1.Diagnostic performance evaluation of artificial intelligence-assisted diagnostic systems in cervical cytopathological examination

Objective:To evaluate the diagnostic performance of artificial intelligence-assisted diagnostic systems in cervical cytopathological examination.Methods:Cervical cytology slide data were retrospectively collected from four hospitals for the external validation of the developed artificial intelligence-assisted diagnostic system. Subsequently, prospective data collection was conducted for human-machine assisted studies.Results:In the retrospective study, a total of 3 162 valid samples were collected as external validation data. The system showed an area under the curve (AUC) of 0.890 (95% CI: 0.878-0.902), accuracy of 0.885 (95% CI: 0.873-0.896), sensitivity of 0.928 (95% CI: 0.914-0.941), and specificity of 0.852 (95% CI: 0.834-0.867). In the prospective study, 212 valid samples were collected, and five junior cytologists participated in the human-machine assisted study. Without artificial intelligence assistance, the average AUC for the five cytologists was 0.686 (95% CI: 0.650-0.722), the accuracy was 0.699 (95% CI: 0.671-0.727), the sensitivity was 0.653 (95% CI: 0.599-0.703), the specificity was 0.719 (95% CI: 0.685-0.750), the Fleiss κ value was 0.510, and the reading time was 223 seconds. With artificial intelligence assistance, the AUC, accuracy, sensitivity, and specificity increased by 0.166, 0.143, 0.225, and 0.107, respectively. Additionally, Fleiss κ was 0.730 and the reading time decreased by 188 seconds. All differences were statistically significant (all P<0.001). Conclusions:Artificial intelligence-assisted diagnosis system shows excellent performance and good generalizability, significantly improving the diagnostic accuracy, consistency, and efficiency of junior cytologists. It can be an effective auxiliary tool for junior cytologists in clinical practice.

3.Stimulating action of capitate phrynium stem and leaf extract on the rodent animals uterus

AIM: To study the exciting effects of ethanol extract of capitate phrynium (Phrynium Capitatum Willd) stem and leaf on the uterine smooth muscle of rat,mice,guinea pig and rabbit pretreated by diethylstilbestrol. METHODS: Conventional method was adopted to prepare animal uterine specimens both in vivo and in vitro. Different doses of the extract were given to the specimens and recorded their tensility, frequency and activity of contraction with the help of BL-420 biological experimental system. IC_(50) of the extract was calculated, with oxytocin as standard comparison. RESULTS: 95% ethanol extract and 50% ethanol extract of capitate phrynium and leaf had obviously stimulating action on the uterine smooth muscle of rat, mice, guinea pig and rabbit in a dose-dependent manner in vitro, whichinclude accelerating contraction frequency, raising tension (P<0.05, P<0.01). In addition, 50% ethanol extract also had a significant stimulating action on uterine smooth muscle of rabbit in vivo(P< 0.01). CONCLUSION: Ethanol extracts of capitate phrynium have an effect on rodent uterine. Smooth muscle indicates that clinical application of Chinese practitioner used to treat the dysfunctional uterine bleeding proves to be effective.
4.Clinical significance for the detection of HLA-B27 in 98 donors' peripheral blood

Objective To establish and optimize flow cytometry method for the detection of human leukocyte antigen B27 (HLA B27) and compare it with serological method. Methods A total of 98 samples from blood donors were entered into the study. HLA B27 was detected with ethylenediaminetetra aceticacid(EDTA), heparin sodium and different dosages of B27 monoclonal antibody, respectively. Other 30 samples were typed by microlymphocytotoxicity test(serological method). Results Genotyping for HLA B27 was successfully performed in 98 samples by flow cytometry, containing 2 cases with B27 positive, 96 cases with B27 negative. The types of anticoagulant agents and dosages of monoclonal antibody(7~30 ?l) had no remarkable effect on the results. The results of serological method for the detection of 30 samples were consistent with those by flow cytometry method. Conclusion Genotyping for HLA B27 by flow cytometry is a rapid and accurate technique with high sensitivity, suitable for clinical application. Detection of HLA B27, a very important basis for the diagnosis and differential diagnosis of ankylosing spondylitis(AS) and other joint diseases of the vertebral column, is of a remarkable clinical significance.
