1.Drying kinetics of Salviae Miltiorrhizae Radix et Rhizoma and dynamics of active components in drying process.

This study explored the drying kinetics of Salviae Miltiorrhizae Radix et Rhizoma(SM), established the suitable models simulating the drying kinetics, and then analyzed the dynamic changes of active components during the drying processes with different methods, aiming to provide a basis for the establishment of suitable drying methods and the quality control of SM. The drying kinetics were studied based on the drying curve, drying rate, moisture effective diffusion coefficient, and drying activation energy, and the appropriate drying kinetics model of SM was established. The drying performance of different methods, such as hot air drying, infrared drying, and microwave drying of SM was evaluated, and the changes in the content of 10 salvianolic acids and 6 tanshinones during drying were analyzed by UPLC-TQ-MS. The Technique for Order Preference by Similarity to an Ideal Solution(TOPSIS) was employed to evaluate the quality of SM dried with different methods. The results showed that the drying rate and moisture effective diffusion coefficient of SM increased with the rise in drying temperature, and the maximum drying rates of different methods were in the order of microwave drying > infrared drying > hot air drying, slice > whole root. The drying rate decreased with the rise in temperature and the extension of drying time. The activation energy of hot air drying was higher than that of infrared drying in SM. The most suitable model for simulating the drying process of SM was the Page model. The TOPSIS results suggested infrared drying at 50 ℃ was the optimal drying method for SM. During the drying process, the content of salvianolic acids increased in different degrees with the loss of moisture, among which salvianolic acid B showed the largest increase of 44 times compared with that in the fresh medicinal material. Tanshinones also existed in the fresh herb of SM, and the content of tanshinone Ⅱ_A increased by 3 times after drying. The results provided a basis for the establishment of suitable drying methods and the quality control of SM.

2.Phenylpropanoids from roots of Berberis polyantha.

The chemical constituents were systematically separated from the roots of Berberis polyantha by various chromatographic methods, including silica gel column chromatography, HP20 column chromatography, polyamide column chromatography, reversed-phase C_(18) column chromatography, and preparative high-performance liquid chromatography. The structures of the compounds were identified by physicochemical properties and spectroscopic techniques(1D NMR, 2D NMR, UV, MS, and CD). Four phenylpropanoids were isolated from the methanol extract of the roots of B. polyantha, and they were identified as(2R)-1-(4-hydroxy-3,5-dimethoxyphenyl)-1-propanone-O-β-D-glucopyranoside(1), methyl 4-hydroxy-3,5-dimethoxybenzoate(2),(+)-syringaresinol(3), and syringaresinol-4-O-β-D-glucopyranoside(4). Compound 1 was a new compound, and other compounds were isolated from this plant for the first time. The anti-inflammatory activity of these compounds was evaluated based on the release of nitric oxide(NO) in the culture of lipopolysaccharide(LPS)-induced RAW264.7 macrophages. At a concentration of 10 μmol·L~(-1), all the four compounds inhibited the LPS-induced release of NO in RAW264.7 cells, demonstrating potential anti-inflammatory properties.

3.Surveillance report of viral diarrhea in children under 5 years old in Wuxi from 2018 to 2023

OBJECTIVE To monitor the common pathogens for viral diarrhea in children under 5 years old in Wuxi area,and to analyze their epidemiological characteristics and trends.METHODS Diarrhea specimens were collected from inpatient and outpatient children with diarrhea in sentinel hospitals in Wuxi from 2018 to 2023.The norovir-us,rotavirus,sapovirus,enteradenovirus and astrovirus were detected by Real-time fluorescence quantitative pol-ymerase chain reaction(PCR),and the monitoring results were statistically analyzed.RESULTS A total of 654 vi-rus-positive cases were detected from 1898 diarrhea specimens with an overall detection rate of 34.46%.Among them,norovirus and rotavirus had the highest detection rates 12.54%and 11.70%,respectively.The detection rate of viruses was the highest in children aged 12 to<24 months,reaching 59.03%.In inpatient children with di-arrhea,rotavirus had the highest detection rate at 14.03%,while in outpatient children with diarrhea,norovirus had the highest detection rate at 11.93%.There was a significant difference in the detection rate of rotavirus be-tween inpatient and outpatient children(P<0.05).CONCLUSION Viral diarrheal diseases exhibit a notably high detection rate among children aged 12 to<24 months in Wuxi,suggesting the need to further optimize prevention and control measures for this age group.
4.Bibliometric analysis of research trends and hotspots on organic solvent induced hearing loss

