1.Construction of glucose oxidase–loaded nanogels and its inhibition effect on the Warburg effect in glioma cells

Objective To construct glucose oxidase(GOx)–loaded nanogels (GONGs), optimize their formulation, and evaluate their capacity to inhibit the Warburg effect in glioma cells. Methods A responsive polymer (HAM) was synthesized and used to self-assemble GONGs, which were then characterized. Encapsulation efficiency and drug loading were determined using fluorescence spectrophotometry. Biocompatibility was tested by measuring cytotoxicity and hemolytic activity. Western blotting was used to evaluate the effects of GONGs on the expression of proteins associated with the Warburg phenotype and oxidative damage in glioma cells. Results GONGs prepared at a drug–to–polymer ratio of 1∶10 exhibited a particle size of 140.3 nm and a zeta potential of −27.2 mV. Compared with free GOx, GONGs markedly reduced cytotoxicity, increased the IC50 in hUVEC cells from 2.150 nmol/L to 74.86 nmol/L, and significantly decreased hemolysis. At a GOx concentration of 2 nmol/L, GONGs effectively downregulated glycolysis-related proteins, such as HK2 and LDHA, and inhibited glutamine metabolism in glioma cells. Conclusion GONGs exhibited high GOx loading capacity, significantly reduced GOx-induced cytotoxicity, inhibited the Warburg effect in glioma cells and induced oxidative damage.
2.Detection of ST2,CA125,and HE4 levels in serum of ovarian cancer patients and their clinical significances

Objective:To discuss the diagnostic value of levels of growth stimulation expressed gene 2 protein(ST2),carbohydrate antigen 125(CA125),and human epididymis protein 4(HE4)in serum of the ovarian cancer patients,and to clarify their relationships with clinicopathological parameters of the ovarian cancer patients.Methods:A total of 136 patients with ovarian benign lesions or primary ovarian malignancies in our hospital confirmed by postoperative histopathology were randomly selected,including 53 cases in ovarian benign ovarian disease group and 83 cases in ovarian cancer group.Additionally,55 healthy female volunteers during the same period were enrolled as healthy control group.The fasting venous blood from all the subjects was collected on the first day of admission,and serum was retained;cyclic enhanced fluorescence immunoassay was used to detect ST2 levels of the subjects in various groups;chemiluminescence method was used to detect serum CA125 and HE4 levels of the subjects in various groups;receiver operating characteristic(ROC)curve was used to assess the diagnostic performance of each indicator;the Cut-off value and area under the ROC curve(AUC)were calculated,with AUC representing the diagnostic performance;Kendall's method was used to analyze the correlations between serum ST2 levels and TNM stage,maximum tumor diameter,distant metastasis,lymphnode metastasis,peritoneal metastasis,carcinoembryonic antigen(CEA),CA125,carbohydrate antigen 199(CA199),HE4,P53,and Kiel-67(Ki67)in the ovarian cancer patients.Results:Compared with healthy control group,the serum CA125 level of the patients in ovarian benign ovarian disease group was significantly increased(P<0.05).Compared with healthy control group and ovarian benign group,the serum levels of ST2,CA125,and HE4 of the patients in ovarian cancer group were significantly increased(P<0.05).The AUC values were 0.719(95%CI:0.616-0.822)for ST2,0.868(95%CI:0.794-0.942)for CA125,and 0.867(95%CI:0.793-0.942)for HE4.The combined detection of ST2+CA125+HE4 yielded an AUC of 0.894(95%CI:0.832-0.955).Significant differences were observed in serum ST2,CA125,and HE4 levels among ovarian cancer patients with different TNM stages,lymph node metastasis,distant metastasis,and peritoneal metastasis(P<0.05),while there were no significant differences in the patients with different ages or maximum tumor diameters(P>0.05).The ST2 level in serm of the ovarian cancer patients was positively correlated with TNM stage,distant metastasis,lymph node metastasis,peritoneal metastasis,CA125 level,HE4 level,and Ki67 level(P<0.05),but was not correlated with maximum tumor diameter,CEA,CA199 level,or P53 level(P>0.05).Conclusion:ST2,CA125,and HE4 are highly expressed in the serum of the ovarian cancer patients.Combined detection of serum ST2,CA125,and HE4 exhibits good sensitivity and specificity for ovarian cancer screening and improves diagnostic efficacy.ST2 is associated with advanced TNM stage and metastasis in ovarian cancer and may participate in the occurrence and progression of ovarian cancer.

Medical equipment management plays a crucial role in enhancing the quality and efficiency of healthcare services. However, traditional management approaches are increasingly inadequate to meet the growing demands of modern healthcare. As intelligent operation and maintenance (O&M) models based on big data, the Internet of Things (IoT), and artificial intelligence (AI) technologies develop, it is imperative to explore their application in medical equipment management. This paper reviews the technical overview of intelligent O&M and discusses the algorithms and challenges of intelligent O&M models based on different technologies. It also proposes issues that need improvement in intelligent O&M models, aiming to provide valuable references for the future development of medical equipment management.

