1.Research on the policy and management system of emergency rescue materials in response to major infectious disease emergencies in China

Objective:To sort out China's emergency rescue material support policies and management systems for responding to major infectious disease emergencies in China,to analyze the effects of policy implementation at each stage,and to improve emergency management capabilities of public hospitals.Methods:The China National Knowledge Infrastructure(CNKI),Wanfang,VIP and he State Council Policy Document databases from 1949 to 2023 were searched,and 538 research literatures related to major infectious disease emergencies and emergency rescue material support were retrieved,and 79 research literatures were finally included according to the inclusion and exclusion criteria.The relevant policy documents,research reports and academic articles were sorted and analyzed using literature analysis methods,emergency management theory and organizational resilience theory.Results:The 79 research literatures on the evolution of emergency rescue material support policies in response to major infectious disease emergencies in China can be divided into four stages and three periods.14 research papers were included in the embryonic period of emergency management,including the two stages from the founding of People's Republic of China to the period before the reform and opening up(7 papers)and from the reform and opening to the SARS epidemic in 2003(7 papers),during which the government was in a single response and decentralized and extensive management mode.30 research papers were included in the comprehensive coordination emergency management model period,which was from the after of the SARS epidemic in 2003 to the beginning of 2018,during which the first"one plan,three systems"(emergency response plan,emergency management system,mechanism and legal system)were proposed and gradually moved towards informatization and intelligence.35 research papers were included in the comprehensive emergency management model period,which began in early 2018,and the main feature was the establishment of a centralized and unified emergency management system at the national level.Public hospitals can learn from the current situation of emergency rescue material support policy and management system in China and enhance the emergency management capabilities from the whole process of before,during,and after emergencies.Conclusion:The policy evolution in three stages has gradually adapted to the complex public health emergencies,which could better realize the mature stage of legalization,intelligence and refinement of emergency management of China.Public hospitals should closely follow the requirements of the national policy system,continuously improve the emergency management mechanisms and enhance the ability to deal with major infectious disease emergencies.
2.Epidemiological investigation and etiological analysis on a case of human infection with avian influenza A (H3N8) virus in Changsha

Objective:To understand the epidemiological and clinical characteristics of human infection with avian influenza A(H3N8) virus and the molecular biological characteristics of the pathogen, and provide scientific evidence for the prevention and control of human infection with avian influenza A(H3N8) virus.Methods:An epidemiological investigation was conducted for a case of human infection with avian influenza A(H3N8) virus in Changsha in May 2022 to collect the information about exposure history, route of infection, onset and treatment, potential contacts and other possible exposures for a descriptive analysis. Nucleic acid detection and gene sequencing were used to detect the pathogen and analyze the genetic characteristics.Results:The case had a history of exposure to live poultry trading market 6 days before the onset of illness. Subtypes H3 and N8 of avian influenza virus were detected in live poultry markets. Deep gene sequencing showed that the virus had adaptive mutations in mammals, reduced sensitivity to alkamine agent, and no resistance mutations related to neuraminidase inhibitors and polymerase inhibitors were detected.Conclusion:The case was infected due to exposure to the live poultry market environment contaminated by avian influenza A(H3N8) virus, and no human to human transmission was found.
3.DAPT attenuates ox-LDL-induced human umbilical vein endothelial cell injury

AIM:To investigate the effect of N-[N-(3,5-difluorophenacetyl)-L-alanyl]-S-phenylglycine t-butyl ester (DAPT) on the Notch signaling pathway in a model of oxidized low-density lipoprotein (ox-LDL)-induced human umbilical vein endothelial cell (HUVEC) damage.METHODS:HUVECs were divided into control group, ox-LDL group, DAPT group and ox-LDL+DAPT group.The morphological changes of the HUVECs with different treatments were observed under light microscope.The viability of the HUVECs was measured by CCK-8 assay.The protein expression levels of Notch1, Notch4 and Jagged1 were determined by Western blot.RESULTS:ox-LDL induced great damage to the HUVECs, evidenced by increased cell death and debris in the culture.However, the cell damage was abolished by adding DAPT into the culture.The viability of the HUVECs was increased by co-treatment with DAPT and ox-LDL.ox-LDL treatment significantly decreased the protein expression levels of Notch1 and Jagged1, and elevated Notch4.However, these changes were totally reversed by DAPT.None of these proteins showed significant change in the HUVECs co-treated with DAPT and ox-LDL as compared with control group.CONCLUSION:ox-LDL is able to induce HUVEC damage in vitro.DAPT attenuates ox-LDL-induced damage in the HUVECs by regulating the Notch signaling pathway.
5.Slow synaptic transmission mediated by 5-HT_(1P) receptor in sympathetic ganalion of guinea pig

AIM To investigate the effects of several 5-hydroxytryptamine(5-HT) subtype receptor antagonists on late slow excitatory postsynaptic potential(LS-EPSP) of the neurons of guinea pig inferior mesenteric ganglion(IMG). METHODS Intracellular recordings were made from neurons of the isolated guinea pig IMG. RESULTS Cyproheptadine and BRL 24924 suppressed LS-EPSP of 5-HT sensitive neurons reversibly, while mianserin, MDL 72222 and spiperone showed no significant effect. Continuous superfusion of IMG with MCPP suppressed LS-EPSP of 5-HT sensitive neurons by 5-HT 1P receptor desensitizated. CONCLUSION LS-EPSP of 5-HT sensitive neuron is mediated by 5-HT 1P subtype receptor.
