1.Improvement effect of rhodopsin on the hepatic function and intestinal mucosal barrier damage in cirrhotic rats

Objective To investigate the improvement effect of rhodopsin on the hepatic function and intes-tinal mucosal barrier damage in cirrhotic rats by regulating the expression of inflammatory factors.Methods Thir-ty-six male rats were randomly divided into normal group,model group and rhodopsin group,12 rats in each group.Except for the normal group,the rats in the remaining groups were used to induce rat cirrhosis model by carbon tetrachloride(CCl4)-corn oil twice/week for a total of 12 weeks.After successful modeling,rhodop-sin(rhodopsin group)and saline(model group)interventions were given once daily for 4 weeks,respectively.The pathological and structural changes in rat liver and intestinal tissues were observed by HE and Sirus Red staining.Serum levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBIL),alpha hydroxypropionic acid(α-LA),diamine oxidase(DAO),and plasma endotoxin(ET)and inter-leukin(IL)-6,tumor necrosis factor alpha(TNF-α),and IL-17 in ileal tissues were detected.Western blot was used to detect the expression levels of Toll like receptor 4(TLR4)and nuclear factor kappa B(NF-κB)pro-teins in intestinal tissue.Results Compared with normal group,the model group showed pathological changes such as hepatocyte necrosis,disordered arrangement,shedding of intestinal mucosal villi,infiltration of inflam-matory factors and so on.Compared with the model group,inflammatory factors and the degree of injury in liver and intestinal tissue of rats in the rhodopsin group were improved,and the serum levels of AST,ALT,TBIL,IL-6,TNF-α,IL-17,α-LA,DAO,ET levels and TLR4 and NF-κP protein expression levels were signifi-cantly reduced(P<0.05).Conclusion Rhodopsin is able to attenuate the inflammatory response by inhibi-ting the release of inflammatory factors,attenuate the inflammatory response thereby improving liver func-tion,decreasing α-LA,DAO,and ET levels,and alleviating intestinal mucosal barrier damage.
2.Cost calculation of centralized dispensing of 4 categories of drugs in pharmacy intravenous admixture service

OBJECTIVE To calculate the cost of centralized dispensing of four categories of drugs (ordinary drugs, antibacterial drugs, hazardous drugs, and parenteral nutrition solutions) in pharmacy intravenous admixture service (PIVAS), and provide reference for setting charging standards for relevant departments. METHODS The operating costs of PIVAS in 12 medical institutions from Shaanxi province were collected through questionnaire survey, including labor costs, medical and health material costs, fixed asset depreciation and repair costs, water and electricity costs, and management costs. The operation time allocation coefficient method and workload allocation coefficient method were comprehensively used to allocate the above costs, and the unit preparation costs of four categories of drugs were calculated. RESULTS The average annual total costs of dispensing ordinary drugs, antibacterial drugs, hazardous drugs, and parenteral nutrition solutions in Shaanxi province were (2 195 900.25±1 680 893.73) yuan, (746 341.59±725 839.39) yuan, (331 420.15±183 258.83) yuan, and (330 322.68±277 281.70) yuan, respectively, with labor costs accounting for the highest proportion, averaging 85.49%. The costs of dispensing a set of ordinary drugs, antibacterial drugs, and hazardous drugs were 5.89, 7.60, and 14.37 yuan, respectively; the cost of dispensing one bag of parenteral nutrition solution was 32.15 yuan (excluding the cost of disposable intravenous nutrition bags). CONCLUSIONS The cost calculation method and data of different types of intravenous drugs obtained in this study can provide reference for relevant departments to formulate and adjust PIVAS fee standards.

4.Relationship between DNA methylation of glucocort icoid receptor gene promotor and glucocorticoid receptor gene mRNA expression in systemic lupus erythematosus patients

Objective To investigate the relatio nship between the DNA methylation status of gluco-corticoid receptor (GR) gene promoter and mRNA expression level of GRα gene of peripheral blood mononuclear cells (PBMCs) in patients with systemic lupus erythematosus (SLE). Methods Fifteen new onset SLE patients and fifteen healthy controls were enrolled in this study. The DNA methylation status of GR gene promoter 1 of PBMCs was detected through bisulfite sequencing polymerase chain reaction (PCR). The mRNA expression of GRα, DNA methyltransferases, growth arrest and DNA damage-induced 45α (GADD45α) of PBMCs was detected using the quantitative real-time polymerase chain reaction method. T-test and χ2-test were used to detect the differences between the two groups, Pearson's correlation coefficient was used to analyze the linear correlation between two variables. Results Compared with healthy controls, the mRNA expression of GRα was signi-ficantly declined in SLE patients (10±5, 17±7, t=2.69, P<0.05), and the mRNA expression of DNMT1 and GADD45α was significantly elevated in SLE patients (t=3.11, P<0.05 and t=2.98, P<0.05). The overall mean methylation status of the 142 CpG islands of the four promoters was significantly elevated in SLE patients [(16±8)%vs (11±6)%, t=2.75, P<0.05]. The global methylation status of PBMCs in SLE patients was obviously lower than healthy controls (t=4.39, P<0.05). Conclusion Hypermethylation of GRα promoter may result in GRαgene low expression in PBMCs of patients with SLE.
5.Effect of travoprost eye drops on visual function, stability of tear film and inflammatory index after anti-glaucoma surgery

Objective To study influence of travoprost eye drops in treatment of after anti-glaucoma surgery and its effects on visual function, the tear film stability and inflammatory index. Methods 90 patients of anti glaucoma who received therapy from August 2014 to August 2015 in the first hospital of Ninghai County Zhejiang Province were selected as research objects. The control group was treated with timolol maleate, while the observation group was treated with travoprost eye drops, then the visual acuity, intraocular pressure, tear film stability (rupture time, schirmer), inflammation (flashlight, cell count), visual field defect percentage range, curative effect after treatment were compared. Results After treatment, the vision in observation group (0.95±0.26)D was better than the control group (0.76±0.21)D, intraocular pressure in observation group (11.29±3.23) mmHg was less than the control group (13.89±3.72)mmHg, the difference was statistically significant (P<0.05), rupture time in observation group (12.93±1.90)s was higher than the control group (10.36±1.80)s, schirmer in observation group (13.01±1.60)mm was higher than the control group (11.10±1.02)mm, the difference was statistically significant (P<0.05), TNF-α, IL-6 in observation group was less than the control group, the difference was statistically significant (P<0.05), field of visual field defect in observation group (38.96±10.21)% was less than the control group (47.37±11.35)%, the difference was statistically significant (P<0.05), the total effective rate of observation group 95.56%(43/45) was statistically higher than that in the control group 77.78%(35/45), the difference was statistically significant (P<0.05). Conclusion Travoprost eye drops can improve glaucoma postoperative visual function, tear film stability, reduce inflammation.