Traumatic brain injury (TBI) is intricately linked to the most severe clinical manifestations of brain damage. It encompasses dynamic pathological mechanisms, including hemodynamic disorders, excitotoxic injury, oxidative stress, mitochondrial dysfunction, inflammation, and neuronal death. This review provides a comprehensive analysis and summary of biomaterial-based tissue engineering scaffolds and nano-drug delivery systems. As an example of functionalized biomaterials, nano-drug delivery systems alter the pharmacokinetic properties of drugs. They provide multiple targeting strategies relying on factors such as morphology and scale, magnetic fields, pH, photosensitivity, and enzymes to facilitate the transport of therapeutics across the blood-brain barrier and to promote selective accumulation at the injury site. Furthermore, therapeutic agents can be incorporated into bioscaffolds to interact with the biochemical and biophysical environment of the brain. Bioscaffolds can mimic the extracellular matrix environment, regulate cellular interactions, and increase the effectiveness of local treatments following surgical interventions. Additionally, stem cell-based and exosome-dominated extracellular vesicle carriers exhibit high bioreactivity and low immunogenicity and can be used to design therapeutic agents with high bioactivity. This review also examines the utilization of endogenous bioactive materials in the treatment of TBI.
5.Application of MXene-based hydrogels in wound repair

BACKGROUND:MXene-based hydrogel is a kind of nano-composite multifunctional hydrogel material,which has broad application prospects in the field of chronic wound repair,such as diabetic foot,pressure sore,cancer,and traumatic ulcer.OBJECTIVE:To review the advantages of MXene-based hydrogels and their application and related action mechanism in wound repair.METHODS:The articles included in CNKI,VIP,PubMed,and Science Direct databases were searched from January 2010 to October 2023.The key words were"MXene,hydrogel,tissue engineering application,wound repair"in Chinese and"MXene,hydrogel,wound repair"in English.Finally,70 articles that met the criteria were selected for review.RESUILTS AND CONCLUSION:MXene-based hydrogels have excellent mechanical,electrical,photo-thermal properties,biocompatibility,and antibacterial effects in wound tissue engineering,and can be combined with other organic and inorganic substances to produce greater therapeutic effects in hydrogel.MXene-based hydrogels can be applied to skin wounds in combination with wound monitoring,drug delivery and release,and photothermal therapy.Despite some progresses in the design and preparation of MXene-based hydrogel composites,they are still in the fundamental research stage.There is a lack of clinical validation regarding their effectiveness and safety,but they hold great potential for development and application.
6.Regulatory effect and mechanism of Yiqi Jiedu Decoction on ionizing radiation-induced macrophage polarization

Objective To investigate the regulatory effect and mechanism of Yiqi Jiedu Decoction(YQJD)on ionizing radiation-induced macrophage polarization and its correlation with the Toll-like receptor 4(TLR4)/myeloid differentiation primary response protein 88(MyD88)/nuclear factor kappa-B(NF-κB)signaling pathway.Methods Fifty-five specific-pathogen-free male Sprague-Dawley rats were randomly divided into blank(n=30),anduolin(n=10),and YQJD groups(n=15).They were respectively gavaged with deionized water,anduolin suspension(0.345 6 g/kg),and YQJD high-dose(20.88 g/kg)at a dose of 0.01 mL/g body weight once a day for seven consecutive days.2 hours after the last gavage,blood was collected from the abdominal aorta to prepare the control rat,andolin rat,and YQJD high-dose sera.Appropriate amounts of YQJD high-dose and control sera were mixed in a ratio of 1∶1 and 1∶3,respectively,to obtain YQJD medium-and low-dose rat serum.RAW264.7 cells were divided into blank(10%blank rat serum),model(10%blank rat serum),anduolin(10%anduolin rat serum),and YQJD-L,YQJD-M,YQJD-H groups(10%YQJD low-,medium-,and high-dose rat serum).Except for the blank group,the cells in other groups were irradiated with 12 Gy60 Co γ-rays once to establish the macrophage radiation injury model.At 24 h after irradiation,cell viability was detected using the CCK-8 method,and the cell migration rate was measured using the scratch test.Cell morphology was observed using phalloidin staining,tumor necrosis factor-alpha(TNF-α)and interleukin-10(IL-10)levels in the cell supernatant were quantified using enzyme-linked immunosorbent assay,and the proportion of M1 macrophages was detected using flow cytometry.TLR4,MyD88,and NF-κB protein expression were detected using Western blotting.Results Twenty-four hours after irradiation,compared with the blank group,the model group exhibited significantly reduced cell viability and migration rate(P<0.01),increased cell volume and pseudopodia formation,elevated TNF-α and IL-10 levels,an increased proportion of M1 macrophages,and upregulated TLR4,MyD88,and NF-κB protein expression(P<0.05,P<0.01).Compared with the model group,each drug-treated group showed improved cell viability and migration rate(P<0.05,P<0.01),decreased cell volume,more regular cell shape,reduced TNF-α levels,lower M1-type macrophage proportion,and downregulated TLR4,MyD88,and NF-κB protein expression(P<0.05,P<0.01).IL-10 level showed an upward trend.Conclusion YQJD can partially inhibit M1 macrophage polarization and suppress inflammatory responses,which may be related to the TLR4/MyD88/NF-κB signaling pathway.
7.Comprehensive Assessment of Cardiovascular Risk in Diabetes Using Atherosclerosis Indicators

