1.A preliminary study on Toxoplasma gondii interfering with copper metabolism pathways in mouse kidney

ObjectiveTo investigate the effect of Toxoplasma gondii infection on copper metabolism in the kidneys of mice. MethodsA total of 80 7-8-week-old C57BL/6 female mice were randomly divided into four groups of 20 mice in each group after one week of adaptation, including Control group, Cu group, TgCtwh6 group and Cu+TgCtwh6 group. Mice that were not infected and fed with normal diet and water were used as the Control group; Mice fed with 1 g/kg of copper chloride processing diet and 0.1% copper chloride water for 60 consecutive days were used as Cu group; Mice infected with 25-30 TgCtwh6 cysts (one of the predominant genotype Chinese 1 in China) fed with normal diet and water were used as the TgCtwh6 group; mice infected with 25-30 TgCtwh6 cysts and fed with a processed diet containing 1 g/kg of copper chloride and water with 0.1% copper chloride for 60 consecutive days were used as the Cu+TgCtwh6 group. ICP-MS was used to determine the changes in copper content in kidney tissues. Hematoxylin-eosin (HE) staining was used to observe the pathological changes of mouse kidney tissue. The number of apoptotic cells was observed by PI staining. Western blot was used to detect the protein expression levels of glutathione peroxidase 4 (GPX4) and superoxide dismutase (SOD1, SOD2). RT-qPCR was used to detect the mRNA expression of cuproptosis-related genes. 2.Constructing a rat animal model of pelvic organ prolapse:a comparison of three modeling methods

BACKGROUND:Currently,there are many modeling methods for pelvic organ prolapse animal models,and the commonly used methods are vaginal balloon dilatation,oophorectomy and the combination of the two.There is no study comparing the three modeling methods in detail.OBJECTIVE:To construct and validate a rat animal model of pelvic organ prolapse using three different methods and to identify the advantages and disadvantages of various models.METHODS:Seventy-two 8-week SPF-grade female Sprague-Dawley rats were selected and randomly divided into four groups,namely,vaginal balloon dilatation group,ovariectomy group,ovariectomy combined with vaginal balloon dilatation group(the combined group),and the sham-operated group(no ovariectomy and no vaginal dilatation).The vaginal wall tissues of rats were collected at 4,8 and 12 weeks after the operation for hematoxylin-eosin staining,Masson staining,EVG staining and immunohistochemical staining of α-smooth muscle actin,Vimentin and matrix metalloproteinase 9 detection,and the pelvic floor muscle tissues were taken at 4,8 and 12 weeks after the operation for hematoxylin-eosin staining,Masson staining and EVG staining.RESULTS AND CONCLUSION:(1)Hematoxylin-eosi staining showed that there was no significant difference in the decrease of vaginal epithelial layer thickness in the vaginal balloon dilatation group compared with the sham-operated group,(P>0.05),while the thickness of the vaginal epithelial layer was significantly reduced in the ovariectomy group and the ovariectomy combined with vaginal balloon dilation group(P<0.001),and the reduction was more significant in the ovariectomy combined with vaginal balloon dilation group,remained stable at 8 weeks after surgery and lasted until 12 weeks.(2)The changes in the content of collagen fibers and elastic fibers in the vaginal wall stained by Masson and EVG staining were the same as the changes in the thickness of the vaginal epithelial layer stained by hematoxylin-eosin,and there were no changes in collagen fibers and elastic fibers in the pelvic floor muscle tissues of the treatment groups.(3)At 4,8 and 12 weeks after treatment,there was no significant difference in the expression levels of α-smooth muscle actin,Vimentin and matrix metalloproteinase 9 in the vaginal wall tissue of the balloon dilation group compared with the control group(P>0.05),whereas the expression levels of α-smooth muscle actin and Vimentin were significantly decreased in the ovariectomy group and ovariectomy combined with vaginal balloon dilation group(P<0.01)and the expression of matrix metalloproteinase 9 showed a significant increase(P<0.01),with a more pronounced increase in the ovariectomy combined with vaginal balloon dilation group,and the increase reached a stable state at 8 weeks after surgery and could persist up to 12 weeks.To conclude,vaginal balloon dilatation could not maintain the degeneration of pelvic organ prolapse formed by the vaginal wall for a long period,and both ovariectomy and the combined method can be used.Ovariectomy combined with vaginal balloon dilatation can significantly accelerate and aggravate the formation of typical histological features of pelvic organ prolapse in vaginal wall tissues,effectively shorten the experimental period,and improve the efficiency.These effects reach a stable state at 8 weeks after surgery and can be sustained up to 12 weeks,which is practical and convenient for the study of pelvic organ prolapse animal models.

