1.Investigation on the effect of job-transfer training for psychiatrists in Henan province

Objective:To evaluate the effect of job-transfer training for psychiatrists in Henan province in 2018 and to compare them with the results of 2016.Methods:Data of the trainees were collected through questionnaires in 2017 and 2019 respectively. The influencing factors of knowledge and skills were determined by Multiple linear regression analysis; baseline data, training intention, training feedback and the proficiency of knowledge and skills were compared by independent sample t test and chi-square test. Results:The overall satisfaction rate for training was 98.3%, and the overall mastery rate of training knowledge and skills was 59.2% in year 2018. Compared with 2016, the willingness to participate in training, the satisfaction rate, the recognition degree of " 1+ 10+ 1" training mode, the degree of mastery and practical application of training knowledge and skills increased( P<0.05). There were statistically significant differences in the distribution of the primary reasons for participating in the training, factors hindering their participation in the training, and the causes for their failure to fully apply their learning to practice( P<0.01). The results showed that scope of practice, title, intention, and interest in psychiatry was related to the mastery of training knowledge and skills( P<0.05). Conclusions:The effect of training in 2018 is better than 2016, and the degree of mastery and practical application of training knowledge and skills should be increased.
2.A study on influencing factors and current status of drug compliance in patients with serious mental disorder in Henan

Objective:To understand the current status of drug compliance in patients with serious mental disorder in Henan,and to study its influencing factors.Methods:Using stratified random sampling method. The baseline data of patients were collected by self-designed questionnaires. Logistic regression analysis was conducted to determine associated factors. Independent sample t-test and chi-square test were performed to compare the baseline data of the two groups. Results:A total of 1 872 valid questionnaires were collected, and the rate of medication compliance in patients with serious mental disorder was 51.34%(961). Through multivariate logistic regression analysis, we found that 10 variables, such as relation ship, household income level, and year treatment cost were significantly associated with drug compliance. Conjugal( Wald χ2=10.491, P=0.001), and sibling relationship ( Wald χ2=10.472, P=0.001) were the risk factors for drug compliance, and it has an increased tendency. Regarding household income level, compared with the level of ‘good’, the level of ‘very good’, ‘better than good’ ( Wald χ2=4.373, P=0.037), and ‘moderate’ ( Wald χ2=4.189, P=0.041) were the protective factors for drug compliance, and it has a reducing tendency. In addition, patients′ age (45-59 years)( Wald χ2=7.060, P=0.008), living status (living together with caregivers)( Wald χ2=32.029, P<0.01), and residency (urban residents)( Wald χ2=21.908, P<0.01) were the protective factors for drug compliance. Conclusions:The current status of drug compliance in patients with serious mental disorder in Henan was rigorous, and its influence factors are complicated. The interaction between factors should be comprehensively considered before applying interventions.
3.An epidemiological survey of schizophrenia in Henan Province

Objective To investigate the morbidity rate,prevalence and influencing factors of schizophrenia among people aged 15 and above in Henan Province.Methods In 2015,a multi-stage stratified random sampling method was adopted,and the revised General Health Questionnaire (GHQ-12) was used as a preliminary screening tool to carry out mental disorder epidemiological investigation in 20 counties of Henan Province.The sampled people were divided into high,medium and low risk groups according to the GHQ-12 scores.The diagnosis of mental disorders was made by attending psychiatrist with DSM-Ⅳ-TR Axis Ⅰ diagnostic clinical interview pattern scale (SCID).The qualitative data were compared by chi-square test,and the influencing factors of schizophrenia were analyzed by Logistic regression analysis.Results A total of 23 846 people were surveyed,including 11 865 (49.76%) men and 11 981 (50.24%) women in which 124 ones were diagnosed with schizophrenia,with the adjusted lifetime prevalence rate of 5.56‰,and time point prevalence of 5.04‰.Of the 124 cases,83 lived in towns and 41 lived in cities,54 were male and 70 were female.The other basic message:level of education:illiterate 26,primary school 39,junior high school 47,high school or technical secondary school 9,junior or regular college 3;marital status:unmarried 31,first marriage 78,remarried 3,divorced 11,widowed 1.Univariate analysis showed statistically significant differences in the prevalence of schizophrenia at different levels of education (x2=33.903,P<0.01) and marital status (x2=137.143,P<0.01).Logistic regression analysis showed that urban and rural areas (Wald x2=37.296,P<0.01),gender (Wald x2=13.131,P<0.01),education level (Wald x2=48.338,P<0.01),marital status (Wald x2=54.495,P<0.01) and family history (Wald x2=107.340,P<0.01) were the influencing factors of schizophrenia.Conclusion The prevalence rate of schizophrenia in Henan Province is basically consistent with the research results at home and abroad,and the prevention and treatment of schizophrenia should be based on different regions,ages,genders,et al.

