1.Impact of curculigoside on neuronal pyroptosis in rats with acute cerebral infarction by regulating cGAS-STING signaling pathway

Objective:To explore effect of curculigoside on neuronal pyroptosis in rats with acute cerebral infarction(CI)by regulating cyclic guanosine-adenosine synthase(cGAS)-interferon gene stimulating factor(STING)signaling pathway.Methods:Fifteen rats were randomly selected as sham operation group,and remaining rats were constructed CI models by modified Longa suture method,CI rats successfully modeled were randomly divided into CI group,curculigoside group(5 mg/kg),SR-717 group(3 mg/kg cGAS-STING signaling pathway activator SR-717)and curculigoside+SR-717 group(5 mg/kg curculigoside+3 mg/kg SR-717),with 15 rats in each group,injected once a day for 7 consecutive days,sham operation group and CI group were given equal amounts of nor-mal saline.Neural function was evaluated by Zea-Longa score;inflammatory factors levels were detected by ELISA;TTC staining was used to evaluate volume of CI;TUNEL staining was used to detect neuronal apoptosis;immunofluorescence staining was used to detect GSDMD-N expression;Western blot was used to detect expression of pyroptosis and cGAS-STING pathway proteins.Results:Com-pared with sham operation group,Zea-Longa score,CI volume,IL-1β and IL-18 levels,apoptosis rate,number of GSDMD-N positive cells,NLRP3,cleaved-Caspase-1/Caspase-1,GSDMD-N,cGAS and STING protein expressions in CI group were significantly increased(P<0.05);compared with CI group,Zea-Longa score,CI volume,IL-1β and IL-18 levels,apoptosis rate,number of GSDMD-N positive cells,NLRP3,cleaved-Caspase-1/Caspase-1,GSDMD-N,cGAS and STING protein expressions in curculigoside group were significantly decreased(P<0.05),while the above indexes in SR-717 group had opposite trend(P<0.05);SR-717 reversed improvement effect of curculigoside on neuronal pyroptosis in CI rats.Conclusion:Curculigoside may improve neuronal pyroptosis in CI rats by down-regulating cGAS-STING signaling pathway.
2.Analysis of the efficacy and influencing factors of radiotherapy after keloid surgery

Objective To explore the efficacy of hypofractionated radiotherapy at different time intervals after surgery for keloid,and to analyze the factors affecting the efficacy.Methods A total of 76 patients who underwent 20 Gy/5 postoperative radiotherapy regimen in the Fifth Affiliated Hospital of Zhengzhou University from January 2021 to June 2023 were selected as study subjects,and a total of 100 keloids were divided into effective group(n=79)and recurrence group(n=21).Regular follow-up and record of the patients after radiotherapy treatment effect and adverse effects,and multivariate Logistic was used to analyze factors of recurrence in keloid patients.Results Multivariate Logistic regression analysis found that postoperative radiotherapy time and scar incision length were related to recurrence after treatment,radiotherapy within 7h of surgery was an independent risk factor for recurrence after treatment(OR>1,P=0.022),and scar incision≤5cm was an independent protective factor for recurrence after treatment(OR<1,P=0.028).Conclusion Surgical excision combined with hypofractionated radiotherapy is one of the effective measures to prevent and treat keloid recurrence,though keloids on the trunk may need more effective treatment options.The recurrence rate of radiotherapy initiated 7-48h after surgery is relatively the lowest,and it is worthy of clinical promotion and application.

5.Study on neuroprotective effect of alpinetin on ischemic stroke rats by regulating Shh/Gli1 signaling pathway

Objective:To investigate neuroprotective effect of alpinetin on ischemic stroke(IS)rats by regulating Shh/Gli1 signaling pathway.Methods:Fifteen rats were randomly collected as control group,remaining rats were used to construct an IS model.Rats that successfully modeling were randomly grouped into model group,alpinetin group(5 mg/kg),Shh/Gli1 signaling pathway inhibitor cycloparamide group(15 mg/kg)and alpinetin+cycloparamide group(5 mg/kg alpinetin+15 mg/kg cycloparamide),with 15 rats in each group,and administered once a day for two consecutive weeks,control group and model group were given equal amounts of physiological saline.Zea-Longa scoring method was applied to evaluate neural function;ELISA was applied to detect inflammatory factors levels;mass of brain tissue was weighed and water content of brain tissue was calculated;TTC staining was applied to measure volume of cerebral infarction;HE staining was applied to observe nerve cell damage;TUNEL staining was applied to detect neuronal apoptosis;qRT-PCR and Western blot were used to detect mRNA and protein expressions of Shh and Gli1.Results:There were no abnormalities in hippocampal tissue of control group,while hippocampal tissue structure of model group rats was abnor-mal,with disordered cell arrangement and nuclear pyknosis of nerve cells,cell damage rate,Zea-Longa score,infarct volume,IL-1β,IL-6,TNF-α,brain tissue water content,and cell apoptosis rate in model group were obviously higher than control group(P<0.05),mRNA and protein levels of Shh and Gli1 were obviously decreased(P<0.05);compared with model group,cells in alpinetin group were arranged more neatly,and phenomenon of neuronal cell nucleus pyknosis was improved,cell damage rate,Zea-Longa score,infarct volume,IL-1β,IL-6,TNF-α,brain tissue water content,and cell apoptosis rate were obviously decreased(P<0.05),mRNA and protein levels of Shh and Gli1 were obviously increased(P<0.05),trend of above indicators in cyclophosphamide group was opposite,ciclopramide reversed neuroprotective effect of alpinetin on IS rats.Conclusion:Alpinetin may exert neuroprotective effects on IS rats by activating Shh/Gli1 signaling pathway.
6.Metabolic reprogramming in idiopathic pulmonary fibrosis