Objective:To explore the research hotspots and trends in the literature related to hearing loss caused by organic solvents, and to provide an effective theoretical basis for further research on the impact of ototoxic organic solvents on the auditory system.Methods:In January 2024, literatures that met the inclusion/exclusion criteria from 1974 to 2023 in the PubMed database were selected. The authors, countries, journal fields and publishing institutions of the literatures were analyzed through Bicomb version 2.01. The author co-occurrence knowledge network graph and the literature publication institution graph were plotted using CiteSpace 6.3R1 Advance, and the co-occurrence clustering map of keywords was drawn using VOS viewer 1.6.19 software.Results:A total of 380 relevant literatures were retrieved, and 256 were finally included. The top three countries in terms of the number of published literature were the United States (accounting for 30.08%, 77/256), the United Kingdom (accounting for 17.97%, 46/256), and the Netherlands (accounting for 10.16%, 26/256). The published authors were represented by Pierre Campo, involving a total of 165 institutions, mainly occupational health research institutions and hearing institutions. It mainly involved related fields such as audiology, occupational medicine, environmental hygiene, toxicology, and otolaryngology. The key groups were painters, aviation industry personnel, gas station workers and agricultural workers. The key solvents included organic solvent mixtures, benzene and its derivatives, trichloroethylene, ethanol, carbon disulfide, etc.Conclusion:Research on hearing loss caused by organic solvents mainly focuses on countries such as the United States, the United Kingdom, and the Netherlands, with a particular emphasis on occupational groups such as painters, aviation industry personnel, gas station workers, and agricultural personnel, as well as the ototoxic effects of organic solvent mixtures, benzene and its derivatives, etc. This provides a basis for subsequent mechanism exploration, protection strategy formulation, and hearing health management of the target population.
5.Sinicization of Evidence-Informed Decision-Making Competence Measure for nurses and its reliability and validity test

Objective To translate Evidence-Informed Decision-Making Competence Measure for Chinese nurses and test its validity and reliability.Methods A research group was set up to use the Brislin translation model to translate the original scale into Chinese,and the back translation,cross-cultural adaptation,pre-experiment and cognitive interview were conducted to finally form the Chinese version of the Evidence-Informed Decision-Making Competence Measure for nurses.A total of 1 247 nurses from 7 tertiary A hospitals in Beijing,Hubei,Hunan and Xinjiang were selected by convenience sampling method in April 2024 to test its reliability and validity.Results 1 026 effective question-naires were collected,with an effective recovery rate of 82.28%.The Chinese version of the Evidence-Informed Decision-Making Competence Measure included 25 items,including knowledge/skill,attitude and behavior.A total of 3 common factors were extracted from exploratory factor analysis,and the cumulative variance contribution rate was 91.725%.The content validity index at the item level was 0.83-1.00;the content validity index at the scale level was 0.988;the calibration association validity was 0.496.The Cronbach's α coefficient of the whole scale was 0.992;the half-point reliability was 0.930;the retest reliability was 0.927.Conclusion The Chinese version of Evidence-Informed Decision-Making Competence Measure for nurses has good reliability and validity,and it can be used to evaluate the evidence-informed decision-making competence of Chinese nurses,provide references for promoting evidence-based nursing practice and evidence-informed decision-making.