6.Analysis of mental health knowledge and associated factors among under-graduates in Henan Province

Objective:
To survey mental health knowledge and its influence factors among undergraduates in Henan provenience and to provide a scientific evidence for further formulating the targeted strategies.
Methods:
Totally 840 agriculture and forestry college students, literature and history students, science students and medical students were selected with stratified multistage random sampling from 45 colleges and universities in Henan provenience,investigation of Mental Health related knowledge by self-designed questionnaire.
Results:
The awareness rate of mental health knowledge was 75.86%(32 806/43 248). Multivariate linear regression analysis showed that score of mental health knowledge of femal students was 1.03 of the of male students; score of medical students was 5.19,3.65,2.65 of that of students in the other majors; score of students who obtained mental health knowledge through other channels(television/movie/network/talking) was 1.42 of that of than those who obtained through formal approaches(slogans/manuals/broadcast/cathedra)(P<0.05).
Conclusion
The awareness rate of mental health knowledge among undergraduates in Henan provenience need to be improved, the education of male, senior students, non-medical students and the explore of diversified forms should be strengthened.
7.The effects of sodium butyrate on ethanol-seeking behavior and H3K9 acetylation in NMDA receptor 2B subunit promotor region in the hippocampus of rats

Objective To investigate the effects of sodium butyrate on ethanol-seeking behavior and H3K9 acetylation levels in NMDA receptor 2B subunit(NR2B) promoter region in the hippocampus of Wistar rats.To explore the epigenetic mechanism underlying ethanol-seeking behavior.Methods According to random number table,48 male Wistar rats were divided into saline group,sodium butyrate group,ethanol group and sodium butyrate + ethanol group,with 12 rats in each group and administered by intraperitioneal injection respectively.Conditioned place preference (CPP)was used to evaluate the ethanol-seeking behavior.Using Western-blot,real-time PCR and chromatin immunoprecipitation assays,the expression of NR2B protein,NR2BmRNA and the relative level acetylated H3K9 in NR2B promoter region in the hippocampus were determined respectively.Results The CPP test and the CPP score in each group were different (P< 0.05).Compared with the CPP test(261.1 ± 102.2) and the CPP score(48.5±94.6) of saline group,the CPP test ((406.8±109.2),(502.7±72.89)) and the CPP score((198.2± 119.4),(277.5±76.2)) of ethanol group and sodium butyrate + ethanol group were significantly higher(P<0.05),the CPP test(193.4±93.8) and the CPP score (9.7±94.0)of sodium butyrate group were not significantly different(P>0.05).Compared with the ethanol group,CPP test of sodium butyrate + ethanol group was significantly higher(P<0.05).The expression of NR2B protein,NR2BmRNA and acetylated level H3K9 in NR2B promoter region in the hippocampus in each group were different (P< 0.05).Compared with the expression of NR2B protein (1.00 ± 0.28),NR2BmRNA(1.00±0.14) and H3K9 acetylation in NR2B promoter region(1.00±0.25)in the hippocampus of saline group the expression of NR2B protein((1.40±0.34),(1.79±0.30)),NR2BmRNA((1.26±0.16),(1.50±0.08)) and aeetylated level H3K9 in NR2B promoter region ((1.68±0.16),(2.35±0.45)) of ethanol group and sodium butyrate ± ethanol group were significantly higher(P<0.05).The expression of NR2B protein(0.85±0.24),NR2BmRNA(1.05±0.13) and acetylated level H3K9 in NR2B promoter region(0.96±0.41) of sodium butyrate group were not significantly different(P>0.05).Compared with the ethanol group,the expression of NR2B protein,NR2BmRNA and acetylated level H3K9 in NR2B promoter region in the hippocampus of ethanol group,these of sodium butyrate + ethanol group were significantly higher (P<0.05).The CPP score were positively correlated with the expression of NR2B protein (r=0.474,P<0.05).The expression of NR2B protein were positively correlated with the expression of NR2BmRNA (r=0.468,P<0.05).The expression of NR2BmRNA were positively correlated with the expression of H3K9 acetylation in NR2B promoter region(r=0.596,P<0.05),and the CPP score were positively correlated with the expression of H3K9 acetylation in NR2B promoter region (r=0.542,P<0.05).Conclusion The increasing acetylation level of H3K9 in NR2B promoter region in the hippocampus may be one of the epigenetic mechanisms of promoting ethanolseeking behavior,and H3K9 deacetylation in NR2B promoter region in the hippocampus is likely to be a new target for controlling ethanol dependence.
8.Influence of maxadilan on human adipose-derived stem cells

