1.Chemical elements pollution characteristics and health risk assessment of PM2.5 during heating season in Xinxiang university town

Objective To detect the PM2.5 pollution situation during heating season in Xinxiang university town, analyze the contents of 10 elements in PM2.5, and evaluate its health risks, so as to provide data support for the subsequent environmental governance of college town. Methods PM2.5 were collected during heating season. The contents of 10 elements were detected by ICP-MS after PM2.5 was digested. The source was analyzed using enrichment factor method. The ecological risk and health risk of PM2.5 was evaluated by applying the potential survival hazard index method and health risk assessment method. Results The average contents of Al, Mg, Mn, Cr, Zn, Se, Cu, Pb, Cd and As were respectively 165.59,203.37,7.75,328.93,133.61,8.24,30.82,7.09,2.77and 9.15 ng/m3. The average contents of Cr, Pb, and As all exceeded their air environment targets. The enrichment factors of Pb, Cu, As, Zn, Cd, Cr and Se were all more than 10, and their sources were affected by human activities. The potential ecological risk index of PM2.5 was 352.42, which was strong ecological hazard factor. PM2.5 had carcinogenic risk to human, and the risk in adults was higher than that in children. Conclusion During heating season in Xinxiang college town mainly, PM2.5 may have short-term health risks on environment and human. When the air quality is poor, it is recommended to reduce outdoor activities while taking personal protective measures to minimize the potential health risks to the population in the university town.
2.Effect of Electroacupuncture on TDP-43 and RhoA/ROCK2 Signaling Pathway in Mouse with Amyotrophic Lateral Sclerosis

Objective To observe the effects of electroacupuncture on RhoA/ROCK2 signaling pathway in cerebral cortex of mice with amyotrophic lateral sclerosis(ALS);To explore the mechanism of electroacupuncture in improving the motor function of ALS mice.Methods The male hSOD1G93A mice were divided into model group and electroacupuncture group,and wild-type male mice in the same litter were set as blank group,with 12 mice in each group.After the mice were 60 days old,"Baihui"and both side of"Tianzhu","Tianshu"were selected for electroacupuncture for 10 min per day,5 days of continuous treatment and 2 days off for 3 weeks.Rotating rod experiment and open field experiment were used to evaluate the motor function of mice,the damage of neurons in cerebral cortex was observed by Nissl staining,Western blot was used to detect the expressions of Tar DNA binding protein-43(TDP-43),tumor necrosis factor(TNF)-α,interleukin(IL)-1β,RhoA and ROCK2 protein in cerebral cortex.Immunofluorescence staining was used to detect the positive expressions of ion calcium binding adapter molecule 1(Iba-1)and glial fibrillary acidic protein(GFAP)in cerebral cortex.Results Compared with the blank group,the incubation period of rod turning and the total distance of open field movement in the model group were reduced(P<0.01),the neuron damage was obvious in the cerebral cortex,with Nissl body shrinkage and reduction in quantity(P<0.01),the relative expressions of TDP-43,TNF-α,IL-1β,RhoA and ROCK2 protein increased(P<0.01),and the positive expression of Iba-1 and GFAP increased(P<0.01).Compared with the model group,the incubation period of rod turning and the total distance of open field movement increased in electroacupuncture group(P<0.05),the damage of neuron in cerebral cortex was reduced,the number of Nissl bodies increased(P<0.05),the expressions of TDP-43,TNF-α,IL-1β,RhoA and ROCK2 decreased(P<0.05),and the positive expression of Iba-1 and GFAP reduced(P<0.05).Conclusion Electroacupuncture can improve motor function in ALS model mice,and the mechanism may be related to the inhibition of RhoA/ROCK2 signaling pathway activity and then relieve neuroinflammation.
3.Development of one kind of rapidly integrated detection vehicle for emergency

Frequent occurrence of disaster rescue and public health emergencies has increased demand for rapidly and efficiently emergent medical rescue.This study modified a closed van which equipped a rapid testing module for physiological indicators,clinical blood transfusion module,detection and sampling module of etiology,and module for health and epidemic prevention.This research designed one kind of emergent vehicle for rapidly comprehensive detection,which integrated test vehicle,blood transfusion vehicle,biosafety testing vehicle,and vehicle for health and epidemic prevention in one vehicle.This vehicle can meet various demands of emergently medical rescue and other tasks in clinical test,which can improve the response capability and emergency handling capability for disasters and public health emergencies,at the same time,it also can save manpower.
4.Perioperative nursing care of a child with Angelman syndrome and moderate scoliosis

