1.Impact and mechanisms of ganoderma lucidum polysaccharide on ETEC-induced enteritis

Objective To study on the effects and mechanisms of ganoderma lucidum polysaccharides(GLP)on enteritis induced by enterotoxigenic Escherichia coli(ETEC)in mice.Methods Twenty-four 6-to 8-weeks-old male SPF Kunming mice were selected.A mouse enteritis model was induced by intraperitoneal injection of ETEC bacterial suspension.Mice were divided into normal group,ETEC infection group,ETEC infection+high-dose GLP group,and ETEC infection+ow-dose GLP group using a random number table method,six mice in each group.Pathological changes in mice small intestine tissue were observed through HE staining.The expression of C-reactive protein(CRP),tumor necrosis factor-(TNF)-α,and interleukin(IL)-6 proteins in mice small intestine tissues were detected by Western blot method,and expression levels of Toll-like receptor 4(TLR4),nuclear factor-KB(NF-κB)p65,and myeloid differentiation factor 88(MyD88)mRNA in mice small intestine tissues were detected by RT-qPCR technique to explore the regulatory effect of GLP on ETEC-induced mice intestinal inflammation through TLR4/NF-κB signaling pathway.Results Compared with control group,mice infected with ETEC showed lethargy,reduced appetite,damaged intestinal villi,significant inflammatory infiltration in tissues,and an increasing trend in disease activity index(DAI)scores(P<0.05).The relative expression levels of TLR4,NF-κB p65,and MyD88 mRNA,as well as protein expression levels of CRP,TNF-α,and IL-6 in small intestine tissues of mice,were significantly increased(P<0.01).Compared with ETEC-infected group,mice in different dosage groups showed improvements in mental status and appetite.The relative expression levels of TLR4,NF-κB p65,and MyD88 mRNA in small intestine tissues of mice were significantly decreased in all dosage groups(P<0.01).In ETEC infection+high-dose group,protein expression levels of CRP,TNF-α,and IL-6 in small intestine tissues of mice were significantly decreased(P<0.01),while the decrease in ETEC infection+low-dose group was not significant(P>0.05).Compared with ETEC infection+low-dose group,ETEC infection+high-dose group showed significant decreases in relative expression levels of TLR4 and MyD88 mRNA,as well as in protein expression level of IL-6 in small intestine tissues of mice(P<0.01 or P<0.05).Conclusion GLP can effectively relieve intestinal inflammation induced by ETEC,and may exert its anti-inflammatory effect through regulating TLR4/NF-κB signaling pathway,which may be proportional to its concentration.

3.Relationship among depression,anxiety and social support in elderly patients from community outpa-tient clinic

Objective To explore the relationship among depression,anxiety and social support in elderly patients in community outpatient clinic. Methods A total of 551 elderly outpatients from two com-munity health service centers of Hongkou District in Shanghai were evaluated with patient health question-naire-9 (PHQ-9),generalized anxiety disorder-7 (GAD-7),perceived social support scale( PSSS) for de-pression,anxiety,physical health and social support. Results The prevalence rates of depression and anxiety were 26. 1% and 17. 2%,respectively. The scores of PHQ-9 and GAD-7 were 2. 0(4. 0) and 1. 0(2. 0). There were statistically significant differences in the scores of family support,friend support,other support and social support among the elderly patients with different degrees of depression or anxiety (P<0. 01). Fam-ily support(B=-0. 196) and friend support(B=-0. 171) were protective factors of depression in elderly pa-tients in community outpatient clinic. Age,family support and friend support were protective factors of anxiety in elderly patients,while gender and fluctuation of physical diseases were protective factors of anxiety(P<0. 05). Con-clusions The depression and anxiety is intimately related to social support in elderly outpatients. Appropriate measures should be taken to optimize social support,mitigate bad mood negative improve their quality of life.
4. Prognostic factors in immune thrombocytopenia in children

Objective:
To explore the prognostic relationship between initial absolute lymphocyte count(ALC) of peripheral blood and primary immune thrombocytopenia (ITP) in children, in order to provide basis for judging the prognosis and treatment of ITP in children.
Methods:
Clinical data of 166 children with primary ITP in children admi-tted at the Affiliated Hospital of Guizhou Medical University from January 2014 to March 2018 were analyzed retrospectively, and they were followed up by clinic and telephone, the prognostic factors (gender, age, ethnicity, inducement, bleeding, initial ALC, platelet count and treatment) were statistically analyzed, and the relationship between ALC of peripheral blood and the prognosis of children with ITP was observed.
Results:
Of 166 children with ITP, 89 cases (53.6%) had remission within 3 months, 18 cases (10.8%) within 3-12 months, 20 cases (12.0%) within 1-4 years, a total of 39 cases (23.5%) were refractory (no remission in 1-10 years), the remission rate within 1 year was 64.5%, the total remission rate was 76.4%.ALC of remission cases was (4.58±2.87)×109/L within 3 months, (4.47±2.04)×109/L within 3-12 months, and (2.86±1.61)×109/L within 1- 4 years.Thirty-nine cases (23.5%) were refractory(no remission in 1-10 years), ALC of them was (2.07±0.98)×109/L, and there were significant differences among different groups (
