1.Effect of hesperetin on inflammatory degeneration of chondrocytes by inhibiting oxidative stress

BACKGROUND:Studies have shown that hesperetin exerts protective effects on chondrocytes through a variety of mechanisms or signaling pathways,but the protective mechanisms have not been fully elucidated. OBJECTIVE:To investigate the effect of hesperetin on lipopolysaccharide-induced inflammatory degeneration of chondrocytes. METHODS:Knee joint chondrocytes from suckling Sprague-Dawley rats were isolated and cultured in vitro,and identified by Safranine O staining.The cytotoxicity of hesperetin on chondrocytes was determined by the MTT assay to ensure the optimal concentration of hesperetin.Chondrocytes were randomly divided into three groups,including the control group,model group,and experimental group.The cellular model of osteoarthritis was established in the latter two groups by simulating chondrocytes with lipopolysaccharide.Chondrocytes in the experimental group was then intervened with hesperetin for 24 hours.Calcein-AM/EthD-I staining was used to detect cell viability.Immunohistochemical staining was performed to determine the expression of type Ⅱ collagen in chondrocytes.Intracellular reactive oxygen species level was detected by a reactive oxygen species detection kit.Total glutathione level was detected by ELISA.Real-time fluorescent PCR was employed to detect the expression of interleukin 1β,interleukin 6,tumor necrosis factor α and type Ⅱ collagen. RESULTS AND CONCLUSION:Safranine O staining results showed that the cells extracted were chondrocytes.Cytotoxicity test results showed 0.5 μmol/L hesperetin had the most significant effect on chondrocyte vitality.Compared with the control group,the model group showed a decrease in chondrocyte proliferation ability,an increase in reactive oxygen species levels,a decrease in total glutathione levels,an increase in type Ⅱ collagen degradation,an increase in the levels of interleukin 1β,interleukin 6,and tumor necrosis factor α,and a decrease in the expression of type Ⅱ collagen(P<0.05).Compared with the model group,in the experimental group,the proliferation ability of chondrocytes increased,reactive oxygen species levels decreased,total glutathione levels increased,and type Ⅱ collagen degradation decreased,levels of interleukin 1β,interleukin 6,and tumor necrosis factor α downregulated,and the expression of type Ⅱ collagen upregulated(P<0.05).To conclude,hesperetin has a protective effect on lipopolysaccharide-induced inflammatory degeneration of osteoarthritic chondrocytes,and the mechanism may be associated with inhibition of reactive oxygen species-mediated oxidative stress.
2.Effect of erythropoietin on hemorrhage of hip replacement surgery

Objective To investigate the effect of erythropoietin on hemorrhage in hip replacement surgery .Methods 48 cases undergoing elective hip replacement surgery in our hospital From June 2014 to October 2016 were selected and devided into control group and experimental group according to the conditional similarity, 24 cases in each group, then the application of erythropoietin in hip replacement surgery was explored.For the experimental group, erythropoietin was injected subcutaneously at three, five and seven days before operation, the dose was 250 U/(kg· times).The control group received 0.9%sodium chloride injection of the same volume of saline, and the other treatments were the same as in the experimental group.Preoperative situation, hemoglobin (Hb) levels, blood pressure (diastolic pressure and systolic blood pressure) at one day before surgey and three, seven and 14 days after surgey were compared and analyzed.Results There were no significant difference in body weight, hemoglobin (Hb), and age between the two groups before operation.The levels of Hb in the experimental group and the control group at 3 and 7 days after operation were compared, the difference was statistically significant (P<0.05), 14 days after the surgey, Hb returned to normal in the experimental group, while the Hb level in the control group was still significantly lower than that in the control group before surgey, the difference was statistically significant (P<0.05).There was no significant difference in diastolic blood pressure and systolic blood pressure between the two groups.Conclusion Preoperative erythropoietin mobilization in patients with hip replacement surgery can reduce the amount of bleeding during and after operation , and the adverse reactions are not obvious.
3.Effects of sodium hyaluronate in combination with compound salvia injection on levels of inflammatory factors and knee joint function in patients with osteoarthritis