Objective To summarize the perioperative nursing experience of a child with Angelman syndrome(AS)complicated with moderate scoliosis undergoing posterior 3-dimensional osteotomy correction,fusion,and internal fixation under general anesthesia.Methods The clinical data of the child with AS and moderate scoliosis who underwent surgical treatment in our hospital on Aug.4,2023,were analyzed.A multidisciplinary team was established upon admission.Relevant literatures and evidences were reviewed to develop and implement a"1+X"nursing plan,which included 2 components:"1"(core perioperative nursing priorities for scoliosis)and"X"(multiple AS-related nursing issues and corresponding strategies).Results The surgery was successful,with stable postoperative conditions and significant improvement in spinal curvature.The child's height increased by 7 cm.Discharge occurred on postoperative day 10,with 100%follow-up compliance.The child regained preoperative independent walking ability during follow-up.Conclusion The"1+X"nursing protocol for AS complicated with scoliosis can effectively ensure perioperative safety and promote recovery,which providing insights for perioperative nursing care of other rare diseases complicated with scoliosis.
5.Expression of Slitrk6 during testicular tissue development in rats

Objective To investigate the transcriptional and translational expression of Slitrk6 in rat spermatogene-sis.Methods The mRNA transcription level and translation level of Slitrk6 were detected by RT-qPCR and West-ern blot,respectively in testis of rats aged 2-65 days(repeat three times at each time point).Results The mRNA expression of Slitrk6 reached the highest on day 4 and day 10.SLITRK6 protein was continuously expressed during testis development in rats.Slitrk6 was found in human testis through PPI network.Conclusions Slitrk6 is potential-ly involved in mitosis and meiosis in spermatogenesis;The Slitrk6 may be one of the key genes in spermatogenesis.SLITRK6 protein is involved in mitosis and the transformation of round spermatids.The results lay a foundation for subsequent research on male infertility.
6.Astragali Radix Polysaccharide Inhibits Proliferation and Migration of Gastric Cancer Cells by Targeting ID1 and Akt

ObjectiveTo explore the regulatory effects and mechanisms of Astragali Radix polysaccharide (APS) on inhibitor of differentiation1 (ID1) and protein kinase B (Akt) in gastric cancer. MethodsImmunohistochemical staining was used to detect the expression of ID1 and Akt in 61 gastric cancer tissue samples and 20 adjacent normal gastric tissue samples. Immunofluorescence was used to detect the localization of ID1 and Akt. The effects of APS at the concentrations of 0.625, 1.25, 2.5, 5, 10, 20 mg·L-1 on the proliferation of gastric cancer MGC-803 cells were examined by the cell counting kit-8(CCK-8) method and the colony formation assay. The target information of APS was retrieved from the Traditional Chinese Medicine Systems Pharmacology and Analysis Platform and Swiss Target Prediction. Keywords such as gastric cancer, gastric tumor, and stomach cancer were searched against GeneCards, UniProt, DisGeNET, and Online Mendelian Inheritance in Man (OMIM) for the screening of gastric cancer-related targets. The online tool jvenn was used to create the Venn diagram to identify the common targets, and STRING and Cytoscape were used to construct the protein-protein interaction network. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses were conducted via R 4.2.2 to predict the potential roles of APS in the development of gastric cancer. The cell scratch assay was employed to assess the effect of APS on the migration of MGC-803 cells. The protein and mRNA levels of ID1 and Akt in the cells treated with APS were determined by Western blot and Real-time PCR, respectively. ResultsCompared with the adjacent normal gastric tissue, the gastric adenocarcinoma tissue showed increased positive expression of ID1 (χ2 =81.00, P<0.01). Immunofluorescence detection showed that ID1 and Akt were mainly located in the cytoplasm of gastric adenocarcinoma cells. Bioinformatics analysis identified 14 common genes shared between APS and gastric cancer. The average degree of protein-protein interaction network nodes was 14.29. GO and KEGG pathway enrichment results showed that ID1 and Akt were significantly enriched in the Rap1 and phosphatidylinositol-3-kinase (PI3K) /Akt signaling pathways. Cell experiments demonstrated that 5-fluorouracil (0.1 mg·L-1) and APS (10, 20 mg·L-1) groups showed decreased cell proliferation, migration, and colony formation. Compared with the control group, 10, 20 mg·L-1 APS inhibited the proliferation of MGC-803 cells (P<0.01), with 10 mg·L-1 APS demonstrating stronger inhibitory effect. In addition, APS at 10, 20 mg·L-1 inhibited the migration (P<0.01) and colony formation (P<0.05, P<0.01) of MGC-803 cells. Compared with the control group, APS at 10, 20 mg·L-1 down-regulated the protein levels of ID1 (P<0.01) and Akt (P<0.05) and the mRNA levels of ID1 (P<0.05, P<0.01) and Akt (P<0.05, P<0.01) in MGC-803 cells. ConclusionID1 and Akt are highly expressed in the gastric adenocarcinoma tissue, which may be related to the development of gastric cancer. APS can down-regulate the protein and mRNA levels of ID1 and Akt to exert anti-tumor effects, which is expected to provide new therapeutic targets for gastric cancer treatment.
