1.Relationship between monocyte to high-density lipoprotein cholesterol ratio and atrial fibrillation in diabetes mellitus patients

Objective To explore the relationship between the monocyte to high-density lipoprotein choles-terol ratio(MHR)and atrial fibrillation(AF)in diabetes mellitus(DM)patients by analyzing the clinical da-ta.Methods A total of 132 patients diagnosed with DM in Linfen People's Hospital,the Seventh Clinical Col-lege of Shanxi Medical University from January 2018 to December 2023 were selected as group A,and 139 pa-tients with DM complicated with AF were selected as group B.The clinical data of the two groups were com-pared,and multivariate Logistic regression analysis was applied to identify the influencing factors of DM com-plicated with AF.The diagnostic value of MHR for AF in DM patients was evaluated by receiver operating characteristic(ROC)curve.Results There were statistically significant differences in age,hypertension,coro-nary heart disease,platelet count,neutrophils,lymphocytes,mean red blood cell volume,red blood cell volume distribution width,total bilirubin,conjugated bilirubin,unconjugated bilirubin,high-density lipoprotein choles-terol,neutrophil to lymphocyte ratio,and MHR between group A and group B(P<0.05).The results of mul-tivariate Logistic analysis showed that MHR,age,hypertension,and coronary heart disease were independent risk factors for DM patients with AF(P<0.05).ROC curve analysis showed that the area under the curve of MHR and the combination of age and MHR for diagnosing DM complicated with AF were 0.578(95%CI:0.510-0.646)and 0.811(95%CI:0.759-0.862),respectively.Conclusion MHR is an independent risk fac-tor for DM complicated with AF,and has good diagnostic value combined with age,which can be used to as-sess the risk of the occurrence of AF in the clinical management of DM patients.

Objective To study the indicators of serum inflammation and depression in patients with anxiety-depression after PCI for coronary heart disease(CHD),who were treated with Chinese herbs based on the"heart-liver"theory of traditional Chinese medicine(TCM),and to analyze the curative effect.Methods A computerized retrieval of clinical randomized controlled trials(RCTs)concerning the Chinese herbs therapy based on the"heart-liver"theory of TCM for patients with anxiety-depression after PCI for CHD from the databases of SinoMed,CNKI,Wanfang,VIP,PubMed and Cochrane Library was conducted.The patients of control group received basic treatment for CHD combined with anti-anxiety and depression treatment,while the patients of TCM group received basic treatment for CHD plus Chinese herbs therapy based on the"heart-liver"theory of TCM or received additional Chinese herbs therapy based on the"heart-liver"theory of TCM on the basis of the same treatment regimen as the control group adopted.The retrieval time period was from the establishment of the database to December 30,2023.RevMan 5.4 software was used to make meta-analysis.TSA 0.9.5.1 Beta software was used to make sequential analysis of trials.Results A total of 8 RCTs were included in this study,involving 919 patients.The results of meta-analysis showed that in TCM group the serum inflammatory indicators including serum hs-CRP content,IL-6 content,TNF content,serum MPO content were lower than those in the control group(P<0.05);the serum depression indicators including serum 5-HT content and BDNF content were higher than those in the control group(P<0.05).Based on the above results of the serum indicator analysis,the total effective rate in TCM group was higher than that in the control group(P<0.05).The assessment of disease improvement degree including TCM syndrome score,anxiety scale score,and depression scale score in TCM group were lower than those in the control group(P<0.05).The trial sequential analysis indicated that the sample size effect strength of the total effective rate met the requirements expected from meta-analysis and excluded the possibility of false positives.Conclusion For patients with anxiety-depression after PCI for CHD,Chinese herbs therapy based on the"heart-liver"theory of TCM is superior to basic treatment for CHD combined with anti-anxiety and depression treatment in improving serum inflammation and depression indicators with reliable therapeutic efficacy.
3.Efficacy and safety of rTMS combined with agomelatine in the treatment of depression:a Meta-analysis and trial sequential analysis