Objective To investigate the clinical curative effect and the influence on knee joint function of knee osteoarthritis treating with sodium hyaluronate combined with compound salvia injection.Methods 78 patients with osteoarthritis treated in our hospital from June 2015 to August 2016 were randomly divided into three groups: sodium hyaluronate group, compound salvia injection group and sodium hyaluronate plus compound salvia injection combination therapy group, 26 cases each group.The levels of serum inflammatory cytokines (IL-6,TNF-αand IL-1β) were measured by ELISA before and after treatment in the three groups, and the improvement of knee joint function and the change of pain score before and after treatment were analyzed as well.Results The total effective rate of sodium hyaluronate combined with compound salvia injection was significantly higher than that of sodium hyaluronate and compound salvia injection alone ( P <0.05 ); after treatment, patients VAS pain score was significantly decreased ( P <0.05), and the VAS pain scores in the combination group were significantly lower than those of the group treated with sodium hyaluronate and the compound salvia injection (P<0.05).The levels of IL-6, TNF-αand IL-1βin the three groups were significantly lower than those before treatment (P<0.05),and the inflammatory factors in the combination group were significantly lower than those in the alone treatment group (P <0.05). Conclusion Sodium hyaluronate combined with compound salvia injection has good clinical curative effect on knee osteoarthritis,improving the function of knee joint, reducing pain and so on.
4.Computer-assisted sperm analysis for assessing sperm mobility parameters in in vitro fertilization.

OBJECTIVETo evaluate the association of sperm mobility parameters assessed by computer-assisted sperm analysis (CASA) with the rates of normal fertilization, oocyte cleavage and excellent embryos in in vitro fertilization (IVF) cycles.
METHODSA total of 288 infertile women undergoing IVF cycles patients were divided into two groups according to the normal fertilization rate (≥50% and <50%), cleavage rate (≥90% and <90%), or excellent embryo rates (≥50% and <50%). The means of the sperm motility parameters analyzed by CASA twice before oocyte retrieval were recorded and analyzed using t-test in relation to the rates of normal fertilization, cleavage and excellent embryos in IVF cycles.
RESULTSThe mean curvilinear velocity (VCL), average path velocity (VAP), and amplitude of lateral head displacement (ALH) were significantly higher in women with a normal fertilization rate of ≥50% than in those with a normal fertilization rate of <50% (P<0.05). Women with an oocyte cleavage rate of ≥90% had significantly higher VCL and VAP than those with a cleavage rate of <90% (P<0.05). The VCL, straight line velocity (VSL), VAP, linearity, straightness, wobble coefficient, ALH, or beat-cross frequency showed no significant differences between women with excellent embryo rates of ≥50% and <50% (P>0.05).
CONCLUSIONThe sperm motility parameters assessed using CASA are associated with normal fertilization and oocyte cleavage rates but not with excellent embryo rate in IVF cycles.

5.Construction of eukaryotic expression vectors for different domains of the extracellular region of RAGE and their expression in prostate cancer cells.

OBJECTIVETo construct eukaryotic expression vectors for different domains (V and VC1) of the extracellular region of the receptor of advanced glycation end products (RAGE) and investigate the roles of these domains in prostate cancer.
METHODSThe coding sequence of V and VC1 domains was amplified from the plasmid pcDNA3-HA-RAGE by PCR and cloned into the pcDNA3-HA vector following routine procedures. After identification by PCR and sequencing, the vectors including V and VC1 domains were transfected into PC-3 cells. Western blotting and immunofluorescence were used to detect the expression and distribution of the expressed products in transfected PC-3 cells.
RESULTSThe expression vectors containing V and VC1 domains of RAGE were successfully constructed as confirmed by PCR and DNA sequence analysis. The V and VC1 domains of RAGE were highly expressed and showed a cytoplasmic distribution in transfected PC-3 cells.
CONCLUSIONThe constructed eukaryotic expression vectors for V and VC1 domains of RAGE can be efficiently expressed in prostate cancer cells.
