1.Transcriptomic differences between the spleens of mice immunized with inactivated antigens of foot-and-mouth disease virus and Senecavirus A.

The aim of this study was to compare the immune responses of C57BL/6 mice immunized with two pathogens, foot-and-mouth disease virus (FMDV) and Senecavirus A (SVA), and to provide clues for revealing the regulatory mechanisms of acquired immunity. Inactivated and purified FMDV and SVA antigens were used to immunize C57BL/6 mice respectively, and the mice immunized with PBS were taken as the control. The percentages of Th1 and Th2 cells in the spleen lymphocytes of mice in each group were analyzed by flow cytometry at 14 and 28 days after immunization. RNA-Seq was performed for the spleen. Mouse macrophages were stimulated with the antigens in vitro to examine the expression of the differentially expressed genes (DEGs) screened out. The results showed that 14 days after immunization, there was no significant difference in the magnitude of the Th1/Th2 immune response elicited by the FMDV and SVA antigens. After 28 days, the magnitudes of the Th1 and Th2 immune responses elicited by the SVA antigen were higher than those elicited by the FMDV antigen. RNA-Seq revealed two common DEGs, Rsad2 and Tspan8, between the two immunization groups, which indicated that the two genes may be involved in the activation of the Th1/Th2 immune responses by FMDV and SVA antigens. FMDV and SVA antigens stimulated macrophages to secrete interleukin (IL)-12 and IL-33 in vitro, and the expression of Tspan8 and Rsad2 was consistent with the RNA-Seq results. The expression of Rsad2 was regulated by type I interferons (IFNα, IFNβ). In this study, we obtained the DEGs involved in the immune responses to the two antigens in mouse spleen, which provides a molecular basis for investigating the immune response mechanisms induced by FMDV and SVA.

2.The effect of vitrectomy in the treatment of different types of chorioretinal coloboma with retinal detachment

Objective:To observe the efficacy of pars plana vitrectomy (PPV) in the treatment of different types of chorioretinal coloboma with retinal detachment (RD).Methods:A single-center, retrospective clinical study. From April 2021 to March 2023, 24 eyes of 23 patients who were diagnosed as chorioretinal coloboma with RD in Henan Provincial Eye Hospital were included in this study. There were 11 males with 12 eyes and 12 females with 12 eyes. The mean age was (33.3±13.7) years old. Best corrected visual acuity (BCVA), spectral domain optical coherence tomography were performed. The BCVA examination was performed using a international standard logarithmic visual acuity chart, which was converted into logarithm of the minimum angle of resolution (logMAR) visual acuity during statistics. According to the types of chorioretinal coloboma, the affected eyes were divided into the coloboma involved the optic disc group and the coloboma not involved the optic disc group, with 15 eyes and 9 eyes. According to whether the RD containing the coloboma area, the affected eyes were divided into RD containing the coloboma area group and the RD not containing the coloboma area group, with 15 eyes and 9 eyes. All eyes underwent standard pars plana three-channel 25G PPV, retinal laser photocoagulation combined with silicone oil tamponade. The follow-up time after surgery was (19.5±16.3) months. The last follow-up was the time point for efficacy determination. The retinal reattachment, BCVA recovery and postoperative complications were observed. Paired t-test or t test was performed for comparison of quantitative data. Fisher's exact test was performed for comparison of qualitative data. Results:At the last follow-up, retinal reattachment was achieved in 20 eyes (83.3%, 20/24). The logMAR BCVA of the coloboma involved the optic disc group before and after surgery were 1.85±0.62 and 1.71±0.71, the difference was no significant ( t=0.845 , P=0.412). The logMAR BCVA of the coloboma not involved the optic disc group before and after surgery were 1.75±0.45 and 0.84±0.26, the difference was statistically significant ( t=6.153 , P<0.001). The improvement of BCVA in the coloboma not involved the optic disc group was significantly higher than that in the coloboma involved the optic disc group after surgery, with statistically significant differences ( t=3.024 , P=0.006). There was no significant difference in the retinal reattachment rate between the two groups ( P=0.615). There was no significant difference in the retinal reattachment rate between the RD containing the coloboma area group and the RD not containing the coloboma area group ( P=0.259). Postoperative complications included elevated intraocular pressure in five eyes, cataract progression in ten eyes, recurrent RD in two eyes, bullous keratopathy in one eye and band-shaped keratopathy in one eye. Conclusion:PPV combined with silicone oil tamponade is safe and effective in the treatment of chorioretinal coloboma with RD, the improvement of visual acuity in the coloboma not involved the optic disc group is better than that in the coloboma involved the optic disc group after surgery.

The second part describes the novel medications and several advanced physical therapies (e.g., transcranial magnetic stimulation and transcranial direct current stimulation) for the negative symptoms and cognitive dysfunction that will optimize the therapeutic strategy for patients with schizophrenia in future.