4.Effects of androgens on cognitive function in castration male mice

ObjectiveTo establish a castrated male mouse model and to preliminarily investigate the effects of testosterone replacement therapy (TRT) on behavior, serum indices, and histopathological changes in castrated mice, as well as to explore the role of androgens in cognitive function. MethodsForty 6-month-old male C57/BL6J mice were randomly divided into sham operation group, castration group, testosterone propionate (0.5,1.0 mg/kg) treated group, with 10 mice in each group. Following castration and subcutaneous administration of testosterone propionate at different doses (0.5 and 1.0 mg/kg) for TRT, learning and memory abilities were assessed using the Morris water maze (MWM) test and the passive avoidance test. Serum testosterone and serum brain-derived neurotrophic factor (BDNF) levels were measured by ELISA, and histopathological changes in the hippocampus were examined using hematoxylin-eosin (HE) staining. ResultsRoutine observations: there were no statistically significant differences in body weight among groups at any time point. MWM test: compared with castration group, sham operation group and testosterone propionate-treated groups (0.5, 1.0 mg/kg) showed significantly reduced escape latency on days 4 and 5 (P0.05), while the number of platform crossings and the time spent in the target quadrant significantly increased (P0.05). Passive avoidance test: the number of passive avoidance errors significantly decreased in sham operation group and testosterone propionate (1.0 mg/kg)-treated group (P0.05), and the passive avoidance latency was significantly prolonged in sham-operated group and testosterone propionate-treated groups (0.5, 1.0 mg/kg) (P0.05). Serum testosterone and serum BDNF assays: serum testosterone levels and serum BDNF concentrations significantly increased in sham operation group and testosterone propionate-treated groups (0.5, 1.0 mg/kg) (P0.01). HE staining: compared with sham operation group, neuronal density in all hippocampal subregions was slightly reduced in castration group; in the testosterone propionate (0.5 mg/kg)-treated group, neuronal arrangement in the CA1 and CA3 regions was improved and apoptotic cells were reduced compared with castration group; in testosterone propionate (1.0 mg/kg)-treated group, the pyramidal cell layer in the CA3 region was more compactly arranged, with fewer apoptotic cells than in castration group. ConclusionTRT improves learning and memory performance in castration male mice, potentially through modulation of hippocampal BDNF signaling pathways.
5.Loss of histone H3K27me3 up-regulates SLC7A11 in diffuse gastric cancer cells