[ ABSTRACT] AIM:To investigate the effect of maxadilan, which specifically activates pituitary adenylate cycla-se-activating polypeptide type I receptor (PAC1 receptor), on the proliferation, apoptosis and differentiation potential of human adipose-derived stem cells ( ASCs) .METHODS:ASCs from human adipose tissue were isolated by enzymatic di-gestion and cultured.ASCs were confirmed by the analysis of the markers for cell phenotypes by flow cytometry ( FCM) and adipogenic/osteogenic induction.The effect of maxadilan on ASCs viability was analyzed by CCK-8 assay and FCM.ASCs were irradiated by ultraviolet C ( UVC) at 254 nm and the absorbance of apoptotic ASCs induced by various doses of UVC was measured by CCK-8 assay.ASCs were exposed to 702 J/m2 UVC for 24 h to induce apoptosis.The effect of maxadilan on ASC apoptosis was analyzed by FCM and the determination of caspase 3 and caspase 9 levels.RESULTS:Adipose-de-rived stem cells were confirmed by the detection of the positive expression of cell phenotypes including CD29, CD44, CD59 and CD105 by FCM.The data of CCK-8 assay revealed that ASCs treated with maxadilan (80 nmol/L) had the strongest ability of proliferation.The data of FCM also demonstrated that the addition of 80 nmol/L maxadilan to ASCs in experimen-tal group markedly improved the proliferation capacity of the cells compared with control group (P<0.05).The apoptosis of ASCs exposed to 702 J/m2UVC was dramatically inhibited by the treatment with maxadilan (80 nmol/L).Such process involved the caspase signaling pathway including caspase 3 and caspase 9.There was statistical significance (P<0.05) between experiment group ( ASCs irradiated by UVC and supplemented with maxadilan) and control group ( ASCs only irra-diated by UVC) .Meanwhile, adipogenic and osteogenic differentiation potentials were both positive in experiment group and control group.CONCLUSION:Maxadilan promotes proliferation and inhibits apoptosis of the ASCs.The differentia-tion potential of ASCs toward adipogenic and osteogenic lineages wouldn’ t be altered by maxadilan.Maxadilan would bene-fit to growth and expansion of ASCs in vitro.
9.Transwell contact co-culture promotes growth and differentiation of sin-gle-dissociated iPSCs

