Objective:To prepare mesenchymal stem cells with antioxidant capacity (AO-MSC ) from human umbilical cords and evaluate its cell biological properties. Methods:In control group, mesenchymal stem cells (MSC) were isolated by digesting human umbilical cord Wharton's Jelly tissues with 0.2% collagenase Ⅱ,and the released cells were collected and cultured in an animal serum-free culture medium. In AO-MSC group, incompletely collagenase Ⅱ-digested tissue debris were allowed to adhere to flusk flat bottoms and the AO-MSC was harvested by adherent culture. The conventional digestion and culture method was used as control.MSC colony forming ability was evaluated by fibroblast colony forming assay (CFU-F).MSC proliferative capacity was evaluated by CCK-8 assay. The MSC surface markers were detected by using flow cytometry and immunofluorescence staining. The adipogenic and osteogenic capacity of MSC was evaluated by multi-differentiation in vitro, and the mRNA expression of genes that control adipogenic and osteogenic differentiation was detected by real-time fluorescence quantitative PCR (RT-qPCR );Moreover, the mRNA expression of antioxidant substances such as SOD-1,GSH,GAT,and NQO1 in MSC was also evaluated by RT-qPCR.Results:The AO-MSC isolated by this strategy reached a confluence of 80%-90% at around 18 days and grew in a swirling pattern.Flow cytometry and immunofluorescence staining assays showed that CD73,CD29,CD105,CD90 were highly expressed and CD31,CD45,HLA-DR were scarcely expressed in AO-MSC.AO-MSC exhibited stronger self-renewal and differentiation ability compared to MSC.However, the in vitro adipogenic-osteogenic capacity of MSC in the control group was stronger than that of AO-MSC.RT-qPCR assay showed that AO-MSC expressed higher mRNA levels of antioxidant substances compared to MSC.Conclusion:Human AO-MSC is successfully prepared from human umbilical cord without animal serum.

Objective:To explore the application effects of a neurosurgical mixed-reality distance teaching (NMDT) model in standardized residency training in neurosurgery. Methods:We built an NMDT system using mixed-reality technology and remote interaction technology, and designed the implementation procedure according to the teaching objectives. After the teaching activities were completed, a teaching satisfaction questionnaire survey was conducted among 20 neurosurgery resident trainees, in which they provided satisfaction scores for the same teaching content with different teaching models (i.e., the NMDT model and traditional teaching model). SPSS 22.0 software was used to perform the t test for data analysis. Results:There were significant differences between the NMDT model and the traditional teaching model in key indicators including the score for "completion of teaching objectives" (9.20±0.68 vs. 8.25±0.70, P<0.001) and the score for "satisfaction with learning gains" score (8.95±0.67 vs. 8.05±0.92, P=0.001). The NMDT model also outperformed the traditional teaching model in the other individual scores and the total score. Conclusions:The NMDT model can improve teaching quality, increase training efficiency, and enrich teaching content, which is worthy of promotion.

As a potent narcotic opioid analgesic, fentanyl has been used as intravenous anesthesia for clinical procedures while also being abused by drug users, resulting in an increasing number of deaths year by year, and thus it is urgent to develop rapid screening methods for fentanyl. In this study,by using a binary metal-organic framework MIL-88B(Fe/Ni)as an electrocatalyst,an electrochemical sensor based on multi-walled carbon nanotubes(MWCNTs)modified glassy carbon electrode(GCE)was constructed for rapid detection of fentanyl. The morphology, crystal structure and elemental characteristics of the synthesized MIL-88B(Fe/Ni)/MWCNTs and MIL-88B(Fe/Ni)composites were confirmed by scanning electron microscopy(SEM),energy dispersive X-ray spectroscopy(EDS),X-ray diffraction(XRD)and X-ray photoelectron spectroscopy(XPS).In addition, the electrochemical characterization also revealed that the synthesized MIL-88B(Fe/Ni)/MWCNTs composites modified GCE possessed low electrochemical impedance and large electrochemical surface area with ultra-high sensitivity to fentanyl. The experimental conditions for the electrochemical detection of fentanyl were optimized and the limit of detection(LOD,3Sb/M)for fentanyl was 0.015 μmol/L with a linear range of 0.04-10 μmol/L,and the reproducibility, selectivity and stability were all favorable. In addition, the developed sensor showed satisfactory performance in real samples, with recoveries ranging from 58.6%to 81.5%and from 82.7%to 97.5%in urine and blood, respectively,at concentration levels of 0.2-1.0 μmol/L.The proposed MIL-88B(Fe/Ni)/MWCNTs composite could be used as a novel electrode material for electrochemical detection of fentanyl, and had great potential for rapid detection of fentanyl in public security anti-drug or medical emergency.

International Journal of Oral Science 2024;16(1):163-172
A decline in mucosal vascularity is a histological hallmark of oral submucous fibrosis(OSF),a premalignant disease that is largely induced by betel quid chewing. However, the lack of available models has challenged studies of angiogenesis in OSF.Here,we found that the expression of thrombospondin 1(THBS1),an endogenous angiostatic protein, was elevated in the stroma of tissues with OSF.Using a fibroblast-attached organoid(FAO)model, the overexpression of THBS1 in OSF was stably recapitulated in vitro. In the FAO model, treatment with arecoline,a major pathogenic component in areca nuts, enhanced the secretion of transforming growth factor(TGF)-β1 by epithelial cells, which then promoted the expression of THBS1 in fibroblasts.Furthermore, human umbilical vein endothelial cells(HUVECs)were incorporated into the FAO to mimic the vascularized component.Overexpression of THBS1 in fibroblasts drastically suppressed the sprouting ability of endothelial cells in vascularized FAOs(vFAOs).Consistently, treatment with arecoline reduced the expression of CD31 in vFAOs,and this effect was attenuated when the endothelial cells were preincubated with neutralizing antibody of CD36,a receptor of THBS1.Finally,in an arecoline-induced rat OSF model,THBS1 inhibition alleviated collagen deposition and the decline in vascularity in vivo.Overall,we exploited an assembled organoid model to study OSF pathogenesis and provide a rationale for targeting THBS1.