Objective To systematically review the efficacy and safety of repetitive transcranial magnetic stimulation(rTMS)combined with agomelatine in the treatment of depression.Methods PubMed,Embase,Cochrane Library,Web of Science,CNKI,VIP,WanFang Data,SinoMed and Yiigle databases were electronically searched to collect randomized controlled trials(RCTs)of comparison between rTMS combined with agomelatine(combined group)and agomelatine or combined pseudostimulation(control group)in the treatment of depression from inception to May 10,2024.Meta-analysis was performed by using RevMan 5.4 software,GRADE grading system for evaluating the level of evidence for observational indicators,and TSA 0.9.5.lBeta software for trial sequential analysis.Results A total of 15 RCTs involving 1 196 patients were included.Meta-analysis showed that compared with control group,combination group had a higher effective rate(RR=1.24,95%CI 1.15 to 1.34,P<0.001),lower depression scale score(SMD=-2.95,95%CI-3.71 to-2.19,P<0.001),anxiety scale score(SMD=-2.21,95%CI-3.29 to-1.14,P<0.001),PSQI score(MD=-1.69,95%CI-2.79 to-0.59,P=0.003).There was no statistical difference between the two groups in terms of NE level,5-HT level and incidence of adverse reactions(P>0.05).GRADE quality grading of the evidence showed moderate-quality evidence for the effectiveness rate and incidence of adverse reactions,with the depression scale score and the NE level of the evidence being low-quality,and the level of evidence for the anxiety scale score,the PSQI score,and the 5-HT content was very low.The trial sequential analysis results showed that in terms of effective rate,the analysis results of combination group superior to control group were reliable,and the conclusion could exclude false positives.Conclusion The rTMS combined with agomelatine in the treatment of depression has advantages in improving effective rate,depression scale score,anxiety scale score and PSQI score compared with agomelatine alone or combined pseudostimulation,incidence of adverse reactions were equivalent to those of agomelatine aloneor or combined pseudostimulation.
4. Pathogen spectrum of viral encephalitis in children living in Hebei province, China from May to December 2017

Objective:
To investigate the pathogenic characteristics of viral encephalitis in children living in Hebei province.
Methods:
We randomly collected cerebrospinal fluid specimens from a total of 399 children diagnosed with viral encephalitis in Hebei Children′s Hospital from May to December 2017. Real-time fluorescence quantitative PCR and Sanger sequencing were used to detect viral nucleic acids in cerebrospinal fluid by an automatic laboratory station. Statistical analysis was performed on the experimental data using SPSS 21.0 software and the clinical data were analyzed. Comparison of infection rates of EV encephalitis in different months, using line × column chi-square test. The MRI and EEG positive rates of different viral encephalitis and viral encephalitis patients not infected with the virus were analyzed by Fisher′s exact probability test. The positive rate of infection with different viruses and non-virus agents was analyzed by Fisher′s exact probability test.
Results:
The result showed that 80 of 399 samples were positive, and the positive rate was 20.05%. It included 22 cases of enterovirus, 4 cases of influenza A virus, 3 cases of mumps virus, 2 cases of herpes simplex virus type 1, 1 case of herpes simplex virus type 2, 4 cases of EB virus, 7 cases of cytomegalovirus, 7 cases of herpes zoster virus, 8 cases of adenovirus, 14 cases of human herpesvirus type 6. Eight cases had combined viral infection. Eight cases had concurrent infections: 3 cases had enterovirus and herpesvirus type 6 concurrent infection, 1 case had enterovirus and Japanese encephalitis virus concurrent infection and 1 case had herpes simplex virus type 2 and adenovirus, 1 case had influenza A virus herpesvirus type 6, 1 case had mumps virus and herpesvirus type 6, 1 case had mumps virus and herpesvirus type 6, 1 case had herpes simplex virus type 1 and herpes zoster virus concurrent infections. 5.Expressions of CD44v6 in acute leukemia and its implication to clinical outcome

Objective To investigate the significance of CD44v6 expression in acute leukemia( AL) and it's relation with the prognosis of AL. Methods Sixty AL patients were treated by enzyme linked immunosorbent assay( ELISA)as initial treatment group. Fourty-seven cases were remission as remission group, and 20 cases with no-remission group. Meanwhile,45 healthy people were served as the control group. The level of CD44v6 was measured by ELISA. Results The serum CD44v6 in initial treatment patients,remission group, no-remission group and control group were( 179. 34 ± 39. 41 )μg/L,( 190. 61 ± 28. 05 )μg/L,( 106. 72 ± 26. 38)μg/L and(98. 31 ± 21. 78)μg/L respectively,and the difference was significant( F =56. 303,P﹤0. 01),and the CD44v6 of initial treatment group and remission group were higher than that of no-remission group and control group(P﹤0. 05). The leukocyte levels was positive related to CD44v6 levels in 60 patients(r=0. 826,P﹤0. 01),and it was also related to disease stage,extramedullary infiltration(( r=0. 485,0. 512;P﹤0. 01). Conclusion The level of CD44v6 is closely related with the occurrence and development of acute leukemia. The assay of CD44v6 in serum of AL patients is helpful in diagnosing and predicting the risk of metastasis and prognosis in AL.
6.Content Determination of Four Effective Constituents in Euphorbiae Semen Decoction

