total RNA was extracted. The variable region of the heavy-chain antibody was amplified via PCR,and recombinant plasmids were constructed and transformed into the E.coli competency strain ER2738.The resulting library contained about 3.5×108 CFU/mL,which increased to 1.15×1012 PFU/mL after phage rescue, with a 100%Nbs gene insertion rate, indicating high diversity. Its Nbs phage output was significantly enriched by four rounds of solid-phase elution with an enrichment rate of 155.9.Four rounds of solid-phase panning yielded 35 positive clones, all of which shared the same amino acid sequence upon sequencing. The selected Nb was ex-pressed in a prokaryotic system, and its binding ability to β-conglycinin was confirmed using Western blotting and ELISA.The results demonstrated excellent specificity and affinity. This research lays the groundwork for developing a rapid and efficient detection method for β-conglycinin using Nbs,potentially enhancing food safety and allergen management.

Objective To assess the efficacy of a novel inguinal tourniquet in healthy individuals and to investigate the relationship between localized inguinal compression and femoral artery blood flow occlusion. Methods A self-controlled study was conducted. From November 9 to November 30,2024,11 volunteers were recruited at the Third Medical Center of Chinese PLA General Hospital.Three compression methods--finger pressure,a novel groin tourniquet, and a SAM junction tourniquet(SJT)—were applied bilaterally to the inguinal region until distal blood flow signals disappeared. Each compression method was tested in 22 trials with a 5-minute interval between operations.Differences in hemostatic efficacy between bilateral inguinal regions and across compression methods were compared.Subsequently, the novel tourniquet was incrementally pressurized in 120 mmHg multiples using an integrated pressure device to analyze trends in popliteal artery blood flow velocity.Observational indicators included the internal pressure of the tourniquet pressurization device, peak systolic velocity(PSV)of popliteal artery, inguinal surface pressure magnitude, inguinal surface pressure distribution, and pain scores(assessed using a single-dimensional numerical rating scale). Results No statistically significant difference was observed in the minimum pressure required to occlude femoral artery blood flow bilaterally(P>0.05).The success rates of femoral artery blood flow occlusion at the inguinal region were 100%for the novel inguinal tourniquet,SJT,and finger pressure. The novel inguinal tourniquet induced the highest pain scores, ranging from 5 to 8.A significant reduction in PSV of popliteal artery was noted when the intra-tourniquet pressure reached 360 mmHg and 480 mmHg(P<0.05),with a 95%hemostasis efficacy observed within the range of 360-600 mmHg.No significant association was observed between the recovery of popliteal artery blood flow after limb movement and inguinal pressure distribution(P>0.05).The PSV of popliteal artery exhibited the strongest negative correlation with the average pressure within the inguinal compression area(r=-0.79,P<0.001),with a linear regression fitting line of y=69.69-0.13x(P<0.001,R2=0.58).Conclusions The novel inguinal tourniquet effectively occludes femoral artery blood flow within a pressure range of 360-600 mmHg,accompanied by moderate-to-severe pain. Its hemostatic mechanism mainly relies on increasing the mean pressure within the inguinal compression area.

Allantoin,as a functional constituent of yam,has an extremely important role in the medical and cosmetic fields. In this study, based on the Raman spectroscopy detection system constructed in the laboratory, the Raman spectra of the powder of allantoin standard and the surface-enhanced Raman spectra of the allantoin extract of fresh yam were analyzed, and the surface-enhanced Raman characteristic displacements of allantoin in raw yam were determined to be 644,1027 and 1398 cm-1. The effects of the adsorption time of allantoin and silver sol and the thickness of the yam on the intensity of Raman feature displacement were investigated, and a method was established to directly obtain the surface-enhanced Raman feature information of allantoin in fresh yam. Based on this method, the surface-enhanced Raman spectra of 32 raw yams were collected, and the Raman feature displacements of allantoin at 644,1027 and 1398 cm-1 were established by unary linear regression (ULR),multivariable linear regression (MLR),and partial least squares regression (PLSR). The results showed that the MLR model was the most effective, with the validation set coefficient of determination (R2V) of 0.93 and the root mean square error of validation (RMSEV) of 0.35 mg/g. However, the allantoin feature shift was susceptible to the changes of solution polarity and substrate, which led to a certain shift of the feature shift affecting the accuracy of the detection, and the quantitative prediction model of PLSR using the full-waveband Raman spectroscopy would improve the model's Robustness. The random frog (RF)-PLSR quantitative prediction model of allantoin was established based on the RF algorithm to screen the feature variables, and the R2V was increased to 0.96,and the RMSEV was reduced to 0.26 mg/g. The model was externally validated using ten raw yam samples which were not involved in the modeling, and the absolute value of maximum residual was 0.74 mg/g. The method could realize the rapid quantitative detection of allantoin content in raw fresh yam,and provided new ideas and technical references for the direct rapid quantitative detection of allantoin in agricultural products.