Objective To investigate the relationship between the atherogenic index of plasma(AIP),carotid intima-media thick-ness(CIMT),and brachial-ankle pulse wave velocity(baPWV)with the 10-year risk of atherosclerotic cardiovascular disease(AS-CVD)in patients with type 2diabetes mellitus(T2DM).Methods This study retrospectively analyzed the clinical data of 131 T2DM pa-tients managed through the National Metabolism Management Center(MMC)at the Affiliated Central Hospital of Shandong First Medical University from January 2020 to December 2022,all of whom had undergone AIP,CIMT and baPWV assessments.The 10-year ASCVD risk for each patient was estimated using the China-PAR model,classifying patients into low-to-moderate and high-risk groups.Uni-variate analysis was employed to identify variables correlated with cardiovascular risk,followed by multivariate Logistic regression analysis to evaluate the independent impact of AIP,CIMT and baPWV on ASCVD risk.The Hosmer-Lemeshow goodness-of-fit test was used to assess the suitability of the combined variables(AIP,CIMT and baPWV).Lastly,the predictive efficacy of the combined variables and individual indicators was assessed using receiver operating characteristic(ROC)curves and the DeLong test.Results Multivariate Logis-tic regression analysis demonstrated that AIP,CIMT and baPWV were independent risk factors for ASCVD in T2DM patients(OR>1,P<0.05).The Hosmer-Lemeshow goodness-of-fit test indicated satisfactory model fit(x2=2.725,P=0.950).The combined variable of AIP,CIMT and baPWV exhibited superior discriminative ability in predicting ASCVD risk among T2DM patients(AUC=0.834),sig-nificantly outperforming individual variables.Conclusion AIP,CIMT and baPWV have significant clinical value in assessing ASCVD risk in T2DM patients.Integrating these indicators enhances predictive performance,offering an effective tool for individualized cardiovascular risk assessment in T2DM patients.

Objective To conduct a clinical comprehensive evaluation of Lu Dangshen Oral Liquid from 6 dimensions,including safety,effectiveness,economic value,innovativeness,suitability and accessibility.Methods The clinical evidence of Lu Dangshen Oral Liquid and seven control drugs(Bufei Huoxue Capsules,Xiangsha Liujun Pills,Jinshuibao Capsules,Shenling Baizhu Granules,Yupingfeng Granules,Zhenqi Fuzheng Granules and Bailing Capsules)were obtained by using quantitative and qualitative methods through literature retrieval,questionnaire survey and data collection.Based on the multi criteria decision analysis(MCDA)model,the clinical value of drugs was comprehensively evaluated.The weights of criterion layer,index layer and alternative scheme were calculated by analytic hierarchy process.The equal frequency discretization rule was used to classify the evaluation results.Results The safety evidence showed that the main adverse reactions were epistaxis and nausea,and there were no serious adverse events.The comprehensive evaluation was grade A(the risk was controllable and the evidence was sufficient).Clinical studies showed that its treatment of lung spleen qi deficiency syndrome reflects the idea of treating different diseases with same method,and its efficacy in stable chronic obstructive pulmonary disease and children with recurrent respiratory tract infection was better than conventional treatment or Spleen Aminopeptide Oral Lyophilized Powder,and its effectiveness was rated as grade A.In terms of economy,the daily drug cost was relatively high,and it was rated as grade C.The preparation process was authorized by one patent.The single Chinese materia medica could improve the diseases and symptoms related to lung spleen qi deficiency syndrome,and the innovation was rated as grade A.In the questionnaire survey,the clinical use and patient compliance were excellent,and the suitability was rated as grade A.The supply of raw materials was stable,and the accessibility was rated as grade B.Conclusion The comprehensive evaluation of the clinical value of Lu Dangshen Oral Liquid in the treatment of lung spleen qi deficiency syndrome is grade A,which can provide references for the adjustment of the national medical insurance catalog and the national essential medicine catalog.