Objective To map the genome-wide distribution profile of histone H3K27me3 modification in diffuse gastric cancer tissues,identify target genes regulated by H3K27me3,and primarily explore the potential mechanism of its modification reprogramming in the occurrence and development of the tumor.Methods Normal gastric mucosal tissues and diffuse gastric cancer tissues were harvested from the patients who underwent examinations or treatments in the departments of gastroenterology and gastrointestinal surgery of our medical center between 2021 and 2023.There were 14 patients in the normal group(6 males and 8 females,average age of 46 years)and 14 patients in the gastric cancer group(8 males and 6 females,average age of 63 years).Cleavage under target and tagmentation(CUT&Tag)technology was employed to capture genomic regions modified by H3K27me3,and analyze the reprogramming characteristics of these modifications.RNA sequencing data,data from high-throughput chromosome conformation capture(Hi-C)technology,and publicly available single-cell data were integrated to investigate the target genes regulated by the reprogramming of H3K27me3 modifications in diffuse gastric cancer cells.Results The quality of the CUT&Tag and RNA sequencing data met the standards required for subsequent analysis.Histone H3K27me3 modifications in normal gastric mucosa and diffuse gastric cancer tissues were primarily distributed in distal intergenic regions and intronic regions.In gastric cancer tissues,compared to normal tissues,there was significant reprogramming of H3K27me3 modifications,characterized by a marked reduction in overall H3K27me3 signal intensity.The loss of 2 912 H3K27me3 signal peaks might lead to the up-regulation of 822 tumor-associated genes.Among them,56 genes displayed the most significant up-regulation(fold change in signal intensity≥2,P<0.05),with notable enrichment in the mammalian target of rapamycin complex 1(mTORC1)signaling pathway.Specifically,the methionine transporter SLC7A5 and the cystine transporter SLC7A11 were found to have the highest expression levels in gastric cancer tissues.Single-cell data revealed that the abnormal overexpression of SLC7A11 in diffuse gastric cancer was primarily observed in tumor epithelial cells.Further validation using public data and immunohistochemical experiments confirmed the elevated expression of SLC7A11 in diffuse gastric cancer,which is associated with poor prognosis in gastric cancer patients.Conclusion The reprogramming of histone H3K27me3 modification is an important epigenetic characteristic in diffuse gastric cancer.Loss of H3K27me3 signal peaks may up-regulate the expression of SLC7A11 in diffuse gastric cancer cells,and thereby promote tumor progression.
6.Quantification of Atmospheric Total Reactive Nitrogen Oxides by Thermal Decomposition-Broadband Cavity Enhanced Absorption Spectroscopy

Nitrogen oxides(NOx=NO+NO2)are important precursors of ozone(O3),and NOx and its oxides together constitute reactive nitrogen oxides(NOy)in the atmosphere.A comprehensive understanding of the total NOy level in the atmosphere is of great significance for a deeper understanding of the atmospheric nitrogen cycle and oxidation,as well as for formulating strategies for air pollution prevention and control.In this work,a thermal decomposition-broadband cavity enhanced absorption spectroscopy(TD-BBCEAS)technique for online measurement of total NOy in the atmosphere was developed.With this method,the NOy was efficiently converted into NO2,and the total NOy concentration in the atmosphere was indirectly obtained by measuring NO2.Focusing on the key factors affecting the measurement of total NOy,the influence of NO titration efficiency and other NOy component TD efficiency on measurement accuracy was emphasized.By changing the oxygen(O2)flow rate through the mercury lamp to alter the O3 concentration for titrating NO,the conversion efficiency of NO was evaluated.At O2 flow rate of 6 mL/min,the conversion efficiency of NO was greater than 99%.TD efficiency testing and analysis on NO2,peroxyacetyl nitrate(PAN),nitric acid(HNO3),and nitrous acid(HONO),which account for a large proportion of atmospheric NOy components,was carried out using 680℃as the optimal TD temperature for efficient conversion of NOy.With NO and HONO sample gases as typical verification gases,the conversion efficiency of NOy and the accuracy of NOy measurement by TD-BBCEAS system were verified by switching the on and off modes of mercury lamp and TD device.At integration time of 60 s,the detection limit of the system for NOy was 2.83×1010 molecules/cm3(60 s,2σ).A comparative measurement of actual atmospheric NOy was conducted between the TD-BBCEAS system and the NOy analyzer.The observation results showed a correlation coefficient(R2)of 0.98 and a slope of 0.93,further verifying the feasibility and accuracy of applying the TD-BBCEAS system to measurement of total NOy.
7.Regional Differences and Source Apportionment of Atmospheric Volatile Organic Compounds in A Typical Industrial City During Summer