Fibrosing alopecia in a pattern distribution (FAPD) is a newly recognized entity of primary lymphocytic cicatricial alopecia with clinical and histopathological characteristics of both androgenetic alopecia and lichen planopilaris. Currently, there is still a lack of full understanding of and standardized treatment protocols for FAPD, which is prone to be clinically underdiagnosed and misdiagnosed. This review systematically summarizes the research progress in FAPD in terms of clinical manifestations, diagnosis, and treatment, in order to facilitate its clinical diagnosis and treatment.
7.Establishment and application of quadruplex RT-qPCR for differentiation of viral pathogens associated with diarrhea in pig herds

A quadruplex RT-qPCR method was developed for rapid identification and diagnosis of transmissible gastroenteritis virus(TGEV)of swine,porcine epidemic diarrhea virus(PEDV),porcine deltacorona virus(PDCoV),and porcine rotavirus type A(PoRVA).The full-length sequences of PEDV(77 strains),TGEV(63 strains),PDCoV(17 strains)that are prevalent in China,as well as the 85 VP6 gene sequences of PoRVA,were downloaded from the NCBI database for homology analysis.Based on the relatively conserved sequences,the corresponding primers and probes for each virus were designed and used to establish the quadruplex RT-qPCR method.After optimization of the probes and the reaction conditions,the specificity,sensitivity,and repeatability were determined.Using the established method,109 clinical samples of diarrhea were tested and further compared with the results by standard method.The results showed that the quadruplex RT-qPCR method established in this experiment has good amplification effect,with the C,value linearly correlated with the copies of templates(R2>0.99).Specificity assay demonstrated that the quadruplex RT-qPCR method can identify TGEV,PEDV,PDCoV,PRoVA strains,and do not de-tect African swine fever virus(ASFV),porcine circovirus type 2(PCV2),porcine reproductive and respiratory syndrome virus(PRRSV),classical swine fever virus(CSFV),pseudorabies virus(PRV)and other epidemic viruses.Sensitivity assay showed that the detection limits for TGEV,PEDV,PDCoV and PoRVA were 10,20,20 and 50 copies/μL,respectively.The method exhibits excellent reproducibility,with coefficients of variation(Cv)for both intra-and inter-assay repli-cates being less than 1%.Detection of 109 samples of diarrhea by this method yielded the coinci-dence rate of 100%with the industry standard,indicating high practical applicability.The devel-oped method possesses the advantages of strong strain compatibility,high sensitivity,strong speci-ficity,good repeatability and stability.It is suitable for virus diagnosis and large-scale clinical sam-ple testing,providing technical support for disease prevention and control as well as epidemiologi-cal investigation.
8.Establishment and application of RT-RAA-CRISPR/Cas13a diagnostic method for porcine Senecavirus

The objective of this study was to develop a rapid and precise detection technique for por-cine Senecavirus A(SVA)employing reverse transcription recombinase polymerase amplification(RT-RAA)in conjunction with CRISPR Cas13a technology.Additionally,the study aimed to opti-mize the assay's reaction conditions to enhance amplification efficiency.Eight RT-RAA primer sets were designed based on the conserved gene sequence of porcine SVA,and a series of reaction condi-tions were evaluated to refine the RT-RAA reaction system.Subsequently,CRISPR-derived RNA(crRNA)sequences were developed and selected to construct the RT-RAA-CRISPR reaction sys-tem.The method's specificity was determined by examining six prevalent porcine pathogenic nucleic acids,while its sensitivity was assessed using SVA cRNA standards quantified by digital PCR.The method's stability and the consistency of clinical sample analysis were also evaluated.The findings revealed that the optimized RT-RAA and CRISPR reaction systems exhibited the highest amplifi-cation efficiency at a reaction temperature of 37 ℃.Among the eight crRNAs,five were identified as exhibiting the strongest detection signals.The formulated RT-RAA-CRISPR Cas13a method demonstrated exceptional specificity,showing no cross-reactivity with other common porcine disea-ses,including ASFV,PRRSV,PEDV,PCV2,CSFV,and PRV.The method achieved high sensitivi-ty,detecting as low as 0.86 copies/μL of SVA,and exhibited stable fluorescence output,robust re-producibility,and the ability to complete clinical sample analysis within 50 minutes.Consistency e-valuation with six positive and 58 negative samples indicated 100%agreement in outcomes.These results substantiate that the study successfully established a rapid and specific RT-RAA-CRISPR Cas13a detection method for the on-site identification of porcine Senecavirus A,demonstrating high specificity and sensitivity,and holds promise for application in SVA monitoring and control initia-tives.