Objective To determine the contents of four effective constituents in Euphorbiae Semen decoction;To provide evidence for Euphorbiae Semen decoction into clinical application. Methods Established quantitative analysis multi-components by single marker method was used to determine the contents of diterpenoids constituents, such as euphorbia storoid, euphorbia factor L2, and euphorbia factor L3. HPLC method was used to determine the contents of aesculetin. Results Contents of euphorbia storoid, euphorbia factor L2, and euphorbia factor L3 in smashed Euphorbiae Semen decoction were 0.015 9%, 0.005 9% and 0.024 1%, respectively. However, the contents of the above three constituents could not be detected in whole Euphorbiae Semen decoction. The content of aesculetin (0.693 6%) in smashed Euphorbiae Semen decoction was more than that in whole Euphorbiae Semen decoction (0.288 2%). Conclusion Decoction digestion effect of diterpenoids constituents in Euphorbiae Semen decoction is not good. Decocting with water is not suitable for the clinical application of Euphorbiae Semen for purgation and diuresis. Aesculetin in smashed Euphorbiae Semen decoction has good decoction digestion effect, in which clinical use for antisepsis and anti-inflammation is effective.
7.Quantitative evaluating the heritability explained by known susceptibility variants of schizophrenia

Objective To evaluate the heritability explanation degree of schizophrenia by all known susceptibility variants in schizophrenia. Methods The GWAS catalog of National Human Genome Research Institute (NHGRI) was queried to retrieve all the susceptible gene variations of schizophrenia. Sixty-two variants with risk allele frequency and odds ratio (OR ) were selected from 347 susceptible gene variants of schizophrenia. The heritability explanation degree of each susceptibility variants was calculated using the multifactorial liability threshold model. Results The total heritabili-ty explanation degree of schizophrenia by 62 known susceptible variants was 25.66%. In contrast, 74.34%of heritability, which could not be explained by known susceptibility variants, were then defined as the missing heritability of schizophre-nia. Conclusions The results demonstrate that the heritability explanation degree of schizophrenia by all known suscepti-bility variants in schizophrenia is low, indicating that there may be many unknown schizophrenia molecular genetic mech-anisms need to be further clarified.
8.Hypothesis of textural composite hydrate system IV: Study on hydration composite ways

BACKGROUND: The water molecules and biological macromolecules in human tissue may combine to form hydrate composite. Different types of macromolecules show different approaches to combine with water molecules. OBJECTIVE: To discuss the basic form of water molecules combined with biological macromolecules. METHODS: The theoretical hypothesis of texture composite hydration system was proposed. The relevant literatures were retrieved through the computer to discuss these issues. RESULTS AND CONCLUSION: According to the role of water in the hydrate composite, the composite ways can be divided into the following 4 groups. ①Surfaca composite: Hydrated membrane layers can form on the surface of globular protein or DNA molecules. The hydrated membrane layer shows that the speed of water molecular and hydrate density were reduced gradually from surface to inside. The hydrated layer makes the shape and physical state of the macromolecules stabilized, and it plays important role in forming of TCHS.②Swelling composite: As a result of the strong hydration of glycosaminoglycans, the hydrated proteoglycan often swells and shows high viscosity and hydrogel character. The hydrated proteoglycan play some mechanicalroles in connective tissue. For example, it can make the cartilage have a strong deformation-resistibility.③Hydrophobic composite: Hydr0phobic effect drives structural rearrangement, including the formation of bilayers (Bio-membrane). This hydrophilic-hydrophobic-hydrophilic "sandwich" structure is conducive to the self-stability in mechanics, combination with membrane protein and transmembrane transport. ④Slit composite: A lot of slit spaces with nano scale were existing inside and outside of the cells. Aqueous solution in the slit spaces shows some abnormal characters such as higher viscosity and lower ice point, because of the rearrangement of water molecules. Slit composite and surface composite have some universality, and it can be regarded the former as a special case of the latter. The classification way mentioned above has not any absolute connotations. However, the rational utilization of these hydration composite ways will be conducive to research the theory on textural composite hydrate system.

The hypoxanthine-guanine phosphoribosyl transferase (HGPRT) gene mutation is responsible for gouty arthritis, kidney stone, and Lesch-Nyhan Syndrome (LNS). It has been reported that the expression of HGPRT is decreased or even absent in these diseases. Rabbits are an ideal model for studying the pathology of these diseases. Therefore, the development of an HGPRT-knockdown rabbit model will be highly beneficial m such studies. Stable HGPRT-knoekdown transgenie fibroblast lines were generated by transfecting rabbit fibroblasts with RNA interference (RNAi) plasmids. Polymerase chain reaction (PCR) analyses indicated that the average positive rate was 83.3%. The mRNA and protein levels of HGPRT in the transgenic fibroblast lines were significantly lower than that in the control. Transgenic rabbit blastocysts were derived after performing nuclear transfer. The results show that RNAi can be used to stably knock down expression of the HGPRT in rabbit fibroblasts and further improvements in related technologies will facilitate the use of this method for the generation of HGPRT-knockdown rabbits.