A method for determining volatile organic compounds(VOCs)in environmental air samples using thermal desorption-gas chromatography/mass spectrometry(TD-GC/MS)was developed.The qualitative and quantitative analyses of 103 kinds of VOC species were achieved under the optimal conditions such as cold trap desorption temperature and time during the thermal desorption process,combined with full-scan data acquisition using an electron impact ionization source.With a sampling volume of 3.0 L,the method exhibited detection limits of 0.1-0.5 μg/m3 and quantitation limits of 0.4-2.0 μg/m3.At spiked concentrations of 1.0 μg/m3 and 10.0 μg/m3,the recoveries ranged from 60.5%to 118.0%,with relative standard deviations varying from 2.37%to 18.70%.Furthermore,all target compounds showed correlation coefficients(R2)exceeding 0.997 across their respective concentration ranges,demonstrating that the method had high accuracy and reliability.Using this method,a study was conducted to investigate the spatial variations and source apportionment of VOCs across urban,suburban,and rural sites in Anyang,a representative industrial city,during the summer season.The results revealed significant differences in VOC concentrations among the three regions.The urban site recorded the highest concentration at 40.1 μg/m3,followed by the rural site at 23.5 μg/m3,while the suburban site had the lowest concentration of 9.74 μg/m3.With regard to compositional characteristics,alkanes were the dominant components in the urban and rural areas,whereas oxygenated VOCs were predominant in the suburban site.The ozone formation potential(OFP)also varied significantly across regions:96.0 μg/m3 in urban areas,72.0 μg/m3 in rural areas,and only 27.6 μg/m3 in suburban areas.Alkenes were identified as the primary contributors to the total ozone formation potential(TOFP)in all regions,highlighting their critical role in atmospheric oxidation processes.Source apportionment analysis using the positive matrix factorization(PMF)model identified combustion sources,natural sources,chemical industry emissions,industrial emissions,solvent use,and vehicle emissions as the major sources of VOCs in Anyang during summer.Notably,chemical industry emissions and combustion sources were dominant in urban and rural areas,whereas combustion sources and natural sources were more prominent in the suburban area,reflecting distinct emission patterns and anthropogenic activities across the regions.
8.Correlation between serum circUSP36,circBPTF and severity of diabetic retinopathy

Objective To explore the correlation between the levels of serum circUSP36 and circBPTF in patients with diabetic retinopathy(DR)and the severity of the disease.Methods Totally 175 DR patients who visited Luzhou People's Hospital from January 2022 to October 2024 were selected as the research sub-jects.According to the international clinical DR classification standard,they were divided into the non-signifi-cant DR group(NDR group),non-proliferative DR group(NPDR group),and proliferative DR group(PDR group),with 72 cases in the NDR group,63 cases in the NPDR group,and 40 cases in the PDR group.Another 50 healthy individuals who underwent physical examinations during the same period were selected as the con-trol group.The levels of serum circUSP36 and circBPTF in each group were determined by fluorescence quan-titative PCR.The correlation between the expression levels of serum circUSP36 and circBPTF in patients of the PDR group was analyzed by Pearson method.Logistic regression analysis was used to analyze the factors of DR.Receiver operating characteristic(ROC)curve was used to analyze the evaluation value of the expres-sion levels of serum circUSP36 and circBPTF in DR patients for the severity of DR.Results There were sta-tistically significant differences in the levels of circUSP36,circBPTF,fasting plasma glucose(FPG),2-hour postprandial blood glucose(2 hPG),glycated hemoglobin(HbA1c)between the NDR group,the NPDR group,the PDR group and the control group(P<0.05).The levels of circUSP36,circBPTF,FPG,2 hPG and HbA1c increased with the severity of DR(P<0.05).The expression levels of serum circUSP36 and circBPTF in patients of the PDR group were positively correlated(r=0.441,P=0.004).The duration of T2DM,high circUSP36,and high circBPTF were all independent risk factors for PDR(P<0.05).The area under the curve(AUC)of serum circUSP36 and circBPTF alone and in combination for predicting NPDR were 0.678,0.698,and 0.775,respectively,and the predictive value of the combination of serum circUSP36 and circBPTF was su-perior to that of their individual detections(P<0.05).The AUCs of serum circUSP36 and circBPTF alone and in combination for predicting PDR were 0.788,0.787,and 0.859,respectively,and the combination of the two was superior to their individual detections(P<0.05).Conclusion The levels of serum circUSP36 and circBPTF are elevated in DR patients,and with the aggravation of DR,the expression levels of both show an upward trend.The combined detection of the two has certain predictive value for the severity of DR.
9.Effects of modified Chaihu Jia Longgu Muli Decoction on the efficacy of Parkinson's disease with liver and kidney yin deficiency syndrome and the phosphatase and tensin homolog-induced putative kinase 1 (PINK1)/E3 ubiquitin ligase (Parkin) signaling pathway induced by phosphatase and tensin homologues

