Objective To monitor rodent species in the southern Fujian ports of China and investigate the natural pathogens they carry. Methods Rodents were captured using night-trapping and cage method across 17 port areas in southern Fujian in 2023. Morphological identification was performed to classify the captured specimens. Tissues from the liver, spleen, lungs, kidneys, and bladder were harvested for nucleic acid extraction. Nested and conventional PCR method were used to detect seven pathogens: Yersinia pestis, Hantavirus, pathogenic Leptospira, Borrelia burgdorferi, Bartonella, Orientia tsutsugamushi, and Anaplasma phagocytophilum. The Chi-square test was used to analyze the distribution of rodents and the status of natural pathogen infections across the study sites. Results A total of 161 rodents across five species belonging to two orders, two families, and three genera were captured. Suncus murinus was the dominant species(72.67 %), followed by Rattus tanezumi(13.04 %). Six pathogens were detected, with an overall positivity rate of 18.01%. Bartonella had the highest positivity rate(11.18%), followed by pathogenic Leptospira(4.97%), Borrelia burgdorferi(2.48%), Orientia tsutsugamushi(1.86%), Seoul virus(1.24%), and Anaplasma phagocytophilum(0.62%). Co-infections were noted in five rodents, representing 17.24% of those that tested positive, and one rodent was infected with four pathogens. No Y. pestis or Hantaan virus was detected in the captured rodents. Conclusions The rodents in the southern Fujian ports are naturally infected with multiple pathogens, including Bartonella, pathogenic Leptospira, and Seoul virus. Enhanced monitoring of rodent populations and the implementation of preventive and control measures for rodent-borne diseases are essential.

Objective To explore the relationship and internal path between activities of daily living(ADL),sleep quality and mental health of community elderly people in Shanghai. Methods A questionnaire survey was conducted among community residents aged 60 years and older seeing doctors in community health care center of five streets in Shanghai during Sept to Dec,2021 using convenience sampling.Activities of Daily Living(ADL),Pittsburgh Sleep Quality Index(PSQI)and 10-item Kessler Psychological Distress Scale(K10)were adopted in the survey.Single factor analysis, correlation analysis and multiple linear regression were used to analyze the data. The effect relationship between the variables was tested using Bootstrap's mediated effects test. Results A total of 1 864 participants were included in the study. The average score was 15.53±4.47 for ADL,5.60±3.71 for PSQI and 15.50±6.28 for K10.The rate of ADL impairment, poor sleep quality, poor and very poor mental health of the elderly were 23.6%,27.3%,11.9%and 4.9%,respectively.ADL and sleep quality were all positively correlated with mental health(r=0.321,P<0.001;r=0.466,P<0.001);ADL was positively correlated with sleep quality(r=0.294,P<0.001).Multiple linear results of factors influencing mental health showed that ADL(β= 0.457,95%CI:0.341-0.573),sleep quality(β =0.667,95%CI:0.598-0.737)and mental health were positively correlated(P<0.001).Sleep quality partially mediated the relationship between ADL and mental health(95%CI:0.078-0.124)with an effect size of 33.0%.Conclusion Sleep quality is a mediator between ADL and mental health among community elderly people.Improving ADL and sleep quality may improve mental health in the population.

AIM:To explore the mechanism by which Qingshen granules(QSG)intervene in the microRNA-4516(miR-4516)targeted regulation of the SIAH3/PINK1 axis, enhancing mitophagy and inhibiting renal fibrosis.METHODS:Male SD rats were randomly divided into three groups:control, model, and QSG groups. The QSG aqueous solution was administered via gavage once daily,4 mL each time, for 8 consecutive weeks.Blood creatinine levels were measured in each group.Hematoxylin-eosin(HE)and Masson staining were utilized to assess the degree of renal patholog-ical damage.Western blot analysis was performed to determine the expression levels of β-actin,PINK1,Parkin,SIAH3,VDAC1,Mfn1,Mfn2,OPA1,LC3B,and P62 proteins in renal tissue.RT-qPCR was used to detect the mRNA expres-sion level of SIAH3 in rat kidney tissue, and transmission electron microscopy was employed to observe mitochondrial dam-age in renal tissue.QSG-containing serum and transforming growth factor-β1(TGF-β1)were used to induce an HK-2 cell fibrosis model. The cells were divided into the following groups:normal cell(NC)group, model cell(MC)group,MC+miR-4516 mimics group,MC+miR-4516 NC+QSG group,MC+miR-4516 mimics+QSG group, and MC+QSG group.Cell activity in each group was detected using the CCK-8 method, and Western blot analysis was performed to determine E-cad-herin and α-SMA protein expression levels. The regulation of SIAH3 by miR-4516 was verified using a dual luciferase re-porter assay.RT-qPCR was used to detect the mRNA expression of miR-4516,SIAH3 mRNA,and PINK1.RESULTS:The results indicated that QSG intervention reduced fibrosis in rat renal tissue and HK-2 cells, decreased SIAH3 mRNA expression, increased PINK1 expression, and activated mitophagy in renal tissue. In vitro results confirmed that QSG can elevate miR-4516 expression, inhibit SIAH3 mRNA expression, promote PINK1 expression in HK-2 cells, and reduce the expression of the fibrosis marker protein α-SMA.CONCLUSION:In summary, this study preliminarily clarified the mechanism by which QSG intervention targets miR-4516 to regulate the SIAH3/PINK1 axis, thereby enhancing mitophagy and inhibiting renal fibrosis.

Chinese journal of integrative medicine 2024;30(11):1035-1044
OBJECTIVE:
To investigate changes of myeloid differentiation factor 2 (MD2) in inflammation-induced pain and acupuncture-mediated analgesia.
METHODS:
Mice were randomly divided into three groups by a random number table method: saline group (n=16), complete Freund's adjuvant (CFA) group (n=24) and CFA+electroacupuncture (EA) group (n=26). Inflammation-induced pain was modelled by injecting CFA to the plantar surface of the hind paw of mice and EA was applied to bilateral Zusanli (ST 36) to alleviate pain. Only mice in the CFA+EA group received EA treatment (30 min/d for 2 weeks) 24 h after modelling. Mice in the saline and CFA groups received sham EA. von-Frey test and Hargreaves test were used to assess the pain threshold. Brain and spinal tissues were collected for immunofluorescence staining or Western blotting to quantify changes of MD2 expression.
RESULTS:
CFA successfully induced plantar pain and EA significantly alleviated pain 3 days after modelling (P<0.01). Compared with the CFA group, the number of MD2+/c-fos+ neurons was significantly increased in the dorsal horn of the spinal cord 7 and 14 days after EA, especially in laminae I - IIo (P<0.01). The proportion of double positive cells to the number of c-fos positive cells and the mean fluorescence intensity of MD2 neurons were also significantly increased in laminae I - IIo (P<0.01). Western blotting showed that the level of MD2 was significantly decreased by EA only in the hippocampus on day 7 and 14 (both P<0.01) and no significant changes were observed in the cortex, thalamus, cerebellum, or the brainstem (P<0.05). Fluorescence staining showed significant decrease in the level of MD2 in periagueductal gray (PAG) and locus coeruleus (LC) after CFA injection on day 7 (P<0.01 for PAG, P<0.05 for LC) and EA significantly reversed this decrease (P<0.01 for PAG, P<0.05 for LC).
CONCLUSION
The unique changes of MD2 suggest that EA may exert the analgesic effect through modulating neuronal activities of the superficial laminae of the spinal cord and certain regions of the brain.