Pulmonary fibrosis is caused by repeated damage to the pulmonary alveolar epithelium,leading to abnormal epithelial-mesenchymal transition and myofibroblast production,resulting in the accumulation of extra cellular matrix and remodeling of the interstitium.Analogous to many tumor cells,pulmonary fibrosis in-volves metabolic reprogramming,encompassing alterations in carbohydrate,lipid,and amino acid metabolism.Notably,this reprogramming is marked by enhanced glycolysis,diminished fatty acid oxidation paired with augmented synthesis,and increased degradation of glutamine.Glycolysis efficiently and rapidly fulfills the en-ergy requirements for the proliferation of macrophages and fibroblasts during fibrotic development.Additionally,the reprogramming of amino acid metabolism in activated fibroblasts not only facilitates collagen synthesis but also intensifies myofibroblast activation by generating reactive oxygen species(ROS)during the production of hydroxyproline.
7.Clinical and pathological research on 148 cases of glomerular minor lesion

Objective To analyze cases diagnosed with glomerular minor lesion (GML) by light microscopy and immunofluorescence,uncover their final pathology diagnosis by electron microscopy,and thereby clarify the pathological and clinical meaning of GML.Methods One hundred and forty-eight patients receiving renal biopsy between 2003 and 2008 in Peking Union Medical College Hospital,with diagnosis of GML described by light microscopy and immunofluorescence examination were retrospectively studied.All the clinical data and pathological observation were collected and analyzed,including intact results of electron microscopic examination which were considered as golden standards of pathological diagnosis.Results The 148 patients with GML had heterogenous clinical features,with isolated hematuria as the most common presentation.Electron microscopy revealed various pathological presentations:thin basement membrane nephropathy (TBMN,66.2%),mesangial proliferative glomerulonephritis (MsPGN,20.3%),Alport syndrome (2.7%),membranous nephropathy (MN,3.4%),normal tissue (4.7%).Among GML patients with isolated hematuria,TBMN ranked as the most common pathology (76.9%).Conclusions GML is only an equivocal description of pathological manifestation by light microscopy and immunofluorescence examination.And electron microscopy is necessary to obtain accurate pathology diagnosis for patients undergoing renal biopsies.
8.Clinical cohort study on renal pathological features and prognosis of type 2 diabetic kidney disease patients

Objective To analyze the relationship between renal pathological characteristics and clinical prognosis in type 2 diabetic kidney disease patients,and discuss predictive value of pathological type and indexes for renal function declining rate and related outcome events.Methods Ninety-two type 2 diabetes patients from PUMC Hospital (with macroalbuminuria and followed up no less than 6 months,excluding patients with non-diabetic renal disease) were divided into typical diabetic glomerulopathy group (DG,n=51) and atypical diabetes-related renal disease group(ADRD,n=41) according to renal pathological findings.A retrospective cohort study was performed to investigate renal pathological features and prognosis.Results Total of 29 renal outcome events and 12 death events occurred in DG group and none in ADRD group; the survival rate and kidney survival rate are different between two groups (P < 0.05); DG group,thick GBM,severe vascular and tubular lesion are predicative indicators for renal outcome event; mesangial volume fraction is predicative indicator for renal outcome events independent of age and serum creatinine.Conclusions DG and ADRD patients have different prognosis and might undergo different pathophysiological mechanisms; renal pathological type and mesangial volume fraction could help predicting outcomes of type 2 diabetic nephropathy patients.
9. Preparation and quality control of Bushen Yinao Dispersible Tablets

Objective: To optimize the preparation technology of Bushen Yinao Dispersible Tablets (BYDT) and establish a method for the quality control of BYDT. Methods: The prescription of BYDT was optimized by single factor test with the disintegrating time as index. The contents of psoralen and isopsoralen were determined by HPLC. The dissolution of BYDT was determined and compared with other two preparations. Results: PVPP was optimized as the disintegrants, MCC was optimized as the stuffing bulking agents, the proportion of PVPP and MCC was 1:2.7, and 80% ethanol was optimized as adhesives. The concentration of psoralen and isopsoralen showed a good linear relationship at the range of 4.2-52.5 and 4.6-57.5 μg/mL (r=0.9998), respectively. The average recoveries of psoralen and isopsoralen were 98.89% with RSD of 0.53%; and 100.65% with RSD of 0.93%, respectively. The total content and dissolution of psoralen and isopsoralen had no significant differences among different batches. BYDT had the higher dissolution speed and could reach the highest dissolution rate in vitro compared with the other preparations. Conclusion: This prescription of BYDT is reasonable and effective. The method is simple and accurate and could be used for the quality control of BYDT.