Objective:To explore the clinical efficacy of modified Chaihu Jia Longgu Muli Decoction in patients with liver and kidney Yin deficiency syndrome of Parkinson's disease (PD) and the effects on phosphatase and tensin homolog-induced putative kinase 1 (PINK1) /E3 ubiquitin ligase (Parkin) signaling pathway.Methods:A randomized controlled trial study was conducted. A total of 120 PD patients from the Department of Encephalopathy of Xi'an Hospital of Traditional Chinese Medicine from May 2020 to May 2024 were selected as the observation objects and divided into 2 groups through random number table method, with 60 cases in each group. The control group took dopashydrazine tablets orally, while the combined group took Chaihu Jia Longgu Muli Decoction on the basis of the control group. Two groups were treated continuously for 2 months. The levels of glial cell-derived neurotrophic factor (GDNF), nerve growth factor (NGF), brain-derived neurotrophic factor (BDNF), and basic fibroblast growth factor 2 (FGF2) were detected by ELISA. The expression levels of PINK1, Parkin and chelate 1 (SQSTM1) mRNA were detected by real-time fluorescence quantitative PCR. The adverse reactions during the treatment were observed and recorded to evaluate the clinical efficacy.Results:The total effective rate was 95.00% (57/60) in the combined group and 83.33% (50/60) in the control group, with statistical significance ( χ2=4.23, P=0.040). After treatment, the levels of serum GDNF [(510.78±31.57) ng/L vs. (431.89±30.45) ng/L, t=13.93], BDNF [(36.47±3.72) ng/L vs. (27.84±3.55) ng/L, t=13.00], NGF [(57.24±4.17) ng/L vs. (50.72±4.11) ng/L, t=8.63] and FGF2 [(26.21±3.71) ng/L vs. (21.63±3.54) ng/L, t=6.92] in the combined group were higher than those in the control group ( P<0.001). After treatment, the mRNA expression levels of PINK1 (2.38±0.07 vs. 2.19±0.05, t=17.11), Parkin (1.89±0.03 vs. 1.74±0.05, t=19.93), and SQSTM1 (1.82±0.07 vs. 1.61±0.05, t=18.91) in the combined group were higher than those in the control group ( P<0.001). During the treatment period, the incidence of adverse reactions was 6.67% (4/60) in the combined group and 10.00% (6/60) in the control group, without statistical significance ( χ2=0.44, P=0.509). Conclusion:Modified Chaihu Jia Longgu Muli Decoction can increase the levels of neurotrophic factors in patients with PD of liver and kidney yin deficiency syndrome, regulate the PINK1/Parkin pathway, protect neurons, improve mitochondrial autophagy function, and enhance clinical efficacy.
10.Efficacy and safety of microwave ablation via different approaches for pulmonary nodules: A retrospective cohort study

Objective To compare the efficacy and safety of computed tomography (CT)-guided percutaneous versus electromagnetic navigation bronchoscopy (ENB)-guided microwave ablation (MWA) for the treatment of pulmonary nodules. Methods A retrospective analysis was conducted on the data of high-risk pulmonary nodule patients who underwent MWA at the Nanjing Drum Tower Hospital between 2022 and 2023. The pathological diagnosis rate, complications, and progression-free survival (PFS) rate were compared between the CT group and the ENB group. Results There were 61 patients in the CT group, including 30 males and 31 females, with an average age of (67.22±9.13) years. There were 53 patients in the ENB group, including 29 males and 24 females, with an average age of (65.29±13.76) years. The pathological diagnosis rate in the CT group was slightly higher than that in the ENB group (88.52% vs. 71.69%, P=0.03). However, the ENB group exhibited a lower incidence of perioperative complications, including pneumothorax (16.39% vs. 3.77%, P=0.03), hemoptysis (19.67% vs. 5.66%, P=0.05), and pain (22.95% vs. 7.55%, P=0.03). There was no statistically significant difference in PFS rate between the two groups [HR=1.17, 95%CI (0.23, 5.81), P=0.85]. Conclusion Both CT-guided and ENB-guided MWA are effective treatment modalities for high-risk pulmonary nodules.