To explore the rule of acupuncture and moxibustion in treating insomnia, and to use the Ancient and ModernMedical Cloud Platform (V1.4), a data mining technology, as a basis for interdisciplinary research ideas to providereference for clinical. Searching for acupuncture therapy insomnia clinical research literature in the past 30 years (January 1, 1987-December 31, 2017 in Chinese Knowledge Resource Base (CNKI), Chinese Science and TechnologyJournal Database (Chongqing Weipu), Chinese Academic Journal Database (Wanfang Data), and China BiomedicalDocumentation Database (CBM) for) by computer, using Excel to establish a database and enter the Ancient and ModernMedical Case Cloud Platform (V1.4), data mining methods such as frequency analysis, association rule analysis, hole pairanalysis, community analysis, and complex network analysis are used to analyze the data from the perspectives of singlehole frequency, cave Association, core prescriptions, and clinical syndrome differentiation prescriptions. The data arevisually displayed using novel and colorful charts to enhance visibility. After screening, a total of 191 articles wereincluded in the literature, involving a total of 149 acupoints and a total of 2139 points. Among them, the top fiveacupoints in the point frequency were Sanyinjiao (143 times), Shenmen (139 times), Baihui (120 times), Neiguan (115 times), and Taichong (107 times) . The first five points are Shenmen-Sanyinjiao (120 times), Neiguan-Sanyinjiao (98 times), Neiguan-Shenmen (96 times), Baihuo-Sanyinjiao (96 times), Baihui-Shenmen (91 times) and so on. The maincore prescriptions are Sanyinjiao, Shenmen, Baihui, Neiguan, Taichong, Xinshu, Taixi, Zusanli, Si shencong, Pi shu, andAn mian. The secondary core prescriptions are Fenglong, Shen shu, Yin tang, Gan shu, Dan shu, Fengchi, Zhong wan, Shenting, Zhaohai, Da ling, and others. Using the Ancient and Modern Medical Cloud Platform (V1.4) to analyze thedata, the clinical study of acupuncture and moxibustion for insomnia is illustrated with charts, thus visibility is enhanced, and it is easier to understand. It can guide clinical practice, but remember to consider specific clinical conditions.
6.Nursing care of patients with malignant tumor treated by 3D printing individualized template and 125I seed implantation

This paper summarized nursing experience of 23 patients with malignant tumor treated by 3D printing individualized template and 125I seed implantation.Nursing points included:preoperative assessment and preparation,reviewing the process of template conduction,assisting the physician to simulate the position of patients,making treatment plans,preparing templates before operation;resetting and maintaining position of patients,performing template alignment,seed implantation,monitoring vital signs and complications during operation;observation of complications,providing radiation protection and discharge guidance after operation.All 23 patients completed 125I seed implantation and no serious complication was observed.All patients recovered well and were discharged after treatment.
7.Discussion on the Treatment of Children with Asthma in Remission Stage from Healthy Qi Deficiency and Pathogenic Factor Excess

Objective] The treatment of pediatric asthma in remission period is summarized from the positive and negative aspects, in order to provide new ideas and methods for clinical treatment. [Methods] By consulting the related literatures on the traditional Chinese medicine treatment of children with asthma in remission phase, the paper mainly summarized the different physicians' understanding of pathogenesis and approaches to the treatment of pediatric asthma in remission stage. [Results]The treatment of physicians for pediatric asthma in remission phase can be divided into asthenia healthy qi and sthenia pathogenic factor syndrome.The treatment of healthy qi deficiency mainly includes treating lung,spleen,kidney seperately;curing lung-spleen,lung- kidney, spleen-kidney ;treating three viscera meanwhile.The therapy of pathogenic excess contains treating wind,phlegm,stasis respectively;curing phlegm-stasis. [Conclusion]The treatment of traditional Chinese medicine for pediatric asthma has a unique advantage in improving children's physique, reducing the frequency of asthma attacks, and improve children's life quality which has significant clinical effect.

Satisfactory effects were achieved with efforts.
9.Change of V-ATPase B subunit in transforming growth factor β1-induced epithelial-mesenchymal transition of rat renal proximal tubular cells

Objecfive To investigate the change of V-ATPase B subunits on epithelial to mesenchymal transition (EMT)in rat renal tubular epithelial cells (NRK52E) stimulated by transforming growth factor β1 (TGF-β1). Methods NRK52E cells were stimulated by TGF-β1 (10 μg/L)for O h(control),12 h,24 h,48 h,72 h after sefrum-free culture for 24 h.The mRNA and protein expression of E-cadherin,α-SMA,B2 and B1 subunits of V-ATPase were detected by real-time PCR,Western blotting and immunofluorescence. Results After stimulated by TGF-β1 (10 μg/L)for 48 h,the expression of α-SMA was markedly increased(P<0.05),but the expression of E-cadherin was dramatically decreased(P<0.05).Meanwhile,the expressions of V-ATPase subunit B2 was significantly increased (P<0.05).However,the B1 subunit distributed rarely in NRK 52E cells,and did not increase after TGF-β1 stimulation.Double-label immunofluoerscence staining also showed that the V-ATPase B2 subunit was increased in the cytosol.tending to accumulate to the cell membrane after TGF-β1 stimulation. Conclusions The main isoform of V-ATPase distributed in NRK52E cells is B2 subunit.B2 subunit is increased alone with TGF-β1-induced EMT.It may suggest that V-ATPase B2 subunit may play a potential role in TGF-β1-induced tubular EMT and renal fibrosis.
10.Clinicopathologic features and diagnosis of 18 patients with angioimmunoblastic T-cell lymphoma.

OBJECTIVE:
To explore the clinical and pathologic features of angioimmunoblastic T-cell lymphoma(AITL) and provide evidence for diagnosis.
METHODS:
Eighteen AITL patients (9 males and 9 females aged from 14 to 70 years) were retrospectively analyzed in Xiangya Hospital of Central South University from July 2002 to September 2007.
RESULTS:
Characteristic features at the presentation of AITL included generalized lymphadenopathy, fever, splenomegaly, and skin rashes with polyclonal hyper-gammaglobulinemia and other hematological abnormalities (such as Coombs-positive hemolytic anemia), which often involved the bone marrow and had well-described histologic features. The positive rate for CXCL13 was 93.3%.
CONCLUSION
Repeated lymphadenbiopsy is helpful for AITL diagnosis. Routine histological and immunohistochemical examinations (especially including CXCL13) play significant role in the diagnosis and differential diagnosis of AITL.