[ABSTRACT]AIM:ToinvestigatethepromotingroleofTranswellcontactco-culturesysteminthegrowthand differentiation of single-dissociated induced pluripotent stem cells (iPSCs).METHODS:Bovine corneal endothelial cells (CECs) at passage 1~2 (P1~2) were seeded on the underside of Transwell inserts placed into culture plates and were cultured in 37 ℃and 5%CO2 for 8 h.Accutase digestion and 40μm filter process disaggregated colony-aggregated iPSCs into single-dissociated iPSCs , and the cells were seeded on the inside of Transwell inserts with CECs in medium of mTeSR 1 for 3 d and then in low-glucose DMEM supplemented with 10% FBS for 2 weeks.The characteristics and differentiation markers were evaluated by real-time fluorescence quantitative polymerase chain reaction ( qPCR ) , immunofluorescence staining, live&dead cell staining and alkaline phosphatase (ALP) staining.The group of iPSCs cultured in conventional medium was used as control group 1.The group of single-dissociated iPSCs co-cultured with CECs was set as experimental group, while single-dissociated iPSCs without co-culture were as control group 2.RESULTS: The bovine CECs showed typical hexagonal cobblestone shape .iPSCs showed colony-like growth , while became single-dissociated cells after Tran-swell contact co-culture with bovine CECs for 3 d.The single-dissociated iPSCs positively expressed the undifferentiated markers, Nanog and Oct4.The mRNA expression levels of Nanog , Oct4 and Sox2 between experimental group and control group 1 were both positive and had no statistical significance difference (P>0.05).The dead cells in experimental group decreased significantly, and there was statistically significant difference compared to control group 2 (P<0.01).After 14 d of induced differentiation co-culture , the single-dissociated iPSCs showed rather uniform polygonal morphology , increased dimension and no obvious colony existence .Negative ALP staining, positive immunofluorescence staining for ZO-1, AQP1 and CD31, and negative for CD34 and CD133 were also observed.The results of qPCR showed that the mRNA expression of Oct4, Nanog and Sox2 significantly decreased , and had statistically significant difference compared with control group 1 (P<0.01).CONCLUSION: When co-cultured with bovine CECs, iPSCs morphologically changed to endothelial-like cells and expressed some markers of CECs .Transwell contact co-culture system not only enhances the growth of single-dis-sociated iPSCs , but also promotes their differentiation .
10.Experimental study on changes of Th1/Th2 cytokines and its relationship with lipopolysaccharide in pretreatment of relieving nonalcoholic steatohepatitis

Objective To investigate the changes of Th1/Th2 cytokines and its relationship with lipopolysaccharide (LPS) in pretreatment of relieving nonalcoholic steatohepatitis (NASH).Methods The 24 male Wistar rats were randomly divided into 3 groups: normal control group, liver injury group and LPS pretreatment group. The rats were given normal diet in normal control group,high-sucrose and high-fat diet both in liver injury group and in LPS pretreatment group, and the rats in LPS pretreatment group were given hypodermic injection of LPS 0. 5 mg/kg every other day. The level of plasma endotoxin (ET), activity of alanine aminotransferase (ALT), content of tumor necrosis factor-α (TNF-α) and interleukin-10 (IL-10) were determined. At the end of week 9, the rats were executed, and the liver tissue slices were prepared to investigate hepatic pathologic change by hematoxylin and eosin (HE) staining.Results The level of plasma ET was significantly higher in liver injury group than in normal control group. The level of plasma ALT and infiltrating lymphocytes in liver tissue were significantly lower in LPS pretreatment group than in liver injury group. The level of plasma TNF-α was significantly lower in LPS pretreatment group compared with liver injury group.In contrast, the level of plasma IL-10 was higher (P<0. 05). Histology with HE staining showed that hepatocyte steatosis was obviously relieved with smaller lipid droplet in LPS pretreatment group than in liver injury group. Conclusions LPS pretreatment can alleviate high-sucrose and high-fat induced NASH. The disequilibrium of Th1/Th2 cytokines may be an important part of mechanism.
